RESUMEN

Transforming growth factor-ß2 (TGF-ß2) induced fibrogenic changes in human trabecular meshwork (HTM) cells have been implicated in trabecular meshwork (TM) damage and intraocular pressure (IOP) elevation in primary open-angle glaucoma (POAG) patients. Silibinin (SIL) exhibited anti-fibrotic properties in various organs and tissues. This study aimed to assess the effects of SIL on the TGF-ß2-treated HTM cells and to elucidate the underlying mechanisms. Our study found that SIL effectively inhibited HTM cell proliferation, attenuated TGF-ß2-induced cell migration, and mitigated TGF-ß2-induced reorganization of both actin and vimentin filaments. Moreover, SIL suppressed the expressions of fibronectin (FN), collagen type I alpha 1 chain (COL1A1), and alpha-smooth muscle actin (α-SMA) in the TGF-ß2-treated HTM cells. RNA sequencing indicated that SIL interfered with the phosphoinositide 3-kinase (PI3K)/protein kinase B (PKB, also known as AKT) signaling pathway, extracellular matrix (ECM)-receptor interaction, and focal adhesion in the TGF-ß2-treated HTM cells. Western blotting demonstrated SIL inhibited the activation of Janus kinase 2 (JAK2)/signal transducers and activators of transcription 3 (STAT3) and the downstream PI3K/AKT signaling pathways induced by TGF-ß2, potentially contributing to its inhibitory effects on ECM protein production in the TGF-ß2-treated HTM cells. Our study demonstrated the ability of SIL to inhibit TGF-ß2-induced fibrogenic changes in HTM cells. SIL could be a potential IOP-lowering agent by reducing the fibrotic changes in the TM tissue of POAG patients, which warrants further investigation through additional animal and clinical studies.

RESUMEN

Long non-coding RNAs (lncRNAs) mediate diverse biological events mainly through the modulation of transcriptional hierarchy. The phytohormone gibberellin (GA) is essential for various aspects of plant growth and development. However, the roles of lncRNAs in the regulation of the GA response remain largely unknown. Through sequencing multiple strand-specific and ribosomal-depleted RNA libraries, we delineated the landscape of lncRNAs in maize (Zea mays). Out of identified lncRNAs, 445 GIBBERELLIN-RESPONSIVE lncRNAs (GARRs) were differentially expressed upon GA application. By the intersection of GARRs from normal-height and dwarf plants from an advanced backcross population, four shared GARRs (GARR1 to GARR4) were identified. Out of these four shared GARRs, GARR2 was derived from a Gypsy LTR retrotransposon. GA-responsive element P-boxes were identified upstream of GARR2. GARR2-edited lines exhibited a GA-induced phenotype. Editing of GARR2 resulted in changes in the transcriptional abundance of GA pathway components and endogenous GA contents. Besides GA, GARR2 affected the primary auxin response. An RNA pull-down assay revealed the HECT ubiquitin-protein ligase family member ZmUPL1 as a potential interaction target of GARR2. GARR2 influenced the abundance of ZmUPL1 in the GA response. Our study uncovers lncRNA players involved in the modulation of the GA response and guides the development of plant height ideotype driven by knowledge of the phytohormone GA.

RESUMEN

Sugar efflux transporter SWEET family is involved in multiple biological processes, from nectar secretion, pollen fertility to seed filling. Although roles of SWEETs in abiotic stress adaption have been revealed mainly in reference organism Arabidopsis, cereal crops SWEETs responses to abiotic stimulation remain largely elusive. Here, we report the characterization of maize SWEET family member ZmSWEET1b, with emphasis on its response to salinity stress. ZmSWEET1b is a canonical sugar transporter, characteristic of seven transmembrane helices and plasma membrane localization. ZmSWEET1b and its rice ortholog OsSWEET1b in phylogenetic clade I underwent convergent selection during evolution. Two independent knockout lines were created by the CRISPR/Cas9 method to functionally characterized ZmSWEET1b. Sucrose and fructose contents are significantly decreased in ZmSWEET1b knockout lines. Mature leaves of ZmSWEET1b-edited lines exhibit chlorosis, reminiscent of senescence-like phenotype. Ears and seeds of ZmSWEET1b knockout lines are small. Upon salinity treatment, ZmSWEET1b-edited lines become more wilted. Transcriptional abundance of genes for Na+ efflux from roots to the rhizosphere, including ZmSOS1, ZmH+-ATPASE 2, and ZmH+-ATPASE 8, is decreased in salt-treated ZmSWEET1b knockout lines. These findings indicate that convergently selected sugar transporter ZmSWEET1b is important for maize plant development and responses to salt stress. The manipulation of ZmSWEET1b may represent a feasible way forward in the breeding of salinity tolerant ideotypes through the optimization of assimilate allocation.

RESUMEN

OBJECTIVE: This study evaluates the correlation between the sonographic features and the nodal fine-needle aspiration thyroglobulin (FNA-Tg) in papillary thyroid carcinoma (PTC) patients with the recurrent/persistent lymph node for the purpose of the reasonable selection of lymph nodes. METHODS: This study prospectively contained PTC patients with the suspicious cervical lymph nodes in one medical center from April 2018 to January 2019. Each suspected lymph node was aspirated with a 22-gauge needle and the value of FNA-Tg was measured as well. RESULTS: There were 136 lymph nodes related to the disease involved. The FNA-Tg levels of 89 (65.44%) metastatic lymph nodes were significantly higher than those of the benign. The median value of the former was 631.550 ng/mL while the latter was 0.056 ng/mL (p = 0.000). The cut-off value of the metastatic lymph nodes diagnosed by FNA-Tg was 2.71 ng/mL, and 6.5 by FNA-Tg/sTg. The suspicious ultrasonographic features including cystic, hyperechoic content, and lack of hilum were closely related to the high level of FNA-Tg value (p < 0.05). However, the round shape (Solbiati index <2) and calcification were not significantly correlated with the positive FNA-Tg (p > 0.05). CONCLUSION: FNA-Tg is an effective supplement to fine-needle aspiration (FNA) cytology in the nodal metastasis diagnosis. The FNA-Tg level was much higher in the metastatic lymph nodes. The reliable sonographic features of lymph nodes suggested the positive FNA-Tg were cystic content, hyperechoic content and lack hilum. Solbiati index <2 and calcification did not show an exact correlation with the result of FNA-Tg.

RESUMEN

Some microbial volatile organic compounds (mVOCs) can act as antagonistic weapons against plant pathogens, but little information is available on the contribution of individual mVOC to biocontrol and how they interact with plant pathogens. In this study, the Bacillus subtilis strain N-18 isolated from the rhizosphere of healthy plants grown in areas where Fusarium crown and root rot (FCRR) of tomato occurs could reduce the 30% of the incidence of FCRR. Moreover, the volatile organic compounds (VOCs) produced by N-18 had inhibitory effects on Fusarium oxysporum f. sp. radicis-lycopersici (FORL). The identification of VOCs of N-18 was analyzed by the solid-phase microextraction coupled to gas chromatography-mass spectrometry. Meanwhile, we conducted sensitivity tests with these potential active ingredients and found that the volatile substances acetoin and 2-heptanol can reduce the 41.33% and 35% of the incidence of FCRR in tomato plants. In addition, the potential target protein of acetoin, found in the cheminformatics and bioinformatics database, was F. oxysporum of hypothetical protein AU210_012600 (FUSOX). Molecular docking results further predicted that acetoin interacts with FUSOX protein. These results reveal the VOCs of N-18 and their active ingredients in response to FORL and provide a basis for further research on regulating and controlling FCRR.

RESUMEN

The high power and variable repetition-rate of Yb femtosecond lasers makes them very attractive for ultrafast science. However, for capturing sub-200 fs dynamics, efficient, high-fidelity and high-stability pulse compression techniques are essential. Spectral broadening using an all-solid-state free-space geometry is particularly attractive, as it is simple, robust and low-cost. However, spatial and temporal losses caused by spatio-spectral inhomogeneities have been a major challenge to date, due to coupled space-time dynamics associated with unguided nonlinear propagation. In this work, we use all-solid-state free-space compressors to demonstrate compression of 170 fs pulses at a wavelength of 1030nm from a Yb:KGW laser to ∼9.2 fs, with a highly spatially homogeneous mode. This is achieved by ensuring that the nonlinear beam propagation in periodic layered Kerr media occurs in spatial soliton modes, and by confining the nonlinear phase through each material layer to less than 1.0 rad. A remarkable spatio-spectral homogeneity of ∼0.87 can be realized, which yields a high efficiency of >50% for few-cycle compression. The universality of the method is demonstrated by implementing high-quality pulse compression under a wide range of laser conditions. The high spatiotemporal quality and the exceptional stability of the compressed pulses are further verified by high-harmonic generation. Our predictive method offers a compact and cost-effective solution for high-quality few-cycle-pulse generation from Yb femtosecond lasers, and will enable broad applications in ultrafast science and extreme nonlinear optics.

RESUMEN

KEY MESSAGE: G protein couples MAPK cascade through maize heterotrimeric Gß subunit MGB1. Heterotrimeric G protein Gß interacts with Gγ subunit to generate Gßγ dimer in modulation of various biological processes. The modulatory events at transcriptome scale of plant Gß subunit remain largely unknown. To reveal the regulatory basis of Gß subunit at transcriptome level, we first identified a canonical maize Gß subunit MGB1 that physically interacted with Type C Gγ protein MGG4. For transcriptome analysis, two independent CRISPR/Cas9-edited MGB1 lines were generated, which all exhibited growth arrest, suggestive of MGB1 essential for maize seedling establishment. Transcriptomic outcomes showed that MGB1 knockout resulted in elevated transcriptional abundance of plant immune response marker PR and immune receptor RPM1. Integrated GO, KEGG, and GSEA analyses pinpointed the enrichment of differentially expressed genes in defense response pathway. Functional association network construction revealed MAPK cascade components and PR protein as hub regulators of MGB1-mediated immune signaling. MGB1 and scaffold protein ZmRACK1 together with MAPK cascade components coordinately modulated maize immune responses. We built a modulatory hierarchy of Gß subunit at transcriptome and interacting scales, which is informative for our understanding of the regulatory basis of G protein signaling.

RESUMEN

BACKGROUND: China faces a serious shortage of competent nurses who can address the healthcare needs of older people in an ageing society. Chinese higher education institutes face serious challenges when it comes to developing new curricula that are capable of educating sufficient numbers of competent gerontological nurses. Therefore, the aim of this research study was to identify and verify competencies for gerontological nurses in China that are needed to provide nursing care for the growing number of older people in all care settings. This study takes into account the possible opportunities that trends and developments may offer in the near future. METHODS: In this study, a two-phase research design was used. The first phase concerned needs analysis, including a situational analysis, a trend analysis and a competence analysis. This process resulted in a draft competence framework. The second phase addressed the verification of the competence framework through a two-round Delphi study with a panel of Chinese and European experts. This process led to the final competence framework. RESULTS: The final competence framework for gerontological nursing in China included six competencies divided into 13 essential and five relevant learning outcomes. The competencies are: 'providing gerontological care', 'communication and collaboration', 'organization of gerontological nursing care', 'health promotion', 'evidence-based nursing and lifelong learning' and 'professional behaviour'. CONCLUSION: The framework comprehensively covers the six core competencies that nurses who care for older people should possess. These competencies are well-embedded in a Chinese context. The framework therefore offers concrete, practical suggestions for the competencies and skills that nursing graduates will need to work in current and future professions related to gerontological nursing education and practice.

RESUMEN

OBJECTIVE: To study the mechanical properties of sciatic nerve in rats with chronic alcoholism (CA) and intervened with bone marrow mesenchymal stem cells (BMSCs) and to provide biomechanical basis for clinical practice. METHODS: the serum of the BMSCs-intervened CA rats was sampled and determined the contents of malondialdehyde (MDA), metallothionein (CAS, MT), and Glutathione/r -glutamyl cysteinyl/glycine (GSH); meanwhile, the rats' sciatic nerve was tested the tensile and observed the histomorphological changes. RESULTS: The mechanical properties of sciatic nerve in BMSCs-intervened CA rats, as well as the serum levels of MT and GSH, were significantly different from those in the basic fibroblast growth factor (bFGF)-intervened CA rats (p < 0.05). CONCLUSIONS: BMSCs intervention can restore the levels of MT, GSH, MDA, histomorphology, and tensile mechanical properties in CA animal model, and its effects on repairing sciatic nerve are obvious.

RESUMEN

BACKGROUND AND OBJECTIVES: The objective of this study is to compare the efficacy and the safety of ultrasound-guided microwave ablation (MWA) and laser ablation (LA) for the treatment of papillary thyroid microcarcinoma (PTMC). STUDY DESIGN/MATERIALS AND METHODS: A total of 67 patients with unifocal PTMC were studied retrospectively, including 33 cases who underwent MWA (MWA group) and 34 cases who received LA (LA group). The follow-up consisted of thyroid function tests, ultrasonography, contrast-enhanced ultrasonography (CEUS), and chest X-ray or computed tomography scan. The treatment response and complications were compared between the two groups. RESULTS: The follow-up time for the MWA and LA group was 23.3 ± 4.4 and 22.8 ± 4.1 months, respectively. All the ablations were successfully performed as planned without complementary ablations, and it was confirmed by CEUS after treatment in both groups. It was observed that, at the last follow-up, the mean largest diameter decreased from 5.0 ± 1.4 mm to 0.1 ± 0.4 mm (MWA group) and from 4.5 ± 1.6 mm to 0.6 ± 1.2 mm(LA group) (P < 0.05 for both). The average volume reduced from 51.9 ± 40.8 to 0.2 ± 1.0 mm3 (MWA group) and from 38.5 ± 43.0 to 1.3 ± 3.8 mm3 (LA group) (P < 0.05 for both). The complication rates did not differ between the MWA group (9.1%) and the LA group (2.9%) (P > 0.05). No local recurrence or distant metastasis occurred in either group. CONCLUSIONS: During the short-term follow-up period, ultrasound-guided MWA and LA were both safe and effective methods in treating patients with unifocal PTMC. Lasers Surg. RESUMEN

OBJECTIVES: Heart failure (HF) progression can be prevented by an inhibitor of the mammalian target of rapamycin and an autophagy enhancer rapamycin. This current study aimed to investigate the effect of rapamycin on HF progression and myocardial cells apoptosis. METHODS: HF rats were injected with low-, middle-, and high-dose rapamycin. Echocardiography, hematoxylin-eosin staining, plasma brain natriuretic peptide, myocardial cells apoptosis, and Akt activation in rapamycin-treated rats were detected. RESULTS: HF rats showed reduced cardiac functions, destructive pathological changes in the myocardium, enhanced Akt activation and myocardial cells apoptosis. However, rapamycin reversed all the changes in a dose-dependent manner. Cardiac functions were enhanced by rapamycin. Myocardial cells apoptotic percentage, Akt expression, and pathological changes of the myocardium in HF rats were inhibited by rapamycin administration. CONCLUSIONS: Rapamycin protected against myocardial hypertrophy and myocardial cells apoptosis in HF rats in a dose-dependent manner.

RESUMEN

Objectives: To compare the clinical outcomes of ultrasound-guided laser ablation (LA) and surgery for treatment of solitary papillary thyroid microcarcinoma (PTMC). Methods: A total of 81 consecutive patients with solitary PTMC were included in this retrospective study. Among them, 36 received LA and 45 underwent surgery. Surgery was performed by hemithyroidectomy with unilateral central neck dissection. The follow-up consisted of a physical examination, neck ultrasonography, chest X-ray or CT scan and thyroid function tests. The procedure time, hospital stay, complication and recurrence rates were compared between the two groups after treatment. Results: The follow-up period for the LA and surgical group were 49.2 ± 4.5 months (range, 30-54 months) and 48.5 ± 6.2 months (range, 24-54 months), respectively. The mean hospital stay and procedure time in the LA group were shorter than those in the surgical group. After LA, the largest diameter and average volume decreased from 4.7 ± 1.4 mm to 0.2 ± 0.8 mm, and from 43.2 ± 38.8 mm3 to 0.7 ± 4.1 mm3 (p < .05 for both), respectively. The complication rates and recurrence rates did not differ between the LA group (2.8% [1 of 36] and 5.6% [2 of 36]) and the surgical group (6.7% [3 of 45] and 6.7% [3 of 45]) (p > .05 for both). No distant metastasis occurred in the either group during the follow-up period. Conclusions: Compared with hemithyroidectomy with unilateral central neck dissection, ultrasound-guided LA was also a safe and effective therapy for treating solitary PTMC, and it may be considered as a treatment alternative for patients who are ineligible or refusal to undergo surgery.

RESUMEN

BACKGROUND: Our purpose is to assess the effectiveness and safety of ultrasound-guided percutaneous microwave ablation (MWA) for lymph node metastases (LNMs) from papillary thyroid carcinomas (PTC). METHODS: In total, 14 patients with recurrent PTC were enrolled in this retrospective study. The vascularity within the ablation zone was evaluated by contrast-enhanced ultrasonography (CEUS) after MWA. Patients were followed up with measurement of the size and volume of tumor, serum thyroglobulin, and clinical evaluation at 7 days, 1, 3, 6 months, and every 6 months thereafter. RESULTS: Twenty-one LNMs were confirmed by biopsy and successfully treated by MWA in a single session. No incomplete ablation was detected by CEUS after treatment. The average largest diameter and volume of the tumors were reduced from 10.1 ± 4.7 mm (range, 3.1-20.0 mm) and 291.9 ± 255.6 mm3 (range, 11.6-766.6 mm3) to 0.9 ± 1.6 mm (range, 0-4.1 mm; p < 0.05) and 4.0 ± 9.0 mm3 (range, 0-31.6 mm3; p < 0.05) at the final follow-up. Neither progression of treated tumors nor newly suspicious LNMs could be detected after treatment. The overall complication rate was 7.1% (1/14). CONCLUSIONS: Ultrasound-guided MWA can effectively control LNMs from PTC, but it is less safe for tumors in the central compartment. MWA may become an alternative therapy in selected PTC patients, who were ineligible or refused to undergo repeated neck explorations.

RESUMEN

We investigate magnetoplasmon resonances and their coupling effects in gyroelectric cylinders. In individual cylinders, the dipole plasmon can be excited by plane wave illumination, and the dipole plasmon splits into lower energy and higher energy rotational magnetoplasmons in the presence of an external magnetic field. With respect to the external magnetic field, the two magnetoplasmons carry either right-handed chirality or left-handed chirality. In addition, originally dark plasmons can also be excited as the magnetic field increases. They are lower-order bulk plasmons (such as the radial breathing mode). In cylindrical dimers, the optically bright modes are combinations of magnetoplasmons with the same chirality. If the magnetic fields are antiparallel, the absorption spectra will be different for light incident from two opposite directions. This asymmetry can be well understood by carrying out eigenstate analysis, where the eigenstate does not possess mirror symmetry respecting the dimer axis. The dark modes engineering and asymmetrical optical behavior could have potential for terahertz device applications.

RESUMEN

Mercury (Hg) contamination in soil-rice systems from industry, mining and agriculture has received increasing attention recently in China. Pot experiments were conducted to research the Hg accumulation capacity of rice under exogenous Hg in the soil and study the major soil factors affecting translocation of Hg from soil to plant. Soil treated with 2 mg kg-1 Hg decreased rice grain yield and inhibited the growth of rice plants. With increased Hg contamination of the rice, the enrichment rate of Hg was significantly higher in the rice grain than that in the stalk and leaf. Soil pH and cation exchange capacity are the key factors controlling Hg bioavailability in soils.

RESUMEN

Transcranial magneto-acoustical stimulation (TMAS), utilizing focused ultrasound and a magnetostatic field to generate an electric current in tissue fluid to regulate the activities of neurons, has high spatial resolution and penetration depth. The neuronal spike-frequency adaptation plays an important role in the treatment of neural information. In this paper, we study the effects of ultrasonic intensity, magnetostatic field intensity and ultrasonic frequency on the neuronal spike-frequency adaptation based on the Ermentrout neuron model. The simulation results show that, the peak time interval becomes smaller, the interspike interval becomes shorter and the time of the firing of the neuron is shortened with the increasing of the magnetostatic field intensity. With the increase of the adaptive variables, the initial spike-frequency is shifted to the right with the magnetostatic field intensity, and the spike-frequency is linearly related to the increase of the magnetostatic field intensity in steady state. The simulation effect with change of the ultrasonic intensity is consistent with the change of magnetostatic field intensity. The change of the ultrasonic frequency has no effect on the neuronal spike-frequency adaptation. Under the different adaptive variables, with the increase of the adaptive variables, the initial spike-frequency amplitude decreased with the increasing of the ultrasonic frequency, and the spike-frequency is linearly related to the increase of the ultrasonic frequency in steady state. These results of the study can help us to reveal the mechanism of transcranial magneto-acoustical stimulation on the neuronal spike-frequency adaptation, and provide a theoretical basis for its application in the treatment of neurological disorders.

RESUMEN

Thanks to the efficient retrieval speed and low storage consumption, learning to hash has been widely used in visual retrieval tasks. However, the known hashing methods assume that the query and retrieval samples lie in homogeneous feature space within the same domain. As a result, they cannot be directly applied to heterogeneous cross-domain retrieval. In this article, we propose a generalized image transfer retrieval (GITR) problem, which encounters two crucial bottlenecks: 1) the query and retrieval samples may come from different domains, leading to an inevitable domain distribution gap and 2) the features of the two domains may be heterogeneous or misaligned, bringing up an additional feature gap. To address the GITR problem, we propose an asymmetric transfer hashing (ATH) framework with its unsupervised/semisupervised/supervised realizations. Specifically, ATH characterizes the domain distribution gap by the discrepancy between two asymmetric hash functions, and minimizes the feature gap with the help of a novel adaptive bipartite graph constructed on cross-domain data. By jointly optimizing asymmetric hash functions and the bipartite graph, not only can knowledge transfer be achieved but information loss caused by feature alignment can also be avoided. Meanwhile, to alleviate negative transfer, the intrinsic geometrical structure of single-domain data is preserved by involving a domain affinity graph. Extensive experiments on both single-domain and cross-domain benchmarks under different GITR subtasks indicate the superiority of our ATH method in comparison with the state-of-the-art hashing methods.

RESUMEN

The rapid development of wearable and portable electronic devices prompts the ever-growing demand for wearable, flexible, and light-weight power sources. In this work, a MXene/GNS/PPy@PEDOT/Cotton nanocomposite electrode with excellent electrochemical performances was fabricated using cotton fabric as a substrate. Poly (3,4-ethylenedioxythiophene): poly (styrenesulfonate) (PEDOT:PSS) was coated on the cotton fabric to obtain a conductive substrate through a controllable dip-drying coating process, while a nanocomposite consisting of MXene, Graphene nanoscroll (GNS), and polypyrrole (PPy) was directly synthesized and deposited on the PEDOT:PSS-modified cotton fabric via a one-step in situ polymerization method. The resultant MXene/GNS/PPy@PEDOT/Cotton electrode delivers excellent electrochemical performances including an ultra-high areal capacitance of 4877.2 mF·cm-2 and stable cycling stability with 90 % capacitance retention after 3000 cycles. Moreover, the flexible symmetrical supercapacitor (FSC) assembled with the MXene/GNS/PPy@PEDOT/Cotton electrodes demonstrates a prominent areal capacitance (2685.28 mF·cm-2 at a current density of 1 mA·cm-2) and a high energy density (322.15 µWh·cm-2 at a power density of 0.46 mW·cm-2). In addition, the application of the FSC for wearable electronic devices was demonstrated.
