[Role of PD153035 in the induction of apoptosis of XG-1 myeloma cell line].

ABSTRACT
OBJECTIVE:
To study the role of epidermal growth factor receptor (EGFR) in the proliferation and survival of human myeloma cells.METHODS:
The inhibitor of EGFR, PD153035, was used to block the signal transduction of EGFR. The proliferation and apoptosis of myeloma cell line, XG-1, were detected by 3H-TCR incorporation assay and Annexin V staining analysis, respectively. The phosphatation of STAT3, the key activate signal to the myeloma cell proliferation, was detected with Western blot.RESULTS:
PD153035 decreased the proliferation of XG-1 and induced an obvious apoptosis in XG-1. The phosphatation of STAT3 induced by HB-EGF but not by IL-6 was blocked by PD153035.CONCLUSION:
The proliferation and survival of myeloma cells may be suppressed by PD153035 due to the blockage of phosphatation of STAT3 induced by the activation of EGFR.
