RESUMO

In pursuit of a sustainable future powered by renewable energy, hydrogen production through water splitting should achieve high energy efficiency with economical materials. Here, we present a nanofluidic electrolyzer that leverages overlapping cathode and anode electric double layers (EDLs) to drive the splitting of pure water. Convective flow is introduced between the nanogap electrodes to suppress the crossover of generated gases. The strong electric field within the overlapping EDLs enhances ion migration and facilitates the dissociation of water molecules. Acidic and basic environments, which are created in situ at the cathode and anode, respectively, enable the use of nonprecious metal catalysts. All these merits allow the reactor to exhibit a current density of 2.8 A·cm-2 at 1.7 V with a nickel anode. This paves the way toward a new type of water electrolyzer that needs no membrane, no supporting electrolyte, and no precious metal catalysts.

C57BL/6 mice are frequently utilized as murine models with the desired genetic background for altertion in multiple research contexts. So far, there is still a lack of comprehensive kidney morphology and single-cell transcriptome atlas at all stages of growth of C57BL/6 mice. To provide an interactive set of reference standards for the scientific community, we performed the current study to investigate the kidney's development throughout the capillary-loop stage until senescence. Eight groups, with five to six mice each, represented embryonic stage (embryos 18.5 days), suckling period (1 day after birth), juvenile stage (1 month old), adulthood (containing 3 months old, 6 months old and 10 months old), reproductive senescence stage (20 months old), and post-senescence stage (30 months old), respectively. With age, the thickness of the glomerular basement membrane (GBM) was increased. Notably, GBM knobs appeared at three months and became frequent with age. Using single-cell transcriptome data, we evaluated how various biological process appear in particular cell types and investigated the potential mechanism of formation of GBM konbs. In conclusion, having access to detailed kidney morphology and single-cell transcriptome maps from C57BL/6 mice at various developmental stages of C57BL/6 mice would be a novel and major resource for biological research and testing of prospective therapeutic approaches.

Suspended sediment (SS) is a natural component of aquatic environments. It is characterized by the adsorption of pollutants, and its physical properties can affect water volume quality. In this study, SS dynamics were simulated using a 2D hydrodynamic model in the Nanji Mountain Nature Reserve (NNR), and the fluxes of pollutants caused by SS were calculated to assess the biological risks during the wet (May-August) and dry (November-March) seasons. High spatial and temporal variability in SS load within the NNR was found in this study. The average SS load in the reserve increased and then decreased during the year, and the SS input from Ganjiang significantly affected the SS load in the NNR (p < 0.01). The SS load uptrend in the NNR occurred later than that of Ganjiang during the wet season because of the SS sedimentation in the NNR. And the suspension of SS in the NNR during the dry season resulted in a later SS load downtrend compared to Ganjiang. High SS load from Ganjiang during the wet season was responsible for the high nutrient and microplastic fluxes in the NNR, which were 8.38 and 10.61 times higher than those in the dry season, respectively. And the pollutant fluxes during the wet season were almost all from Ganjiang. In contrast, higher waterbird diversity and population during the dry season is the main reason for the increased biological risk of contaminants. Therefore, monitoring and managing SS and its contamination concentrations in rivers entering the lake is helpful for the protection of ecologically sensitive areas and key species in the lake.

As a critical source of marine microplastics (MPs), estuarine MPs community varied in movement due to particle diversity, while tide and runoff further complicated their transport. In this study, a particle mass gradient that represents MPs in the surface layer of the Yangtze River estuary was established. This was done by calculating the masses of 16 particle types using the particle size probability density function (PDF), with typical shapes and polymers as classifiers. Further, Aschenbrenner shape factor and polymer density were embedded into drag coefficients to categorically trace MP movement footprints. Results revealed that the MPs in North Branch moved northward and the MPs in South Branch moved southeastward in a spiral oscillation until they left the model boundary under Changjiang Diluted Water front and the northward coastal currents. Low-density fibrous MPs are more likely to move into the open ocean and oscillate more than films, with a single PE fiber trajectory that reached a maximum oscillatory width of 16.7 km. Over 95 % of the PVC fiber particles settled in nearshore waters west of 122.5°E. Elucidating the aggregation and retention of different MPs types can provide more accurate environmental baseline reference for more precise MP exposure levels and risk dose of ingestion for marine organisms.

Aims: Endothelial cells are the critical targets of injury in diabetic nephropathy (DN), and endothelial cell lesions contribute to the disease progression. Neurite outgrowth inhibitor B (Nogo-B), an endoplasmic reticulum (ER)-resident protein, plays a pivotal role in vascular remodeling after injury, and maintains the structure and function of the ER. Yet, the role of Nogo-B in the regulation of ER stress and endothelial cell injury remains largely unknown. Herein, we tested the hypothesis that Nogo-B activates ER stress-mediated autophagy and protects endothelial cells in DN. Results: The level of Nogo-B was decreased in glomerular endothelial cells in biopsy specimens from DN patients. In vivo and in vitro studies have shown that silencing Nogo-B activated ER stress signaling, and affected the expression of autophagy-related marker early growth response 1 and microtubule-associated protein light chain 3 (LC3) in endothelial cells in hyperglycemic condition. Conclusion and Innovation: These results denote that Nogo-B contributes to ER stress-mediated autophagy and protects endothelial cells in DN, providing new evidence for understanding the role of ER stress-mediated autophagy in endothelial cells of DN.
