RESUMEN

A tick survey was conducted to document tick-host associations with Florida (USA) wildlife, and to determine the relative abundance and distribution of ixodid ticks throughout the state. The survey was conducted using collection kits distributed to licensed Florida hunters as well as the examination of archived specimens from ongoing state wildlife research programs. Collected tick samples were obtained from 66% of Florida counties and were collected from nine wildlife hosts, including black bear, bobcat, coyote, deer, gray fox, Florida panther, raccoon, swine, and wild turkey. In total, 4176 ticks were identified, of which 75% were Amblyomma americanum, 14% Ixodes scapularis, 8% A. maculatum, 3% Dermacentor variabilis, and < 1% were I. affinis and I. texanus. americanum, D. variabilis, and I. scapularis had the broadest host range, while A. maculatum, D. variabilis, and I. scapularis had the widest geographic distribution. While the survey data contribute to an understanding of tick-host associations in Florida, they also provide insight into the seasonal and geographic distribution of several important vector species in the southeastern USA.

Mosquito surveillance in remote areas with limited access to canisters of CO2 or dry ice will benefit from an effective alternative CO2 source, such as the natural production of CO2 from yeast fermentation. In this study, we investigate differences in mosquito capture rates from the Centers for Disease Control and Prevention (CDC) light traps baited with dry ice compared with traps baited with yeast fermentation of several carbohydrate sources over 23 trap-nights. Results demonstrated the ability of yeast-generated CO2 to effectively attract mosquitoes to a CDC trap, regardless of carbohydrate source. Total collections of mosquitoes using dry ice were significantly higher than collections from yeast-generated CO2 sources. However, mosquito community structure, i.e., the species and relative capture rate of each species, was represented comparably across collections regardless of CO2 source. Volatiles produced by yeast fermentation were analyzed by carbohydrate source, revealing a suite of compounds, possibly synergistic, enhancing effects with CO2 on mosquito collection capability compared with the amount of CO2 used to attract mosquitoes.

Voluntary collections of ticks from domestic dogs and cats by veterinary practitioners across Florida, USA, were conducted over a 10 month period. Of the 1337 ticks submitted, five species of ixodid ticks were identified and included Rhipicephalus sanguineus, Amblyomma americanum, A. maculatum, Dermacentor variabilis, and Ixodes scapularis. Most ticks were collected from dogs (98.4%) with the most predominant species being R. sanguineus (94.3%). Of the ticks collected from cats (1.6%), A. americanum were the most common (74%). Only R. sanguineus were collected throughout the state, with the other species collected only in central and north Florida. The tick species collected from dogs and cats represent a risk to these domestic species as well as associated humans for a range of tick-borne diseases in Florida.

The brown dog tick, Rhipicephalus sanguineus (Latrielle), can be a serious residential pest due to its unique ability, among ticks, to complete its lifecycle indoors. A single engorged and fertilized female tick can oviposit around 4,000 eggs, allowing indoor establishment to be rapid and easy to miss in early-stage infestations. Acaricide treatment is currently the primary method of control, but can be costly and can lead to the development of acaricide resistance in tick populations. Traps of various designs can be used to help monitor and manage populations of indoor pests, such as cockroaches and bed bugs, but there are currently no commercially available traps for use with brown dog tick infestations. This study included a comparison of four commercially available bed bug traps (NightWatch [BioSensory Inc., Putnam, CT], Bed Bug Beacon [PackTite, Fort Collins, CO], ClimbUp [Susan McKnight Inc., Memphis, TN], and Verify [FMC Corporation, Philadelphia, PA]) with regard to their efficacy in capturing brown dog ticks, and also compared tick attraction to ClimbUp traps baited with several stimuli including CO2. Significantly more ticks were captured and attracted to the NightWatch and CO2-baited ClimbUp traps than the other two trap models. The results suggest that bed bug traps may be useful in brown dog tick monitoring, and CO2 will likely be an important component of a trapping system employed in the future.

Permethrin is a commonly used acaricide for tick control on domestic animals and in residential environments, while fipronil use is restricted to on-animal treatment. Following widespread reports of permethrin and fipronil application failures to control indoor infestations of Rhipicephalus sanguineus (Latreille), the brown dog tick, 31 tick populations were obtained from Florida and Texas for acaricide resistance screening. These field-collected ticks from kennels and residential facilities were challenged with technical grade permethrin and fipronil to create dose response curves that were compared with an acaricide-susceptible strain. Permethrin resistance was successfully screened in nine populations, all of which were resistant or highly resistant. Tick susceptibility to fipronil was conducted on four populations, which were found to be tolerant, with resistance ratios below 10. This is the first documentation of R. sanguineus permethrin resistance and fipronil tolerance in the United States. Potential causes of resistance development and recommendations on future brown dog tick control management plans are discussed.

The attraction of female Aedes aegypti to single compounds and binary compositions containing L-lactic acid and an additional saturated compound from a set of ketones, sulfides, and chloroalkanes was studied using a triple-cage dual-port olfactometer. These chemical classes were studied because of their structural relation to acetone, dimethyl disulfide, and dichloromethane, which have all been reported to synergize attraction to L-lactic acid. Human odors, carbon dioxide, and the binary mixture of L-lactic acid and CO2served as controls for comparison of attraction responses produced by the binary mixtures. All tested mixtures that contained chloroalkanes attracted mosquitoes at synergistic levels, as did L-lactic acid and CO2. Synergism was less frequent in mixtures of L-lactic acid with sulfides and ketones; in the case of ketones, synergistic attraction was observed only for L-lactic acid combined with acetone or butanone. Suppression or inhibition of attraction response was observed for combinations that contained ketones of C7-C12 molecular chain length (optimum in the C8-C10 range). This inhibition effect is similar to that observed previously for specific ranges of carboxylic acids, aldehydes, and alcohols.

Euxesta eluta Loew and Chaetopsis massyla Walker (Diptera: Ulidiidae) are primary pests of sweet corn in Florida. Attraction of adult flies to various visual stimuli was evaluated in the laboratory to provide insight into the potential development of enhanced trapping strategies. In assays evaluating different colored sticky traps, more E. eluta were collected on light blue, mid blue, lime green, and orange yellow traps, whereas attraction of C. massyla was greater to lime green and fluorescent green traps. In a comparison of yellow 3-dimensional shapes, more E. eluta were collected on yellow cylinders than on spheres or cubes; however, more C. massyla were collected on cylinders than on cubes or spheres. When colored traps were placed against a white background, more E. eluta were collected on lime green compared with yellow, blue, and orange traps; however, when placed against a black background, attraction to the lime green trap was reduced. Against the white background attraction of C. massyla, was strongest to yellow, then lime green and orange traps, followed by blue, but with a black background, differences between traps collections were reduced. The addition of ultraviolet (UV) reflectance to yellow increased the attraction of E. eluta, but C. massyla were more attracted to yellows without UV reflection or fluorescence. Black patterns on yellow traps affected attraction, with E. eluta more attracted to wide stripes, a large square, or many small squares, whereas C. massyla was more strongly attracted to stripes. Utilization of these visual attributes could be useful in improving surveillance for these species.

Ultralow volume droplets of DUET, prallethrin, and sumithrin at a sublethal dose were applied to unfed (nonbloodfed) and bloodfed female Aedes aegypti L. and Aedes albopictus (Skuse) in a wind tunnel. Control spray droplets only contained inert ingredients. Individual mosquitoes were videotaped before, during, and after spraying and various behaviors analyzed. During the spray periods of all three pesticide treatments, mosquitoes spent a greater percentage of time moving, and the distance moved was greater than for mosquitoes in the control treatments. In the postspray period, the percent of time moving increased for mosquitoes exposed to all pesticide treatments compared with the controls. After treatment, all females spent more time walking compared with controls, with unfed Ae. aegypti females walking more after exposure to DUET and sumithrin than after exposure to prallethrin and the control. Pesticide exposure increased flying in both species. Sumithrin exposure increased activity and velocity of unfed mosquitoes more than bloodfed mosquitoes. DUET and sumithrin treatments enhanced activity of Ae. aegypti females more than Ae. albopictus females.

A high-throughput bioassay system to evaluate the efficacy of residual pesticides against mosquitoes and muscid flies with minimal insect handling was developed. The system consisted of 4 components made of readily available materials: 1) a CO2 anaesthetizing chamber, 2) a specialized aspirator, 3) a cylindrical flat-bottomed glass bioassay chamber assembly, and 4) a customized rack.

Corn silk flies, or picture-winged flies (Diptera: Ulidiidae), are important pests of fresh market sweet corn in commercial production areas in southern Florida. Issues with pest management related to insecticide resistance, problems in insecticide application, and alternate crop population sources constitute a significant challenge for the protection of developing corn ears. Developed larvae leave cobs and pupate in the soil; however, relatively little is known about these behaviors. In this study, two soil types collected from fields were compared with sand under six different moisture levels in the laboratory to determine the pupation depths of the larvae. Comparisons were carried out concerning the pupation depth of Chaetopsis massyla and Euxesta eluta, which are major pest species in Florida. Both soil type (muck, loamy sand, and sand) and moisture levels (0, 10, 25, 50, 75, 100% field holding capacity) significantly affected pupation depth, with shallow pupation observed under dry or saturated wet conditions. The addition of structures such as pipe cleaners simulating corn roots resulted in deeper pupation under most conditions.

Effective entomological surveillance planning stresses a careful consideration of methodology, trapping technologies, and analysis techniques. Herein, the basic principles and technological components of arthropod surveillance plans are described, as promoted in the symposium "Advancements in arthropod monitoring technology, techniques, and analysis" presented at the 58th annual meeting of the Entomological Society of America in San Diego, CA. Interdisciplinary examples of arthropod monitoring for urban, medical, and veterinary applications are reviewed. Arthropod surveillance consists of the three components: 1) sampling method, 2) trap technology, and 3) analysis technique. A sampling method consists of selecting the best device or collection technique for a specific location and sampling at the proper spatial distribution, optimal duration, and frequency to achieve the surveillance objective. Optimized sampling methods are discussed for several mosquito species (Diptera: Culicidae) and ticks (Acari: Ixodidae). The advantages and limitations of novel terrestrial and aerial insect traps, artificial pheromones and kairomones are presented for the capture of red flour beetle (Coleoptera: Tenebrionidae), small hive beetle (Coleoptera: Nitidulidae), bed bugs (Hemiptera: Cimicidae), and Culicoides (Diptera: Ceratopogonidae) respectively. After sampling, extrapolating real world population numbers from trap capture data are possible with the appropriate analysis techniques. Examples of this extrapolation and action thresholds are given for termites (Isoptera: Rhinotermitidae) and red flour beetles.

Cornsilk flies are serious pests of sweet corn through damage to cobs and secondary fungal establishment. As pupae are generally outside the infested cob on the ground, there can be potential for use of pupal parasitoids for control. Two species of gregarious parasitoids, Muscidifurax raptorellus and Nasonia vitripennis, and three species of solitary parasitoids, Spalangia endius, Spalangia cameroni and Muscidifurax raptor, were evaluated against pupae of the two cornsilk fly species, Euxesta eluta and Chaetopsis massyla. House fly pupae, the most common host for most of the parasitoids, were included for comparison. All of the parasitoids killed and successfully parasitized pupae of the two cornsilk fly species at rates that were similar to house fly pupae. Adult parasitoids that emerged from cornsilk fly hosts were somewhat smaller than parasitoids reared from house flies and had proportionally fewer females. These parasitoids, which are widely and commercially available for filth fly control, warrant further consideration for their potential against cornsilk flies in the field.

The effects of colonization on host-seeking behavior of mosquitoes was examined by comparing attraction responses of newly colonized Aedes aegypti (L.) from field-collected eggs in Puerto Rico to that of the Gainesville (Florida) strain, originally from Orlando (Florida) and in colony since 1952. Females from the Orlando and the F0 through F10 generations of the Puerto Rico strain were evaluated using attractant odors in a triple-cage dual-port olfactometer. Two attractant sources were used: odors from the hand of a volunteer and a standard blend of L-lactic acid, acetone, and dimethyl disulfide. Convergence of the percentage of attraction responses occurred around the F4-F6 generations of the Puerto Rico strain. Both the Orlando and Puerto Rico strains exhibited similar responses for tests with the remaining F7-F10 generations. A temporal effect on mosquito responses was observed for both strains regardless of the attractant blend used in tests. This study indicates that Ae. aegypti host-seeking behavior changes significantly over the first four to six generations after introduction into the laboratory, whereas the field-collected strain increases in attraction response until it stabilizes at a new level.

We sought to visually enhance the attractiveness of a standard black ovitrap routinely used in surveillance of the Asian tiger mosquito, Aedes albopictus, and now being used as lethal ovitraps in Ae. aegypti dengue control programs. Black plastic drinking cups (ovitraps) were visually altered to offer field populations of gravid female Ae. albopictus 6 different oviposition site choices. Trials were conducted at 3 field locations in Gainesville, Orange Park, and Jacksonville, FL, during July-August 2009. A black glossy cup served as the control and was tested against 5 cup choices consisting of white, blue, orange, or black-and-white contrasting patterns (checkered or vertically striped). Means (SE) of eggs collected over 6 wk for each choice were: black 122.53 (9.63) > blue 116.74 (10.74) > checkered 101.84 (9.53) > orange 97.15 (7.95) > striped 84.62 (8.17) > white 81.84 (8.74). Black ovitraps outperformed competing colored and contrasting patterned ovicups with respect to choice from gravid Ae. albopictus seeking artificial oviposition sites.

Recent experiments suggest spatial repellents may significantly reduce biting pressure from host-seeking riceland mosquitoes, such as Anopheles quadrimaculatus, in a warm-humid open-field habitat. However, little is known regarding efficacy of these formulations in partially enclosed spaces where US military personnel may be sheltered or concealed in an operational environment. In this study we investigated the capability of 3 spatial repellents-metofluthrin, linalool, and d-cis/trans allethrin-to reduce mosquito incursion into small open-top enclosures of US military camouflage netting. We found that metofluthrin was more effective in partially enclosed spaces compared with the open field, whereas both linalool and d-cis/trans allethrin provided superior protection in the open. These findings support strategic selection of spatial repellents depending on the environment immediately surrounding the host.

Two commercial formulations of Beauveria bassiana (Balsamo-Crivelli) Vuillemin were applied to residential sites in Old Lyme, CT, for the control of nymphs of the blacklegged tick, Ixodes scapularis, in 1999 and 2000. The pyrethroid bifenthrin was applied to other premises for comparison with B. bassiana. A wood chip barrier was installed and maintained at six of the treated properties. In 1999, control of I. scapularis nymphs ranged from 74.5 to 83.0% on lawns without wood chips and from 88.9 to 90% on lawns with wood chip barriers. As a control check, no ticks (n = 23) collected at pretreatment or control sites died from B. bassiana, although 15 of 18 nymphs from treated lawns developed mycoses. Control of I. scapularis on the lawns in 2000 with the two B. bassiana products was lower, as follows: 38.0 and 58.7% without the barrier and 56.9 and 55.1% with the wood chip barrier. Posttreatment differences in nymphal numbers between treatments and control were significant (P = 0.005 and P = 0.039, 1999 and 2000, respectively). The bifenthrin provided 86 and 87% control each year, respectively. The application of Metarhizium anisopliae (Metschnikoff) Sorokin strain F52 to 9 residential sites in Westport and Weston, CT, in 2002 provided significant (P = 0.034; P = 0.039) reductions in nymphal tick abundance with 55.6 and 84.6% fewer ticks on lawn and woodland plots, respectively. These results suggest the application of entomopathogenic fungi could provide another approach for the control of I. scapularis nymphs in residential or similar landscapes.

Female Culex quinquefasciatus Say (Diptera: Culicidae) mosquitoes were exposed to sublethal amounts of prallethrin, sumithrin, and piperonyl butoxide applied as ultralow volume (ULV) droplets in a wind tunnel. Mosquitoes were video recorded before, during, and after treatment, and the number and size of droplets on their bodies were later determined using a compound microscope. A positive correlation was found between mosquitoes that spent more time flying during the time of spraying and number of droplets on their bodies. Excitation, in the form of increased speed and duration of flight, was immediate in mosquitoes exposed to prallethrin, whereas exposure to sumithrin did not increase their exposure to the ULV droplets. The location of droplets on mosquitoes, the effects of droplet volume, and subsequent mortality are discussed.

The fall armyworm (Spodoptera frugiperda) (J. E. Smith) (Lepidoptera: Noctuidae), a major agricultural pest in the Western Hemisphere, has recently become established in Africa and Asia. This highly polyphagous species has potential to economically harm multiple crops. Contributing to this host range are two fall armyworm populations historically called 'host strains' that differ in host specificity. Understanding behaviors of the two strains is crucial to effective management of this pest. A major difficulty in such studies is that strains have long been considered morphologically indistinguishable, with molecular markers the only reliable means of identification. However, studies of fall armyworm in Colombia reported strain differences in wing morphology sufficiently large to potentially provide a more economical alternative method to determine strain. This study tested whether a similar phenotypic difference was present in Florida populations using geometric morphometric analysis of 15 anatomical landmarks on forewings of 182 specimens from three habitats associated with different host plants. Principle component and linear discriminant analyses identified significant differences in wing size and shape in comparison of strains from different habitats, but not between strains within the same habitat. Data indicate that apparent strain distinctions in wing phenotype are most likely a secondary consequence of differences in developmental growth patterns on different host plants combined with strain-biased host choice. Furthermore, Florida specimens showed much larger phenotypic overlap than observed for strains from Colombia. Together these findings suggest that wing morphology is probably not a reliable indicator of strain identity in field populations where different host plants are available.

Asian citrus psyllid, Diaphorina citri Kuwayama, negatively impacts the citrus industry as it transmits Candidatus Liberibacter spp., the causal agent of citrus greening. Monitoring D. citri population levels is critical for management of vectors and citrus greening and is usually through use of yellow sticky traps. In our study, use of odors, odor blends, and visual targets were evaluated to determine whether attraction to yellow sticky traps could be improved. Methyl salicylate consistently increased D. citri attraction to decoy yellow but not to other yellow or yellow/green traps. Addition of a visual target did not enhance attraction to sticky traps. While several chemical blends were evaluated, they did not increase psyllid attraction to decoy yellow traps. The increased attraction to traps with methyl salicylate is promising and may contribute to trapping efficiency under field conditions.

Laboratory assays were conducted to evaluate responses of Diaphorina citri to various aspects of visual cues associated with traps in an effort to improve trap effectiveness. Addition of white or UV violet but not yellow light-emitting diodes (LEDs) increased attraction to standard yellow adhesive traps moderately (11-17%), with no difference in attraction between white or UV violet LEDs. Addition of a black border on yellow traps enhanced collections. However, there were no differences between attraction to black patterns on traps. Comparisons were made between different commercial paints, some with UV-reflecting properties or fluorescence. A yellow paint with UV reflectance, used for painting bird decoys (decoy yellow), was more attractive than the standard yellow Olson sticky trap. Addition of white or green pigment to increase intensity or enhance green reflectance, respectively, did not increase attraction. Alteration of reflectance of Olson traps with addition of UV-reflecting or fluorescent pigments did not enhance attraction of D. citri. In field comparisons, decoy yellow and fluorescent yellow sticky traps were more attractive to D. citri than Olson yellow.
