ABSTRACTPerching or standing on one foot is commonly reported in birds but the level of consistency in using one foot over the other has been less-well documented in most species, particularly birds of prey. For birds experiencing colder temperatures, unipedal perching has been attributed to limiting heat loss through unfeathered legs and feet; individuals should spend longer periods of time perched on one foot as temperatures decrease. Using radio tracking, I collected 486 hours of observations on nine overwintering, free-living merlins (Falco columbarius) in Saskatoon, Canada. Five merlins displayed clear preferences to perch on one foot, however the direction of preference was not consistent and four birds were ambidextrous. There was a curvilinear response in the proportion of time spent in unipedal posture versus temperature, with a peak of ∼22% of the time at moderate temperatures (-10 to -19°C), but lower values at warmer and colder temperatures; the main effect of the squared term for temperature was highly influential while individual foot preference had no impact on the use of unipedal perching. Although preferential use of one foot for perching was displayed by some individuals, thermoregulation may not be the primary driver of this behaviour at colder temperatures.

Populations of some fish- and meat-eating birds suffered dramatic declines globally following the introduction of organochlorine pesticides during the late 1940s and 1950s. It has been hypothesised that these population declines during the 1950s-1970s were largely driven by a combination of reproductive failure due to eggshell-thinning, egg breakage and embryonic death attributable to DDT and its metabolites, and to enhanced mortality attributable to the more toxic cyclodiene compounds such as aldrin and dieldrin. Using 75 years (1946-2021) of Peregrine falcon (Falco peregrinus) monitoring data (315 unique nest-sites monitored for 6110 nest-years), we studied the breeding performance of a resident Peregrine population in southern Scotland relative to the spatiotemporal pattern of organochlorine pesticide use. We show that (i) Peregrine breeding success and measures of breeding performance increased substantially following the reduction in, and subsequently a complete ban on, the use of organochlorine pesticides; (ii) improvements in Peregrine breeding performance were more dramatic in southeastern Scotland where agriculture was the predominant land use than in southwestern Scotland where there was less arable and more forested land; (iii) Peregrines nesting closer to the coast generally had higher fledging success (that is, a higher proportion of clutches that produced at least one fledgeling) than those nesting inland farther away from the coast; (iv) low temperatures and excessive rain in May negatively affected Peregrine fledging success; and (v) Peregrine abundance increased in parallel with improvements in reproductive performance following the reduction and then complete ban on the use of organochlorine pesticides in the UK. However, recovery was gradual and occurred over four decades, and rate of recovery varied among measures of reproductive performance (egg, nestling and fledgeling production). Our results suggest that the temporal pattern of organochlorine pesticide use strongly influenced Peregrine reproductive parameters but that the pattern of influence differed regionally. Overall results are consistent with the hypothesis that reproductive failure caused by organochlorine pesticides was an important driver of the decline in the south Scottish Peregrine population, and that improvements in all measures of breeding performance following a reduction and eventual ban on organochlorine use facilitated the observed increase in this population.

A 3-year-old male saker falcon (Falco cherrug) was presented with a history of hyperflexion of the first digit of the left pelvic limb with what appeared to be a hypercontracted distal end of the deep digital flexor tendon (m. flexor hallucis longus). Conservative treatment, bandaging, and splinting techniques failed to resolve the hypercontraction of the deep digital flexor tendon and improve digit function. Intraoperatively, the deep digital flexor tendon was found to have formed adhesions with the tendon sheath as well as the bone and soft tissue structures underneath it, impeding its ability to move freely within the tendon sheath. The adhesions were surgically resolved, and the tendon was lengthened by hemisectioning the deep digital flexor tendon at its distal end. Postsurgically, the digit was maintained in an extended position by bandaging. Mild physiotherapy was provided at 5-day intervals to retain function and prevent adhesions. Digit extensibility and function were restored to nearly normal levels after 4 weeks.

Endothermic animals that live permanently in hot deserts must avoid harmful hyperthermia when their body temperature increases from heat gained through external and internal sources. This is true particularly for endotherms that are exclusively diurnal. We investigated the Grey Falcon (Falco hypoleucos), a predatory Australian endemic restricted to the hot arid/semi-arid zone. To understand how this species' entire population persists exclusively and permanently in this extreme environment we examined its activity levels and compared these with equivalent variables from the Peregrine Falcon (F. peregrinus), a cosmopolitan species that inhabits similar environments without being restricted to them. Further, we compared, across a selected group of Falco species, specific plumage characteristics (measured on museum specimens) that we anticipated would enhance the Grey Falcons' ability to cope with high heat loads. We found no morphological or physiological characteristics that would allow them to cope with heat better than other birds, but the chicks seem to have unusually high thermal tolerances. Grey Falcons do, however, possess a suite of unusual behavioural adaptations that, as we propose, enable them to cope with climatic extremes in arid environments. Specifically, throughout their lives Grey Falcons keep activity levels and thus physical exertion low. This behaviour contrasts strikingly with that of the Peregrine Falcon, which also actively hunts birds in flight. Keeping activity levels low is expected to minimize endogenous heat production and thus ease the Grey Falcon's thermoregulation during periods of high heat load. These birds may rely on low levels of relative humidity for efficient evaporative cooling, and this may explain their absolute restriction to hot arid/semi-arid zones.

At a routine health check of a female peregrine falcon, 23 trematodes preliminary identified as Prosthogonimus sp. were removed from the bursa of Fabricius. Based on morphological and molecular examination, a new species, Prosthogonimus falconis, was described. The pear-shaped flukes were 4.3-6.9 mm long, with greatest width posterior to testes. Tegumental spines measuring between 17 and 21 µm long covered the whole body. Length and width ratio of oral to ventral suckers were 1:1.3. Extracaecal, multifollicular vitelline glands commenced prior to acetabulum and terminated posterior to testes. Eggs in the distal uterus measured 21 × 12 µm. Molecular analysis of internal transcribed spacer 2, cytochrome c oxidase subunit 1 and NADH dehydrogenase subunit 1 gene regions revealed that the new species described here is phylogenetically closest to Prosthogonimus cuneatus and Prosthogonimus pellucidus clusters.

Species distribution models (SDMs) generate predicted distribution maps which can be used as effective tools for conservation purposes. The persistence of isolated populations at the margin of a large distributional area depends on local threats which may differ from those faced by the main population. Environmental predictors can indicate suitable areas for these species and, indirectly, evaluate the impact on peripheral populations due to fragmentation and isolation. The Lanner Falcon (Falco biarmicus) is an Afro-tropical and Mediterranean polytypic species considered critically endangered (CR) in Arabian Peninsula by IUCN, but a lack of published information about its distribution persists. Here, we model the distribution of the Lanner Falcon in the Arabian Peninsula using nest-site data and map its core area and their habitat suitability using a robust algorithm with good prediction accuracy even at low sample sizes (MaxEnt). The predictive map suggests a potential distribution of the Lanner Falcons that runs from north to south along the eastern coast of the Red Sea. The Terrain Roughness Index contributed the most to the breeding range model predictions (57.6%), followed by isothermality (Bio3, 15.3%). The model suggests a tendency by Lanner Falcons to occupy areas with a low terrain complexity according to their behavioural patterns and breeding strategies. In addition, this falcon is highly sensitive to climate occupying high isothermal regions in order to avoid extreme heating events. Overall, predictive models indicate a narrow range of suitable environmental conditions for breeding as well restricted favourable areas during dispersal and migration. Thus, these small and fragmented populations are more likely prone to anthropogenic factors and must be buffered against them.

The aerial hunting behaviours of birds are strongly influenced by flight morphology and ecology, but little is known of how this relates to the behavioural algorithms guiding flight. Here, we used GPS loggers to record the attack trajectories of captive-bred gyrfalcons (Falco rusticolus) during their maiden flights against robotic aerial targets, which we compared with existing flight data from peregrine falcons (Falco peregrinus). The attack trajectories of both species were well modelled by a proportional navigation (PN) guidance law, which commands turning in proportion to the angular rate of the line-of-sight to target, at a guidance gain N However, naive gyrfalcons operate at significantly lower values of N than peregrine falcons, producing slower turning and a longer path to intercept. Gyrfalcons are less manoeuvrable than peregrine falcons, but physical constraint is insufficient to explain the lower values of N we found, which may reflect either the inexperience of the individual birds or ecological adaptation at the species level. For example, low values of N promote the tail-chasing behaviour that is typical of wild gyrfalcons and which apparently serves to tire their prey in a prolonged high-speed pursuit. Likewise, during close pursuit of typical fast evasive prey, PN will be less prone to being thrown off by erratic target manoeuvres at low guidance gain. The fact that low-gain PN successfully models the maiden attack flights of gyrfalcons suggests that this behavioural algorithm is embedded in a guidance pathway ancestral to the clade containing gyrfalcons and peregrine falcons, though perhaps with much deeper evolutionary origins.

Many falcons (Falco spp.) exhibit a distinct dark plumage patch below the eye, termed the malar stripe. This stripe is hypothesized to reduce the amount of solar glare reflected into the eyes while foraging, thereby increasing hunting efficiency in bright conditions. Here, we use a novel, global-scale correlative approach to test this 'solar glare hypothesis' in peregrine falcons (Falco peregrinus), the most widespread falcon species, using web-sourced photographs from across the species' global range. We found that the size and prominence of the malar stripe were positively associated with average annual solar radiation, but not with other environmental variables, such as temperature and rainfall. Our results provide the first published evidence for the hypothesis that this plumage feature functions to reduce the amount of solar glare reflected into the falcon's eyes, thereby improving the ability to pinpoint and target agile prey in bright conditions.

Air sac nematodes from birds are known for more than 200 years now and Filaria attenuata was the first described species from falcons, owl and corvid birds. The superficial description and the loss of the original material made F. attenuata a species inquirenda. Seurat (1915) redescribed the species with material from lanner falcon and pallid harrier from Algeria and based on this description Bain and Mawson, Rec S Aust Mus 18:265-28, (1981) created a new species, Serratospiculum seurati, by adding some, slightly divergent, measurements. The current paper is based on light and scanning electron microscopy of five male and 10 female S. seurati specimens from a Peregrine falcon that acquired the infection in Pakistan. The length of the slender male and female nematodes varied between 42-70 and 165-221 mm, respectively, spicules of unequal shape and length measured 292-325 and 638-785 µm. S. seurati was also found in Saker, Barbary and crossbreed falcons.

Dispersal is a critical process influencing population dynamics and responses to global change. Long-distance dispersal (LDD) can be especially important for gene flow and adaptability, although little is known about the factors influencing LDD because studying large-scale movements is challenging and LDD tends to be observed less frequently than shorter-distance dispersal (SDD). We sought to understand patterns of natal dispersal at a large scale, specifically aiming to understand the relative frequency of LDD compared to SDD and correlates of dispersal distances. We used bird banding and encounter data for American kestrels (Falco sparverius) to investigate the effects of sex, migration strategy, population density, weather, year and agricultural land cover on LDD frequency, LDD distance and SDD distance in North America from 1961 to 2015. Nearly half of all natal dispersal (48.9%) was LDD (classified as >30 km), and the likelihood of LDD was positively associated with the proportion of agricultural land cover around natal sites. Correlates of distance differed between LDD and SDD movements. LDD distance was positively correlated with latitude, a proxy for migration strategy, suggesting that migratory individuals disperse farther than residents. Distance of LDD in males was positively associated with maximum summer temperature. We did not find sex-bias or an effect of population density in LDD distance or frequency. Within SDD, females tended to disperse farther than males, and distance was positively correlated with density. Sampling affected all responses, likely because local studies more frequently capture SDD within study areas. Our findings that LDD occurs at a relatively high frequency and is related to different proximate factors from SDD, including a lack of sex-bias in LDD, suggest that LDD may be more common than previously reported, and LDD and SDD may be distinct processes rather than two outcomes originating from a single dispersal distribution. To our knowledge, this is the first evidence that LDD and SDD may be separate processes in an avian species, and suggests that environmental change may have different outcomes on the two processes.

Merlins, Falco columbarius, breed throughout temperate and high latitude habitats in Asia, Europe, and North America. Like peregrine falcons, F. peregrinus, merlins underwent population declines during the mid-to-late twentieth century, due to organochlorine-based contamination, and have subsequently recovered, at least in North American populations. To better understand levels of genetic diversity and population structuring in contemporary populations and to assess the impact of the twentieth century decline, we used genomic data archived in public databases and constructed genomic libraries to isolate and characterize a suite of 17 microsatellite markers for use in merlins. We also conducted cross-amplification experiments to determine the markers' utility in peregrine falcons and gyrfalcons, F. rusticolus. These markers provide a valuable addition to marker suites that can be used to determine individual identity and conduct genetic analyses on merlins and congeners.

Centrorhynchus globocaudatus (Zeder, 1800) Lühe, 1911 (Centrorhynchidae) was reported in birds of prey. Our population from Falco tinnunculus Linnaeus (Falconidae) and Buteo buteo Linnaeus (Accipitridae) in northern Italy was morphologically distinct from others described elsewhere. The worms are elongate and cylindrical. Proboscis long, apically truncated and bare, with wider base and variably faint constriction at point of attachment of receptacle. Large anterior hooks well rooted; posterior spiniform hooks with reduced roots; transitional hooks with scutiform roots in-between. Four tubular cement glands extend into prominent ducts overlapping a large Saefftigen's pouch. Bursa large, with sensory plates. Vagina with laterally slit orifice in sub-ventral pit of globular terminal extension. Thick-shelled eggs ovoid without polar prolongation of fertilization membrane. In our specimens, proboscis hooks, receptacle, male reproductive system, and lemnisci especially in males varied in size from those from Ukraine, India, Egypt, Kyrgystan, Russia, Georgia, Armenia and Asian Soviet Republics. Our description of the Italian specimens includes new morphological information supported by scanning electron microscopy and microscope images, molecular analysis and energy dispersive X-ray analysis (EDXA) of hooks. Additional new details of proboscis hook roots, micropores and micropore distribution are described. Metal composition of hooks (EDXA) demonstrated high levels of calcium and phosphorous, and high levels of sulphur in core and cortical layers of eggs. The molecular profile based on sequences of 18S and cytochrome c oxidase 1 genes is also provided, as well as phylogenetic reconstructions including all available sequences of the family Centrorhynchidae, although further sequences are needed in order to clarify their phylogenetic relationships.

A total of 356 gyrfalcon (Falco rusticolus) fecal, fomite, and environmental samples were collected from a breeding center located in the United Arab Emirates to assess the prevalence of Caryospora species oocysts in the environment. These included 136 samples (38%) from fomites and fecal samples from chicks at 0 to 10 days old, 29 samples (8%) at 15 days old, 23 samples (6%) at 60 days old, 7 samples (2%) at 67 days old, and 24 samples (7%) at 70 days old. In addition, 105 samples (29%) were collected from the environment of 13 breeding chambers, and 32 samples (9%) from the environment of 17 juvenile falcons. The prevalence of Caryospora species oocysts in fomites and fecal samples from the chicks had negative results from 10 to 60 days old. However, at 67 and 70 days old, the prevalence increased to 71.42% (5 of 7) and 95.83% (23 of 24), respectively. The prevalence of Caryospora species in the environment of 13 pairs of falcons housed in 13 breeding chambers was 0.15 oocyst/m2 in the sand, whereas, in the environment of 17 juvenile falcons housed in the free-flying aviary, the prevalence was 0.00086 oocyst/m2 in the sand and 0.15 oocyst/L in contaminated water. These results indicate that oocysts of Caryospora species may be found in the environment and in areas of poor and substandard hygiene. Caryospora species is an important protozoon parasite affecting captive falcons maintained in breeding centers and those used for falconry in the Middle East.

Pododermatitis is prevalent in falcons and is characterized by inflammation and infection of the plantar aspect of the feet, particularly at the central metatarsal pad. Suboptimal perch design and increased weight loading on the plantar skin have been proposed as major risk factors for the development of pododermatitis. Prevention and treatment are challenging, but weight load reduction on the affected area of the foot is an accepted goal of initial treatment. To date, to our knowledge no studies have compared the performance of different bandage-perch surface combinations on weight redistribution away from the central metatarsal pad. An ex vivo factorial experiment was designed using the feet from a peregrine falcon cadaver to quantify weight load reduction on the central metatarsal pad with different combinations of perch surfaces (wood, neoprene, artificial turf) and bandages (none, interdigital, silicone shoe). Feet were attached to a digital force gauge mounted on a manual test stand for compression testing. Weight loading at the central metatarsal pad was measured using a small force sensor. Perch-surface combinations in randomized order were tested at 250 g, 500 g, and 1 kg with 9 replicates per foot. At 250 g, all combinations reduced measured metatarsal pad forces, but otherwise performed similarly. As compression forces increased, differences emerged with the shoe combinations performing better overall, followed by a group including the neoprene and artificial turf-interdigital bandage combinations, and a third group including the interdigital/wood and no bandage-artificial turf. All combinations performed better than control (no bandage on wood). This study may assist veterinarians in selecting appropriate perching surface/bandage combinations when treating falcons with pododermatitis.

BACKGROUND: Read alignment and transcript assembly are the core of RNA-seq analysis for transcript isoform discovery. Nonetheless, current tools are not designed to be scalable for analysis of full-length bulk or single cell RNA-seq (scRNA-seq) data. The previous version of our cloud-based tool Falco only focuses on RNA-seq read counting, but does not allow for more flexible steps such as alignment and read assembly. RESULTS: The Falco framework can harness the parallel and distributed computing environment in modern cloud platforms to accelerate read alignment and transcript assembly of full-length bulk RNA-seq and scRNA-seq data. There are two new modes in Falco: alignment-only and transcript assembly. In the alignment-only mode, Falco can speed up the alignment process by 2.5-16.4x based on two public scRNA-seq datasets when compared to alignment on a highly optimised standalone computer. Furthermore, it also provides a 10x average speed-up compared to alignment using published cloud-enabled tool for read alignment, Rail-RNA. In the transcript assembly mode, Falco can speed up the transcript assembly process by 1.7-16.5x compared to performing transcript assembly on a highly optimised computer. CONCLUSION: Falco is a significantly updated open source big data processing framework that enables scalable and accelerated alignment and assembly of full-length scRNA-seq data on the cloud. Birds primarily create and control the forces necessary for flight through changing the shape and orientation of their wings and tail. Their wing geometry is characterised by complex variation in parameters such as camber, twist, sweep and dihedral. To characterise this complexity, a multi-view stereo-photogrammetry setup was developed for accurately measuring surface geometry in high resolution during free flight. The natural patterning of the birds was used as the basis for phase correlation-based image matching, allowing indoor or outdoor use while being non-intrusive for the birds. The accuracy of the method was quantified and shown to be sufficient for characterising the geometric parameters of interest, but with a reduction in accuracy close to the wing edge and in some localised regions. To demonstrate the method's utility, surface reconstructions are presented for a barn owl (Tyto alba) and peregrine falcon (Falco peregrinus) during three instants of gliding flight per bird. The barn owl flew with a consistent geometry, with positive wing camber and longitudinal anhedral. Based on flight dynamics theory, this suggests it was longitudinally statically unstable during these flights. The peregrine falcon flew with a consistent glide angle, but at a range of air speeds with varying geometry. Unlike the barn owl, its glide configuration did not provide a clear indication of longitudinal static stability/instability. Aspects of the geometries adopted by both birds appeared to be related to control corrections and this method would be well suited for future investigations in this area, as well as for other quantitative studies into avian flight dynamics.

Tetrabromobisphenol A bis(2,3-dibromopropyl ether) (TBBPA-BDBPE) and 1,2-bis(2,4,6-tribromophenoxy)ethane (BTPBE) are both brominated flame retardants (BFRs) that have been detected in birds; however, their potential biological effects are largely unknown. We assessed the effects of embryonic exposure to TBBPA-BDBPE and BTBPE in a model avian predator, the American kestrel (Falco sparverius). Fertile eggs from a captive population of kestrels were injected on embryonic day 5 (ED5) with a vehicle control or one of three doses within the range of concentrations that have been detected in biota (nominal concentrations of 0, 10, 50 or 100 ng/g egg; measured concentrations 0, 3.0, 13.7 or 33.5 ng TBBPA-BDBPE/g egg and 0, 5.3, 26.8 or 58.1 ng BTBPE/g egg). Eggs were artificially incubated until hatching (ED28), at which point blood and tissues were collected to measure morphological and physiological endpoints, including organ somatic indices, circulating and glandular thyroid hormone concentrations, thyroid gland histology, hepatic deiodinase activity, and markers of oxidative stress. Neither compound had any effects on embryo survival through 90% of the incubation period or on hatching success, body mass, organ size, or oxidative stress of hatchlings. There was evidence of sex-specific effects in the thyroid system responses to the BTBPE exposures, with type 2 deiodinase (D2) activity decreasing at higher doses in female, but not in male hatchlings, suggesting that females may be more sensitive to BTBPE. However, there were no effects of TBBPA-BDBPE on the thyroid system in kestrels. For the BTPBE study, a subset of high-dose eggs was collected throughout the incubation period to measure changes in BTBPE concentrations. There was no decrease in BTBPE over the incubation period, suggesting that BTBPE is slowly metabolized by kestrel embryos throughout their ∼28-d development. These two compounds, therefore, do not appear to be particularly toxic to embryos of the American kestrel.

An adult female gyrfalcon (Falco rusticolus) was presented with a right-wing droop and weight loss. Radiographic images revealed osteolysis and osseous proliferation of the right shoulder and the mobile vertebra between the notarium and synsacrum. The tentative diagnosis was vertebral osteomyelitis secondary to septic arthritis. The bird did not respond to antibiotic and anti-inflammatory therapy and represented 10 days later, with feathers soiled with feces, an impacted, dilated cloaca, and an inability to stand due to spastic paralysis of the hind legs. The bird's condition did not improve with 24 hours of supportive care and its quality of life was considered poor; therefore, the patient was euthanatized and submitted for postmortem examination. Multicentric septic osteomyelitis and arthritis were confirmed in the mobile vertebra between the notarium and synsacrum and the right shoulder. Despite 10 days of antibiotic therapy, Staphylococcus aureus was isolated from within the 2 locations in which septic osteomyelitis and arthritis were identified. This report describes the clinical features, diagnosis, and pathologic findings of septic osteomyelitis and arthritis caused by S aureus in a falcon.

Distraction osteogenesis was used in 2 wild raptor patients for the repair of tibiotarsal fractures. The first case was a hatching year female peregrine falcon (Falco peregrinus) admitted with an open oblique right distal tibiotarsal fracture. The fracture was surgically managed with the external skeletal fixator intramedullary pin tie-in technique (ESF-IM pin tie-in). Appropriate healing of the fracture site occurred with the ESF-IM pin tie-in. However, there was significant pelvic limb shortening of the affected leg, and the bird consequently developed pododermatitis on the contralateral foot. The second case was an adult female great horned owl (Bubo virginianus) that was admitted with a closed, partially healed, overriding right tibiotarsal fracture. Because of the advanced stage of this fracture, which presented with a fibrous callus, and the already apparent pelvic limb shortening, the ESF-IM pin tie-in was not used. Both patients were fitted with a circular external skeletal fixator (CESF), and distraction osteogenesis was performed until the length of the pelvic limb was deemed anatomically adequate. It is critical that rehabilitated raptors be released without any physical conditions that may reduce their ability to survive and reproduce in the wild. Pelvic limb shortening can potentially predispose a raptor patient to pododermatitis, even with fatal consequences, in both captive and wild environments. The orthopedic technique used here proved useful to repair the limb shortening in both raptor cases, and each bird fully recovered and was released.

In this study, we sequenced the complete mitogenome of Falco amurensis (Falconiformes, Falconidae). The F. amurensis mitogenome is 17,464 bp long, and contains 37 genes, including 13 protein-coding genes (PCGs), two rRNAs, 22 tRNAs, and two non-coding regions (control region and pseudo-control region). Most PCGs initiate with ATG and terminate with TAA. atp8 exhibits the highest evolutionary rate, with cox1 showing the lowest. rrnS and rrnL contain three domains with 46 helices and six domains with 59 helices, respectively. All tRNAs have a typical cloverleaf secondary structure, except that trnS(agy) lacks the dihydrouracil arm. The control region is located between trnT and trnP and the pseudo-control between trnE and trnF. Phylogenetic relationships of 23 species from Falconiformes were analyzed based on the nucleotide sequences of the 13 PCGs and two rRNAs. The results support Falco as a monophyletic taxon, and F. amurensis has a close relationship with the clade containing F. cherrug/F. rusticolus/F. peregrinus.
