RESUMEN

The cosmetic industry is constantly searching for bioactive ingredients, namely, those obtained from natural sources with environmentally friendly connotations and less toxic effects. A previous study of our research group optimized the extraction of phenolic compounds from Juglans regia by heat-assisted extraction. Due to its richness in different phenolic compounds, the present work aimed to develop a formulation containing J. regia leaf extract. The extract's antioxidant, anti-tyrosinase, antimicrobial, anti-inflammatory, wound healing, cytotoxicity, and photostability properties were evaluated. The extract was then incorporated into an O/W base cream, followed by characterization of the final formulation in terms of its antioxidant properties, phenolic composition, and stability over time and at different storage conditions. The most abundant compounds in the hydroethanolic extract were 3-O-caffeoylquinic acid (18.30 ± 0.04 mg/g), quercetin-O-pentoside (9.64 ± 0.06 mg/g), and quercetin 3-O-glucoside (6.70 ± 0.19 mg/g). Besides those, the extract presented antioxidant, anti-inflammatory, wound closure, and antibacterial effects against several skin pathogens. In addition, HaCaT cell viability was maintained up to 98% at 400 µg/mL. Within Proteus vulgaris-infected HaCaT cells, the extract also presented an over 40% bacterial mortality rate at its nontoxic concentration (200 µg/mL). After incorporating the extract, the obtained formulation presented a good physicochemical profile over time and at different storage conditions while also maintaining its antioxidant effect; as such, it can be considered stable for topical application. Future work to evaluate its performance in terms of skin permeation and detailed toxicological studies with a focus on regulatory requirements, involving skin irritation, eye irritation, genotoxicity, photo-irritation, and dermal absorption, should be conducted, as the prepared formulation demonstrated relevant properties that deserve to be further explored.

RESUMEN

Phenolic compounds have attracted much attention in recent times as their dietary intake has been associated with the prevention of some chronic and degenerative diseases that constitute major causes of death and incapacity in developed countries, such as cardiovascular diseases, type II diabetes, some types of cancers or neurodegenerative disorders like Alzheimer's and Parkinson's diseases. Nowadays it is considered that these compounds contribute, at least in part, for the protective effects of fruit and vegetable-rich diets, so that the study of their role in human nutrition has become a central issue in food research. This chapter reviews the current knowledge on the phenolic compounds as food components, namely their occurrence in the diet, bioavailability and metabolism, biological activities and mechanisms of action. Besides, the approaches for their extraction from plant matrices and technological improvements regarding their preparation, stability and bioavailability in order to be used as functional food ingredients are also reviewed, as well as their legal situation regarding the possibility of making "health claims" based on their presence in food and beverages.

Sanguisorba minor Scop. is a perennial plant native to the Mediterranean region which exhibits several medicinal activities. In the present study, plant growth, chemical composition, and antitumor and antimicrobial properties of S. minor plants were evaluated under different growing conditions. In particular, plants were grown on different substrates, namely (A) peat, (B) peat : perlite (1 : 1) and (C) peat : perlite (2 : 1). The dry weight of the aerial parts and roots was higher for the peat treatment (A), whereas plant growth was severely affected in the peat : perlite (1 : 1) treatment (B). The major detected sugars were fructose and glucose, while the peat : perlite (2 : 1) treatment resulted in a significant increase of α-tocopherol, glucose and total sugars content, especially in the aerial parts. Oxalic acid and citric acid were the major organic acids detected, with a varied effect of growth substrate on organic acid profile. Growth substrate and plant part also had a significant effect on fatty acid composition, especially on linoleic acid and α-linolenic acid content which was higher in the peat : perlite (1 : 1) treatment in both aerial parts and roots. An impact on the composition of phenolic compounds was also observed, where the plants grown on peat : perlite (2 : 1) and peat : perlite (1 : 1) substrates had the highest content of individual and total phenolic compounds in their aerial parts and roots, respectively. Moreover, roots contained significantly higher amounts of phenolic compounds than the aerial parts. Moreover, plant extracts exhibited antiproliferative activity against four tumor cell lines (HeLa, HepG2, MCF-7 and NCI-H460) and a primary culture of porcine liver cells (PLP2), as well as significant antimicrobial properties. In conclusion, S. minor presented significant bioactive properties, while growth substrates affected the nutritional value, chemical composition, antitumor and antimicrobial properties of the species which could be probably attributed to the higher phenolic compound content and different compositions of phenolic compounds. Therefore, although the species is commonly found in the Mediterranean region, it is underexploited yet and its plant tissues could be a potential source of natural bioactive compounds with further use in pharmaceutical and medicinal applications.

The extraction of triterpenoids and phenolic compounds from Ganoderma lucidum was optimized by using the response surface methodology (RSM), using heat and ultrasound assisted extraction techniques (HAE and UAE). The obtained results were compared with that of the standard Soxhlet procedure. RSM was applied using a circumscribed central composite design with three variables (time, ethanol content, and temperature or ultrasonic power) and five levels. The conditions that maximize the responses (extraction yield, triterpenoids and total phenolics) were: 78.9 min, 90.0 °C and 62.5% ethanol and 40 min, 100.0 W and 89.5% ethanol for HAE and UAE, respectively. The latter was the most effective, resulting in an extraction yield of 4.9 ± 0.6% comprising a content of 435.6 ± 21.1 mg g-1 of triterpenes and 106.6 ± 16.2 mg g-1 of total phenolics. The optimized extracts were fully characterized in terms of individual phenolic compounds and triterpenoids by HPLC-DAD-ESI/MS. The recovery of the above-mentioned bioactive compounds was markedly enhanced using the UAE technique.

Leaf fractions of Garcinia brasiliensis were evaluated concerning their antioxidant, antimicrobial, anti-inflammatory and cytotoxic properties, and the most active fraction was then fully characterized regarding its phenolic composition using HPLC-DAD-ESI/MSn. The ethyl acetate fraction from partitioning of the methanolic leaf extract revealed a strong antioxidant activity that was comparable to Trolox, the positive control. This fraction was also able to show a significant antimicrobial activity against Gram-positive and Gram-negative bacteria and the fungus Candida albicans. However, the dichloromethane fraction was found to present the highest anti-inflammatory (83 ± 9 µg mL-1) and cytotoxic activities, thus presenting slight toxicity using a non-tumor cell line. Regarding the phenolic profile, the ethyl acetate fraction presented twelve flavonoids, with morelloflavone-7''-O-glucoside (52.1 ± 0.4 mg g-1) and gardinia biflavonoid 2a glucoside (27.5 ± 0.2 mg g-1) being the major compounds identified. These results indicate that leaves of G. brasiliensis might be a potential source of natural biomolecules for pharmaceutical and medicinal applications.
