Purpose: Elevations in intraocular pressure (IOP) are associated with the development of glaucoma and loss of sight. High transforming growth factor-ß (TGF-ß) 1 levels in the eye's anterior chamber can lead to dysfunctional contractions through RhoA signaling in trabecular meshwork (TM) cells and IOP spikes. Sustained high TGF-ß levels leads to TM fibrosis and sustained increases in IOP. We investigated whether inhibiting RhoA, using a siRNA-mediated RhoA (siRhoA), controls IOP by altering TM expression of fibrosis and contractility-related proteins in a rodent model of glaucoma. Methods: TGF-ß was injected intracamerally twice a week into adult Sprague Dawley rats, and IOP was recorded with tonometry. Animals were euthanized on day 7 and 35 with TM expression of fibrosis and contractility-related proteins, as well as survival of retinal ganglion cells (RGCs) assessed with immunohistochemistry. siRNA against RhoA or enhanced green fluorescent protein (EGFP) was also injected intracamerally into select animals. Successful RhoA knockdown was determined with quantitative reverse transcription polymerase chain reaction (RT-PCR) and immunohistochemistry, and the effects of the knockdown on the parameters above analyzed. Results: TGF-ß caused increased TM contractile proteins and IOP spikes by day 7, sustained increases in IOP from day 15, and TM fibrosis at day 35. siRhoA abolished the transient 7 day IOP rise but not the later sustained IOP increase (due to fibrosis). At 35 days, TGF-ß-related RGC loss was not prevented with siRhoA treatment. Conclusions: We conclude that RhoA signaling mediates the early IOP rise induced by TM cellular changes associated with contractility but not the sustained IOP elevation caused by TM fibrosis. Thus, RhoA therapies offer a clinically relevant opportunity for IOP management, likely through the modulation of TM contractility, but appear to be ineffective in the amelioration of fibrosis.

The sustained decline in marine fisheries worldwide underscores the need to understand and monitor fisheries trends and fisher behavior. Recreational fisheries are unique in that they are not subject to the typical drivers that influence commercial and artisanal fisheries (e.g., markets or food security). Nevertheless, although exposed to a different set of drivers (i.e., interest or relaxation), recreational fisheries can contribute to fishery declines. Recreational fisheries are also difficult to assess due to an absence of past monitoring and traditional fisheries data. Therefore, we utilized a nontraditional data source (a chronology of spearfishing publications) to document historical trends in recreational spearfishing in Australia between 1952 and 2009. We extracted data on reported fish captures, advertising, and spearfisher commentary and used regression models and ordination analyses to assess historical change. The proportion of coastal fish captures reported declined approximately 80%, whereas the proportion of coral reef and pelagic fish reports increased 1750% and 560%, respectively. Catch composition shifted markedly from coastal temperate or subtropical fishes during the 1950s to 1970s to coral reef and pelagic species in the 1990s to 2000s. Advertising data and commentary by spearfishers indicated that pelagic fish species became desired targets. The mean weight of trophy coral reef fishes also declined significantly over the study period (from approximately 30-8 kg). Recreational fishing presents a highly dynamic social-ecological interface and a challenge for management. Our results emphasize the need for regulatory agencies to work closely with recreational fishing bodies to observe fisher behavior, detect shifts in target species or fishing intensity, and adapt regulatory measures.

Marine ecosystems are in decline. New transformational changes in governance are urgently required to cope with overfishing, pollution, global changes, and other drivers of degradation. Here we explore social, political, and ecological aspects of a transformation in governance of Chile's coastal marine resources, from 1980 to today. Critical elements in the initial preparatory phase of the transformation were (i) recognition of the depletion of resource stocks, (ii) scientific knowledge on the ecology and resilience of targeted species and their role in ecosystem dynamics, and (iii) demonstration-scale experimental trials, building on smaller-scale scientific experiments, which identified new management pathways. The trials improved cooperation among scientists and fishers, integrating knowledge and establishing trust. Political turbulence and resource stock collapse provided a window of opportunity that triggered the transformation, supported by new enabling legislation. Essential elements to navigate this transformation were the ability to network knowledge from the local level to influence the decision-making processes at the national level, and a preexisting social network of fishers that provided political leverage through a national confederation of artisanal fishing collectives. The resultant governance scheme includes a revolutionary national system of marine tenure that allocates user rights and responsibilities to fisher collectives. Although fine tuning is necessary to build resilience of this new regime, this transformation has improved the sustainability of the interconnected social-ecological system. Our analysis of how this transformation unfolded provides insights into how the Chilean system could be further developed and identifies generalized pathways for improved governance of marine resources around the world.

Leucine rich repeat proteins have gained considerable interest as therapeutic targets due to their expression and biological activity within the central nervous system. LINGO-1 has received particular attention since it inhibits axonal regeneration after spinal cord injury in a RhoA dependent manner while inhibiting leucine rich repeat and immunoglobulin-like domain-containing protein 1 (LINGO-1) disinhibits neuron outgrowth. Furthermore, LINGO-1 suppresses oligodendrocyte precursor cell maturation and myelin production. Inhibiting the action of LINGO-1 encourages remyelination both in vitro and in vivo. Accordingly, LINGO-1 antagonists show promise as therapies for demyelinating diseases. An analogous protein to LINGO-1, amphoterin-induced gene and open reading frame-3 (AMIGO3), exerts the same inhibitory effect on the axonal outgrowth of central nervous system neurons, as well as interacting with the same receptors as LINGO-1. However, AMIGO3 is upregulated more rapidly after spinal cord injury than LINGO-1. We speculate that AMIGO3 has a similar inhibitory effect on oligodendrocyte precursor cell maturation and myelin production as with axogenesis. Therefore, inhibiting AMIGO3 will likely encourage central nervous system axonal regeneration as well as the production of myelin from local oligodendrocyte precursor cell, thus providing a promising therapeutic target and an area for future investigation.

A major difficulty in managing wildlife trade is the reliance on trade data (rather than capture data) to monitor exploitation of wild populations. Collected organisms that die or are rejected before a point of sale often go unreported. For the global marine aquarium trade, identifying the loss of collected fish from rejection, prior to export, is a first step in assessing true collection levels. This study takes a detailed look at fish rejections by buyers before export using the Papua New Guinea marine aquarium fishery as a case study. Utilizing collection invoices detailing the species and quantity of fish (Actinopteri and Elasmobranchii) accepted or rejected by the exporting company it was determined that, over a six month period, 24.2% of the total fish catch reported (n = 13,886) was rejected. Of the ten most collected fish families, rejection frequency was highest for the Apogonidae (54.2%), Chaetodontidae (26.3%), and Acanthuridae (18.2%) and lowest for Labridae (6.6%) and Hemiscylliidae (0.7%). The most frequently cited reasons for rejection were fin damage (45.6% of cases), undersized fish (21.8%), and fish deemed too thin (11.1%). Despite fishers receiving feedback on invoices explaining rejections, there was no improvement in rejection frequencies over time (r = -0.33, P = 0.15) with weekly rejection frequencies being highly inconsistent (range: 2.8% to 79.4%; s = 16.3%). These findings suggest that export/import statistics can greatly underestimate collection for the marine aquarium trade as additional factors such as fisher discards, escapees, post-collection mortalities, and unregulated domestic trade would further contribute to this disparity.

Small-scale fisheries are important to livelihoods and subsistence seafood consumption of millions of fishers. Sea cucumbers are fished worldwide for export to Asia, yet few studies have assessed factors affecting socioeconomics and wellbeing among fishers. We interviewed 476 men and women sea cucumber fishers at multiple villages within multiple locations in Fiji, Kiribati, Tonga and New Caledonia using structured questionnaires. Low rates of subsistence consumption confirmed a primary role of sea cucumbers in income security. Prices of sea cucumbers sold by fishers varied greatly among countries, depending on the species. Gender variation in landing prices could be due to women catching smaller sea cucumbers or because some traders take advantage of them. Dissatisfaction with fishery income was common (44% of fishers), especially for i-Kiribati fishers, male fishers, and fishers experiencing difficulty selling their catch, but was uncorrelated with sale prices. Income dissatisfaction worsened with age. The number of livelihood activities averaged 2.2-2.5 across countries, and varied significantly among locations. Sea cucumbers were often a primary source of income to fishers, especially in Tonga. Other common livelihood activities were fishing other marine resources, copra production in Kiribati, agriculture in Fiji, and salaried jobs in New Caledonia. Fishing other coastal and coral reef resources was the most common fall-back livelihood option if fishers were forced to exit the fishery. Our data highlight large disparities in subsistence consumption, gender-related price equity, and livelihood diversity among parallel artisanal fisheries. Improvement of supply chains in dispersed small-scale fisheries appears as a critical need for enhancing income and wellbeing of fishers. Strong evidence for co-dependence among small-scale fisheries, through fall-back livelihood preferences of fishers, suggests that resource managers must mitigate concomitant effects on other fisheries when considering fishery closures. That is likely to depend on livelihood diversification programs to take pressure off co-dependent fisheries.

Over 1.3 billion people live on tropical coasts, primarily in developing countries. Many depend on adjacent coastal seas for food, and livelihoods. We show how trends in demography and in several local and global anthropogenic stressors are progressively degrading capacity of coastal waters to sustain these people. Far more effective approaches to environmental management are needed if the loss in provision of ecosystem goods and services is to be stemmed. We propose expanded use of marine spatial planning as a framework for more effective, pragmatic management based on ocean zones to accommodate conflicting uses. This would force the holistic, regional-scale reconciliation of food security, livelihoods, and conservation that is needed. Transforming how countries manage coastal resources will require major change in policy and politics, implemented with sufficient flexibility to accommodate societal variations. Achieving this change is a major challenge - one that affects the lives of one fifth of humanity.

Inhabitants of Lihir Island, Papua New Guinea, have traditionally relied on reef fishing and rotational farming of slash-burn forest plots for a subsistence diet. However, a new gold mine has introduced a cash economy to the island's socioeconomic system and impacted the fringing coral reef through sedimentation from the near-shore dumping of mine wastes. Studies of the Lihirian people have documented changes in population size, local customs, health, education, and land use; studies of the reef have documented impacts to fish populations in mine affected sites. Indirect effects from these impacts are complex and indecipherable when viewed only from isolated studies. Here, we use qualitative modelling to synthesize the social and biological research programs in order to understand the interaction of the human and ecological systems. Initial modelling results appear to be consistent with differences in fish and macroalgae populations in sites with and without coral degradation due to sedimentation. A greater cash flow from mine expansion is predicted to increase the human population, the intensity of the artisanal fishery, and the rate of sewage production and land clearing. Modelling results are being used to guide ongoing research projects, such as monitoring fish populations and artisanal catch and patterns and intensity of land clearing.
