RESUMEN

PURPOSE: In patients with acute necrotizing pancreatitis (ANP), the site, size, and the number of acute necrotic collections (ANC) may determine the outcome of patients. The current study aimed to correlate the nature of ANC with the adverse outcomes in ANP patients. METHODS: This was a single-center, prospective study (August 2019-August 2022) recruiting patients with ANP, correlating the site, size, and number of ANC with the length of hospital stay, intensive care unit (ICU) stays, development of organ failure and infection, need for intervention, and mortality. RESULTS: A total of 114 patients (mean age: 37.3 ± 13.4 years, 85.1% males) with ANP were included in the study. The number and maximum diameter of collections significantly correlated with the length of the hospital and ICU stay and the need for intervention. Taking a cut-off size of 8 cm, the sensitivity and specificity for predicting the need for intervention were 82.7% and 74.2%, respectively. ANCs located in the perinephric, paracolic, subhepatic, and epigastric regions had a significant correlation with two or more adverse outcomes. Additional points were added to the modified CT severity index (mCTSI) based on the present study's findings. The new score had significantly higher AUROC than mCTSI for predicting infection, need for intervention, ICU stay > 1 week, and mortality. CONCLUSION: The site, size, and number of EPNs have a significant correlation with adverse clinical outcomes in patients with ANP. The inclusion of these parameters, along with present scoring systems, will help further improve the prognostication of patients.

RESUMEN

MAIN CONCLUSION: Genome editing offers revolutionized solutions for plant breeding to sustain food production to feed the world by 2050. Therefore, genome-edited products are increasingly recognized via more relaxed legislation and community adoption. The world population and food production are disproportionally growing in a manner that would have never matched each other under the current agricultural practices. The emerging crisis is more evident with the subtle changes in climate and the running-off of natural genetic resources that could be easily used in breeding in conventional ways. Under these circumstances, affordable CRISPR-Cas-based gene-editing technologies have brought hope and charged the old plant breeding machine with the most energetic and powerful fuel to address the challenges involved in feeding the world. What makes CRISPR-Cas the most powerful gene-editing technology? What are the differences between it and the other genetic engineering/breeding techniques? Would its products be labeled as "conventional" or "GMO"? There are so many questions to be answered, or that cannot be answered within the limitations of our current understanding. Therefore, we would like to discuss and answer some of the mentioned questions regarding recent progress in technology development. We hope this review will offer another view on the role of CRISPR-Cas technology in future of plant breeding for food production and beyond.

The Polygonaceae weed, Rumex dentatus L. grows in association with wheat, mustard and potato, while Polygonum glabrum Willd. grows in association with rice in India. Both larvae and adults of Galerucella placida Baly (Coleoptera: Chrysomelidae) voraciously consume these weeds. Applications of synthetic herbicides to control weeds are harmful to the environment including beneficial organisms. We propose to find volatile organic compounds (VOCs) from both weeds causing attraction of the biocontrol agent, G. placida, in order to attempt to use the insect as a biological weed control. Behavioral responses of G. placida towards volatile blends characteristic of undamaged (UD), insect-damaged (ID), jasmonic acid-treated (JA) or mechanically-damaged (MD) plants were conducted by Y-tube olfactometer bioassays. Cuminaldehyde was predominant in VOCs of UD R. dentatus, ID P. glabrum, and both JA and MD R. dentatus and P. glabrum. Geraniol was predominant in VOCs of UD P. glabrum, while 1,3-diethylbenzene predominated in VOCs of ID R. dentatus. Females were more attracted towards volatile blends of ID plants compared to UD or JA plants. Females did not show attraction towards volatile blends of JA plants. We identified two bioactive synthetics blends, one comprised of seven compounds - 16.65 µg 1,3-diethylbenzene, 10.72 µg acetophenone, 6.52 µg 2,6-(E,Z)-nonadienal, 2.46 µg 1-nonanol, 4.19 µg decanal, 9.86 µg 4-ethylacetophenone and 3.34 µg 1-hexadecene dissolved in 25 µl CH2Cl2 and the other containing five compounds - 2.50 µg 2-octanol, 6.84 µg limonene, 0.64 µg dodecane, 6.63 µg 4-ethylacetophenone and 0.24 µg geranyl acetone dissolved in 25 µl CH2Cl2. These two blends of volatile compounds could be used to attract the biocontrol agent during early vegetative period of these two weeds, which could lead to eradication of weeds from crop fields.

Background & objectives: Data on neonatal COVID-19 are limited to the immediate postnatal period, with a primary focus on vertical transmission in inborn infants. This study was aimed to assess the characteristics and outcome of COVID-19 in outborn neonates. Methods: All neonates admitted to the paediatric emergency from August 1 to December 31, 2020, were included in the study. SARS-CoV-2 reverse transcription- (RT)-PCR test was done on oro/nasopharyngeal specimens obtained at admission. The clinical characteristics and outcomes of SARS-CoV-2 positive and negative neonates were compared and the diagnostic accuracy of a selective testing policy was assessed. Results: A total of 1225 neonates were admitted during the study period, of whom SARS-CoV-2 RT-PCR was performed in 969. The RT-PCR test was positive in 17 (1.8%). Mean (standard deviation) gestation and birth weight of SARS-CoV-2-infected neonates were 35.5 (3.2) wk and 2274 (695) g, respectively. Most neonates (11/17) with confirmed COVID-19 reported in the first two weeks of life. Respiratory distress (14/17) was the predominant manifestation. Five (5/17, 29.4%) SARS-CoV-2 infected neonates died. Neonates with COVID-19 were at a higher risk for all-cause mortality [odds ratio (OR): 3.1; 95% confidence interval (CI): 1.1-8.9, P=0.03]; however, mortality did not differ after adjusting for lethal malformation (OR: 2.4; 95% CI: 0.7-8.7). Sensitivity, specificity, accuracy, positive and negative likelihood ratios (95% CI) of selective testing policy for SARS-CoV-2 infection at admission was 52.9 (28.5-76.1), 83.3 (80.7-85.6), 82.8 (80.3-85.1), 3.17 (1.98-5.07), and 0.56 (0.34-0.93) per cent, respectively. Interpretation & conclusions: SARS-CoV-2 positivity rate among the outborn neonates reporting to the paediatric emergency and tested for COVID-19 was observed to be low. The selective testing policy had poor diagnostic accuracy in distinguishing COVID-19 from non-COVID illness.

Two Polygonaceae weeds, Rumex dentatus L. and Polygonum glabrum Willd. are abundant in wheat- and rice-fields, respectively, in India. Galerucella placida Baly (Coleoptera: Chrysomelidae) is a biocontrol agent of these two weeds. The importance of long-chain alkanes and free fatty acids present in leaf surface waxes of these weeds was assessed as short-range attractant and ovipositional stimulant in G. placida females. Extraction, TLC, GC-MS and GC-FID analyses demonstrated 19 n-alkanes from n-C14 to n-C35 and 14 free fatty acids from C12:0 to C22:0 in leaf surface waxes. Hentriacontane was predominant among alkanes in both weeds, while oleic acid and docosanoic acid were predominant among free fatty acids in R. dentatus and P. glabrum, respectively. Females of G. placida were attracted toward one leaf equivalent surface wax of both weeds against the control solvent (petroleum ether) in a short Y-tube olfactometer bioassay. But, the insect could not differentiate between one leaf equivalent surface wax of R. dentatus and P. glabrum, indicating that both weed leaves were equally attractive in females. A synthetic blend of either 2.44, 35.57 and 23.58 µg ml-1 of octadecane, heptacosane and nonacosane, respectively, resembling the amounts present in one leaf equivalent surface wax of R. dentatus or 4.08, 19.54 and 23.58 µg ml-1 of octadecane, palmitoleic acid and docosanoic acid, respectively, resembling the amounts present in one leaf equivalent surface wax of P. glabrum acted as short-range attractant and ovipositional stimulant in G. placida. These results could be a basis for host plant specificity of the biocontrol agent.

Cytotoxic, mutagenic, and carcinogenic contaminants, such as heavy metals and radionuclides, have become an alarming environmental concern globally, especially for developed and developing nations. Moreover, inefficient prevalent wastewater treatment technologies combined with increased industrial activity and modernization has led to increase in the concentration of toxic metals and radioactive components in the natural water bodies. However, for the improvement of ecosystem of rivers, lakes, and other water sources different physicochemical methods such as membrane filtration, reverse osmosis, activated carbon adsorption, electrocoagulation, and other electrochemical treatment are employed, which are uneconomical and insufficient for the complete abatement of these emerging pollutants. Therefore, the application of bioremediation employing aquatic macrophytes and microphytes have gained considerable importance owing to the benefits of cost-effectiveness, eco-friendly, and higher energy efficiency. Thus, the present review aims to enlighten the readers on the potential application of algae, cyanobacteria, plant, and other aquatic micro- and macrophytes for the elimination of carcinogenic metals and radioactive isotopes from wastewater. Additionally, the use of transgenic plants, genetically modified species, algal-bacterial symbiosis for the enhancement of removal efficiency of mutagenic contaminants are also highlighted. Furthermore, species selection based on robustness, mechanism of different pathways for heavy metal and radionuclide detoxification are elucidated in this review article.

RESUMEN

Gene editing and/or allele introgression with absolute precision and control appear to be the ultimate goals of genetic engineering. Precision genome editing in plants has been developed through various approaches, including oligonucleotide-directed mutagenesis (ODM), base editing, prime editing and especially homologous recombination (HR)-based gene targeting. With the advent of CRISPR/Cas for the targeted generation of DNA breaks (single-stranded breaks (SSBs) or double-stranded breaks (DSBs)), a substantial advancement in HR-mediated precise editing frequencies has been achieved. Nonetheless, further research needs to be performed for commercially viable applications of precise genome editing; hence, an alternative innovative method for genome editing may be required. Within this scope, we summarize recent progress regarding precision genome editing mediated by microhomology-mediated end joining (MMEJ) and discuss their potential applications in crop improvement.

Larval Diaphania indica (Saunders) (Lepidoptera: Crambidae) cause complete defoliation of Trichosanthes anguina L. and reduce crop yield in India. Females lay eggs on the leaf surface, and therefore leaf surface waxes are potentially involved in host selection. Alkanes and free fatty acids are the major constituents of leaf surface waxes, so a study was conducted to determine whether these wax constituents from three T. anguina cultivars (MNSR-1, Baruipur Long, and Polo No.1) could act as short-range attractants and oviposition stimulants in D. indica females. Twenty n-alkanes from n-C14 to n-C36 and 13 free fatty acids from C12:0 to C21:0 were detected in the leaf surface waxes of these cultivars. Heptadecane and stearic acid were predominant among n-alkanes and free fatty acids, respectively, in these cultivars. Females showed attraction towards one leaf equivalent surface wax of each of these cultivars against solvent controls (petroleum ether) in Y-tube olfactometer bioassays. A synthetic blend of heptadecane, eicosane, hexacosane, and stearic acid, a synthetic blend of hexacosane and stearic acid, and a synthetic blend of pentadecane and stearic acid comparable to amounts present in one leaf equivalent surface wax of MNSR-1, Baruipur Long, and Polo No.1, respectively, were short-range attractants and oviposition stimulants in D. indica. Female egg laying responses were similar to each of these blends, providing information that could be used to developing baited traps in integrated pest management (IPM) programs.

KEY MESSAGE: CRISPR/Cas9-based multiplexed editing of SlHyPRP1 resulted in precise deletions of its functional motif(s), thereby resulting in salt stress-tolerant events in cultivated tomato. Crop genetic improvement to address environmental stresses for sustainable food production has been in high demand, especially given the current situation of global climate changes and reduction of the global food production rate/population rate. Recently, the emerging clustered regularly interspaced short palindromic repeats (CRISPR)/CRISPR-associated protein (Cas)-based targeted mutagenesis has provided a revolutionary approach to crop improvement. The major application of CRISPR/Cas in plant genome editing has been the generation of indel mutations via error-prone nonhomologous end joining (NHEJ) repair of DNA DSBs. In this study, we examined the power of the CRISPR/Cas9-based novel approach in the precise manipulation of protein domains of tomato hybrid proline-rich protein 1 (HyPRP1), which is a negative regulator of salt stress responses. We revealed that the precise elimination of SlHyPRP1 negative-response domain(s) led to high salinity tolerance at the germination and vegetative stages in our experimental conditions. CRISPR/Cas9-based domain editing may be an efficient tool to engineer multidomain proteins of important food crops to cope with global climate changes for sustainable agriculture and future food security.

Herbivorous insects employ host plant volatile blends as cue for host recognition. Adults of Aulacophora foveicollis Lucas (Coleoptera: Chrysomelidae) feed on leaves, flowers, and fruits of Solena amplexicaulis (Lam.) Gandhi (syn: Melothria heterophylla) (Cucurbitaceae), commonly known as creeping cucumber. Currently, this pest is controlled by insecticides application. Hence, it is necessary to find out volatile components from fruits attracting the insect, which might be used for eco-friendly pest management program. behavioral responses of females were measured by Y-tube olfactometer bioassays towards volatile blends from undamaged (UD), insect-damaged (ID), and mechanically damaged (MD) fruits with the aim to identify the compounds responsible for host fruit location. Volatile organic compounds were identified and quantified by GC-MS and GC-FID analyses, respectively. Nonanal was predominant in volatile blends of UD, ID, and MD fruits. 1-Octen-3-ol, 3-octanone, 2-octanol, heptadienal (2E,4E), 1-pentadecanol, and 1-hexadecanol were present in volatile blends of ID and MD fruits, but females did not show response to these six compounds. 1-Octanol and 1-heptadecanol were unique in volatile blends of UD fruits after 4 hr of damage, but females did not show response to these compounds. Females were more attracted to volatile blends from UD fruits after 4 hr of damage in comparison to volatile blends released by UD fruits, due to increased emissions of (E,Z)-2,6-nonadienal and 2E-nonenal. A synthetic blend of 3.35 µg (E,Z)-2,6-nonadienal and 1.72 µg 2E-nonenal dissolved in 25 µl CH2Cl2 could be used for the development of baited traps to control this insect pest in integrated pest management strategies.

Plants are the richest source of specialized metabolites. The specialized metabolites offer a variety of physiological benefits and many adaptive evolutionary advantages and frequently linked to plant defense mechanisms. Medicinal plants are a vital source of nutrition and active pharmaceutical agents. The production of valuable specialized metabolites and bioactive compounds has increased with the improvement of transgenic techniques like gene silencing and gene overexpression. These techniques are beneficial for decreasing production costs and increasing nutritional value. Utilizing biotechnological applications to enhance specialized metabolites in medicinal plants needs characterization and identification of genes within an elucidated pathway. The breakthrough and advancement of CRISPR/Cas-based gene editing in improving the production of specific metabolites in medicinal plants have gained significant importance in contemporary times. This article imparts a comprehensive recapitulation of the latest advancements made in the implementation of CRISPR-gene editing techniques for the purpose of augmenting specific metabolites in medicinal plants. We also provide further insights and perspectives for improving metabolic engineering scenarios in medicinal plants.

A microbial fuel cell (MFC) is a cutting-edge bioelectrochemical technology, which demonstrates power and other valuables recovery while treating wastewater by cultivating electroactive microbes. However, rampant biofilm growth over the cathode surface of air cathode MFC exacerbates the oxidation-reduction reaction rate, triggering a dip in the overall performance of MFC. In this sense, biosynthesized silver nanoparticles (AgNPs) have garnered a plethora of potential applications as cathode catalysts as well as anti-biofouling agent for MFCs without harming nature. The MFC equipped with the mixture of aloe vera and algae (@5 mg/cm2) synthesized AgNPs on cathode generated a maximum power density of 66.5 mW/m2 and chemical oxygen demand removal efficiency of 85.2%, which was ca. 5.6 times and 1.2 times higher compared to control MFC operated without any catalyst on cathode. Thus, this investigation paves the way for using eco-amiable, low-cost bioderived organic compounds to assist MFC in achieving high power output and other valuables with minimal reliance on chemicals.

Cytotoxic, malignant, and mutagenic pollutants like heavy metals have emerged as a serious global threat to the ecosystem. Additionally, the quantity of noxious metals in water bodies has increased due to expanding industrial activities and the application of incompetent wastewater treatment techniques. Owing to the benefits of eco-friendly phytoremediation, the utilization of algae in photosynthetic microbial fuel cell (PMFC) for removal of heavy metals has attracted increasing attention among researchers. Therefore, a successful fabrication and operation of a modular PMFC for simultaneous algal biomass production was exhibited, thus resulting in significant removal efficiency of Cu(II) (94 %) and Co(II) (88 %). Moreover, Co(II)-accumulated algal biochar after thermal activation was utilized as a cathode catalyst for the first time and attained 64.2 mW/m2 of power density through PMFC. Hence, this easily synthesised green cathode catalyst proved its ability to enhance the overall performance of PMFC by attaining higher power output while treating wastewater.

BACKGROUND: Patients with hepatocellular carcinoma (HCC) and cirrhosis can present with features of severe portal hypertension, which can be worsened further by portal vein tumoral thrombosis (PVTT). Due to the technical difficulties and short survival of these patients, HCC was traditionally considered a relative contra-indication for transjugular intrahepatic portosystemic shunt (TIPS). However, there is an increasing body of evidence, mainly from China, supporting the use of TIPS in HCC. The present study aimed at analyzing the efficacy and safety of TIPS in patients with HCC. METHODS: From 2000 through May 2023, MEDLINE, Embase and Scopus were searched for studies analyzing the outcome of TIPS in HCC. Technical and clinical success, adverse events (AE) and mortality were the main outcomes assessed. With the use of a random effects model, the event rates were combined. RESULTS: Total 19 studies with 1498 patients were included in the final analysis. The pooled technical and clinical success rates with TIPS in HCC were 98.8% (98.0-99.7) and 94.1% (91.2-97.0), respectively. After TIPS, ascites was controlled in 89.2% (85.1-93.3) of the cases, while rebleeding was observed in 17.2% (9.4-25.0) of cases on follow-up. The pooled incidence of overall AE, serious AE and post-TIPS hepatic encephalopathy (HE) was 5.2% (2.5-7.9), 0.1% (0.0-0.4) and 25.1% (18.7-31.5), respectively. On follow-up, 11.9% (7.8-15.9) of the patients developed shunt dysfunction requiring re-intervention. CONCLUSION: The present analysis supports the feasibility, safety and efficacy of TIPS in the management of portal hypertension in patients with HCC.

Introduction/Purpose: Patients with cirrhosis and hepatocellular carcinoma (HCC) can develop both benign and malignant portal vein thrombosis (PVT). Characterising the nature of PVT is important for planning an optimal therapeutic strategy. In the absence of typical findings or contraindications to computed tomography (CT) or magnetic resonance imaging (MRI), contrast-enhanced ultrasound (CEUS) could help in this differentiation. The present meta-analysis aimed to evaluate the performance of CEUS for characterising PVT in patients with HCC. Methods: Electronic databases of PubMed, Embase and Scopus were searched from inception to 31 December 2022 for studies analysing the role of CEUS in the differentiation of benign and malignant PVT in HCC. Using the bivariate random effect model, pooled sensitivity and specificity were calculated, and the summary receiver operating characteristic (sROC) curve was plotted. Results: A total of 12 studies with data from 712 patients were included in the meta-analysis. The pooled sensitivity and specificity of CEUS for the diagnosis of tumour in vein were 97.0% (95% CI: 93.0-98.7) and 96.8% (95% CI: 92.1-98.7), respectively, without significant heterogeneity. A sROC curve was plotted, and the area under the receiver operating characteristic was 0.99 (95% CI: 0.98-1.00). Despite the presence of publication bias, sensitivity analysis did not show any change in sensitivity and specificity. Discussion: Our meta-analysis summarises the accuracy data from 12 studies, including >700 subjects. Contrast-enhanced ultrasound had excellent diagnostic accuracy with pooled sensitivity and specificity of 97.5% (95% CI: 93.5-99.1) and 98.2% (95% CI: 91.5-99.6), respectively, without any significant heterogeneity. Additionally, the pooled positive LR, negative LR and DOR were 54.6 (95% CI: 11.1-25.6), 0.02 (0.01-0.07) and 2186.8 (318.3-15022.2), respectively. A positive result increases the pretest probability of malignant PVT from 50% to 98%, whereas a negative result decreases it from 50% to 2%. Most of the studies included in our meta-analysis used identical techniques and 6-12-month follow-up scans to check for thrombus progression or regression. Our analysis showed no significant heterogeneity in the studies, and area under receiver operating characteristic curve (AUROC) with 95% CI was 1.00 (95% CI: 0.99-1.00). This critical meta-analysis thus propels CEUS to the forefront for differentiating benign from tumoural PVT and suggests routinely using CEUS in patients presenting with HCC and evidence of thrombus on greyscale ultrasound. Conclusion: Contrast-enhanced ultrasound is an effective diagnostic modality differentiation of benign and malignant PVT in patients with HCC and can be an alternative modality to CT or MRI. Further studies are required to study the role of CEUS as initial diagnostic modality for the characterisation of PVT in HCC.

Introduction: During last few decades, radiological interventions have played crucial role in the management of the patients with chronic liver diseases. Various procedures including transjugualar intrahepatic portosystemic shunt (TIPS), transjugular liver biopsy (TJLB), transarterial chemoembilization (TACE)/transarterial radioembolization (TARE), balloon retrograde transvenous obliteration (BRTO) and plug-assisted retrograde transvenous obliteration (PARTO) are being performed safely and have significantly improved clinical outcomes in these patients. The technical and clinical success depend on appropriate patient selection along with thorough knowledge and experience to perform these procedures. On the other hand, few adverse events may also be associated with these procedures. The intervention radiologist and hepatologists should identify and treat these complications at the earliest so as to improve outcome of the patient. Materials and methods: About 25 hepatic intervention radiology procedures were performed in our center from January 2022 to 2023 May. Among these we have selected five patients who underwent TACE/TIPS/DIPS in our institute. We have selected these cases as in each of these cases we encountered some interesting outcomes/complications which were managed successfully. Results: The first case describes 33-year-old male with POEM syndrome and Budd Chiari Syndrome (BCS) who underwent TIPS and immediately had blockade of the stent. The second case is of a 43 years old male having BCS, refractory ascites with umbilical and inguinal hernia. The third case is of a 40 years old female with decompensated cirrhosis who underwent TIPS for portal hypertensive gastropathy. The fourth case is of a 51-years' female with decompensated cirrhosis with sarcopenia. Finally, the fifth case describes 24-year-old female with BCS and hepatocellular carcinoma. In this article we discuss the procedure and clinical course of the patients following the procedure. Conclusion: Hepatic radiological interventions though widely used can be associated with unusual albeit life threatening complications. Appropriate patient selection and thorough knowledge of procedure along with early diagnosis and management of these complications are key to obtain satisfying long term outcomes.

Cupric oxide mesospheres composed of its nanoparticles have been synthesized by a simple template free chemical route at different temperatures. Thermal aging followed by higher temperature (350 degrees C, 6 hours) annealing on these architects transformed them into hollow mesospheres consisting of sharp needle like structures with high aspect ratio (- 10(3)). A detailed analysis of field emission scanning electron microscopy confirmed a uniform registry of the prepared nanostructures. High resolution transmission electron microscopy showed that the as-grown mesospheres have hollow inner cavity with a thin outer shell. X-ray photoelectron spectroscopic analysis showed no obvious changes in the chemical composition of the nanostructures after annealing, confirming that the elements in the final products were in the proper oxidation states. Electron emission under electric field was investigated from these interesting structures. It was found that both of these nanostructures showed electron emission, but emission performance of the hollow mesospheres consisting of nanoneedles exhibited excellent performance with turn-on field as low as 2.8 V/microm and high enhancement factor (beta) - 5537.

The emission of CO2 from industrial (24%) and different anthropogenic activities, like transportation (27%), electricity production (25%), and agriculture (11%), can lead to global warming, which in the long term can trigger substantial climate changes. In this regard, CO2 sequestration and wastewater treatment in tandem with bioenergy production through photosynthetic microbial fuel cell (PMFC) is an economical and sustainable intervention to address the problem of global warming and elevating energy demands. Therefore, this review focuses on the application of different PMFC as a bio-refinery approach to produce biofuels and power generation accompanied with the holistic treatment of wastewater. Moreover, CO2 bio-fixation and electron transfer mechanism of different photosynthetic microbiota, and factors affecting the performance of PMFC with technical feasibility and drawbacks are also elucidated in this review. Also, low-cost approaches such as utilization of bio-membrane like coconut shell, microbial growth enhancement by extracellular cell signalling mechanisms, and exploitation of genetically engineered strain towards the commercialization of PMFC are highlighted. Thus, the present review intends to guide the budding researchers in developing more cost-effective and sustainable PMFCs, which could lead towards the commercialization of this inventive technology.

Recently, CRISPR-Cas9-based genome editing has been widely used for plant breeding. In our previous report, a tomato gene encoding hybrid proline-rich protein 1 (HyPRP1), a negative regulator of salt stress responses, has been edited using a CRISPR-Cas9 multiplexing approach that resulted in precise eliminations of its functional domains, proline-rich domain (PRD) and eight cysteine-motif (8CM). We subsequently demonstrated that eliminating the PRD domain of HyPRP1 in tomatoes conferred the highest level of salinity tolerance. In this study, we characterized the edited lines under several abiotic and biotic stresses to examine the possibility of multiple stress tolerance. Our data reveal that the 8CM removal variants of HK and the KO alleles of both HK and 15T01 cultivars exhibited moderate heat stress tolerance. Similarly, plants carrying either the domains of the PRD removal variant (PR1v1) or 8CM removal variants (PR2v2 and PR2v3) showed better germination under osmosis stress (up to 200 mM mannitol) compared to the WT control. Moreover, the PR1v1 line continuously grew after 5 days of water cutoff. When the edited lines were challenged with pathogenic bacteria of Pseudomonas syringae pv. tomato (Pto) DC3000, the growth of the bacterium was significantly reduced by 2.0- to 2.5-fold compared to that in WT plants. However, the edited alleles enhanced susceptibility against Fusarium oxysporum f. sp. lycopersici, which causes fusarium wilt. CRISPR-Cas9-based precise domain editing of the SlHyPRP1 gene generated multi-stress-tolerant alleles that could be used as genetic materials for tomato breeding.

Background: This study aimed to evaluate the safety and efficacy of USG-guided percutaneous drainage in liver abscesses of >5 cm. A lot of literature is available on the minimally invasive treatment of liver abscesses since its introduction in the early 1980s. This study focuses on the eastern Indian population and the outcome of treatment of liver abscess of >5 cm by means of catheter drainage and the use of antibiotics. Patients and Methods: This is a retrospective study conducted on a total of fifty patients over a period of 1 year, 1 month (from June 2017 to June 2018). Only patients with liver abscess with size >5 cm were included in the study. The demographic characteristics; comorbidities; and clinical, radiological, and bacteriological characteristics of liver abscesses in the eastern Indian population and the safety and efficacy of catheter drainage were evaluated. Results: It was found that because of preprocedural empirical antibiotic intake, 70% of the patients had no growth in the pus, whereas 12% had Entamoeba histolytica, 8% had Escherichia coli, and 6% had Klebsiella pneumoniae as the causative agent. The total duration of hospital stay ranged from 3 to 22 days, and the duration of intravenous antibiotics ranged from 1 to 9 days with a clinical success rate of 96% without any drainage-related complications. Conclusion: In contradiction to the earlier belief, percutaneous drainage is a safe and effective means of treatment in liver abscesses of >5 cm with high clinical success rate and reduced duration of intravenous antibiotic requirement as well as hospital stay.


Résumé Résumé Contexte: Cette étude visait à évaluer l'innocuité et l'efficacité du drainage percutané guidé par USG dans les abcès du foie de > 5 cm. De nombreuses publications sont disponibles sur le traitement mini-invasif des abcès du foie depuis son introduction au début des années 1980. Cette étude se concentre sur la population de l'est de l'Inde et sur les résultats du traitement des abcès du foie > 5 cm au moyen d'un drainage par cathéter et de l'utilisation d'antibiotiques. Patients et Méthodes: Il s'agit d'une étude rétrospective menée sur un total de cinquante patients sur une période de 1 an, 1 mois (de juin 2017 à juin 2018). Seuls les patients présentant un abcès hépatique de taille > 5 cm ont été inclus dans l'étude. Les caractéristiques démographiques ; comorbidités ; et les caractéristiques cliniques, radiologiques et bactériologiques des abcès du foie dans la population de l'est de l'Inde et la sécurité et l'efficacité du drainage par cathéter ont été évaluées. Résultats: Il a été constaté qu'en raison de la prise d'antibiotiques empiriques préopératoires, 70 % des patients n'avaient pas de croissance dans le pus, alors que 12 % avaient Entamoeba histolytica, 8 % avaient Escherichia coli et 6 % avaient Klebsiella pneumoniae comme agent causal. La durée totale d'hospitalisation variait de 3 à 22 jours, et la durée d'antibiothérapie intraveineuse variait de 1 à 9 jours avec un taux de réussite clinique de 96 % sans aucune complication liée au drainage. Conclusion: Contrairement à la croyance antérieure, le percutané Le drainage est un moyen sûr et efficace de traitement des abcès hépatiques > 5 cm avec un taux de réussite clinique élevé et une durée réduite des besoins en antibiotiques intraveineux ainsi que du séjour à l'hôpital.
