RESUMEN

Gastric cancer is the leading cause of cancer-related death worldwide. The aim of present study was to investigate the anti-tumor effect of purified Omphalia lapidescens protein (pPeOp) in gastric cancer. Microarray analysis was performed to find out differentially expressed genes in pPeOp-treated MC-4 gastric cancer cells. The Janus kinase (JAK)/signal transducer and activator of transcription (STAT) three signaling pathway was most likely to be altered based on bioinformatics analysis. Interleukin-6 (IL-6) and NSC74859 were used as the agonist and inhibitor of the JAK/STAT3 signaling pathway, respectively. Flow cytometry and MTS assay were used for cell proliferation and viability analysis in pPeOp-treated gastric cancer cell lines with IL-6 or NSC74859. The anti-tumor effect was increased when pPeOp were co-treated with IL-6, while decreased in inhibitor treatment. The expression of the crucial members in the pathway of MC-4 cells, including glycoprotein 130 (GP130), JAK1, JAK2, STAT3, p-STAT3, suppressor of cytokine signaling SOCS1 and SOCS3, was detected by western blotting. pPeOp exhibited promising anticancer effect in the xenograft nude mice model, established by STAT3 knock down gastric cancer cells.Thus, JAK/STAT3 inhibition partially contributed to the anticancer effect of pPeOp, which may serve as a novel strategy for gastric cancer.Supplemental data for this article is available online at https://doi.org/10.1080/01635581.2021.1960385.

Gastric cancer is the fifth most frequently occurring and the fourth most lethal malignant cancer worldwide. A bioactive protein (pPe Op) from Omphalia lapidescens exhibits significant inhibitory effects on gastric cancer cells. miRNA deep sequencing analysis shows that miR-30b-5p is significantly upregulated in HGC-27 cells treated with pPe Op. Verification results show that the expression level of miR-30b-5p is significantly increased in HGC-27 cells after pPe Op treatment. Additionally, miR-30b-5p is significantly downregulated in clinical gastric cancer tissues compared to that in adjacent normal tissues. Following pPe Op treatment and/or transfection with miR-30b-5p mimic, the proliferation, migration, and invasion of HGC-27 cells are significantly impaired. Immunofluorescence microscopy shows that pPe Op and/or miR-30b-5p destroy(s) microfilaments and microstructures and inhibit(s) the formation of pseudopodia. Bioinformatics analysis, dual-luciferase reporter assay, and western blot analysis confirm that miR-30b-5p downregulates Rac1/Cdc42 expression and activation by targeting RAB22A. Available data indicate that miR-30b-5p plays an anti-gastric cancer role in mediating pPe Op. pPe Op upregulates miR-30b-5p expression, which in turn inhibits RAB22A expression, resulting in a reduction in the expression and activation of Rac1 and Cdc42 and their downstream targets, thus destroying the cytoskeletal structure and inhibiting the proliferation, migration, and invasion of cancer cells.

RESUMEN

We investigated 16 Japanese spotted fever cases that occurred in southeastern China during September-October 2015. Patients had fever, rash, eschar, and lymphadenopathy. We confirmed 9 diagnoses and obtained 2 isolates with high identity to Rickettsia japonica strain YH. R. japonica infection should be considered for febrile patients in China.

We established a deep learning-based dynamic light scattering (DLS) microscopy sensing mitochondria dynamic for label-free identification of triple-negative breast cancer (TNBC) cells. The capacity of DLS microscopy to detect the intracellular motility of subcellular scatters was verified with the analysis of the autocorrelation function. We also conducted an in-depth examination of the impact of mitochondrial dynamics on DLS within TNBC cells, employing confocal fluorescent imaging to visualize the morphology of the mitochondria. Furthermore, we applied the DLS microscopy incorporating the two-stream deep learning method to differentiate the TNBC subtype and HER2 positive breast cancer subtype, with the classification accuracy achieving 0.89.

Rehabilitation is essential for post-stroke body function recovery. Supported by the mirror neuron theory, motor imagery (MI) has been proposed as a potential stroke therapy capable of facilitating the rehabilitation. However, it is often quite difficult to estimate the degree of the participation of patients during traditional MI training as well as difficult to evaluate the efficacy of MI based rehabilitation methods. The goal of this paper is to develop a virtual reality (VR) based MI training system combining electromyography (EMG) based real-time feedback for poststroke rehabilitation, with the immersive scenario of the VR system providing a shooting basketball training for bilateral upper limbs. Through acquiring electroencephalography (EEG) signal, the brain activity in alpha and beta frequency bands was mapped and the correlation analysis could be achieved. Furthermore, EMG data of each patient was collected and calculated as threshold with root-mean-square algorithm for feedback of the performance score of the shooting basketball training in virtual environment. To investigate the feasibility of this newly-built rehabilitation training system, four experiments namely initial assessment experiment, motor imagery (MI), action observation (AO), and combined motor imagery and action observation (MI+AO) were carried out on stroke patients at different recovery stages. The result shows that MI+AO can generate more pronounced event-related desynchronization (ERD) in alpha band compared to other cases and induce relatively obvious ERD in beta band compared to AO, which demonstrates that VR-based observation has ability to facilitate MI training. Furthermore, it has been found that the muscle strength from MI+AO is the highest through the EMG analysis. This proves that the feedback of EMG can be used to quantify patient's training engagement and promote MI training at a certain extent. Hence, by incorporating such an EMG feedback, a VR-based MI training system has the potential to achieve higher efficacy for post-stroke rehabilitation.

Droplet-based microfluidics with the characteristics of high throughput, low sample consumption, increasing reaction speed, and homogeneous volume control have been demonstrated as a useful platform for biomedical research and applications. The traditional fabrication methods of droplet microfluidics largely rely on expensive instruments, sophisticated operations, and even the requirement of an ultraclean room. In this manuscript, we present a 3D printing-based droplet microfluidic system with a specifically designed microstructure for droplet generation aimed at developing a more accessible and cost-effective method. The performance of droplet generation and the encapsulation capacity of the setup were examined. The device was further applied to measure the variation in cell viability over time and monitor the cell's blebbing activity to investigate its potential ability and feasibility for single-cell analysis. The result demonstrated that the produced droplets remained stable enough to enable the long-time detection of cell viability. Additionally, cell membrane protrusions featuring the life cycle of bleb initiation, expansion, and retraction can be well-observed. Three-dimensional printing-based droplet microfluidics benefit from the ease of manufacture, which is expected to simplify the fabrication of microfluidics and expand the application of the droplet approach in biomedical fields.

miR-30b, which is encoded by the gene located on chromosome 8q24.22, plays an important role in a variety of diseases. In most types of tumors, miR-30b significantly inhibits the proliferation, migration, and invasion of cancer cells through the regulation of target genes. Moreover, miR-30b can inhibit the PI3K/AKT signaling pathway through the regulation of EGFR, AKT, Derlin-1, GNA13, SIX1, and other target genes, thus inhibiting the EMT process of tumor cells and promoting apoptosis. In addition, miR-30 plays a significant role in alleviating drug resistance in tumor cells. Although the use of miR-30b as a clinical diagnostic indicator or anticancer drug is still facing great difficulties in the short term, with the deepening of research, the potential application of miR-30b is emerging.

Motor imagery combining virtual reality (VR) technique has recently been reported to have an increasingly positive impact on post-stroke rehabilitation. However, there is a common problem that the engagement of patients cannot be confirmed during motor imagery training due to a lack of effective feedback control. This paper proposes a VR-based motor imagery training system for post-stroke rehabilitation, using surface electromyographic (EMG)-based real-time feedback to enable the personalized training and quantitative assessment of participation degree. Three different experiments including assessment experiment, action observation (AO), combined motor imagery and action observation (MI+AO) experiment were performed on 4 post-stroke patients to verify the system. The immersive scenario of the VR system provides a shooting basketball training for bilateral upper limbs. The EMG data of assessment of each participant was collected to calculate the thresholds, which was utilized in the subsequent experiments based on real-time feedback of EMG. The result reveals significant differences of the muscle strength between AO and MI+AO experiments. This demonstrates that the EMG-based feedback is effective to be of use in assessment of participation degree. The primary application shows that VR-assisted motor imagery system has potential to provide personalized and more engaged training for post-stroke rehabilitation.

OBJECTIVES: The study aimed to examine the anti-diabetic effects of Gynura divaricata (GD) and the underlying mechanism. METHODS: Information about the chemical compositions of GD was obtained from extensive literature reports. Potential target genes were predicted using PharmMapper and analyzed using Kyoto Encyclopedia of Genes and Genomes (KEGG) and Gene Ontology (GO). To validate the results from bioinformatics analyses, an aqueous extract of GD was administered to type 2 diabetic rats established by feeding a high-fat and high-sugar diet followed by STZ injection. Key proteins of the PI3K/AKT signaling pathway and fatty acid metabolism signaling pathway were investigated by immunoblotting. RESULTS: The blood glucose of the rats in the GD treatment group was significantly reduced compared with the model group without treatment. GD also showed activities in reducing the levels of alanine aminotransferase (ALT), aspartate aminotransferase (AST), blood urea nitrogen (BUN), and creatinine (CREA). The levels of urine sugar (U-GLU) and urine creatinine (U-CREA) were also lowered after treatment with GD. Bioinformatics analysis showed that some pathways including metabolic pathways, insulin resistance, insulin signaling pathway, PPAR signaling pathway, bile secretion, purine metabolism, etc. may be regulated by GD. Furthermore, GD significantly increased the protein expression levels of PKM1/2, p-AKT, PI3K p85, and GLUT4 in the rat liver. In addition, the expression levels of key proteins in the fatty acid metabolism signaling pathway including AMPK, p-AMPK, PPARα, and CPT1α were significantly upregulated. The anti-apoptotic protein BCL-2/BAX expression ratio in rats was significantly upregulated after GD intervention. These results were consistent with the bioinformatics analysis results. CONCLUSIONS: Our study suggests that GD can exert hypoglycemic effects in vivo by regulating the genes at the key nodes of the PI3K/AKT signaling pathway and fatty acid metabolism signaling pathway.

OBJECTIVE: To explore the effect of proteins extraceed from mycelia of Omphalia lapidescens on inhibiting H22 liver cancer in vivo. METHODS: 50 hepatoma 22 tumor bearing mice models were divided into five groups randomly:control group( CG), cyclophosphamide group, and 3 groups of incremental Hepatoma 22 dosages (5, 3, 1 mg/kg). All groups were i.v. with drugs once a day. After 8 consecutive days, the concentrations of interferon-gamma (IFN-gamma) and interleukin-4 (IL-4) in serum were detected by enzyme-linked immunoabsorbent assay (ELISA). The weight changes of tumor, thymus, liver, heart, spleen, lung and kidney were observed. RESULTS: It showed the tumors' weight were significant heavier in CG than in EGs. The tumor-inhibition rate (IR) was 36.4% in high dosage group,which was lower than 43.2% in cyclophosphamide group. The spleen mass of proteins groups increased significantly. The concentration of IFN-gamma in serum of proteins groups increased as CG, but IL-4 in inverse direction. The observations of thymus, liver, heart, lung and kidney in EGs were the same as CG. CONCLUSION: The proteins extracted from mycelium of Omphalia lapidescens can inhibit the growth of tumour and enhance the immune function of H22 tumor-bearing mice.

Label-free microfluidic cytometry is of increasing interest for single cell analysis due to its advantages of high-throughput, miniaturization, as well as noninvasive detection. Here we develop a next generation label-free light-sheet microfluidic cytometer for single cell analysis by two-dimensional (2D) light scattering measurements. Our cytometer integrates light sheet illumination with a disposable hydrodynamic focusing unit, which can achieve 3D hydrodynamic focusing of a sample fluid to a diameter of 19 micrometer without microfabrication. This integration also improves the signal to noise ratio (SNR) for the acquisition of 2D light scattering patterns from label-free cells. Particle sizing with submicron resolution is achieved by our light-sheet flow cytometer, where Euclidean distance-based similarity measures are performed. Label-free, automatic classification of senescent and normal cells is achieved with a high accuracy rate by incorporating our light-sheet flow cytometry with support vector machine (SVM) algorithms. Our light-sheet microfluidic cytometry with a microfabrication-free hydrodynamic focusing unit may find wide applications for automatic and label-free clinical diagnosis.

OBJECTIVE: To study on the relationship between the endophyte and the life cycle of Ophiopogon japonicus in Zhejiang. METHODS: Sample roots of Ophiopogon japonicus at different growth stages were thoroughly washed and cut into small fragments, then cleared (removing cytoplasmic contents from cells) using hot 10% KOH and stained with acid fuchsin (alternative stain). The hyphae, the arbuscular and the vesicular of endophyte were examined. RESULTS: The hyphae appeared and grew in the seedling stage, the hyphae grew into arbuscular in the root tuber generating stage and vesicular in the stage of root tuber expanding period. CONCLUSION: The endophytes in Ophiopogon japonicus appeared in forms of hyphae, arbuscular and vesicular at different growth stages to meet the needs of Ophiopogon japonicus developing.

OBJECTIVE: To study the fungi isolated from Cordyceps sobolifera and its fermentation culture. METHODS: The fungi was isolated and identified by its hypha and spores. Three liquid media were used in the culture. RESULTS: Pure culture was gained and the fungi was identified to be Paecilomyces cicadae. The fungi can grow best in liquid media: egg 110 g + silkworm powder 30 g + VB1 2 tahlets + MgSO4 x 7H2O 0.5 g + K2HPO4 1 g + H2O 1000 ml, in which every litre can produce 135 g wet hypha after cultured 4 d. CONCLUSION: Paecilomyces cicadae from Cordyceps sobolifera can be cultured in liquid media.

A light-sheet-based 2D light scattering cytometer is developed for label-free characterization of senescent cells. The light-sheet provides an illumination beam with controlled thickness for single cell excitation, and 2D light scattering patterns are obtained by using a defocused imaging method. The principle of this cytometer is validated by distinguishing microspheres with submicron resolution. Automatic classification of senescent and normal cells is achieved at single cell level by using the support vector machine (SVM) algorithm, where a sensitivity of 89.1% and a specificity of 96.4% are obtained. Our results suggest that the light-sheet-based 2D light scattering label-free cytometry has the capability to perform size differentiation of beads with submicron resolution and to classify different groups of cells without fluorescent labeling, showing the potential for clinical diagnosis of senescence-related diseases.

METHODS: The tender stem segments with roots as explants were cultured on various media with different kinds of hormones, a lot of bud clusters were induced and grew into seedlings. RESULTS: A great quantity of whole aseptic seedlings can be formed quickly by the tender stem segments on the media.

The endophytic bacterium, MD-b1, was isolated from the medicinal plant Ophiopogon japonicas and identified as the Bacillus amyloliquefaciens sp. with 99% similarity based on the partial sequence analysis of 16S rDNA. Exopolysaccharides were extracted from the endophyte for the evaluation of its antitumor activity against gastric carcinoma cell lines (MC-4 and SGC-7901). 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assays and microscopy were performed to estimate the cell viability and morphological changes of the MC-4 and SGC-7901 cells following treatment with the exopolysaccharides at 14, 22 and 30 µg/µl. The results revealed that the exopolysaccharides displayed concentration-dependent inhibitory effects against the MC-4 and SGC-7901 cells, with an IC50 of 19.7 and 26.8 µg/µl, respectively. The exopolysaccharides also induced morphological abnormalities in the cells. These effects indicated the the exopolysaccharides had an antitumoral mechanism of action associated with the mitochondrial dysfunction of the treated cells. This is the first study to investigate the endophytic microorganism isolated from O. japonicas and also the first discovery of such antitumoral exopolysaccharides derived from the genus Bacillus. This provides a promising and reproducible natural product source with high therapeutic value for anticancer treatment, thereby facilitating the development of new anticancer agents.

Omphalia lapidescens is an important medicinal fungus as well as traditional Chinese medicine used for disease treatment. It is mainly used as a vermifuge for anthelmintic therapy, but it has not been hitherto reported to possess antitumor activity. In this study, a purified bioactive protein in O. lapidescens (pPeOp) was obtained using polyvinylpyrrolidone (PVP) followed by gel filtration chromatography. To evaluate the in vitro antitumor activity of pPeOp in human gastric tumor cells (MC-4 and SGC-7901) and normal cells (MC-1), MTT assay and FCM assay were used and the morphological changes, cell viability, cell death rate and cell apoptosis rate of MC-4, SGC-7901 and MC-1 cells were estimated. The results showed that pPeOp could significantly reduce the cell viability of MC-4 and SGC-7901 cells in a concentration-dependent manner, with IC50 values of 236.05 and 156.28 µg/ml, respectively. The morphological observation also indicated a similar result. In FCM assays, a significant increase of cell death rate and cell apoptosis rate of the tumor cells were observed, indicating probable necrosis-inducing effects and/or apoptosis-inducing effects of pPeOp. Importantly, there was no significant effect of pPeOp on MC-1 cells in each assay, showing that pPeOp has no adverse effects on the normal cells. In conclusion, pPeOp is a newly discovered bioactive protein in O. lapidescens and this is the first report on antitumor activity of such a fungal protein. This may provide a meaningful basis for developing a new protein drug for treatment against cancer, especially gastric cancer.
