OBJECTIVES: Furocoumarins (psoralens and angelicins) have been already used under ultraviolet A light (UVA) for the treatment of skin diseases and cutaneous T-cell lymphoma. Besides their high anti-proliferative activity, some severe long-term side effects have been observed, for example genotoxicity and mutagenicity, likely strictly related to the formation of crosslinks. It has been demonstrated that blue light (BL) activation of 8-methoxypsoralen, an FDA-approved drug, leads to less mutagenic monoadducts in the DNA. So far, in this work the less toxic and more penetrating BL is proposed to activate 4,6,4'-trimethylangelicin (TMA), an already known UVA photoactivatable compound. MATERIALS AND METHODS: Photocleavage, crosslink formation and oxidative damage were detected in pBR322 plasmid DNA treated with 300.0 µmol/L TMA activated with various exposures of BL. Anti-proliferative activity, reactive oxygen species (ROS) formation and activation status of some signalling pathways involved in cell growth and apoptosis were verified on DU145 cells treated with 5.0 µmol/L TMA plus 2.0 J/cm2 of BL. RESULTS: Under BL-TMA, no mutagenic crosslinks, no photocleavage and neither photooxidative lesions were detected on isolated plasmid DNA. TMA showed high anti-proliferative activity on DU145 cells through induction of apoptosis. Besides ROS generation, the proapoptotic effect seemed to be related to activation of p38 and inhibition of p44/42 phosphorylation. Interestingly, the decrease in nuclear ß-catenin was coupled with a significant dropping of CD44-positive cells. CONCLUSION: Overall, our results indicate that TMA can be activated by BL and may be considered for targeted phototherapy of prostate cancer lesions.

RESUMO

Two multidisciplinary field surveys, one in winter and the other in summer have monitored the indoor microclimate, air pollution, deposition and origin of the suspended particulate matter and microorganisms of the Correr Museum, Venice. In addition, this study was focused to identify the problems caused by the heating and air conditioning system (HAC) and the effects due to the presence of carpets. Heating and air conditioning systems (HACs), when chiefly designed for human welfare, are not suitable for conservation and can cause dangerous temperature and humidity fluctuations. Improvements at the Correr Museum have been achieved with the assistance of environmental monitoring. The carpet has a negative influence as it retains particles and bacteria which are resuspended each time people walk on it. The indoor/outdoor pollutants ratio is greater in the summertime, when doors and windows are more frequently open to allow for better ventilation, illustrating that this ratio is mainly governed by the free exchange of the air masses. The chemical composition, size and origin of the suspended particulate matter have been identified, as well as the bacteria potentially dangerous to the paintings. Some general suggestions for improving indoor air quality are reported in the conclusions.

RESUMO

A multi-disciplinary approach was used to investigate the indoor environment of a modern museum building, and its suitability for the conservation of the collection therein. Climate, gaseous and particulate pollution and the concentrations of bacteria were measured in summer and winter campaigns. While the environment overall was found to be an acceptable one, a number of drawbacks were highlighted, the most serious of these being the large temperature and humidity fluctuations that occurred in the summer.

Urine samples from 10 workers that had been exposed to n-heptane were analysed by the GC/MS technique to verify the concentrations and the relative abundances of its metabolites. The procedure of sample preparation has undergone some modifications with respect to the Perbellini method and the mass spectrometric detection was carried out in selected ions monitoring conditions. The analyses of samples collected during three different workshifts showed that 2 heptanol was not the main metabolite and that the remains of 2 heptanone, valerolactone and 2,5 heptanedione were present at the beginning of the successive work week at 12, 34 and 39 of the average values found at the end of the previous week. Overall, a very slow excretion rate was detected for the last metabolite. The main and significant metabolite at the end of the two workshifts was 2 heptanone which was detected in urine at average values of 413 and 238 µg g(-1) creatinine. This urinary ketone correlated better than other metabolites with respect to the airborne n-heptane at the end of both the workshift and work week. These preliminary data suggest that further studies should be carried out to confirm whether 2 heptanone is really useful as an n-heptane marker in biological monitoring.
