Asymmetric conjugate additions of phosphonates to trans-crotonophenone and chalcone derivatives using a diaminomethylenemalononitrile organocatalyst resulted in the generation of the corresponding chiral γ-ketophosphonates in high yields with excellent enantioselectivities (up to 95% ee). This report is the first successful example of asymmetric 1,4-additions of phosphonate to α,ß-unsaturated ketones using an organocatalyst.

Herein, a facile method is developed for the synthesis of vicinal bisphosphine derivatives based on a cascade of hydrophosphinylation, elimination, and hydrophosphinylation of secondary phosphine oxides with nitroalkenes. This cascade reaction provides step-economy access to a series of vicinal bisphosphine derivatives with high to excellent yields (up to 99%). This method was further extended to prepare, in one-pot, regioselective vicinal bisphosphine derivatives that incorporated two different phosphorus functional groups.

An improved diaminomethylenemalononitrile organocatalyst, bearing a N,N-disubstituted structure, promoted enantioselective conjugate addition reaction of α-branched aldehydes with vinyl sulfone, affording adducts with excellent enantioselectivities (up to 96% ee). Mechanistic studies revealed that the diaminomethylenemalononitrile motif holds the vinyl sulfone substrate using a single hydrogen bond accompanied by multiple weak interactions, including electrostatic C-Hâ â â O interactions.

A diaminomethylenemalononitrile organocatalyst efficiently promotes the asymmetric direct vinylogous conjugate additions of α-angelica lactones to benzoyl acrylonitrile derivatives, resulting in the corresponding addition products bearing vicinal tertiary and quaternary stereogenic centers with excellent enantioselectivities (up to 99% ee). This report is the first successful example of the asymmetric conjugate additions of α-angelica lactone to benzoyl acrylonitriles. The chiral γ,γ-disubstituted γ-butenolides obtained can be readily transformed to the bicyclic γ-lactam derivative as a valuable synthetic intermediate.

The aim of this study was to investigate the relationship between postprandial remnant-like particle (RLP) metabolism and insulin resistance (IR). The study group consisted of 52 randomly selected subjects. To evaluate postprandial hyperlipidemia, serum lipid and lipoprotein concentrations during fasting and 4h after the fat-loading test were measured in each subject. IR was assessed using the index of homeostasis model assessment (HOMA-R). The subjects were divided into two groups according to the value of HOMA-R: an IR group (n=17) with a HOMA-R value >/=1.73, and a normal (NR) group (n=35) with a HOMA-R value <1.73. Both fasting and postprandial RLP-cholesterol (RLP-C) concentrations were higher in the IR group than in the NR group (6.2+/-2.6 versus 4.1+/-1.7mg/dl fasting value, and 9.7+/-4.0 versus 5.8+/-2.9mg/dl postprandial value). The changes in RLP-C concentration during the fat-loading test were twice as high in the IR group compared with the NR group (3.5+/-2.4 versus 1.6+/-1.6mg/dl, P=0.0022). The HOMA-R correlated significantly with both fasting and postprandial triglyceride (r=0.41 and 0.43, respectively) and RLP-C (r=0.36 and 0.50, respectively) in all subjects. Multiple regression analysis indicate that postprandial RLP-C concentration was an independent predictor of HOMA-R regardless of age, BMI, and other lipid profiles. Thus, postprandial RLP metabolism is closely related to IR. Atherosclerotic proliferation in IR syndrome may be caused by the accumulation of postprandial remnant lipoproteins after the daily fat intake.

This study compared the effects of long-term administration of nicorandil and isosorbide dinitrate (ISDN) on vascular endothelial function and the progression of arteriosclerosis. Forty-two patients with ischemic heart disease were randomly allocated to receive nicorandil (N group; 15 mg/d) or ISDN (I group, 40 mg/d). Twelve normal subjects served as controls. Vascular endothelial function and the progression of arteriosclerosis (intima-media thickness, IMT), as determined by carotid vascular ultrasound, were assessed 1 week before and 3 months after drug administration. Reactive hyperemia was induced in the forearm for 5 minutes, and the percentage change in the diameter of the brachial artery (% change in flow-mediated dilation, %FMD) was calculated. FMD was significantly lower in CAD groups than in controls. The %FMD significantly decreased (7.2 +/- 1.9 to 4.2 +/- 2.8) in the I group, while rising from 6.8 +/- 1.6 to 8.0 +/- 2.0 in the N group. IMT increased by 0.036 +/- 0.015 mm in the I group but showed no significant change in the N group (-0.01 +/- 0.012 mm). Thus, ISDN deteriorates IMT and FMD, whereas a beneficial effect of nicorandil is seen on FMD with no effect on IMT. Long-term treatment with nicorandil may be desirable for prevention of cardiovascular events.
