RESUMEN

The functional capacity of organisms declines in the process of aging. In the case of breast tissue, abnormal mammary gland development can lead to dysfunction in milk secretion, a primary function, as well as the onset of various diseases, such as breast cancer. In the process of aging, the terminal duct lobular units (TDLUs) within the breast undergo gradual degeneration, while the proportion of adipose tissue in the breast continues to increase and hormonal levels in the breast change accordingly. Here, we review changes in morphology, internal structure, and cellular composition that occur in the mammary gland during aging. We also explore the emerging mechanisms of breast aging and the relationship between changes during aging and breast-related diseases, as well as potential interventions for delaying mammary gland aging and preventing breast disease.

The impact of a liquid drop on a granular medium is a common phenomenon in nature and engineering. The possible splashing droplets and ejected particles could pose a risk of pathogen transmission if the water source or granular medium is contaminated. This work studies the liquid drop impact on the granular medium using high-speed photography and considers the effects of liquid properties, drop impact characteristics, and granular medium properties. Four flow regimes, including direct penetration, prompt splashing, spreading, and corona splashing, are observed and a regime map is created to identify their thresholds. The spreading regime can eject a large number of particles, and the corona splashing regime can produce splashing droplets in addition to the ejected particles. For the splashing droplets, their median diameters and velocities are in the ranges 0.11 to 0.21 and 0.15 to 0.37 of the diameter and velocity of the impact drop, and their median splashing angles range from 14° to 27°. Two particle ejection mechanisms are observed, falling squeeze and forward collision, driven by the collapsing and forward spreading of the liquid lamella, respectively. The particles ejected by the latter mechanism have larger ejection velocities, angles and distances from the impact center, which can facilitate their long-range transmission. In addition, the process of spreading and retracting of the lamella formed by the drop impact is also studied, and it is found that the maximum spreading diameter of the lamella is proportional to the crater diameter. These results improve the understanding of the phenomenon after the drop impact on the granular medium and the characteristics of the splashing droplets and ejected particles, contributing to the prediction and risk assessment of contaminated particle transmission.

Breast cancer in the elderly presents distinct biological characteristics and clinical treatment responses compared with cancer in younger patients. Comprehensive Geriatric Assessment is recommended for evaluating treatment efficacy in elderly cancer patients based on physiological classification. However, research on molecular classification in older cancer patients remains insufficient. In this study, we identified two subgroups with distinct senescent clusters among geriatric breast cancer patients through multi-omics analysis. Using various machine learning algorithms, we developed a comprehensive scoring model called "Sene_Signature," which more accurately distinguished elderly breast cancer patients compared with existing methods and better predicted their prognosis. The Sene_Signature was correlated with tumor immune cell infiltration, as supported by single-cell transcriptomics, RNA sequencing, and pathological data. Furthermore, we observed increased drug responsiveness in patients with a high Sene_Signature to treatments targeting the epidermal growth factor receptor and cell-cycle pathways. We also established a user-friendly web platform to assist investigators in assessing Sene_Signature scores and predicting treatment responses for elderly breast cancer patients. In conclusion, we developed a novel model for evaluating prognosis and therapeutic responses, providing a potential molecular classification that assists in the pre-treatment assessment of geriatric breast cancer.

As an important drinking water source for North China, the Middle Route of China's South-to-North Water Diversion Project (MRP) must provide high-quality water to maintain the health and safety of more than 60 million people. However, different water transfer operation modes may affect the water quality status, and the spatiotemporal characteristics of water quality in the MRP, with high water transfer volumes, remain poorly understood. In this study, the differences in water quality in the MRP between the initial stage (Nov. 2015 to Oct. 2017, low transfer volumes) and the current stage (Nov. 2017 to Oct. 2020, high transfer volumes) were compared, and the spatiotemporal water quality variations in the current stage were evaluated using multivariate statistical methods. For this purpose, approximately 12,528 observations, including the datasets of 12 water quality parameters collected from 29 monitoring sites, were used. The results showed that the water quality status improved significantly during the current stage. Based on principal component analysis (PCA), physical parameters (natural), nutrients, organic matter and microbes (anthropogenic), and heavy metals (natural and anthropogenic) were the key factors influencing water quality variations. Based on hierarchical cluster analysis, 12 months were classified into two groups: the high-flow period (HFP, Jun.-Oct.) and the low-flow period (LFP, Nov.-May). Additionally, 29 sampling sites were grouped into three sections: the Henan section (HN; S1-S16), Hebei section (HB; S17-S24), and Tianjin-Beijing section (TB; S25-S29). From the perspective of water quality regulation, the total nitrogen concentration and permanganate index in the HB and TB sections of the MRP should be considered throughout the year, and the faecal coliform concentrations in these two sections should also be considered during the HFP. The results of this study could be helpful for local administrations to understand and control pollution and better protect the quality of water in the MRP.
