RESUMO

Western Eurasia witnessed several large-scale human migrations during the Holocene1-5. Here, to investigate the cross-continental effects of these migrations, we shotgun-sequenced 317 genomes-mainly from the Mesolithic and Neolithic periods-from across northern and western Eurasia. These were imputed alongside published data to obtain diploid genotypes from more than 1,600 ancient humans. Our analyses revealed a 'great divide' genomic boundary extending from the Black Sea to the Baltic. Mesolithic hunter-gatherers were highly genetically differentiated east and west of this zone, and the effect of the neolithization was equally disparate. Large-scale ancestry shifts occurred in the west as farming was introduced, including near-total replacement of hunter-gatherers in many areas, whereas no substantial ancestry shifts happened east of the zone during the same period. Similarly, relatedness decreased in the west from the Neolithic transition onwards, whereas, east of the Urals, relatedness remained high until around 4,000 BP, consistent with the persistence of localized groups of hunter-gatherers. The boundary dissolved when Yamnaya-related ancestry spread across western Eurasia around 5,000 BP, resulting in a second major turnover that reached most parts of Europe within a 1,000-year span. The genetic origin and fate of the Yamnaya have remained elusive, but we show that hunter-gatherers from the Middle Don region contributed ancestry to them. Yamnaya groups later admixed with individuals associated with the Globular Amphora culture before expanding into Europe. Similar turnovers occurred in western Siberia, where we report new genomic data from a 'Neolithic steppe' cline spanning the Siberian forest steppe to Lake Baikal. These prehistoric migrations had profound and lasting effects on the genetic diversity of Eurasian populations.

Members of the genus Ortholinea are among the worldwide distributed myxozoan parasites that mainly infect marine fish. In this study, a new myxosporean species, Ortholinea hamsiensis n. sp., was isolated from the urinary bladder of European anchovy Engraulis engrasicolus collected from the Sinop coasts of the Black Sea. The prevalence and density values of infection were 1.4% and 1­5 individuals in the field of view (1 + ), respectively. Mature myxospores are subspherical with slight tapering down to the less pronounced tip in the frontal view and subspherical in the sutural view. Myxospores measured 9.1 ± 0.25 (8.8­9.9) µm in length, 9.2 ± 0.11 (8.9­9.4) µm in thickness, and 8.4 ± 0.33 (8.2-9.1) µm in width. Two polar capsules equal in size measured 3.1 ± 0.11 (3.0­3.3) µm in length and 2.7 ± 0.11 (2.6­2.9) µm in width. The polar tubule had 3­4 coils. Along with morphological peculiarities, the results of the 18S rDNA also revealed it to be a new species for science compared to the other species of the genus. In this study, another myxosporean species O. gobiusi was also detected in round goby Neogobius melanostomus with a prevalence of infection value of 4.8% and a density of 1­5 individuals in the field of view (1 + ). The present study also provided the first data of 18S rDNA of O. gobiusi from N. melanostomus and type species of the genus O. divergens from Gobius niger and the phylogenetic relationships of these species with other Ortholinea species have been revealed.

The study of the element accumulation in marine plants against the backdrop of permanently increasing environmental pollution is of particular importance due to the participation of these plants in biogeochemical cycles. The element abundances are highly variable and depend on both the macrophyte species and environment. The purpose of this study was to analyze the elemental composition of widespread marine plants of different taxonomic affiliations collected in the same area of the Black Sea coast. The contents of 74 elements in three species of lower (red, brown, green algae) and one species of higher plants (seagrass) were analyzed using inductively coupled plasma mass spectrometry. High contents of most elements were found in the red alga Ceramium ciliatum and in rhizomes of the seagrass Zostera noltei. In C. ciliatum, high metal bioaccumulation factors were found, which are dependent also on their concentration in the environment. Compared to the higher plant, all the macroalgae accumulated increased amounts of As and I. The seagrass proved to be a good concentrator of Mo and Sb, and relatively high contents of Mn, Co, Ni, Zn, Cd and Ir were registered in its leaves. High contents of Mg, S, Ge, Se and Ta were found in the green alga Ulva rigida, and elevated levels of Al, As, Sr, Zr, Ru, Rh, Pd, Ag, Ba and Re were noted in the brown alga Gongolaria barbata. The enrichment factors for most elements in the sediments were well above 1 with respect to both the local Late Pleistocene sediments and the upper continental crust. The strength of correlations between the element contents in the plants and sediments was found to decrease with the specific surface area growth and appeared to have a lower asymptotic limit of the sediments-seawater correlation strength.

A 3-D transport and dispersion model was applied to study the recent past and future dynamics of perfluorooctanoic acid (PFOA) and perfluorooctane sulfonic acid (PFOS) concentrations in the Black Sea for the 2016-2030 period. The modelled surface concentrations show a distinct seasonal behaviour, shaped by winter to spring convective mixing. A significant increasing long-term trend in PFOS concentrations is established, with concentrations in water layers 200 m below the surface increasing at 4-8% per year. Driving mechanisms for PFOA and PFOS transport and accumulation in the subsurface and deeper layers are the cooling of the surface water in winter and the transport of water masses from the North Western Shelf (NWS) of the Black Sea. A simulated 50% phase-out of PFOA and PFOS from 2020 to 2030 shows a 21% reduction in PFOA, while PFOS continues to increase.

Monitoring and protecting freshwater habitats are paramount for a sustainable water management perspective. This study investigated potentially toxic elements (PTEs) in the potamic water of the Anday Stream Basin (Türkiye), Black Sea Region, for a hydrological year (from May 2020 to April 2021). Among PTEs, the highest average values were recorded for sodium (Na) at 41.3 mg/L and the lowest for mercury (Hg) at 0.009 µg/L and noted under quality guidelines. The stream was found to be at the level of "Low Heavy Metal Pollution" and "Low Contamination" based on the ecotoxicological risk indices. The highest calculated hazard quotient (HQ) value of 1.21E-02 for Cd was noted in the children via the dermal pathway and the lowest of 6.91E-06 for Fe in adults via the ingestion pathway. Results revealed a higher hazard index (HI) value of 1.50E-02 for Cd to children and the lowest of 1.98E-05 for Fe to adults. As a result of applying agricultural risk indices, the stream showed sodium adsorption ratio values less than 6 and was found to be "Excellent" for agriculture. However, the sodium percentage values were less than 20 and found "Permissible" and the magnesium hazard > 50 and noted as "Unsuitable" for agriculture. Statistical analysis revealed that natural factors mainly attributed to PTE contamination of the Anday Stream Basin.

Spatial and temporal variations in marine litter density and composition along the Southeastern Black Sea Coast were investigated. A total of 156,371 litter items weighing 327,258.3 kg were collected. The highest frequency of litter material by number was 15,869 ± 103.88 items/m2 16 and 74.466 ± 7.23 by weight. The highest litter concentrations (77,768 items; 81,737.1 kg) were observed in autumn, mainly comprising single-use items, with plastic being the most abundant (54.05 %), followed by metal (15.69 %), and paper (10.45 %). The subcategories of plastic litter items bags, caps/lids, cigarette lighters, cosmetic packages, gloves, and plastics pieces were found to be the most abundant litter in number. According to Principal Component Analysis (PCA) and Kruskal-Wallis statistical tests (p < 0.005), significant differences in marine litter were identified among the stations and seasons. These findings offer insights for modeling studies, advocating restrictions on single-use products, and enacting legal regulations for local governance.

The Black Sea is a unique environment with strong and permanent vertical stratification, with a thin layer of oxic zone above and a permanent anoxic zone below. Few high-throughput genomic surveys have been conducted to examine microbiota in the Black Sea. Yet, there is no study on the seasonal and vertical variation in microbial community compositions, driving forces and mechanisms of community assembly. In this study, seasonal, vertical, and spatial microbial assemblages were studied in terms of diversity, abundance, and community structure using 16S rRNA metabarcoding. 16S rRNA metabarcoding confirmed seasonal changes in microbial communities and the presence of distinct microbial groups among different water layers. Taxa belonging to Cyanobiaceae contributed a large fraction of the total biomass and were the most abundant autotrophic bacteria found across the whole water column, including hydrogen sulfide-containing anoxic zone. Temperature, salinity, water density, conductivity, light, chlorophyll-a, O2, NO3, NH3, PO4, Si, and H2S had a significant influence on the vertical bacterial community assemblages. The copper mine discharge system at 180 m did not affect microbial community structure and composition. Temperature seemed to be a primary factor in the variance between shallow depths. In conclusion, the lack of light, low dissolved oxygen levels, and low temperature do not restrict microbial diversity, as proven by the higher diversity observed in deeper zones. Wastewater in Black Sea region may be discharged into the Black Sea to depth of 180 m or deeper without impacting microbial ecology.

This study reports, for the first time, the baseline levels for fifteen trace metals in M. galloprovincialis tissue from around the Mediterranean, Marmara and Black Seas. The environmental quality in the surrounding seawater was assessed i.e., a mussel farm was investigated by using CF and DC indices, and the water quality was qualified as good for the aquacultural activities. A strong Cu-regulation in the transplanted mussels was observed and it ranged between 3.20 and 3.60 µg/g d.w. The highest bioavailability and bioconcentration of the particulate Fe fraction could present a health risk to consumers with a low risk level (1 < THQ < 9.9). Cr is considered the limiting metal for mussel consumption (< 2 kg/day). The metal contamination gradient was assessed using TEPI and TESVI indices that identified seven reference stations on the large scale and revealed that Cd is the most investigated metal in the literature databases, and found that Pb was the most bioavailable contaminant in the areas examined.

Rivers are the principal route for terrestrial microplastics to reach the marine environment. The Black Sea exhibits a notable representation because it has a drainage zone almost six times the surface area and is semi-closed, meaning that microplastics tend to gather there. To mitigate MP pollution, it is necessary to identify the contamination sources and then raise public awareness. Thus, the current study focused on the MP presence in the sediment of streams running into the SE Black Sea. 594 MPs were observed in sediment samples from 16 stations along the 350 km coastline. The abundance of MP was higher, particularly in streams that pass through locations with high tourism and industrial activity levels. Detected MPs ranged between 0.1 and 5 mm, while the overall density was smaller than 1 mm. Fragments and fiber MPs were regularly detected, although the presence of films was rarely recorded. The polymer structures that were most commonly observed in the analyzed pollutants were PET and PE. The current study uncovered MP contamination in stream sediments originating from Türkiye's Eastern Black Sea basin and might be a baseline work for future inland water studies.

The Black Sea is a permanently anoxic, marine basin serving as model system for the deposition of organic-rich sediments in a highly stratified ocean. In such systems, archaeal lipids are widely used as paleoceanographic and biogeochemical proxies; however, the diverse planktonic and benthic sources as well as their potentially distinct diagenetic fate may complicate their application. To track the flux of archaeal lipids and to constrain their sources and turnover, we quantitatively examined the distributions and stable carbon isotopic compositions (δ13 C) of intact polar lipids (IPLs) and core lipids (CLs) from the upper oxic water column into the underlying sediments, reaching deposits from the last glacial. The distribution of IPLs responded more sensitively to the geochemical zonation than the CLs, with the latter being governed by the deposition from the chemocline. The isotopic composition of archaeal lipids indicates CLs and IPLs in the deep anoxic water column have negligible influence on the sedimentary pool. Archaeol substitutes tetraether lipids as the most abundant IPL in the deep anoxic water column and the lacustrine methanic zone. Its elevated IPL/CL ratios and negative δ13 C values indicate active methane metabolism. Sedimentary CL- and IPL-crenarchaeol were exclusively derived from the water column, as indicated by non-variable δ13 C values that are identical to those in the chemocline and by the low BIT (branched isoprenoid tetraether index). By contrast, in situ production accounts on average for 22% of the sedimentary IPL-GDGT-0 (glycerol dibiphytanyl glycerol tetraether) based on isotopic mass balance using the fermentation product lactate as an endmember for the dissolved substrate pool. Despite the structural similarity, glycosidic crenarchaeol appears to be more recalcitrant in comparison to its non-cycloalkylated counterpart GDGT-0, as indicated by its consistently higher IPL/CL ratio in sediments. The higher TEX86 , CCaT, and GDGT-2/-3 values in glacial sediments could plausibly result from selective turnover of archaeal lipids and/or an archaeal ecology shift during the transition from the glacial lacustrine to the Holocene marine setting. Our in-depth molecular-isotopic examination of archaeal core and intact polar lipids provided new constraints on the sources and fate of archaeal lipids and their applicability in paleoceanographic and biogeochemical studies.

This study aimed to assess the suitability of the potential solid waste landfill sites in seven provinces (Samsun, Ordu, Giresun, Trabzon, Gümüshane, Bayburt and Artvin) in the Eastern Blacksea Region of Türkiye. The earthquake hazard analysis for two major earthquakes which occurred in the region was first carried out. Then, the geophysical methods including seismic refraction tomography (SRT), electrical resistivity tomography (ERT) and Multichannel Analysis of Surface Waves (MASW) were conducted to find out the structural and physical properties of the subsurface which include the layering, soil classification based on VS30 and the groundwater content at 25 locations of 13 in target provinces. The integrated interpretation of whole data sets demonstrates that Isiktepe, Esence, Çamburnu and Kazantas which are characterized by VP > 1200 m/s, VS30 ≥ 400 m/s, ρ > 70 Ohm-m, low earthquake hazard and seismicity are more suitable among others. Vezirköprü, Sebinkarahisar, Yenice, Bayburt-Center, Balkaynak and Murgul will be suitable after a geotechnical reclamation due to moderate seismic velocities and electrical resistivity which are 900 < VP ≤ 1200 m/s, 200 < VS30 < 400 m/s and 10 < ρ ≤ 70 Ohm-m representing stiff and wet soils. In addition, Bafra, Agalik and Ovacik were considered to be unsuitable due to the presence of thick, water-saturated soft soil and extremely weathered rocks. Finally, this study shows that the joint interpretation of seismicity and geophysical data in potential waste landfill sites, extremely important for the planning and development of a city, can provide the valuable information which will enable to prevent possible deformations, environmental problems and economic losses after waste landfill.

The TvV-SM2 virus, isolated from the coastal waters of the Black Sea, causes lysis of its host, the algae Tetraselmis viridis (Chlorophyta). Under optimal conditions for nutrients, an increase in the initial abundance of algae cells by four times caused a 3-fold reduction in the latent period of viral infection. During the period of the most rapid cell lysis of T. viridis , nitrogen deficiency leads to a decrease in the average daily rate of death of cells affected by the virus by 3.2times relative to the replete conditions, while in the case of phosphorus deficiency, this process slows down by up to 2.4times. Under deplete conditions, the rate of cell death was only 34% lower than under replete conditions. The effect of copper ions (100µgL-1 ) on the viral suspension for 6h led to the complete suppression of its activity. In the presence of the host of this virus, its activity is only partially suppressed. As a result, cell lysis under the influence of a viral infection occurred in two stages. The first stage was noted only during the first 6h of the experiment. The second main stage took place within 78-170h. This study showed that in conditions of nutrient deficiency and in the presence of copper ions in seawater, the impact of viruses on microalgae will be weaker.

Microbial lipids, used as taxonomic markers and physiological indicators, have mainly been studied through cultivation. However, this approach is limited due to the scarcity of cultures of environmental microbes, thereby restricting insights into the diversity of lipids and their ecological roles. Addressing this limitation, here we apply metalipidomics combined with metagenomics in the Black Sea, classifying and tentatively identifying 1623 lipid-like species across 18 lipid classes. We discovered over 200 novel, abundant, and structurally diverse sphingolipids in euxinic waters, including unique 1-deoxysphingolipids with long-chain fatty acids and sulfur-containing groups. Sphingolipids were thought to be rare in bacteria and their molecular and ecological functions in bacterial membranes remain elusive. However, genomic analysis focused on sphingolipid biosynthesis genes revealed that members of 38 bacterial phyla in the Black Sea can synthesize sphingolipids, representing a 4-fold increase from previously known capabilities and accounting for up to 25% of the microbial community. These sphingolipids appear to be involved in oxidative stress response, cell wall remodeling, and are associated with the metabolism of nitrogen-containing molecules. Our findings underscore the effectiveness of multi-omics approaches in exploring microbial chemical ecology.

BACKGROUND: Recent studies have reported the identity and functions of key anaerobes involved in the degradation of organic matter (OM) in deep (> 1000 m) sulfidic marine habitats. However, due to the lack of available isolates, detailed investigation of their physiology has been precluded. In this study, we cultivated and characterized the ecophysiology of a wide range of novel anaerobes potentially involved in OM degradation in deep (2000 m depth) sulfidic waters of the Black Sea. RESULTS: We have successfully cultivated a diverse group of novel anaerobes belonging to various phyla, including Fusobacteriota (strain S5), Bacillota (strains A1T and A2), Spirochaetota (strains M1T, M2, and S2), Bacteroidota (strains B1T, B2, S6, L6, SYP, and M2P), Cloacimonadota (Cloa-SY6), Planctomycetota (Plnct-SY6), Mycoplasmatota (Izemo-BS), Chloroflexota (Chflx-SY6), and Desulfobacterota (strains S3T and S3-i). These microorganisms were able to grow at an elevated hydrostatic pressure of up to 50 MPa. Moreover, this study revealed that different anaerobes were specialized in degrading specific types of OM. Strains affiliated with the phyla Fusobacteriota, Bacillota, Planctomycetota, and Mycoplasmatota were found to be specialized in the degradation of cellulose, cellobiose, chitin, and DNA, respectively, while strains affiliated with Spirochaetota, Bacteroidota, Cloacimonadota, and Chloroflexota preferred to ferment less complex forms of OM. We also identified members of the phylum Desulfobacterota as terminal oxidizers, potentially involved in the consumption of hydrogen produced during fermentation. These results were supported by the identification of genes in the (meta)genomes of the cultivated microbial taxa which encode proteins of specific metabolic pathways. Additionally, we analyzed the composition of membrane lipids of selected taxa, which could be critical for their survival in the harsh environment of the deep sulfidic waters and could potentially be used as biosignatures for these strains in the sulfidic waters of the Black Sea. CONCLUSIONS: This is the first report that demonstrates the cultivation and ecophysiology of such a diverse group of microorganisms from any sulfidic marine habitat. Collectively, this study provides a step forward in our understanding of the microbes thriving in the extreme conditions of the deep sulfidic waters of the Black Sea. Video Abstract.

OBJECTIVE: A large number of patients applying to the dermatology clinics are affected by fungal diseases, and a significant portion of which are superficial fungal infections. Dermatophyte infections are a notable public health concern and frequently encountered in clinical practice. Dermatophytosis not only compromises the quality of life but also predisposes individuals to various comorbidities due to its role as a gateway for secondary bacterial agents. This study aims to determine the species distribution of dermatophytes prevalent and assess their susceptibility to antifungal drugs. PATIENTS AND METHODS: Skin, nail, and hair samples were obtained from patients with a clinical diagnosis of dermatophytosis. Samples were all cultured to isolate and identify the species. In vitro liquid microdilution tests were conducted to assess the susceptibility of the isolated strains against terbinafine, fluconazole, griseofulvin, and butenafine. RESULTS: A total of 353 samples were obtained from the hair, skin, and nail lesions of 326 patients. Dermatophyte was isolated in 71 of the samples (20.1%). The cultured dermatophyte subtypes included Trichophyton rubrum (13.8% in 49 samples), Microsporum audouini (5.7% in 20 samples), and Trichophyton mentagrophytes (0.6% in 2 samples). Antifungal susceptibility testing revealed that terbinafine was the most effective antifungal drug against all dermatophyte species, while fluconazole exhibited the highest resistance. CONCLUSIONS: The most common dermatophytosis agent in our region is T. rubrum. The least antifungal resistance was found against terbinafine. Conducting antifungal susceptibility tests is crucial for selecting effective treatment regimens and early detection of resistance development.

To date, 59 species (61 including infraspecific taxa) of the heterotrophic genus Protoperidinium have been found in the Black Sea. With rare exceptions, the records of most species were not confirmed by either line drawings or microphotographs, and lacked indications of important diagnostic traits. To partially correct these deficiencies, we conducted morphological examinations of the dominant Protoperidinium species in the Black Sea. The study provides original descriptions, line drawings, and microphotographs of 22 species of Protoperidinium with their global distribution maps. Protoperidinium quadrioblongum is reported from the Black Sea for the first time.

The examination of benthic materials collected in September 2022 at six stations off Kyky (south-west coast of the Black Sea, Trkiye) both on hard (three stations) and soft substrata (three stations) at depths ranging from 8 and 27 m revealed a total of 57 polychaete species belonging to 22 families. Among the species, the polynoid Malmgrenia polypapillata is a new record for the Black Sea fauna, and three species belonging to the Syllidae (Erinaceusyllis defneae n. sp.) and Terebellidae (Polycirrus karadenizicus n. sp. and P. rhombolabiatus n. sp.) are new to science. Algae dominated (Cystoseira cinita, Cladophora sp. and Phyllophora crispa) rocky substrata had 30 polychaete species and a maximum polychaete density of 7325 individuals/m2, while soft bottom substrata (fine sand and muddy sand with shell fragments) possessed 38 species and a maximum polychaete density of 3520 individuals/m2. The family Nereididae (3176% of the total number of individuals), and the species Platynereis dumerilii and Polyphthalmus pictus dominated hard bottom stations, whereas the families Nephtyidae, Spionidae and Dorvilleidae, and the species Micronephthys longicornis, Protodorvillea kefersteini and Prionospio maciolekae were represented by high number of individuals at soft-bottom stations.

ABSTRACT Aim: Although prostate cancer (PCa) is the most common cancer type in men, a replaceable risk factor has not yet been established. In our study, we assessed the relationship between the number of sexual partners, age of first sexual experience and age of first masturbation and prostate cancer incidence. Materials and Methods: In Ordu University Department of Urology between January 2013 and September 2016, in PSA elevation and rectal examination, patients with prostate biopsy were evaluated due to nodule palpation in the prostate. At younger ages and at present, their first masturbation ages, first sexual debut ages, and total sexual partner numbers were recorded. The correlation between the obtained data and PCa frequency was evaluated. Results: The study included 146 patients with PCa identified on biopsy and 171 patients with benign biopsy results who answered the questions. 66.7% of the ones whose biopsy results were benign and 40.6% of cancer suspects had only one sexual partner. The median number of sexual partners was 1±4 (1-100) in the benign group and 2±6 (1-500) in the malignant group (p=0.039). There was a negative correlation between age of first sexual debut and number of partners (r: −0,479; p <0.001). Conclusion: In our study, it appears that there may be an association between the number of sexual partners and prostate cancer in the patient group with PSA level above 4ng/mL. Avoidance of sexual promiscuity or participation in protected sex may be beneficial to protect against prostate cancer.
