RESUMO

The primary objective of this study was to quantify the effects of residential fertilizer use on groundwater quality, which have not been well defined by field-scale investigations. Concentrations of constituents associated with fertilizer use were monitored in shallow groundwater in residential areas in Orange and Seminole Counties of Central Florida. The study area is within the Wekiva River basin, a river that is primarily spring fed. Sampling locations were selected to represent land in residential use for more than 5 yr and to avoid septic systems and areas recently used for citrus production. Twenty-six wells were installed in the surficial aquifer, screened within approximately 3 m of the water table, which was encountered between 0.3 and 11.5 m below land surface. Of these wells, 24 were in residential areas, scattered over an area of about 2600 ha, and two were in nearby undeveloped areas. Samples were collected four times between October 2008 and July 2009. Concentrations of nitrate plus nitrite nitrogen (NO-N) averaged 2.0 ± 0.2 mg L in the residential areas and were significantly higher ( < 0.01) than observed in undeveloped areas (0.3 ± 0.1 mg L). Groundwater was also analyzed for stable isotopes of nitrogen and oxygen and has been analyzed for bacteria (by others), which corroborated that these wells were not affected by human or animal waste. Levels of NO-N in the residential areas are primarily attributed to residential fertilizer use, considering the control for and exclusion of other plausible sources.

A potentially important exposure route for humans is the ingestion of chemicals via drinking water. If comprehensive exposure assessments are to be completed for either existing or proposed new chemicals and cost effective control strategies developed, then a quantitative understanding of multimedia transport and fate of specific chemical pollutants must be achieved. Mathematical models provide a powerful framework into which quantitative relationships may be placed to provide guidance in reaching water quality goals. Existing, state-of-the-art media-specific toxic organic transport and fate models for atmospheric (DiDOT), land surface (NPS) and surface water processes (EXAMS) and potable water treatment (WTP) have been linked to demonstrate the technical feasibility of such an approach. Limited application and sensitivity testing of this linked modeling system has shown that the impact of various source loadings and control strategies on drinking water quality can be estimated.

RESUMO

Stabilization for the treatment of a pars defect frequently involves fusion with sacrifice of a motion segment. Intrasegmental stabilization has been described, however, with preservation of the motion segment by using various constructs. We describe a method of obtaining rigid fixation across a pars defect without sacrificing a motion segment.

Gramicidins A, B, and C are the three most abundant, naturally occurring analogues of this family of channel-forming antibiotic. GB and GC differ from the parent pentadecapeptide, GA, by single residue mutations, W11F and W11Y, respectively. Although these mutations occur in the cation binding region of the channel, they do not affect monovalent cation specificity, but are known to alter cation-binding affinities, thermodynamic parameters of cation binding, conductance and the activation energy for ion transport. The structures of all three analogues incorporated into deuterated sodium dodecyl sulfate micelles have been obtained using solution state 2D-NMR spectroscopy and molecular modeling. For the first time, a rigorous comparison of the 3D structures of these analogues reveals that the amino acid substitutions do not have a significant effect on backbone conformation, thus eliminating channel differences as the cause of variations in transport properties. Variable positions of methyl groups in valine and leucine residues have been linked to molecular motions and are not likely to affect ion flow through the channel. Thus, it is concluded that changes in the magnitude and orientation of the dipole moment at residue 11 are responsible for altering monovalent cation transport.

RESUMO

New analogues of the venerable antimalarial drug primaquine have been synthesized and bioassayed in vivo against Pneumocystis carinii, a life-threatening infection common among immunosuppressed patients. Two of these new compounds are significantly more active than primaquine itself, and provide new information for future drug design and development in this area.
