RESUMEN

Overcoming the limitations for efficient and selective drug delivery is one of the most challenging obstacles for newly designed anticancer agents. In this study, we present two types of platinum-based nanoparticles (NP), ultrasmall 2 nm PtNPs and core-shell 30 nm Au@Pt, which can be highly cytotoxic in an oxidative environment and remain biologically inactive in cells with lower oxidative status. Our research highlighted the differences in platinum nanoparticle-induced chemotoxicity and is the first study examining its mechanism as a substantial aspect of Au@Pt/PtNPs biological activity. Selectively induced oxidative stress was found to be a primary trigger of NPs' toxicity. Significant differences between Au@Pt and PtNPs were observed especially during 24 h treatment, due to successful intranuclear PtNPs location (~13% of internalized fraction). Reactive oxygen species (ROS)-level induced from both NPs types were similar, while reduction of reduced glutathione (GSH) intracellular content was stronger after treatment with PtNPs. Any biological activity was found in HER2+ breast cancer cells, which have only slightly increased oxidative status. Platinum-containing nanoparticles are an interesting tool for the improvement of selectivity in anticancer therapies against hepatocellular carcinoma (HCC). Due to intranuclear uptake, 2 nm PtNPs seems to be more promising for further research for HCC therapy.

Chronic heart failure (HF) is often accompanied by systemic iron deficiency (ID). However, effects of ID on cardiac iron status and progression of HF are unknown. To investigate these effects rats underwent LAD ligation to induce post-myocardial infarction HF or sham operation. After 3 weeks the animals from both groups were randomized into three subgroups: control, moderate ID and severe ID+anemia (IDA) by a combination of phlebotomy and low iron diet for 5 weeks. Serum and hepatic iron content were reduced by 55% and 70% (ID) and by 80% and 77% (IDA), respectively, while cardiac iron content was unchanged in HF rats. Changes in expression of all cardiomyocyte iron handling proteins indicating preserved cardiomyocytes iron status in HF and ID/IDA. Contractile function of LV cardiomyocytes, Ca2+ transient amplitude, sarcoplasmic reticulum Ca2+ release and SERCA2a function was augmented by ID and IDA and it was accompanied by an increase in serum catecholamines. Neither ID nor IDA affected left ventricular (LV) systolic or diastolic function or dimensions. To sum up, systemic ID does not result in cardiac ID and does not affect progression of HF and even improves contractile function and Ca2+ handling of isolated LV cardiomyocytes, however, at the cost of increased catecholamine level. This suggests that intravenous iron therapy should be considered as an additional therapeutic option in HF, preventing the increase of catecholaminergic drive with its well-known long-term adverse effects.

193mPt and 195mPt radionuclides are therapeutically attractive Auger electron emitters with notably high Auger electron yield per decay. The present paper summarizes the first step of research on the applications of core-shell (Au@Pt) nanoparticles for electron Auger therapy of HER2+ (human epidermal growth factor receptor 2) breast cancer and hepatocellular carcinoma. Gold nanoparticles (30 nm) were synthesized covered with a platinum shell at high efficiency (>80%) and were further evaluated for in vitro studies such as binding affinity, internalization and cytotoxicity. To find the mechanism(s) responsible for platinum cytotoxicity in HepG2 cells, the platinum concentration in isolated cell nuclei and cytoplasm was determined using ICP-MS (inductively coupled plasma mass spectrometry). Lack of platinum in cell nuclei suggests that the cytotoxic effect is associated with the generation of reactive oxygen species (ROS) and reactive nitrogen species (RNS). Studies carried out on the SKOV-3 cell line with the use of a synthesized targeting bioconjugate (Au@Pt-PEG-trastuzumab) revealed a high affinity of this preparation to HER2+ cells, its internalization, its placement in the perinuclear area and partial intranuclear location. The specific binding for HER2 negative cells, MDA-MB-231, was negligible and Au@Pt-PEG-trastuzumab did not enter these cells. The results obtained are promising and warrant future investigation of Auger electron therapy using 193mPt and 195mPt based radiopharmaceuticals.

Literature on the use of four chelating agents namely: ethylenediaminetetraacetic acid, nitrilotriacetic acid, diglycolic acid and iminodiacetic acid for the chromatographic separation of micro and macro amounts of rare earth elements was critically reviewed and supplemented with some new unpublished data from our Laboratory. Advantages and disadvantages of ion exchange chromatography both in cation and anion mode as well as ion interaction chromatography techniques, which were used for rare earth elements separation, are discussed. The usefulness of some of the chromatographic systems for micro-macro separations was discussed and demonstrated. The importance of resilience of the separation method to column overloading in some analytical and larger scale separations was emphasized. The methods described in this article might suit well for recovering of individual lanthanides and yttrium from e-waste and other industrial wastes which were fast accumulating in recent years.

The preparation, certification, and characterization of two new biological certified reference materials for inorganic trace analysis have been presented. They are based on two different varieties of tobacco leaves, namely, Oriental Basma Tobacco Leaves (INCT-OBTL-5), grown in Greece, and Polish Virginia Tobacco Leaves (INCT-PVTL-6), grown in Poland. Certification of the materials was based on the statistical evaluation of results obtained in a worldwide interlaboratory comparison, in which 87 laboratories from 18 countries participated, providing 2568 laboratory averages on nearly 80 elements. It was possible to establish the certified values of concentration for many elements in the new materials, that is, 37 in INCT-OBTL-5 and 36 in INCT-PVTL-6, including several toxic ones like As, Cd, Hg, Pb, and so forth. The share and the role of instrumental analytical techniques used in the process of certification of the new CRMs are discussed.

Arsenic and its inorganic species: As (III), As (V), dimethylarsenic acid (DMA) and monomethylarsenic acid (MMA) were determined in hypoallergenic formulas and grain porridges commercially available on Polish market, dedicated for infant 0-8 months. After quantitative extraction with 0.5% HNO3, separation of individual species was performed by high performance liquid chromatography (HPLC), and their determination by neutron activation analysis (NAA) and inductively coupled plasma mass spectrometry (ICP-MS). Due to relatively low content of As in the analysed samples, it was only possible to determine DMA using the HPLC-ICP-MS mode. HPLC separation coupled with off-line determination by NAA enabled the determination of more extracted As species (especially inorganic) with good accuracy. Certified reference material (CRM) Rice Flour SRM 1568b (NIST) was used for the validation of both procedures.

The aim of this study was to assess the levels of 13 essential and toxic elements (As, Cd, Co, Cr, Cu, Fe, Mn, Mo, Ni, Pb, Se, V, Zn) in the infant formulas, available on Polish market. Selected food samples were of animal (cow- and goat-based milks) and plant (soy-based milk, hypoallergic products, grain porridges) origin. Two analytical techniques, inductively coupled plasma mass spectrometry (ICP-MS) and neutron activation analysis (NAA), have been complementarily applied to analyze elemental content of 16 formulas dedicated for infants between 0 and 8 months. For arsenic determinations, the radiochemical mode of NAA was also used. The daily intake of some micronutrients in the age 0-8 months for non-breastfed infants was estimated and compared with present safety limits. Certified reference materials (CRMs) have been used for the validation of the methods: Non-fat Milk Powder 1549 (National Bureau of Standards-NBS), Soya Bean Flour INCT-SBF-4 (Institute of Nuclear Chemistry and Technology-INCT), Rice Flour SRM 1568b (National Institute of Standards and Technology-NIST).

Nanotechnology is a dynamically developing field of scientific and industrial interest across the entire world, and the commercialization of nanoparticles (NPs) is rapidly expanding. Incorporation of nanotechnologies into a range of manufactured goods results in increasing concern regarding the subsequent release of engineered NPs into the environment. One of the biggest threats of using NPs is the transfer and magnification of these particles in the trophic chain. The aim of the studies was the evaluation of the distribution of TiO2 NP contamination in the aquatic ecosystem under laboratory conditions. Bioaccumulation of TiO2 NPs by plants (Elodea canadensis) and fish (Danio rerio) in the source of contamination was investigated. The studies were focused on the consequences of short-term water contamination with TiO2 NPs and the secondary contamination of the components of the investigated model ecosystem (plants, sediments). It was found that in the fish and the plants exposed to NP contamination, the amount of Ti was higher than in the control, indicating an effective bioaccumulation of NPs or ions originating from NPs. It was clearly shown that the NPs present in the sediments are available to plants and fish. Additionally, the aquatic plants, an important trophic level in the food chain, can accumulate NPs and be a source of NPs for higher organisms. It was concluded that even an incidental contamination of water by NPs may result in long-term consequences induced by the release of NPs.

Iron deficiency (ID) commonly occurs in chronic heart failure (HF) and is associated with poor prognosis. Neither its causes nor pathophysiological significance are clearly understood. We aimed to assess iron status and the effect of iron supplementation in the rat model of post-myocardial infarction (MI) HF. Four weeks after induction of MI to induce HF or sham surgery, rats received intravenous iron (ferric carboxymaltose) or saline, 4 doses in 1-week intervals. HF alone did not cause anemia, systemic or myocardial ID, but reduced myocardial ferritin, suggesting depleted cardiomyocyte iron stores. Iron therapy increased serum Fe, ferritin and transferrin saturation as well as cardiac and hepatic iron content in HF rats, but did not increase myocardial ferritin. This was accompanied by: (1) better preservation of left ventricular (LV) ejection fraction and smaller LV dilation, (2) preservation of function of Ca2+ handling proteins in LV cardiomyocytes and (3) reduced level of inflammatory marker, CRP. Furthermore, iron supplementation did not potentiate oxidative stress or have toxic effects on cardiomyocyte function, but increased activity of antioxidant defenses (cardiac superoxide dismutase). Despite lack of systemic or myocardial ID we found evidence of depleted cardiomyocyte iron stores in the rat model of HF. Furthermore we observed positive effect of iron supplementation and confirmed safety of iron supplementation in this setting.

In this work a determination of selected elements in the infant formulas commercially available on the Polish market was done. 14 different materials (milk-based formulas and grain porridges) were analyzed. Both, inductively coupled plasma mass spectrometry (ICP-MS) and instrumental neutron activation analysis (INAA) were applied for the determination of As, Cr, Fe and Se, which are recognized as the problematic elements for ICP-MS. For As and Se, the radiochemical NAA was also used. The daily intake of Se and Fe in the age 0-6 months for non-breast fed infants was estimated and compared with present safety limits.

90Sr is a widely determined radionuclide for environmental purposes, nuclear waste control, and can be also monitored in coolants in nuclear reactor plants. In the developed method, the ICP-MS detection was employed together with sample processing in sequential injection analysis (SIA) setup, equipped with a lab-on-valve with mechanized renewal of sorbent bed for solid-phase extraction. The optimized conditions of determination included preconcentration of 90Sr on cation-exchange column and removal of different type of interferences using extraction Sr-resin. The limit of detection of the developed procedure depends essentially on the configuration of the employed ICP-MS spectrometer and on the available volume of the sample to be analyzed. For 1L initial sample volume, the method detection limit (MDL) value was evaluated as 2.9ppq (14.5BqL-1). The developed method was applied to analyze spiked river water samples, water reference materials, and also simulated and real samples of the nuclear reactor coolant.

The recovery of uranium and other valuable metals from Polish Peribaltic sandstones were examined. The solid-liquid extraction is the first stage of the technology of uranium production and it is crucial for the next stages of processing. In the laboratory experiments uranium was leached with efficiencies 71-100 % by acidic lixiviants. Satisfactory results were obtained for the alkaline leaching process. Almost 100 % of uranium was leached with alkaline carbonate solution. In post leaching solutions only uranium and small amounts of vanadium were present.

Raising role of the nuclear power industry, including governmental plans for the construction of first nuclear power plant in Poland, creates increasing demand for the uranium-based nuclear fuels. The project implemented by Institute of Nuclear Chemistry and Technology concerns the development of effective methods for uranium extraction from low-grade ores and phosphorites for production of yellow cake-U3O8. The Liqui-Cel® Extra-Flow 2.5 × 8 Membrane Contactor produced by CELGARD LLC (Charlotte, NC) company is the main component of the installation for liquid-liquid extraction applied for processing of post leaching liquors. In the process of membrane extraction the uranyl ions from aqueous phase are transported through the membrane into organic phase. The flow of two phases in the system was arranged in co-current mode. The very important element of the work was a selection of extracting agents appropriate for the membrane process. After preliminary experiments comprising tests of membrane resistivity and determination of extraction efficiency, di(2-ethylhexyl)phosphoric acid was found to be most favourable. An important aspect of the work was the adjustment of hydrodynamic conditions in the capillary module. To avoid the membrane wettability by organic solvent and mixing two phases equal pressure drops along the membrane module to minimize the transmembrane pressure, were assumed. Determination of pressure drop along the module was conducted using Bernoulli equation. The integrated process of extraction/re-extraction conducted in continuous mode with application of two contactors was designed.

The paper deals with experimental design and optimization of leaching process of uranium and associated metals from low-grade, Polish ores. The chemical elements of interest for extraction from the ore were U, La, V, Mo, Yb and Th. Sulphuric acid has been used as leaching reagent. Based on the design of experiments the second-order regression models have been constructed to approximate the leaching efficiency of elements. The graphical illustrations using 3-D surface plots have been employed in order to identify the main, quadratic and interaction effects of the factors. The multi-objective optimization method based on desirability approach has been applied in this study. The optimum condition have been determined as P=5 bar, T=120 °C and t=90 min. Under these optimal conditions, the overall extraction performance is 81.43% (for U), 64.24% (for La), 98.38% (for V), 43.69% (for Yb) and 76.89% (for Mo) and 97.00% (for Th).

A liquid chromatography-tandem mass spectrometry (LC-MS/MS) method was developed for selenium speciation in dietary supplements. Chromatographic separation was performed on a TSK-Gel ODS-100V column using a mixture of 5mM ammonium acetate water solution and methanol as a mobile phase. Conditions chosen for this process allowed to separate all investigated chemical compounds of selenium: seleno-l-methionine, methyl-seleno-l-cysteine, l-selenocystine, methaneseleninic acid, selenite and selenate. A tandem mass spectrometer with an ion trap operating in negative or positive ion mode according to the selenium form being determined was used as a detector. Three extraction procedures: water extraction, enzymatic hydrolysis and sequential extraction were used for preparation of samples for the determination of the actual forms of selenium in diet supplements. The developed method was used for analysis of six dietary supplements containing selenium bought in a pharmacy and supermarket. Apart from speciation analysis of selenium content in supplements total selenium content was determined using instrumental neutron activation analysis (INAA). All expected forms of selenium except for selenite were determined using LC-MS/MS technique. It should be stressed that amounts of selenate were smaller than expected.

A new radiochemical separation scheme based on extraction chromatography has been proposed for isolation of arsenic from selenium. The distribution coefficients of As and Se on prepared sorbents: (selected aromatic o-diamines supported on polystyrene adsorbents) were determined in order to find the best condition for separation of both the elements. Batch experiments were verified by column studies. This work, together with earlier results from this Laboratory, enabled to elaborate a new separation scheme for selective and quantitative separation of arsenic from selenium. Proposed approach insures high selectivity and radionuclide purity of separated arsenic fraction; it is also characterized by high elution efficiency (>95%) using small volume (2 mL) of 0.9% NaCl with very low breakthrough (<0.01%) of selenium.
