RESUMEN

Inflammatory bowel disease (IBD) is a non-specific chronic intestinal inflammatory disease, often presenting with abdominal pain, diarrhea, bloody stool, anorexia, and body loss. It is difficult to cure completely and a promising treatment is urgently needed. Natural compounds can offer promising chemical agents for treatment of diseases. Polydatin is a natural ingredient extracted from the dried rhizome of Polygonum cuspidatum, which has anti-inflammatory, anti-tumor, and dementia protection activities. The purpose of this study was to evaluate the therapeutic effect of polydatin on IBD and explore its possible mechanism. We found that polydatin could effectively suppress the differentiation of Th17 cells in vitro, but had no effect on the differentiation of Treg cells. Polydatin significantly alleviated colitis induced by dextran sulfate sodium (DSS) and 2, 4, 6-trinitrobenzenesulfonic acid (TNBS) in mice, and dramatically decreased the proportion of Th17 cells in spleen and mesenteric lymph nodes. Mechanism investigations revealed that polydatin specifically inhibited signal transducer and activator of transcription 3 (STAT3) phosphorylation by directly binding to STAT3, leading to Th17 cell reduction and thereby alleviating colitis. These findings provide novel insights into the anti-colitis effect of polydatin, which may be a promising drug candidate for the treatment of IBD.

Huangqin is the dried root of Scutellaria baicalensis Georgi, which has been widely utilized for heat-clearing (Qingre) and dewetting (Zaoshi), heat-killed (Xiehuo) and detoxifying (Jiedu) in the concept of Traditional Chinese Medicine and is used for treating inflammation and cancer in clinical formulas. Neobaicalein (NEO) is of flavonoid isolated from Huangqin and has been reported to possess prominent anti-inflammatory effects in published work. Th17/Treg balance shift to Th17 cells is an essential reason for autoimmune inflammatory diseases. However, the role NEO plays in Th17 and Treg and the underlying mechanism has not been elucidated yet. Network pharmacology-based study revealed that NEO predominantly regulated IL-17 signaling pathway. Moreover, our result shown that NEO (3-30 µmol/L) down-regulated Th17 differentiation and cellular supernatant and intracellular IL-17A level and tumor necrosis factor α production in a concentration-dependent manner. The further mechanism research revealed that NEO also specifically inhibited phosphorylation of STAT3(Tyr725) and STAT4 (Y693) without influence on activation of STAT5 and STAT6 in splenocytes. Immunofluorescence results illuminated that NEO effectively blocked STAT3 translocated into nucleus. Interestingly, NEO at appreciated dose could only inhibit Th17 cell differentiation and have no effect on Treg differentiation. The present study revealed that NEO effectively inhibited Th17 cell differentiation through specifically blocking the activation of STAT3 signaling without inactivation of STAT5 and STAT6. Additional inhibitory effect on activation of STAT4 by NEO also suggested the potential for antagonism against Th1 differentiation. All work suggested that NEO may be a potential candidate for immunoregulation and treating autoimmune inflammatory diseases through inhibiting immune cell viability and T cell differentiation.

Terracing and rainfall characteristics notably influence the water erosion processes. However, an extensive long term in situ quantitative evaluation of the approaches to control the water erosion in different orchard terraces has not yet been performed, especially considering the increasing frequency and severity of extreme rainfall events due to the global climate change. In this study, six types of orchard terraces, including slope land as the control (SLck), level terrace with bare (LTb) and vegetation taluses (LTv), outward (OTv) and inward terrace (ITv) with vegetation taluses and level terrace having front mounds and back ditches with vegetation taluses (MDLTv), were used to analyze the effects of extreme and ordinary rainfall events on the surface runoff and soil erosion. According to the measured data for twelve consecutive years, 356 natural rainfall events were divided into extreme and ordinary rainfall, based on the World Meteorological Organization standard. The results indicated that more severe surface runoff and sediment loss occurred under extreme rainfall: the runoff coefficient and soil loss under extreme rainfall were 2.6 and 11.5 times those under ordinary rainfall, respectively. The sediment yield (contribution rate, 42.9%) exhibited a higher sensitivity to extreme rainfall events compared to that of the surface runoff generation (contribution rate, 16.4%). Moreover, the reduction in the surface runoff and sediment in the extreme rainfall case differed for different orchard terraces. The average surface runoff coefficient and soil loss amount decreased in the following order: SLck>LTb>OTv>LTv>ITv>MDLTv. Nevertheless, the highest and lowest contributions of the extreme rainfall to the sediment yield occurred in the LTb (64.8%) and MDLTv (21.7%) plots, respectively. Therefore, severe talus erosion caused by extreme rainfall should be monitored, and a combination of vegetation taluses and front mounds and back ditches on the platforms is recommended as a sustainable strategy to prevent extreme water erosion when transforming slope land into orchard terraces.

OBJECTIVE: To explore the relationship between the -344T/C polymorphism of aldosterone synthase (CYP11B2) gene and essential hypertension in Chinese Mongolian population. METHODS: By cluster-sampling method, a total of 1575 Mongolian people in Tongliao city of Inner Mongolia were included in this study. And 417 subjects were normotension, 596 subjects were prehypertension and 562 subjects were essential hypertension. A survey was conducted to collect data by personal interview using a standard questionnaire, meanwhile fasting blood samples were drawn. Height, weight, waist circumference, blood pressure, blood-fat indexes and fasting plasma glucose were measured. The variant genotypes of CYP11B2 were identified by PCR assays. The relationship between the -344T/C polymorphism of CYP11B2 gene and essential hypertension were analyzed by multinomial logistic regression model. RESULTS: Crude prevalence of prehypertension among Mongolian people was 37.84% (596/1575) and hypertension was 35.68% (562/1575). The age-standardized prevalence of prehypertension was 38.57% and hypertension was 31.53%. The frequency of the T and C allele was 0.66 (481/728) and 0.34 (247/728) for normotension group, 0.69 (696/1042) and 0.33 (346/1042) for prehypertension group, 0.71 (706/998) and 0.29 (292/998) for hypertension group. The multiple logistic models showed CYP11B2 variant genotypes were associated with prehypertension (TT/CC, OR = 1.33, 95%CI: 0.87 - 2.01; TC/CC, OR = 1.74, 95%CI: 1.13 - 2.67; TC + TT/CC, OR = 1.49, 95%CI: 1.01 - 2.22); CYP11B2 variant genotypes were associated with hypertension (TT/CC, OR = 1.70, 95%CI: 1.07 - 2.70; TC/CC, OR = 1.59, 95%CI: 0.98 - 2.50; TC + TT/CC, OR = 1.66, 95%CI: 1.06 - 2.58). CONCLUSION: CYP11B2 gene -344T/C polymorphism were associated with essential hypertension in Chinese Mongolian population.

Vegetation recolonization has often been used to control roadside slope erosion, and in this paper, four restoration models - Natural Restoration, Grass, Grass & Shrub, Sodded Strip - were chosen to recolonize the plants on a newly built unpaved roadside slope in the Three Gorges Reservoir Area. After eight months growth, eight rainfall simulations (intensity of 90 mm h(-1) for 60 min) and in-situ soil shear strength test were then carried out to identify the impacts of vegetation on roadside slope erosion and soil shear strength. The erosion on cutslopes was higher than that on fillslopes. The runoff coefficient and soil detachment rate were significantly lower on the Grass & Shrub model (4.3% and 1.99 g m(-2) min(-1), respectively) compared with the other three, which had the highest surface cover (91.4%), aboveground biomass (1.44 kg m(-2)) and root weight density (3.94 kg m(-3)). The runoff coefficient and soil detachment rate on roadside slopes showed a logarithmic decrease with the root weight density, root length density and aboveground biomass. The soil shear strength measured before and after the rainfall was higher on Grass & Shrub (59.29 and 53.73 kPa) and decreased on Grass (46.93 and 40.48 kPa), Sodded Strip (31.20 and 18.87 kPa) and Natural Restoration (25.31 and 9.36 kPa). Negative linear correlations were found between the soil shear strength reduction and aboveground biomass, root weight density and root length density. The variation of soil shear strength reduction was closely related to the roadside slope erosion, a positive linear correlation was found between runoff coefficient and soil shear strength reduction, and a power function was shown between soil detachment rate and soil shear strength reduction. This study demonstrated that Grass and Grass & Shrub were more suitable and highly cost-effective in controlling initial period erosion of newly built low-volume unpaved road.

One of the main impacts of road construction is the creation of bare and steep slopes when lack of vegetation protection, which usually generates serious soil loss, a major erosion source in mountainous areas. In this study, six treatments including planting grass (GRA), grass and bush (GBC), building terrace combined with grass and bush (TGB), planting sod strip (SS), building sloping farmland (SFL), and building sloping farmland combined with grass (SFLG) were installed on the newly constructed unpaved roadside slopes in Three Gorges Reservoir Area to investigate the effects of these measures on the erosion prevention of the unpaved roadside slopes under natural rainfall. Among the six treatments, GBC had the best effect, which had the quickest revegetation rate (the mean vegetation coverage increased from 20.2% to 91.6%), decreased the soil bulk density by 9.5%, and increased the saturated hydraulic conductivity by 678.1%, followed by GRA and SS. TGB had the greatest potential in mitigating soil loss and runoff, with the efficiency in trapping sediment and runoff reached 80.2% and 72.3%, respectively, being appropriate for the cut slopes with a gradient greater than 45 degrees, followed by GBC and GRA. GRA had a higher efficiency on fill slope than on cut slope, while GBC showed greater applicability on cut slope. On the fill slope with a moderate gradient 15 degrees, SFLG had a potential of trapping 23.0% of sediment, which was equivalent to that of GRA and GBC, but the efficiency in trapping runoff was only 2.0%. When the fill slopes with a gradient less than 15 degrees were utilized as farmland, a grass buffer strip of 2 m length was needed on the upslope for erosion control.

OBJECTIVE: To evaluate the acceptability of self-management project for patients with hypertension and the short term result, and provide the evidence of preventing and controlling hypertension using chronic disease self-management. METHODS: Voluntary patients with hypertension were grouped into the control and experimental groups after matching age, sex, education levels and the number of chronic diseases between the two groups. Data collected through questionnaire after intervention were compared with the baseline data through analysis of covariance and chi(2) test. RESULTS: Rate of awareness for the diagnostic criteria of hypertension, risk factors and associated diseases increased by 56.4%, 50.4% and 37.6% respectively, and the rate of high salt diet intake, body mass index and the times of visiting doctors decreased by 27.1%, 0.8 kg/m(2) and 2.7 times on average in experimental group, as compared with control group. The health status (energy, fatigue, shortness of breathe, pain) improved (P < 0.05). In the similar comparison, the rate in which the hypertensive patients took antihypertensive medicine increased by 20%, and the scores of cognitive symptoms management and of patient communication with physician increased by 3.9 and 1.1 points respectively. The rate of compliance to medication increased by 21.5% compared with themselves. CONCLUSION: The awareness rate of hypertensive patients on knowledge and on unhealthy life style improved after implementing the self-management program. Their emotion, healthy behavior, health status and symptoms related to hypertension improved. Data showed that it was effective for hypertensive patients to use chronic diseases self-management.
