RESUMEN

BACKGROUND: Due to increasing preclinical study using the rhesus monkeys, the physiological data of these monkeys are very important for the objective evaluation of experimental results. Here, we report the physiological values of the Chinese rhesus monkeys acclimated under a well-controlled laboratory environment. METHODS: Seventy healthy rhesus monkeys of both genders (29 males and 41 females) were used in this study. Available data on hematology, serum biochemistry, electrolytes, blood gas, coagulation time, urinalysis, water consumption, urine volume, and body weight were examined. RESULTS: The lymphocyte values were 1.5-2.5 times higher than the neutrophil values in both genders. In serum biochemistry, there was no significant difference between the two genders. Interestingly, the values of alkaline phosphatase from the monkeys were very high. According to isoenzyme analysis, the percentage of alkaline phosphatase that originated from the bone was 60%, and this value was significantly higher than that from the liver (39%, P = 0.020). CONCLUSION: Physiological data are very important parameters that can be directly and indirectly related to organ function (e.g. islet, kidney, liver, heart, etc.) in the transplantation model. In this respect, our data constitute valuable references for preclinical study using the Chinese rhesus monkeys.

RESUMEN

BACKGROUND: Vascular endothelial growth factor (VEGF) has been demonstrated to play an important role in tumor angiogenesis and to influence prognosis in many cancers. But, its significance in salivary gland carcinomas has not been elucidated. The authors investigated the association between VEGF expression and clinicopathological factors, p53, and Ki-67 to verify its validity as a prognostic factor. METHODS: Surgical specimens from 45 patients with salivary gland carcinoma were examined for VEGF, p53, and Ki-67 expression by immunohistochemical staining. The results were compared with the clinicopathological factors and the relationships were correlated. RESULTS: VEGF expression was low in 14 cases, moderate in 15 cases, and high in 16 cases. It was significantly correlated with a variety of clinicopathological factors such as TNM stage, perineural and vascular invasion, and recurrence. VEGF showed significant association with the expression of p53 but not with that of Ki-67. Univariate analysis showed that age, gender, lymph node metastasis, vascular invasion, p53, Ki-67, and VEGF expression correlate with prognosis. Multivariate analysis demonstrated that VEGF is an independent prognostic factor for patients with salivary gland carcinomas. CONCLUSIONS: The results of this study suggest that VEGF expression is correlated with p53 expression and that it may have prognostic value in salivary gland carcinomas.

RESUMEN

We have identified a novel chordin-like protein, CHL2, which is structurally most homologous to CHL/neuralin/ventroptin. When injected into Xenopus embryos, CHL2 RNA induced a secondary axis. Recombinant CHL2 protein interacted directly with BMPs in a competitive manner to prevent binding to the type I BMP receptor ectodomain, and inhibited BMP-dependent induction of alkaline phosphatase in C2C12 cells. Thus, CHL2 behaves as a secreted BMP-binding inhibitor. In situ hybridization revealed that CHL2 expression is restricted to chondrocytes of various developing joint cartilage surfaces and connective tissues in reproductive organs. Adult mesenchymal progenitor cells expressed CHL2, and its levels decreased during chondrogenic differentiation. Addition of CHL2 protein to a chondrogenic culture system reduced cartilage matrix deposition. Consistently, CHL2 transcripts were weakly detected in normal adult joint cartilage. However, CHL2 expression was upregulated in middle zone chondrocytes in osteoarthritic joint cartilage (where hypertrophic markers are induced). CHL2 depressed chondrocyte mineralization when added during the hypertrophic differentiation of cultured hyaline cartilage particles. Thus, CHL2 may play negative roles in the (re)generation and maturation of articular chondrocytes in the hyaline cartilage of both developing and degenerated joints.
