This study was conducted to determine the seroprevalence of anti-Neospora caninum antibodies and to investigate the risk factors related to seroprevalence in dogs from urban and rural areas with distinct economic activities (milk and coffee production) in Minas Gerais state, Brazil. For this purpose, blood samples from 703 dogs were collected and questionnaires addressing epidemiological aspects were completed by dog-owners. The sera were analysed for anti-N. caninum antibodies by indirect fluorescent antibody tests (IFAT ≥ 1:50). Association between epidemiological aspects and seropositivity in dogs was evaluated with multivariate logistic regression models. A total of 80 (11·4%) dogs tested positive for N. caninum. In the multivariate logistic regression models, dogs aged >4 years, dogs used as guard dogs, dogs that spontaneously hunt, and history of bovine abortion were found to be greater risk factors for canine N. caninum infection. When we considered only dogs from rural areas, an association with seroprevalence was seen for milk farms, dogs not fed with commercial food, dogs that hunt, and dogs used as guard dogs.

Carassius auratus is a teleost fish that has been largely used in behavioral studies. However, little is known about potential environmental influences on its performance of learning and memory tasks. Here, we investigated this question in C. auratus, and searched for potential correlation between exercise and visuospatial enrichment with the total number of telencephalic glia and neurons. To that end, males and females were housed for 183 days in either an enriched (EE) or impoverished environment (IE) aquarium. EE contained toys, natural plants, and a 12-hour/day water stream for voluntary exercise, whereas the IE had none of the above. A third plus-maze aquarium was used for spatial and object recognition tests. Different visual clues in 2 of its 4 arms were used to guide fish to reach the criteria to complete the task. The test consisted of 30 sessions and was concluded when each animal performed three consecutive correct choices or seven alternated, each ten trials. Learning rates revealed significant differences between EE and IE fish. The optical fractionator was used to estimate the total number of telencephalic cells that were stained with cresyl violet. On average, the total number of cells in the subjects from EE was higher than those from subjects maintained in IE (P=0.0202). We suggest that environmental enrichment significantly influenced goldfish spatial learning and memory abilities, and this may be associated with an increase in the total number of telencephalic cells.

Autosomal dominant tubulointerstitial kidney disease (ADTKD) is characterized by autosomal dominant inheritance, progressive chronic kidney disease, and a bland urinary sediment. ADTKD is most commonly caused by mutations in the UMOD gene encoding uromodulin (ADTKD-UMOD). We herein report the first confirmed case of a multi-generational Brazilian family with ADTKD-UMOD, caused by a novel heterozygous mutation (c.163G>A, GGCâAGC, p.Gly55Ser) in the UMOD gene. Of 41 family members, 22 underwent genetic analysis, with 11 individuals found to have this mutation. Three affected individuals underwent hemodialysis, one peritoneal dialysis, and one patient received a kidney transplant from a family member later found to be genetically affected. Several younger individuals affected with the mutation were also identified. Clinical characteristics included a bland urinary sediment in all tested individuals and a kidney biopsy in one individual showing tubulointerstitial fibrosis. Unlike most other reported families with ADTKD-UMOD, neither gout nor hyperuricemia was found in affected individuals. In summary, we report a novel UMOD mutation in a Brazilian family with 11 affected members, and we discuss the importance of performing genetic testing in families with inherited kidney disease of unknown cause.

Studies have shown that nuclear factor erythroid 2-related factor 2 (Nrf2) can be modulated by physical exercise. However, the impact of resistance exercise has never been investigated in patients with chronic kidney disease (CKD). The aim of this study was to evaluate the effects of resistance exercise programs on the expression of transcription factors Nrf2 and nuclear factor κB (NF-κB) in CKD patients on hemodialysis (HD). Patients on an HD program were randomly assigned to an exercise group of 25 patients (54.5% women, aged 45.7±15.2years and time on dialysis=71.2±45.5months) or a control group of 19 patients who had no exercise intervention (61.5% women, aged 42.5±13.5years and time on dialysis=70.1±49.9months). A strength exercise program was performed 3 times a week during the HD sessions. Peripheral blood mononuclear cells were isolated and processed for the expression of Nrf2 and NF-κB by quantitative real-time polymerase chain reaction 3months before and after the exercise program. Using an enzyme-linked immunosorbent assay, the activity of glutathione peroxidase (GPx) as well as the products of high-sensitivity C-reactive protein and nitric oxide (NO) were assessed. Nrf2 expression (ranging from 0.86±0.4 to 1.76±0.8) and GPx activity were significantly increased after exercise intervention. In the exercise group, no difference in the levels of NO was observed; however, there was a significant reduction in the control group. In conclusion, these data suggest that resistance exercises seem to be capable of inducing Nrf2 activation in CKD patients on HD.

Patients with chronic kidney disease (CKD) have various metabolic disorders caused by a chronic state of oxidative stress and inflammation, and recently, nuclear factor-erythroid 2-related factor 2 (Nrf2) has emerged as a factor that plays a significant role in cellular protection against oxidative stress and inflammation. This transcription factor when activated can regulate antioxidant and anti-inflammatory cellular responses leading to the expression of detoxifying enzymes. Studies have shown that Nrf2 expression can be modulated by several factors, such as bioactive compounds and physical exercise. In fact, exercise in CKD patients can bring many benefits; however, there are no studies correlating physical activity and Nrf2 expression in CKD patients. This review aims to discuss whether there is any evidence to justify a recommendation of physical exercise in CKD patients as a non-pharmacological option to activate the Nrf2 pathway.

OBJECTIVE: The acute lymphocytic leukemia is a hematopoietic cancer that occurs predominantly in children. Methotrexate is one of the most useful drugs in cancer chemotherapy. The aim of the study was to develop and validate the methodology of high performance liquid chromatography (HPLC) with ultraviolet detection for methotrexate dosage and to determine its concentration in plasma samples from children with leukemia. PATIENTS AND METHODS: The study included patients from the outpatient care of pediatric oncology at the Faculty of Medicine of ABC carriers in treatment of leukemia. The study was conducted in chromatographic model Agilent 1100 with UV detector at 302 nm and by the method of ELISA microplate reader capable of reading absorbance at 450 nm. RESULTS: We obtained satisfactory results of selectivity, accuracy, linearity, limit of quantification (LOQ), limit of detection (LOD), precision and robustness and apply the basic criteria for validation as RE No. 899, of May 29, 2003 Guide validation of analytical and bioanalytical National Agency Health Surveillance (ANVISA). CONCLUSIONS: We conclude that results for linearity/concentration range, precision, robustness, limit of quantification and detection limits are within the acceptance criteria defined by ANVISA and that the developed analytical method is valid and feasible to be used as a tool in monitoring therapy of methotrexate.

The use of Kumakhov capillary optics will significantly enhance the performance of near-monochromatic, Compton backscattered x-ray programs. The Vanderbilt University Medical Free-Electron Laser Center is developing the capability to create these tunable x rays for medical imaging. The present transport has only reflection optics, and the beam is quite large in diameter at the laboratory. Low loss collimation of this beam would allow higher x-ray intensities after transport. This article describes experimental and computer simulation results which predict the expected performance for a multifiber Kumakhov collimator for use in the x-ray beam transport. Estimates from our research are that a multifiber optic formed of individual polycapillary fibers could be used to capture the full 7 mrad of the Vanderbilt x-ray beam and collimate it to a 1-2 mrad divergence with approximately 40%-50% transmission efficiency. This optic should increase the x-ray intensity at the laboratory level by a factor of > or = 5 by decreasing the beam divergence and subsequent spot size. Additionally, analysis of monolithic optics of fused multicapillary fibers predicts an increase in the intensity of the x rays at the laboratory by a factor of 55. These optics can have tapered channels that greatly decrease their exit divergence. This will greatly enhance the capabilities of this unique x-ray source. This article reports the initial results from a collaboration between Vanderbilt, The Center for X-Ray Optics at University at Albany, SUNY, and X-Ray Optical Systems in Albany, NY.

Capillary optic arrays are bundles of hollow glass capillaries which guide x rays in a manner similar to the way fiber optics guide light. Focused postpatient capillary optic arrays have the potential to significantly improve both contrast and resolution of mammographic images compared to conventional antiscatter grids. Contrast can be improved by the nearly total scatter rejection of the optic. Effective resolution can be improved by geometric magnification without increased focal spot blurring. The best results were found for borosilicate glasses, with transmissions in excess of 60% for 22-cm-long fibers. To evaluate the scatter rejection properties, the transmission of off-axis radiation was measured. Transmission drops to < 1% at an angular displacement of 2.7 mrad. Transmission of a bulk capillary array dropped to near zero if the source was at an angle of 2.5 mrad. This implies excellent scatter rejection capabilities. To evaluate whether unchanneled photons might still reach the detector, absorption measurements were also performed on fibers and arrays. Absorption was found to be adequate for scatter rejection. All of the data agreed well with numerical simulations. Performance calculations for two potential optics geometries gave promising results.

Computed radiography (CR) has shown promise in digital mammographic screening due to its good low spatial frequency MTF and its relatively wide exposure latitude. The CR image format has not gained acceptance clinically because of reduced high spatial frequency resolution as compared to film-screen images. X-ray capillary optics, aligned between the breast and CR phosphor imaging plate, will capture primary x-ray photons almost exclusively. Due to the very small angle of acceptance, scattered photons angled more than about 1.6 x 10(-3) radians from primary trajectory will not be accepted at the capillary optic entrance. The virtual elimination of detected scatter means almost 100% of the possible primary contrast should be visible in the image. In addition, the image can be magnified without focal spot blurring. Effective resolution of CR images can be increased by a factor equal to that magnification. Clinical implementation of future capillary optics are expected to be either in the form of a large, stationary, post-patient optic that accepts primary from the entire breast or a fan-shaped optic that is scanned across the breast. Measurements of a test capillary optic showed a reduction of scatter fraction to 0.018. Images of a lucite contrast detail phantom revealed a corresponding increase in image contrast when compared to anti-scatter grid and no grid methods. Spectral transmission measurements using a high-purity germanium detector showed good primary transmission (45%-50%) in the mammographic energy range. The MTF measurements of both stationary and scanned capillary optics showed improvement at the 5% MTF level to 8.4 mm-1 for scanned optics and 9.2 mm-1 for stationary optics representing a 68% and 84% respective increase over the CR MTF without magnification or capillary optics.

Undifferentiated carcinoma of the pancreas is a malignant neoplasm that is uncommon among domestic species, especially cockatiels (Nymphicus hollandicus), one of the most popular birds kept as a pet throughout the world. The aim of this study was to describe the occurrence of an undifferentiated carcinoma in the pancreas of a cockatiel. A bird, an adult male that died naturally with swelling in the abdominal region, was referred to necropsy. Macroscopic examination showed poor body condition, the coelomic cavity filled with liquid and a white mass attached to the pancreas and other smaller masses attached to the duodenum. Tissue samples and organs were harvested and fixed in 10% buffered formalin, then routinely processed for histopathology and stained with hematoxylin and eosin. Microscopic analysis demonstrated an epithelial neoplasia with a predominantly solid pattern, lymphatic invasion and involvement of the intestinal serous membrane. These findings indicate the occurrence of an undifferentiated pancreatic carcinoma in a cockatiel that was diagnosed by histopathology.(AU)
O carcinoma indiferenciado de pâncreas é uma neoplasia maligna, incomum entre as espécies domésticas, especialmente em calopsitas (Nymphicus hollandicus), uma das aves mais populares como animal de companhia no mundo. O objetivo deste trabalho foi descrever a ocorrência de carcinoma indiferenciado de pâncreas em Nymphicus hollandicus. Uma ave, macho adulto, com morte natural e com aumento de volume em região abdominal, foi encaminhada para necropsia. Ao exame macroscópico foram observados mau estado corporal, cavidade celômica repleta de líquido e massa esbranquiçada aderida ao pâncreas e outra menor aderida ao duodeno. Amostras de tecidos e órgãos foram colhidas em formol 10% tamponado, processadas rotineiramente para histopatologia e coradas por hematoxilina e eosina. Na microscopia foi observada neoplasia epitelial com padrão predominantemente sólido, com invasão linfática e implantação na serosa intestinal. Com esses achados, comprovou-se a ocorrência de carcinoma indiferenciado de pâncreas em Nymphicus hollandicus, o qual pode ser diagnosticado por meio de histopatologia.(AU)

Cytoplasmic inclusion bodies in adrenal medullary chromaffin cells have been described in various species including humans. These inclusions are believed to be related to certain infectious, toxic and neurodegenerative diseases. No reports concerning such adrenal inclusions have been described in bovines. Adrenal glands from twenty bovines were evaluated in a retrospective study. Seven of these exhibited inclusions - three cases of rabies, two cases of chronic suppurative bronchopneumonia, one case of chronic suppurative peritonitis, and one case of gangrenous mastitis. The inclusions were present in higher numbers especially in cases of rabies and also in one case of chronic suppurative bronchopneumonia. The inclusions were intracytoplasmic, eosinophilic, rounded, single or multiple, of various sizes, strongly stained by PAS and were present in higher numbers in the external layer of the adrenal medulla. The inclusions were negative when subjected to immunohistochemistry for detection of viral antigens in the cases of rabies. Although inclusion bodies were present in adrenal glands devoid of other histological alterations, they were more abundant in cases in which the adrenal gland had other alterations. The correlation between certain diseases and the development of inclusion bodies is not known, which highlights the importance of further studies on these inclusions in adrenal glands of bovines.
