RESUMEN

The present study aimed to investigate the long-term effect of feeding barley grain steeped in lactic acid (La) with or without thermal treatment on reticuloruminal pH dynamics and metabolic activity of the liver in 12 primiparous and 18 multiparous early-lactating dairy cows. All cows were included on d 21 postpartum and sampled until d 90 postpartum. Cows were fed a diet based on differently processed ground barley grain: untreated grain (control diet, CON), or grain treated with 1% La alone for 24 h before feeding (La), or with an additional oven-heating at 55°C for 12 h (LaH). The reticuloruminal pH and temperature were measured via indwelling sensors that allowed for continuous (every 10min) and long-term measurement from d 21 to 80 postpartum. Blood samples were taken on d 21, 40, and 90 of lactation and analyzed for liver enzymes aspartate aminotransferase (AST), gamma-glutamyltransferase, and glutamate dehydrogenase, as well as bilirubin, bile acids, and serum amyloid A. Dry matter intake was higher in multiparous cows (20.7±0.27 kg/d) compared with primiparous cows (18.2±0.33 kg/d), but was not affected by dietary treatment. Overall, the relatively short duration (51±5min/d) of reticuloruminal pH <5.8 suggests low risk of subacute ruminal acidosis throughout the experiment. Results indicated that La treatment of barley, with or without heat, lowered the time duration of pH <5.8 compared with CON, but only in primiparous cows (from 118±13 to 46±11 and 25±11min/d for CON, La, and LaH, respectively). In multiparous cows, the opposite effect of feeding the La-treated barley on time duration of pH <5.8 (11±8 vs. 46±9 vs. 57±9min/d for CON, La, and LaH, respectively) was observed. Multiparous cows generally showed higher pH readings and shorter periods in which the ruminal pH dropped below the threshold of pH 5.8. The reticuloruminal temperature was not affected by dietary treatment, whereas parity affected the time duration of reticuloruminal temperature >39.5°C, being 60±19min/d shorter in primiparous cows. The measured activities of the liver enzymes AST, gamma-glutamyltransferase, and glutamate dehydrogenase, as well as bilirubin, bile acids, and the acute phase protein serum amyloid A, were not affected by grain feeding. Additionally, only one small effect of parity on investigated serum variables was noticed, showing slightly but significantly higher values of AST in multiparous (80.5±1.4 U/L) compared with primiparous cows (76.0±1.7 U/L). In conclusion, our results indicate greater risk for primiparous cows to develop subacute ruminal acidosis-like conditions during early lactation than multiparous cows. The study also suggests limited benefits of feeding processed barley grain with La with or without thermal treatment to modulate ruminal tolerance of grain feeding, whereby differing effects in primiparous cows were observed compared with multiparous cows.

This study was performed to investigate the effect of the transition from barn feeding to pasture on the pattern of reticuloruminal pH values in 8 multiparous dairy cows. A indwelling wireless data transmitting system for pH measurement was given to 8 multiparous cows orally. Reticuloruminal pH values were measured every 600 s over a period of 42 days. After 7 days of barn feeding (period 1), all of the animals were pastured with increasing grazing times from 2 to 7 h/day over 7 days (period 2). From day 15 to day 21 (period 3), the cows spent 7 h/day on pasture. Beginning on day 22, the animals had 20 h/day access to pasture (day and night grazing). To study reticuloruminal adaptation to pasture feeding, the phase of day and night grazing was subdivided into another 3 weekly periods (periods 4-6). Despite a mild transition period from barn feeding to pasture, significant effects on reticuloruminal pH values were observed. During barn feeding, the mean reticuloruminal pH value for all of the cows was 6.44 ± 0.14, and the pH values decreased significantly (p < 0.001) during period 2 and 3 to 6.24 ± 0.17 and 6.21 ± 0.19 respectively. During periods 4, 5 and 6, the reticuloruminal pH values increased again (pH 6.25 ± 0.22; pH 6.31 ± 0.17; pH 6.37 ± 0.16). Our results showed that the animals had significantly lowered reticuloruminal pH during the periods of feed transition from barn to pasture feeding. Despite these significant changes, the decrease was not harmful, as indicated by data of feed intake and milk production.

Metabolic disorders, especially hyperketonemia, are very common in dairy sheep. The whole-blood concentrations of ß-hydroxybutyrate (BHBA) and glucose can be determined by commercially available electronic hand-held devices, which are used in human medicine and for the detection of ketosis in dairy cows. The aim of this study was to evaluate the suitability of the hand-held device Precision Xceed (PX; Abbott Diabetes Care Inc., Abbott Park, IL) to detect hyperketonemia in ewes. An additional objective of this study was to evaluate the agreement between samples obtained by minimal invasive venipuncture of an ear vein and measurements of whole-blood samples from the jugular vein (vena jugularis, v. jug.). Blood samples taken from the v. jug. were collected from 358 ewes on 4 different farms. These samples and a blood drop obtained from an ear vein were analyzed simultaneously on farm with the PX. For method comparison, the samples obtained from the v. jug. were also analyzed by standard methods, which served as the gold standard at the Central Laboratory of the University of Veterinary Medicine Vienna, Austria. The correlation coefficients between the serum BHBA concentration and the concentrations measured with the hand-held meter in the whole blood from an ear vein and the v. jug. were 0.94 and 0.96, respectively. The correlation coefficients of plasma and whole-blood glucose concentration were 0.68 for the v. jug. and 0.47 for the ear vein. The mean glucose concentration was significantly lower in animals classified as hyperketonemic (BHBA ≥ 1.6 mmol/L) compared with healthy ewes. Whole-blood concentrations of BHBA and glucose measured with the PX from v. jug. showed a constant negative bias of 0.15 mmol/L and 8.4 mg/dL, respectively. Hence, a receiver operating characteristic analysis was performed to determine thresholds for the PX to detect hyperketonemia in ewes. This resulted in thresholds for moderate ketosis of BHBA concentrations of 0.7 mmol/L in blood from an ear vein and the v. jug. Cutoffs of 1.0 mmol/L (ear vein) and 1.1 mmol/L (v. jug.) BHBA were determined to detect animals at greater risk to develop severe hyperketonemia. Applying these thresholds, excellent test characteristics, with sensitivities of 1.00 for both samples and specificities of 0.98 for the ear vein and 0.97 for the v. jug. were determined. These results demonstrate that the PX is a useful tool for detection of hyperketonemia in ewes.

The objective of this study was to evaluate the suitability of the electronic handheld devices FreeStyle Precision (FSP; Abbott Germany, Wiesbaden, Germany) and GlucoMen LX Plus (GML; A. Menarini GmbH, Vienna, Austria) for the measurement of ß-hydroxybutyrate (BHBA) in whole blood in dairy goats. Additionally, glucose concentration was analyzed with the FSP device. For method comparison, the samples were also analyzed in the laboratory by standard methods, which served as the gold standard in our study. A further objective was to evaluate the agreement between samples obtained by minimal invasive venipuncture of an ear vein and measurements of whole blood samples from the jugular vein (vena jugularis). In total, 173 blood sample pairs collected from 28 goats were obtained from an ear vein and from the jugular vein. The Spearman correlation coefficients (rsp) for BHBA concentrations determined with the FSP or GML and the gold standard were 0.95 and 0.85 for the ear vein and 0.98 and 0.88 for the jugular vein, respectively. Bland-Altman plots of differences showed a positive bias of 0.12 (ear vein) and 0.21 (jugular vein) when determination was performed with the FSP and a negative bias of 0.21 (ear vein) and 0.24 (jugular vein) when using the GML. For the FSP, applying the adjusted thresholds determined by ROC analysis of 0.9 (ear vein) and 1.0 mmol/L (jugular vein) allowed to distinguish between healthy goats and animals with hyperketonemia with sensitivities (Se) and specificities (Sp) for samples from the ear vein of 0.98 and 0.85, and from the jugular vein of 0.99 and 0.94, respectively. For the GML, adjusted thresholds were 0.5 mmol/L for the ear vein (Se=0.94, Sp=0.75) and 0.6 mmol/L for the jugular vein (Se=0.88, Sp=0.91). Repeated analyses of defined whole blood samples showed average inter- and intraassay coefficients of variation of 6.6 and 7.3% for FSP, and of 35.6 and 35.4% for GML, respectively. Test characteristics for determining glucose concentration with the FSP compared with the gold standard were poor (ear vein: rsp=0.41; jugular vein: rsp=0.51), with low validity to distinguish between hypo- and normoglycemia (Se=0.71, Sp=0.66). The present study showed good test characteristics for the FSP and moderate for the GML device for determining BHBA concentrations in whole blood. Additionally the results demonstrate the suitability of testing BHBA concentration in a blood drop obtained from an ear vein. Based on the results of this study, the FSP device is not suitable to differentiate normo- from hypoglycemia in goats.

Twelve cows (mean age 6.4 years) and eight sheep (mean age 0.8 years) were fed silage containing 70 to 90 per cent vitamin D3-effective Trisetum flavescens for a period of 14 weeks. At the beginning and the end of this period, the animals were examined and their kidneys, abdominal aortas and heart valves were examined by ultrasonography; the animals were then slaughtered and these organs were examined histologically, and the specificity and sensitivity of the ultrasonography were calculated. After the feeding period, all the cows had a decreased body temperature and all but one had raised hair, movement disorders and difficulty in rising and lying down. Ultrasonography of the abdominal aorta had a specificity of 90 per cent and a sensitivity of 75 per cent, and ultrasonography of the heart valves had a specificity of 100 per cent and a sensitivity of 50 per cent. In the sheep, cardiac arrythmia was the only pathological finding at the end of the feeding period, and the ultrasonographic examination of the kidneys had a specificity and sensitivity of 100 per cent.

The objective of the study was to compare the performance of five types of stomach tubes for rumen fluid sampling. Rumen fluid was sampled in rumen fistulated cows assigned to a 5×5 Latin square study design. The pH values of samples taken by stomach tubes and via fistulas were measured; the results were compared with indwelling sensor measurements. The practicability of the stomach tubes for regular use was tested in the field. Rumen fluid samples were obtained rapidly. Volumes for transfaunation could be obtained. The pH-values of samples taken with the four out of the five tubes (Dirksen, Geishauser, tube 4 and a simple water hose used with a gag) did not show significant differences to samples taken via rumen fistulas. Mean differences ranged between -0.02 and +0.09. Samples taken with tube 4 and the water hose showed also no significant differences to pH-sensor measurements. This study demonstrates that stomach tubes are suitable for rumen fluid sampling. Tube 4 seems to be the best probe for work in the field. It was well tolerated by the animals, saliva contamination is negligible. We, therefore, conclude that the evaluation of rumen acid base status in the field is possible.

Serum samples from healthy, infected (n=11) and diseased (n=2) cattle as well as positive (n=17) and negative (n=41) reference sera were tested for antibodies to Mycobacterium avium subsp. paratuberculosis with two ELISA-methods (A-ELISA, Allied Monitors, Fayette, USA; H-ELISA, Institute of Microbiology and Animal Diseases, Veterinary University Hannover). Fecal samples of these animals were examined by PCR and culture. Also field serum samples found to be positive (n=664) or inconclusive (n=1589) by A-ELISA during a survey done on 11028 cattle of 2757 farms at different districts in Austria were retested with H-ELISA (Gasteiner et al., 1999). In both ELISA-methods total agreement between antibody detection and shedding of M. avium subsp. paratuberculosis (PCR, culture) in cases of diseased animals during the testing period was found. In subclinically infected animals H-ELISA showed a better correlation with the results of PCR and fecal culture. Reference serum samples of culturally negative cattle were negative in 98% by H-ELISA and in 82% by A-ELISA, and those of positive animals were positive in 59% by H-ELISA and in 82% by A-ELISA. The 664 A-ELISA positive field serum samples were positive in 20.5%, inconclusive in 32.5% and negative in 47% by H-ELISA. A-ELISA inconclusive sera gave positive reactions by H-ELISA in 5.2%, negative in 74.8% and inconclusive results in 20%. The highest prevalence of antibodies (7.9% by A- and 2.2% by H-ELISA) against M. avium subsp. paratuberculosis were found in cattle at the age of six and seven years. Seropositive animals were found at all tested ages. The A-ELISA gave two to three times more positive reactors than the H-ELISA. Also both tests showed the highest prevalence of reagents among Holstein Friesian (6.2% by A-ELISA, 2.5% by H-ELISA) followed by other cattle breeds. Seropositive cattle were observed in all districts of Austria in 3. 3-7.1% and in 0.5-1.8% of herds according to A- and H-ELISA, respectively.

RESUMEN

Atrophic rhinitis was diagnosed in a 10-month-old heifer with the assistance of computed tomography. The aetiology of the condition in cattle is uncertain and the clinical disease may be overlooked owing to the lack of signs such as the shortening or deformity of the snout observed in pigs.

The aim of the present study was the continuous measurement of ruminal pH in grazing dairy cows to monitor the diets effects on ruminal pH value. A novel indwelling pH-measurement and data transmitting system was given to 6 multiparous cows orally. Ruminal pH was measured every 600 sec over a 40 d period. After barn feeding and changeover to pasture, the following 3 treatments (2 cows/treatment) were included in the measurement period: continuous grazing (G), continuous grazing plus 4 kg/d of hay fed twice daily (GH), and continuous grazing plus 4 kg/d of concentrate (GC). Ruminal pH decreased significantly (P < 0.05) from 6.58 ± 0.15 to pH 6.19 ± 0.19 during feed changeover to pasture. Mean ruminal pH for G, GH, and GC was 6.36, 6.56, and 6.01. Mean 24-h minimum pH was 5.95, 6.20 and, 5.58. The time pH was below 6.3, 6.0, 5.8, and 5.5, for G it was 583, 91, 26, and 3 min/d, for GH it was 97, 12, 0, and 0 min/d and for GC it was 1126, 621, 347, and 101 min/d, respectively. Results were significantly influenced by the diet. The indwelling pH-measurement and data transmitting system is a very useful and proper tool for long-term measurement of ruminal pH in cows.

O objetivo do presente experimento foi medir continuamente valores de pH e temperatura em vacas leiteiras usando uma unidade interna de transmissão de dados sem fio. Valores de pH retículo-ruminais foram medidos automaticamente a cada 600 segundos por um período de 50 dias em três vacas leiteiras Holandesas, no pós-parto recente. Valores de pH retículo-ruminais médios diferiram (P<0,05) entre as três vacas (5,69±0,20; 6,10±0,18; 5,99±0,15), assim como o tempo em minutos por dia (332; 23; 18) mantido abaixo de pH 5,5. A variação diurna de pH nas vacas 2 e 3 demonstrou um padrão circadiano e frequente, como consequência dos momentos de fornecimento da alimentação e da ingestão alimentar, respectivamente. Esse padrão diário não pode ser observado no padrão de pH da vaca 1. Os picos e os valores baixos de pH na vaca 1 eram aleatórios, sem relação evidente com os momentos de alimentação, e as amplitudes de pH eram igualmente desordenadas. O valor de pH retículo-ruminal permaneceu anormalmente baixo nesta vaca durante todo o período de observação, caracterizando uma acidose ruminal subaguda. A temperatura retículo-ruminal da vaca 1 foi mais baixa (38.8°C; 39.1°C; 39.0°C) e ela bebeu mais frequentemente por dia (9,5; 6,4; 7,0) quando comparada com as vacas 2 e 3 (P<0,05). O exame clínico revelou um deslocamento de abomaso à esquerda (DAE). Pela literatura consultada, este é o primeiro relato indicando um padrão de pH e temperatura em uma vaca com DAE.(AU)

From 1995 to 1997, the prevalence of serum antibodies against Mycobacterium avium subspecies paratuberculosis (M. av. ssp. ptbc.)--the causal agent of paratuberculosis (Johne's Disease)--was examined in 11,028 Austrian cattle. Samples from the four oldest cattle on 2757 farms were collected according to a specific sampling schedule for this epidemiological study. District, age and breed of animals were included as variables in this study. For antibody screening against M. avium subspecies paratuberculosis, a modified, commercially available ELISA (ALLIED Monitors, Fayette, USA) was employed. A total of 2253 samples that were found to be positive or questionable were subjected to further testing with a more specific ELISA (Institute of Microbiology and Infectious Animal Diseases). Results of this study were used for statistical analysis. The average prevalence of antibodies to M. avium subspecies paratuberculosis was 1.99% in Austria. The highest prevalence was seen in 6-year-old cattle (2.84%) and Holstein Frisian cattle (3.51%). Sero-positive animals were found on 6.96% of farms tested, and the prevalence was highest in Vorarlberg, followed by Salzburg, the Tyrol, Styria and Carinthia. This study is unique in Europe in the use of an adequate random sampling plan for an investigation of this magnitude.
