RESUMEN

Pharmacology has broadened its scope considerably in recent decades. Initially, it was of interest to chemists, doctors and pharmacists. In recent years, however, it has been incorporated into the teaching of biologists, molecular biologists, biotechnologists, chemical engineers and many health professionals, among others. Traditional teaching methods, such as lectures or laboratory work, have been superseded by the use of new pedagogical approaches to enable a better conceptualization and understanding of the discipline. In this article, we present several new methods that have been used in Spanish universities. Firstly, we describe a teaching network that has allowed the sharing of pedagogical innovations in Spanish universities. A European experience to improve prescribing safety is described in detail. The use of popular films and medical TV series in biomedical students shows how these audiovisual resources can be helpful in teaching pharmacology. The use of virtual worlds is detailed to introduce this new approach to teaching. The increasingly important area of the social aspects of pharmacology is also considered in two sections, one devoted to social pharmacology and the other to the use of learning based on social services to improve understanding of this important area. Finally, the use of Objective Structured Clinical Evaluation in pharmacology allows to know how this approach can help to better evaluate clinical pharmacology students. In conclusion, this article allows to know new pedagogical methods resources used in some Spanish universities that may help to improve the teaching of pharmacology.

BACKGROUND: Some studies have been performed to assess the effects of levosimendan on cardiac function when administered to cardiac surgery patients with low cardiac output syndrome (LCOS) in the immediate postoperative period. Levosimendan is an inotropic agent for the treatment of low cardiac output syndrome that seems to have a protective effect on renal function. METHODS: It is a quasi-experimental study. A total of 100 patients with LCOS received either beta-agonists or levosimendan. We assessed the incidence of postoperative kidney failure in cardiac surgery patients. In patients who had kidney failure at diagnosis of LCOS, we examined whether differences existed in the evolution of kidney failure based on the treatment administered for LCOS. The parameters measured included haemodynamics, oxygen supply, and renal function as assessed by the AKI scale. ANOVA, Student's t-test and Wilcoxon or Friedman tests were used. RESULTS: Up to 30% of cardiac surgery patients had kidney failure at diagnosis of LCOS. Kidney failure at discharge from the ICU was more frequent in patients who received beta-agonist drugs as compared to those who received levosimendan (p < 0.05). CONCLUSION: The incidence of kidney failure decreased with the postoperative administration of levosimendan to cardiac surgery patients with LCOS, as compared to beta-agonists. OBJECTIVES: To evaluate the possibility of determining the genetic profile of primary malignant tumors of the breast from specimens obtained by ultrasound-guided percutaneous biopsies during the diagnostic imaging workup. MATERIAL AND METHODS: This is a retrospective study in 13 consecutive patients diagnosed with invasive breast cancer by B-mode ultrasound-guided 12 G core needle biopsy. After clinical indication, the pathologist decided whether the paraffin block specimens seemed suitable (on the basis of tumor size, validity of the sample, and percentage of tumor cells) before sending them for genetic analysis with the MammaPrint® platform. RESULTS: The size of the tumors on ultrasound ranged from 0.6cm to 5cm. In 11 patients the preserved specimen was considered valid and suitable for use in determining the genetic profile. In 1 patient (with a 1cm tumor) the pathologist decided that it was necessary to repeat the core biopsy to obtain additional samples. In 1 patient (with a 5cm tumor) the specimen was not considered valid by the genetic laboratory. The percentage of tumor cells in the samples ranged from 60% to 70%. In 11/13 cases (84.62%) it was possible to do the genetic analysis on the previously diagnosed samples. CONCLUSION: In most cases, regardless of tumor size, it is possible to obtain the genetic profile from tissue specimens obtained with ultrasound-guided 12 G core biopsy preserved in paraffin blocks.

OBJECTIVE: To review the key points that are essential for the correct staging of cervical cancer by magnetic resonance imaging. CONCLUSION: Magnetic resonance imaging is the method of choice for locoregional staging of cervical cancer. Thorough evaluation of prognostic factors such as tumor size, invasion of adjacent structures, and the presence of lymph node metastases is fundamental for planning appropriate treatment.

This study evaluated whether phenotypic appearance (PA) alteration during two developmental phases in laying hens, reared in two different group sizes, affects stress and immune responses. After hatching, 750 chicks were randomly assigned to 30 pens at a group size of either 10 or 40 birds. Then, the appearance of 0, 30, 50, 70 or 100% of the chicks in each pen was altered by blackdyeing their head feathers (marked); remaining chicks were unmarked. At 32 weeks, basal and postacute stress plasma corticosterone concentration, leukocyte counts, phytohemagglutinin-p lymphoproliferative and primary antibody responses were measured in six birds/pen. Analysis of variances (ANOVAs) showed no differences among treatment combinations. In a second phase, birds within initially homogeneous pens were sequentially either marked or had dye bleached to alter PA of 70% of hens in each flock (= group in a pen). Hens within initially heterogeneous pens remained unaltered as controls. The above variables were remeasured. Hens in phenotypically manipulated pens showed modified leukocyte counts compared to hens in control pens, indicating a chronic stress reaction in all penmates (whether individual PA was altered or not). Social isolation increased plasma corticosterone concentration. However, within groups of n = 40, phenotypically unaltered hens had lower responses than their altered penmate counterparts, suggesting that remaining in a stable PA group aids better coping with challenges. Although all hens in manipulated pens showed modified leukocyte counts, their antibody and lymphoproliferative responses did not differ from controls suggesting that all groupmates were able to immunologically cope with the challenges presented, within the timeframe evaluated.

RESUMEN

We present both experimentally and theoretically the transformation of radially and azimuthally polarized vector beams when they propagate through a biaxial crystal and are transformed by the conical refraction phenomenon. We show that, at the focal plane, the transverse pattern is formed by a ring-like light structure with an azimuthal node, this node being found at diametrically opposite points of the ring for radial/azimuthal polarizations. We also prove that the state of polarization of the transformed beams is conical refraction-like, i.e., that every two diametrically opposite points of the light ring are linearly orthogonally polarized.

Due to the increasing interest of polarimetric information in numerous applications, different Stokes polarimeter designs are provided in the literature for the measure of light beam polarization. Recently, the concept of polarimeters based on the conical refraction (CR) phenomenon, occurring in biaxial crystals, was proposed. CR polarimeters are snapshot polarimeters that allow controlling the volume of data redundancy without an increase in the acquisition time. We present the implementation, calibration, and analysis of an incomplete CR-polarimeter optimized for the measure of linear polarizations. A simpler and cheaper experimental configuration is achieved, if compared with other polarimeters proposed in the literature. The suitability of the polarimeter is experimentally demonstrated and some of its benefits, such as its performance in low-intensity conditions, are discussed.

The prevalence of alcohol intake in women who become pregnant is similar to that found in the general population, especially in cases of unplanned pregnancies. Consequently, foetal exposure is high during the period of maximum vulnerability. The present study was carried out to determine the prenatal level of exposure to alcohol in Málaga, a Mediterranean region whose economy is based on the touristic sector (Costa del Sol). A cross-sectional, observational design was used to investigate the consumption of alcohol during pregnancy, based on a self-reporting questionnaire. A total of 451 women in the first, second or third trimesters of pregnancy were recruited. Consumption prevalences in each trimester were 40.7%, 25.5% and 17.1%. A higher educational level was associated with greater exposure to alcohol (risk ratio, 1.87 [1.30-2.69]). These results should alert the providers of obstetric care in touristic areas to the need for the adoption of adequate preventive measures.

Alterations of birds' phenotypic appearance (PA) may lead to unwanted behaviors, potentially impairing poultry welfare, health, and productive performance. Likewise, group size may play an important role modulating the expression of adaptive behaviors. This study evaluates whether changes in the PA of Hy-line Brown laying hens may affect their BW and egg production, and if so, whether these effects depend on group size. A total of 1,050 one-day-old chicks were randomly assigned to 1 of 45 pens. Groups were of 10, 20, or 40 individuals (8 hens/m(2)). At arrival, the PA of 0, 30, 50, 70, or 100% of the birds within each group was artificially altered by marking the back of their heads black. The remaining birds within groups were unaltered. The 30% marked hens within groups of 10 individuals had a lower BW at 24 wk of age than their 70% unmarked counterparts, whereas the other groups showed similar BW. No differences were detected in egg laying performance during this phase. Next, within the initially homogeneous groups (0 and 100%), 30, 50, and 70% of the hens were either marked or unmarked (PA changed) sequentially at 34, 38, and 44 wk of age. Hens within the initially heterogeneous groups of 30, 50, and 70% marked birds remained unchanged and were used as controls. Groups of 40 individuals showed a reduction in BW gain and weekly hen-day-egg production after 30% PA changes, as compared with control counterparts. No differences were found in pens of 10 hens, and the groups of 20 showed intermediate results. A transient reduction in egg production was found after 50% PA changes. No further productive effects were observed after 70% changes. Our findings suggest that differences in hen appearance, which may occur due to variations in health status, injuries, and other natural causes, can be critical for production and welfare management practices depending both on the flock size and the birds' previous experience in exposure to group phenotypic heterogeneity.

The comb is an ornament involved in signalling condition in domestic fowl. We hypothesised that comb size, comb shape complexity (i.e., rugosity, the comb perimeter jaggedness), and comb laterality of laying hens would be influenced by the degree of environmental enrichment experienced during juvenile development in the form of resource choice. We conducted a 2 × 2 factorial crossover experiment with pullets reared in pens containing four perches of equal length and four litter areas of equal size. Pullets were exposed to a single choice vs multiple choices of perch and litter types (i.e., all the same vs all different) during Weeks 1-4 (Period 1) and/or Weeks 5-15 (Period 2) of rearing (n = 4 pens/treatment combination) prior to transfer to standard adult laying pens for Weeks 16-27 (Period 3). In Week 27, combs were photographed, and comb laterality (hanging on left or right side) was noted. Using a custom-made image analysis programme, we captured comb area (mm2), perimeter length (mm), and rugosity ((perimeter length - horizontal length) / horizontal length) from comb photographs of 6-7 randomly selected hens/pen. We predicted that hens reared in the multi-choice environment during Periods 1 and 2 would have larger, more complex, and left-side-biased combs than those in the other treatment groups, reflecting lower allostatic load. The predicted comb side bias was based on a possible bias in head posture/movements associated with greater right eye/ear use and left-brain hemispheric dominance. Contrary to our predictions, we detected an overall right-side bias in comb laterality, and no associations between resource choice treatment in Period 1 or Period 2 and comb area, perimeter length, rugosity, or laterality of the adult hens. Thus, variation in allostatic load resulting from the rearing treatments was insufficient to modify the trajectory of comb morphological development, possibly due to a ceiling effect when comparing environmental treatments on the positive end of the welfare spectrum. We found that left-lopping combs had shorter perimeters than right-lopping combs. However, among hens with left-lopping combs, those with larger combs were heavier and had less feather damage, while among hens with right-lopping combs, those with longer-perimeter combs were heavier and tended to have less comb damage. In conclusion, comb characteristics were related to physical condition at the individual level but did not serve as sensitive integrated indicators of hen welfare in response to basic vs enhanced resource choice during rearing.

Current scientific approaches to welfare assessment in broilers are based on individual sampling that can be time consuming under field conditions. On the other hand, farmers conduct routine checks based on walks through the house to screen birds' health condition. We adapted the walks through following line transect methodology used in wildlife studies to explore their feasibility as a welfare assessment tool. The aim of this study was to compare broiler welfare assessed by individual sampling and transect walks. We evaluated 6 identically managed flocks. For individual sampling, we collected measures on 150 birds, including weight, breast dirtiness, hock and footpad dermatitis, lameness, and immobility. Transect observations were conducted by slowly walking on randomized paths within each house recording: immobility, lameness, back dirtiness, sickness, agony, and dead. Transect walks allowed detection of small variations (P < 0.003) in the prevalence of most welfare indicators considered with consistency in interobserver reliability (P ≥ 0.05). In addition, assessments across transects were highly consistent (P ≥ 0.05). Individual sampling was also sensitive to differences across houses (P < 0.01) with the exception of immobility (P = 0.783). No differences were found across sampling locations (P ≥ 0.05). However, both methods differed greatly in the frequency of the incidence of the parameters considered. For example, immobility varied from 0.2 ± 0.02% for transect walks to 4 ± 2.3% for individual sampling, whereas lameness varied between 0.8 ± 0.07% and 24.2 ± 4.7% for transect and samplings, respectively. It is possible that the transect approach may have overlooked walking deficiencies because a large number of birds were scored, although if this was the case, the consistency obtained in the scoring across observers and transects would be surprising. Differences may also be related to possibly biased individual sampling procedures, where less mobile and passive individuals may be more likely to be caught. Furthermore the procedure may cause fatigue and fear reactions reducing mobility. Current study provides new insights into constraints and advantages of broiler on-farm assessment methods, which should be considered for designing on-farm welfare assessment protocols.

In modern rearing systems, turkey producers often face economic losses due to increased aggression, feather pecking, cannibalism, leg disorders, or injuries among birds, which are also significant welfare issues. The main underlying causes appear to relate to rapid growth, flock size, density, poor environmental complexity, or lighting, which may be deficient in providing the birds with an adequate physical or social environment. To date, there is little information regarding the effect of these factors on turkey welfare. This knowledge is, however, essential to ensure the welfare of turkeys and to improve their quality of life, but may also be beneficial to industry, allowing better bird performance, improved carcass quality, and reduced mortality and condemnations. This paper reviews the available scientific literature related to the behavior of turkeys as influenced by the physical and social environment that may be relevant to advances toward turkey production systems that take welfare into consideration. We addressed the effects that factors such as density, group size, space availability, maturation, lightning, feeding, and transport may have over parameters that may be relevant to ensure welfare of turkeys. Available scientific studies were based in experimental environments and identified individual factors corresponding to particular welfare problems. Most of the studies aimed at finding optimal levels of rearing conditions that allow avoiding or decreasing most severe welfare issues. This paper discusses the importance of these factors for development of production environments that would be better suited from a welfare and economic point of view.

At 17 wk of age, 1,800 Lohman brown hens were housed in 8 pens of an experimental aviary system, specifically set up for the purposes of the present study, and kept until 26 wk without or with nest lighting (lights inside the nest 1.5 h before the lighting of the installation) for training in the nest use. Then, at 27 wk, 4 combinations of nest curtains were adopted to evaluate the effects on hens' distribution, that is, nests with red (RR) or yellow (YY) curtains at all tiers; nests with red and yellow curtains at the first and second tier, respectively (RY); or nests with yellow and red curtains at the first and second tier, respectively (YR). The use of enlightened compared to dark nests at housing increased the oviposition rate (P < 0.001) and decreased the rate of broken (P < 0.001) and dirty eggs (P < 0.05) from 27 to 45 wk, while increasing the rate of eggs laid inside the nests (P < 0.001). The presence of yellow nest curtains increased the rate of hens on the floor in pens YY and YR compared to pens RR and RY (35.3 and 35.5% vs. 34.1 and 33.3%, respectively; P = 0.05) and the rate of floor eggs in pens YR (2.23% vs. 1.63 and 1.65% in pens RR and RY; P < 0.05). In pens RY, a higher rate of eggs was always found on the second tier compared to the first one with the most inhomogeneous distribution compared to pens RR, YY, and YR (+10.8 vs. +3.4, +1.9, and +4.6 percentage points of eggs laid on the second tier compared to the first one, respectively). In conclusion, nest lighting at housing trained hens to the use of nests while improving egg production in terms of quantity and quality. The use of yellow curtains on nests moved hens between the different levels of the aviary but this was not associated with an increased nest use for laying.

Astringency is a highly complex sensation which involves multiple mechanisms occurring simultaneously, such as the interaction between flavan-3-ols and salivary proteins (SP). Moreover, astringency development can be affected by the presence of polysaccharides such as mannoproteins (MP). The aim of this work was to evaluate the molecular mechanisms whereby MP could modulate the astringency elicited by tannins, using a cell-based model of the oral epithelium (TR146 cells), and the effect of salivary proteins on these interactions. The binding of flavan-3-ols to oral cells was evaluated by DMACA assay, while the content of unbound flavan-3-ols after the interactions was assessed by means of HPLC-DAD-MS. Results obtained confirm the existence of cell-tannin interactions, that can be partially inhibited by the presence of SP and/or MP. The most significant decrease was obtained in the system containing MPF (38.16%). Both mannoproteins assayed seem to have modulating effect on flavan-3-ol-SP interactions, acting by two different mechanisms: MPF would lead to the formation of SP/MPF/flavan-3-ols ternary soluble aggregates, while MPL seems to prevent flavan-3-ol-saliva interaction by a competitive mechanism, i.e., MPL would reduce cell-tannin interactions, similar to SP. This study suggests that mannoproteins with different compositional characteristics could exhibit preferential interaction with distinct flavan-3-ol families.

Deterioration of footpad and hock condition and increased stress are important welfare concerns in high-density broiler production. It has been argued that environmental enrichment may help address these issues. We hypothesized that increasing environmental complexity by providing access to barrier perches across a range of densities should improve footpad and hock health, reduce leg fluctuating asymmetry (FA), and mitigate fear in broilers without compromising bird performance. In this study, 2,088 one-day-old broiler chicks were randomly assigned to one of the following barrier and density treatment combinations over 4 replications: simple barrier, complex barrier, or no barrier (control) and low (8 birds/m(2)), moderate (13 birds/m(2)), or high (18 birds/m(2)) density. In wk 7, data were collected on average tibia width and length, FA of tibia width and tibia length, footpad and hock lesions, tonic immobility, feed conversion, final BW, and mortalities. Broilers at higher densities had more severe footpad (P < 0.0001) and hock lesions (P < 0.0001). Their tibias were also longer (P < 0.0001) and less symmetric in length (P < 0.05). The tibias of birds in complex barrier pens were more symmetric in length compared with controls (P < 0.05). Barrier treatment had no effect on hock lesions, although there was a trend for simple barriers to reduce the severity of footpad lesions compared with the control treatment (P = 0.089). Birds kept in simple barrier pens were more susceptible to tonic immobility induction (P < 0.05). Final BW, feed conversion, and mortalities were neither affected by stocking density nor by barrier inclusion. In conclusion, this study shows a negative effect of high density on broiler footpad health and FA. Although barrier perches did not appear to reduce fearfulness, the improvement in footpad health suggests that simple barriers may provide key welfare benefits to broiler chickens.

In captivity, the positioning of structural enrichment and food resources influences behavior and space use. The aim of this experiment was to examine the influence of cover panels and the positioning of food resources on the movement and space use of domestic fowl (Gallus gallus domesticus). Eight groups of 45 male chickens were used for this study. Each group was temporarily divided into 2 groups of 20 birds; each group was used to investigate the influence of cover panels and the effects of food resources. In the cover panel treatments, 20 birds were placed in the 10-m(2) testing enclosures that contained one 2-m cover panel in the center, four 0.5-m panels in a zigzag fashion, or had no panels (controls). In the food resource treatments, the position of the feed trays varied, with 1 feed tray in the center; 2 feed trays, one at each edge; or 4 feed trays, one at each corner of the enclosure. Locations of focal birds were collected through instantaneous scan sampling that was recorded as X,Y coordinates. From these X,Y coordinates, we calculated net and total distance moved, mean and maximum step lengths, and angular dispersion of the path of movement. To calculate long-term space use, 3 replications for each of 3 cover panel and food resource treatments were placed in nine 10-m(2) enclosures for 1 wk. Locations of focal birds in each group were collected by ad libitum scan sampling and data were used to calculate core areas by kernel estimates. Mixed model ANOVA was used to determine the effects of the distribution of cover panels and positioning of food resources on movement parameters during the study period, whereas 1-way ANOVA was used for core areas. Surprisingly, our analyses showed that long-term and short-term movement was not affected by changing the location of cover panels or food resources. Only net distance seemed to be affected to a certain degree by the presence of the cover and the distinctive availability of food resources.

One of the most accepted mechanisms of astringency consists of the interaction between polyphenols and some specific salivary proteins. This work aims to obtain further insights into the mechanisms leading to a modulation of astringency elicited by polyphenols. The effect of the presence of different chemical species (present in food and beverages as food additives) on the polyphenol-protein interaction has been evaluated by means of techniques such as sodium dodecyl sulfate polyacrylamide gel electrophoresis and cell cultures using a cell-based model of the oral epithelium. Results obtained showed that several chemicals, particularly sodium carbonate, seem to inhibit polyphenol binding to salivary proteins and to oral epithelium. These results point out that polyphenol-saliva protein interactions can be affected by some food additives, which can help to better understand changes in astringency perception.

The potential of the transect method was tested for early detection of welfare problems associated with bird age and genetic line, litter quality, and transect location. On-farm welfare impairment and its consequences on slaughter outcomes were evaluated to test the method's predictive ability. A total of 31 commercial Ross, Cobb, and mixed RC broiler flocks were evaluated at 3, 5, and 6 wk of age. Two observers evaluated 3 transects each, simultaneously and in the same house by detecting welfare indicators including lame, immobile, sick, small, dirty, tail wounds, other wounds (head and back wounds), featherless, terminally ill, and dead birds. Increasing lame, immobile, sick, and terminally ill birds according to bird age (P < 0.001) was detected. Higher incidences of small and sick birds were detected in C and RC (P < 0.001) as compared to R flocks, whereas more dead and tail wounded were observed in RC compared to R and C flocks at week 5 (P < 0.001). Dirty incidence increased as litter quality deteriorated (P < 0.001). A higher incidence of immobile, small, sick, dirty, and dead was registered near house walls (P < 0.001). Differences across observers were detected for lame, immobile, and terminally ill birds (P < 0.001). For the observer by bird age interaction, differences were detected for dirty, tail wounds, and other wounds (P < 0.05). Pearson correlations between welfare indicators at week 3 and those at final weeks of age (P < 0.05) ranged between r values of -0.2 and 0.654 (P < 0.05). Correlations between welfare indicators and slaughter outcomes showed a relationship between flock mortality and dead on arrival, footpad dermatitis, leg problems, and illness (P < 0.05). Litter quality positively correlated with downgrades (P < 0.001). This study showed the potential of transects to detect differences in welfare indicators according to factors that effects were previously reported. It demonstrated the transect potential for detecting and predicting the consequences of welfare impairment on slaughter outcomes. This would make the transect method a useful tool for notifying and rectifying welfare deterioration as early as at 3 wk of age.

In this study, we have evaluated by HPLC-DAD, DLS and MALDI-TOF a synergic effect of the coexistence of two salivary-PRP fractions (basic-PRPs and acidic PRPs) on the interaction with flavanols. Results obtained showed noticeable enhancement of the interaction between (epi)catechin and PRPs when both types of proteins are blended. Up to 30 soluble aggregates have been tentatively identified with molecular weight from 4680 to 35,851. (epi)Catechins seem to bind preferentially bPRPs than aPRPs, although the medium size aggregates flavanol-bPRPs formed could favour the interaction with aPRPs giving rise to soluble mixed aggregates.

Designs to enrich the environment are crucial in the effort to fully address the biological needs of domestic animals. Although enrichment programs have been shown to improve health and welfare, little is known of their potential for application to commercial broiler breeder environments. We investigated the potential benefits of cover panels for broiler breeder reproductive performance in a commercial setting. This demonstration trial occurred on 5 commercial broiler breeder farms, each with a control and panel treatment room containing approximately 7,000 females and 800 males. Reproductive performance was measured from 25 to 60 wk by the number of eggs laid per female per week as well as weekly fertility and hatchability rates. The location of marked males was recorded weekly to quantify male movement. Access to cover panels improved egg production by 2.1% and maintained better hatchability and fertility throughout the breeding cycle (significant interactions of age and panel treatment) leading to an additional 4.5 chicks/female. Male home ranges, based on minimum convex polygons, were larger in the enriched (259 +/- 24.4 m(2)) vs. control flocks (184 +/- 23.1 m(2)). Providing enrichment in the form of cover panels improved reproductive performance, most likely by increasing males' mating opportunities and reducing female stress. We found a clear economic benefit to providing enrichment, an estimated $3 million if all breeder houses within the participating company were outfitted with the panels. These results demonstrate that environmental enrichment is not only beneficial for broiler breeder welfare, but can also be economically advantageous, resulting in a win-win situation for poultry welfare and production.
