RESUMEN

A comprehensive and sensitive method combining ultra-performance liquid chromatography with tandem mass spectrometry was developed for the quantification of characteristic triterpenoids in Ganoderma mycelia. Eight ganoderic acids previously isolated from the mycelia of Ganoderma lingzhi were separated with a binary mobile phase on a reversed-phase C18 column. A triple quadrupole mass spectrometer equipped with an electrospray ionization source was used as the detector in the negative ion mode. Identification and quantitation of target ganoderic acids were accomplished using the dynamic multiple reaction monitoring mode. The developed method was validated in terms of linearity, precision, accuracy, stability, and recovery. The method was first applied to quantify the contents of eight ganoderic acids in the mycelia of G. lingzhi at different times to determine the optimum fermentation conditions. Subsequently, the distribution of triterpenoids and the contents of eight ganoderic acids in sixteen different Ganoderma species were investigated. The results indicated that UV chromatography combined with dynamic multiple reaction monitoring quantification was an effective chemotaxonomy method for Ganoderma species identification. This study also provided a helpful analytical methodology for both scientific and industrial applications in the quality control of Ganoderma triterpenoids.

Ectomycorrhizal fungi play a key role in forests by establishing mutualistic symbioses with woody plants. Genome analyses have identified conserved symbiosis-related traits among ectomycorrhizal fungal species, but the molecular mechanisms underlying host specificity remain poorly known. We sequenced and compared the genomes of seven species of milk-cap fungi (Lactarius, Russulales) with contrasting host specificity. We also compared these genomes with those of symbiotic and saprotrophic Russulales species, aiming to identify genes involved in their ecology and host specificity. The size of Lactarius genomes is significantly larger than other Russulales species, owing to a massive accumulation of transposable elements and duplication of dispensable genes. As expected, their repertoire of genes coding for plant cell wall-degrading enzymes is restricted, but they retained a substantial set of genes involved in microbial cell wall degradation. Notably, Lactarius species showed a striking expansion of genes encoding proteases, such as secreted ectomycorrhiza-induced sedolisins. A high copy number of genes coding for small secreted LysM proteins and Lactarius-specific lectins were detected, which may be linked to host specificity. This study revealed a large diversity in the genome landscapes and gene repertoires within Russulaceae. The known host specificity of Lactarius symbionts may be related to mycorrhiza-induced species-specific genes, including secreted sedolisins.

This article is the 13th contribution in the Fungal Diversity Notes series, wherein 125 taxa from four phyla, ten classes, 31 orders, 69 families, 92 genera and three genera incertae sedis are treated, demonstrating worldwide and geographic distribution. Fungal taxa described and illustrated in the present study include three new genera, 69 new species, one new combination, one reference specimen and 51 new records on new hosts and new geographical distributions. Three new genera, Cylindrotorula (Torulaceae), Scolecoleotia (Leotiales genus incertae sedis) and Xenovaginatispora (Lindomycetaceae) are introduced based on distinct phylogenetic lineages and unique morphologies. Heimuer, Auricularia heimuer, is one of the most famous traditional Chinese foods and medicines, and it is the third most important cultivated mushroom worldwide. The aim of this study is to develop genomic resources for A. heimuer to furnish tools that can be used to study its secondary metabolite production capability, wood degradation ability and biosynthesis of polysaccharides. The genome was obtained from single spore mycelia of the strain Dai 13782 by using combined high-throughput Illumina HiSeq 4000 system with the PacBio RSII long-read sequencing platform. Functional annotation was accomplished by blasting protein sequences with different public available databases to obtain their corresponding annotations. It is 49.76Mb in size with a N50 scaffold size of 1,350,668bp and encodes 16,244 putative predicted genes. This is the first genome-scale assembly and annotation for A. heimuer, which is the third sequenced species in Auricularia.

Three new species of Coltricia from southern China are described on the basis of morphological characters and molecular analyses. Phylogenetic analyses based on nuc rDNA ITS1-5.8S-ITS2 and partial 28S rDNA, and partial sequences of genes for RNA polymerase II subunits 1 and 2 confirmed the generic placement of the three new species. Coltricia lateralis is characterized by small, laterally stipitate basidiocarps, obtuse pileus margin, cinnamon-buff to honey yellow pore surface, and broadly ellipsoid basidiospores that measure 7-8 × 5-6 µm. Coltricia rigida is characterized by large, woody-hard fan-shaped basidiocarps, small pores (7-8 per mm), presence of septate sclerified hyphae, and subglobose to globose basidiospores that measure 6-7 × 5-6.5 µm. Coltricia wenshanensis is characterized by centrally stipitate basidiocarps, a distinctly concentrically zonate and sulcate pileal surface, large pores (0.5-2 per mm), and broadly ellipsoid to subglobose basidiospores that measure 7.5-8 × 6-7 µm.

Fomitiporella accommodates polypores producing annual to perennial basidiocarps with an indistinct subiculum (very thin to almost lacking), mostly a dimitic hyphal structure, lacking any kind of setae, with brownish, thick-walled basidiospores, and causing a white rot. Previously, only a few samples of Fomitiporella were studied on the basis of morphological and nuc 28S rDNA (28S)-based phylogenetic analyses. In this study, we made a comprehensive study on Fomitiporella on the basis of collections from Central America, USA, Europe, and China. The phylogenetic analysis, including 28 nuc 28S rDNA and 29 nuc rDNA ITS1-5.8S-ITS2 (internal transcribed spacer [ITS]) sequences newly generated, discovered 14 new lineages. Combined with morphological evidence, 4 new lineages are described and illustrated as new species, viz., Fomitiporella americana, F. micropora, F. sinica, and F. subinermis; 10 other new lineages, each with a single collection, are still treated as unidentified taxa; three new combinations, viz., Fomitiporella tenuissima, F. chinensis, and F. resupinata, are proposed. In addition, F. inermis is redescribed. A key to the 12 known species of Fomitiporella is provided.

Onnia includes white rotting polypores with annual basidiocarps, a duplex context, monomitic hyphal structure, hymenial setae, and hyaline, thin-walled, smooth basidiospores. Specimens of Onnia, originating mainly from East Asia, Europe, and North America, were studied using both morphology and phylogenetic analyses. Our concatenated data set was derived from 25 collections and included (i) 25 nuc rDNA internal transcribed spacer region sequences (ITS1-5.8S-ITS2 = ITS), 17 generated in this study; and (ii) 14 nuc rDNA 28S rDNA sequences, including the D1-D2 domains, 11 of them generated in this study. The resulting maximum likelihood and Bayesian phylogenies recovered all sampled collections of Onnia as a well-supported clade. In this clade, three previously accepted species, viz., Onnia leporina, O. tomentosa, and O. triquetra, received strong support, whereas three additional lineages with strong support represent the new species described in this paper, O. subtriquetra, O. microspora, and O. tibetica. Of the six Onnia species occurring on gymnosperms, O. tomentosa and O. leporina grow mainly on Picea and have circumboreal distribution in the Northern Hemisphere. In contrast, other species that mostly grow on Pinus are geographically restricted to limited regions, viz., O. triquetra in Europe, O. subtriquetra in North America, and O. microspora and O. tibetica in Asia.

Neomensularia (Hymenochaetaceae) is a new genus introduced for N. duplicata sp. nov. (generic type), N. crocitincta and N. kanehirae combs. nov., based on a combination of distinct morphological characters and phylogenetic analyses inferred from nuc rDNA partial 28S and ITS datasets. The new genus is characterized by annual, pileate, brown to fuscous basidiocarps, duplex context, dimitic hyphal structure, ventricose, and hooked hymenial setae, golden yellow, thick-walled, smooth basidiospores that are negative in both Melzer's reagent and Cotton Blue and a growth habit in tropical forests. Neomensularia duplicata closely resembles Mensularia radiata by hooked hymenial setae, but M. radiata has a homogenous context, larger pores (5-7 per mm), monomitic hyphal structure, hyphoid setae, larger cyanophilic basidiospores and occurs in temperate forests. Central American specimens labeled as Inonotus crocitinctus and Asian specimens identified as Fulvifomes kanehirae were re-examined. The duplex context, hooked hymenial setae and colored basidiospores are similar to N. duplicata Phylogenetic inferences based on 28S and ITS sequence data confirm their affinities with N. duplicata and result in the proposal of new combinations, Neomensularia crocitincta and N. kanehirae.

The Phellinus igniarius complex corresponds to Phellinus s.s., a well supported clade in the polyphyletic Phellinus s.l. studied worldwide. Recently a molecular phylogeny of this complex was performed for the European and North American species. In this study we expand the taxon samplings of the P. igniarius complex to include 59, seven and 12 additional collections originating from China, Czech Republic and USA, respectively. We generated 78 nuc rDNA ITS1-5.8S-ITS2 (ITS) and 42 translation elongation factor 1-α gene (tef1α) sequences. Based on the morphological and phylogenetic (combined ITS and tef1α dataset) analyses, Phellinus monticola, P. orientoasiaticus, P. padicola, P. parmastoi and P. pomaceoides are newly described and illustrated from China and USA. Phellinus pseudoigniarius is treated as a later synonym of P. igniarius, whereas Phellinus betulinus subsp. betulinus is accepted as P. betulinus. A total of 15 species are accepted in the P. igniarius complex worldwide. Of them, 10 species are distributed in eastern Asia, eight in Europe and six in North America. The taxonomy, phylogeny, host associations and geographic distributions of these 15 species are briefly discussed.

Bondarzewia is a remarkable polypore genus due to its relatively large poroid basidiocarps and belongs to order Russulales according to recent phylogenetic analyses. Two species, B. berkeleyi and B. mesenterica, are commonly reported in North America and Europe but the genus is poorly known elsewhere. We explored the phylogeny and species diversity of Bondarzewia based on a larger number of samples covering a wider geographic range, including eastern Asia, Europe, North America, Oceania and South America. Sequences were generated from the nuc rDNA region encompassing the internal transcribed spacers 1 and 2, along with the 5.8S rDNA (ITS) and nuc 28S rDNA D1-D2 domains (28S), and the final dataset included 28 samples of which 21 ITS and 28S sequences were newly generated. As a result of phylogenetic analyses and comparison of morphological features we propose a new classification of the genus recognizing 10 species. Most of the novel taxonomic concepts are geographically restricted in contrast to the past broad species concepts, especially of B. berkeleyi In addition, a degree of host specificity is observed with B. mesenterica, B. occidentalis, B. podocarpi, B. propria and B. submesenterica apparently restricted to gymnosperms and all others on angiosperm hosts where known. Three new species, B. kirkii, B. occidentalis and B. submesenterica, are described and illustrated. Three new combinations (B. dickinsii, B. propria, B. retipora) are revived from synonymy under B. berkeleyi A key to known species of Bondarzewia is provided.

Fungal interspecific interactions enhance biosynthesis of phenylpropanoid metabolites (PM), and production of nitric oxide (NO) is known to be involved in this process. However, it remains unknown which signaling pathway(s) or regulator(s) mediate fungal PM biosynthesis. In this study, we cocultured two white-rot fungi, Inonotus obliquus and Phellinus morii, to examine NO production, expression of the genes involved in phenylpropanoid metabolism and accumulation of phenylpropanoid-derived polyphenols by I. obliquus. Coculture of the two fungi caused an enhanced NO biosynthesis followed by increased transcription of the genes encoding phenylalanine ammonia lyase (PAL) and 4-coumarate CoA ligase (4CL), as well as an upregulated biosynthesis of styrylpyrone polyphenols in I. obliquus. Addition of the NO synthase (NOS) selective inhibitor aminoguanidine (AG) inhibited NO production by more than 90% followed by cease in transcription of PAL and 4Cl. Treatment of guanylyl cyclase inhibitor 1H-[1,2,4]oxadiazolo[4,3-a]quinoxalin-1-one did not affect NO production but suppressed transcription of PAL and 4CL and reduced accumulation of total phenolic constituents. Genome-wide analysis of I. obliquus revealed two genes encoding a constitutive and an inducible NOS-like protein, respectively (cNOSL and iNOSL). Coculture of the two fungi did not increase the expression of the cNOSL gene but triggered expression of the iNOSL gene. Cloned iNOSL from Escherichia coli shows higher activity in transferring L-arginine to NO, and this activity is lost upon AG addition. Thus, iNOSL is more responsible for NO production in I. obliquus and may act as an important regulator governing PM production during fungal interspecific interactions.

A laminated root rot on Qilian juniper (Sabina przewalskii) caused by a species of Phellinidium (Basidiomycota) was observed in northwest China. Seventeen fungal samples collected from Qinghai Province during 2012 and 2013 were used for taxonomic and pathogenicity tests. The fungal pathogen was identified by morphological characters and phylogenetic analyses based on nLSU sequences. A new fungus is described herein as Phellinidium qilianense sp. nov. One-year-old Qilian juniper seedlings were wound-inoculated under controlled conditions to test pathogenicity of the fungal species. The fungus was successfully reisolated from decayed tissue of tested seedlings. P. qilianense is a new forest pathogen on coniferous trees in China.

Cyanosporus is a cosmopolitan genus characterized by effused-reflexed to pileate basidiomata with a bluish tint and allantoid to cylindrical basidiospores which are negative to weakly positive in Melzer's reagent and Cotton Blue, causing a brown rot. Three new species of Cyanosporus, namely, C.linzhiensis, C.miscanthi and C.tabuliformis are described and illustrated. Phylogenies on Cyanosporus are reconstructed with seven loci DNA sequences including ITS, nLSU, nSSU, mtSSU, RPB1, RPB2 and TEF1 based on phylogenetic analyses combined with morphological examination. The description for the new species is given. The main morphological characteristics of all 38 accepted species in Cyanosporus are summarized.

Phylogenetic and morphological analyses on Perenniporia s.l. were carried out. Phylogenies on Perenniporia s.l. are reconstructed with two loci DNA sequences including the internal transcribed spacer (ITS) regions and the large subunit (nLSU). Two new species from Yunnan Province, southwest China, Perenniporiaprunicola and P.rosicola in Perenniporia s.l., are illustrated and described. Perenniporiaprunicola is characterised by the perennial and resupinate basidiomata with a clay pink pore surface when fresh, a trimitic hyphal system, the presence of clavate to fusiform hymenial cystidia, ellipsoid to broadly ellipsoid basidiospores measuring 4.8-6.2 × 3.6-4.5 µm. Perenniporiarosicola is characterised by annual and resupinate basidiomata with a white pore surface when fresh, a dimitic hyphal system, the presence of dendrohyphidia, broadly ellipsoid to subglobose basidiospores measuring 5-5.8 × 4-5.2 µm. In addition, Crassisporus is a genus in Perenniporia s.l., in which two new combinations Crassisporusminutus and C.mollissimus are proposed. Main morphological characteristics of species related to new taxa are also provided.

RESUMEN

Two taxonomically controversial polypore genera with reddish brown to orange basidiomata that stain reddish with KOH solution, Aurantiporus and Hapalopilus, are revised based on additional sampling, morphological examination, and phylogenetic analysis of a combined dataset of ITS1-5.8S-ITS2-nLSU sequences. Hapalopilus is a monophyletic genus belonging to Phanerochaetaceae, whereas Aurantiporus is a polyphyletic genus belonging to Meruliaceae. Hapalopilus and Aurantiporus s. str. are circumscribed, and two new species - Aurantiporusorientalis and Hapalopilustabuliformis - are described and illustrated from temperate China. In addition, four new combinations, viz. Aurantiporusalboaurantius, A.mutans, A.tropicus and Luteoporiaalbocitrina, are proposed based on morphology and phylogenetic analysis. The relationships between Aurantiporus and Hapalopilus are discussed.

The genus Favolaschia within the family Mycenaceae is characterised by the gelatinous basidiomata with poroid hymenophore and most species inhabit monocotyledonous plants. In this study, many samples covering a wide geographic range in China were examined morphologically and phylogenetically using concatenated ITS1-5.8S-ITS2-nLSU sequence data. Three new species clustering in Favolaschiasect.Anechinus, namely Favolaschiaimbricata, F.miscanthi and F.sinarundinariae, are described. Favolaschiaimbricata is characterised by imbricate basidiomata with pale grey to greyish colour when fresh and broadly ellipsoid basidiospores measuring 7-9 × 5-6.8 µm; F.miscanthi is characterised by satin white basidiomata when fresh, broadly ellipsoid basidiospores measuring 7.5-10 × 5.5-7 µm and inhabit rotten Miscanthus; F.sinarundinariae is characterised by greyish-white basidiomata when fresh, dark grey near the base upon drying, broadly ellipsoid to subglobose basidiospores measuring 7-9 × 5-7 µm and inhabit dead Sinarundinaria. The differences amongst the new species and their morphologically similar and phylogenetically related species are discussed. In addition, an updated key to 19 species of Favolaschia found in China is provided.

Amyloporia pinea sp. nov. is described and illustrated on the basis of collections from southern China. Morphology and phylogenetic analysis of rDNA ITS sequences support this new species. Morphologically, it is characterized by resupinate, annual basidiocarps, cream to yellowish buff pore surface when fresh, which becomes yellowish brown to clay-buff upon drying, a dimitic hyphal system with clamped generative hyphae and inamyloid skeletal hyphae, fusoid cystidioles, and cylindrical basidiospores; moreover, it causes a brown rot. Molecular phylogeny inferred from ITS sequence data suggested a close relationship between A. pinea and Amyloporia crassa sensu lato. Antrodia subxantha has amyloid skeletal hyphae, and grouped within the Amyloporia clade, hence, it is transferred to Amyloporia, and a new combination Amyloporia subxantha is proposed.

The phylogeny of taxa with poroid and lamellate hymenophores in Auriculariales as yet has not been well studied. Here we sequenced the nLSU and ITS regions of poroid and lamellate representatives from the genera Elmerina, Protodaedalea and Protomerulius to establish the phylogenetic position of these morphologically similar taxa. Our analyses suggest that (i) Elmerina foliacea is divergent from all other Elmerina species sampled; (ii) Protodaedalea hispida (the type of monotypic genus Protodaedalea) and Protomerulius efibulatus should be transferred to Elmerina as E. hispida and E. efibulata respectively; (iii) after exclusion of P. efibulatus, Protomerulius becomes monophyletic and represents the core of a "Protomerulius family" clade that potentially includes species from Tremellodendropsis, Heterochaetella and Protodontia; and (iv) the presence or absence of gloeocystidia serves as the most reliable morphological feature in delimiting Elmerina s.l. and Protomerulius. Concepts of Elmerina s.l. and Protomerulius are redefined, and diagnostic keys for these two genera are provided.

Russulales comprises a highly diverse group of species with respect to basidiocarp morphology and hymenophore type. We reconstructed evolutionary histories of Russulales using ITS and nLSU rDNA sequence data with an emphasis on phylogeny of wood-inhabiting hydnoid species in this order. Based on the combination of morphological and molecular evidence, two new genera, three new species and two new combinations are proposed. Dentipellis microspora and D. coniferarum, newly described, were placed outside Hericiaceae but are provisionally treated in Dentipellis due to their morphological similarity to the generic type. Dentipellis leptodon and D. taiwaniana are transferred to the new genus Dentipellicula as D. leptodon and D. taiwaniana, which is designated as the generic type. The new genus Dentipellopsis was erected to accommodate a newly described species D. dacrydicola. Characters are provided in a generic key to distinguish Dentipellicula, Dentipellis and Dentipellopsis that morphologically are highly similar in addition to a key to the current species in Dentipellis. A new species of Gloeodontia was described based on both molecular and morphological data, and a key to this genus also is provided.

RESUMEN

Two new triterpenoids, 3ß-(2-methoxy-oxalyloxy)-24-methylene-lanost-8-en-21-oic acid (1) and 3ß-(2-methoxy-oxalyloxy)-15α-hydroxy-24-methylene-lanost-8-en-21-oic acid (2), were isolated from cultures of the fungus Perenniporia maackiae, together with two previously known triterpenoids (3 and 4). Their structures were determined by spectroscopic methods, including 1D and 2D NMR, and MS analyses. All compounds were evaluated for their cytotoxic activities on five tumor cell lines.
