Emerging scientific studies have supported the hypothesis that exposure to environmental chemicals increases the incidence of diverse human metabolic disorders. Therefore, this study aimed to evaluate the association between fluoride exposure and metabolic syndrome (MetS) prevalence in people from the Central Region of Mexico. This research included 575 adult individuals. Urinary fluoride concentrations were determined using a potentiometric method. Anthropometric measurements and blood pressure were also acquired. Serum fasting glucose and lipid levels were quantified. For the MetS screening, we used the International Diabetes Federation (IDF) and National Cholesterol Education Program Adult Treatment Panel III (NCEP ATP III) criteria. The mean urinary fluoride levels were 3.50 ± 2.50 mg/L. The prevalence of MetS was approximately 27 and 25% according to NCEP ATP III and IDF criteria, respectively. A multivariate logistic regression analysis showed significant associations (p < 0.05) between urinary fluoride concentrations and MetS occurrence using NCEP ATP III criteria (OR = 1.60; 95% CI 1.05-2.10) and IDF criteria (OR = 1.35; 95% CI 0.95-1.45). These findings emphasize the need to discover the underlying mechanisms that fluoride exposure has on MetS pathogenesis.

The lower basin of Coatzacoalcos River is one of the most polluted regions of the southern Gulf of Mexico. Organochlorine compounds, polybrominated diphenyl ethers, polycyclic aromatic hydrocarbons, and heavy metals have been registered in this region. In the present study, genotoxicity was evaluated in the blood of giant toads (Rhinella marina) from Coatzacoalcos' rural and industrial zones, and compared with laboratory toads. Determination of the frequency of micronucleus and erythrocyte nuclear abnormalities by the light microscope and cell cycle and apoptosis by flow cytometry were used as biomarkers of genotoxicity. We found more variability in micronucleus and more nuclear buds in toads from industrial zones. Also, cell cycle alterations and an increase of apoptosis in erythrocytes were found in toads from rural and industrial zones. Multivariate statistics show that the toads from the industrial zone were more affected than toads from laboratory and rural zones.

Human paraoxonase-1 (PON1) is a hydrolytic enzyme related in the onset, establishment, and progression of inflammatory illnesses, including cardiovascular diseases (CVD). Therefore, the primary goal of this investigation was to determinate the influence of the PON1 gene polymorphism (Q192R) on prognostic biomarkers of CVD in women (n = 350) from San Luis Potosi, Mexico. For everyone enrolled, anthropometric, clinical, and biochemical measurements were acquired. Q192R polymorphism of the PON1 gene was assessed by real-time PCR using TaqMan probes. Also, quantification and/or calculation of suggested predictive CVD biomarkers were completed. The association between PON1(Q192R) polymorphism with clinical predictive CVD biomarkers was assessed using univariate and multivariate logistic regression models. Genotype PON1 (Q192R) frequencies were: 18%, 47% and 35% for QQ, QR, and RR, respectively. Besides, the frequency of the variant R allele was 0.58. Furthermore, robust significant associations were found between PON1 (Q192R) polymorphism with serum ADMA [OR (95% CI); 3.50 (1.20-5.00), p < 0.01]; FABP4 [2.50 (2.15-3.95), p < 0.01]; and miR-126 [1.50 (1.15-2.00), p < 0.01] levels after adjusting for probable confounders. Data found in this research strongly support the notion that proposes a crucial role of PON1 enzyme (Q192R polymorphism) as a genetic determinant in the CVD risk in Mexican women.

A major public health concern in Mexico is the natural contamination of groundwater with fluoride and arsenic. Therefore, this work aimed to evaluate the magnitude of human health risk after determining fluoride and arsenic concentrations in groundwater samples (n = 50) from the Metropolitan area of the city of San Luis Potosi, Mexico. Fluoride levels in water were determined via a potentiometric method using an ion-selective electrode. Arsenic concentrations in water samples were determined with an Atomic Absorption technique. Subsequently, a probabilistic health risk assessment was developed (Monte Carlo Analysis). Fluoride levels in water ranged from 0.20 to 3.50 mg/L. For arsenic, the mean level found in the assessed water samples was 15.5 ± 5.50 µg/L (range: 2.50-30.0 µg/L). In addition, when the probabilistic health risk assessment was completed, a mean HI (cumulative hazardous index) of higher than 1 was detected, indicating a high NCR (non-carcinogenic risk) for children and adults. According to the results found in this study, exposure protection campaigns are imperative in the Metropolitan area of the city of San Luis Potosí, Mexico, to successfully diminish exposure to arsenic and fluoride and, as a consequence, decrease the NCR in the population living in that region of Mexico.

An appropriate and precise identification of high-risk individuals to develop cardiovascular diseases (CVD) is of high importance to reduce these kinds of diseases, a major health concern worldwide. Therefore, the aim of this research was to evaluate prognostic CVD biomarkers in Mexican women exposed to inorganic arsenic via drinking water. Then, a cross-sectional study including 190 women was achieved. Urinary arsenic (UAs) levels were analyzed as exposure biomarker to that metalloid. While, plasma asymmetric dimethylarginine (ADMA), adipocyte fatty acid-binding protein (FABP4), adiponectin, and chemerin levels, hypertriglyceridemic waist (HW) phenotype, atherogenic index of plasma (AIP), and Framingham risk score (FRS) were assessed as prognostic CVD biomarkers. Mean UAs level detected in the evaluated urinary samples was 45.0 ±â€¯40.0 µg/g creatinine. In addition, mean plasma ADMA, FABP4, chemerin and adiponectin levels were 0.68 µmol/L, 20.3 ng/mL, 12.5 µg/mL, and 255 ng/mL, correspondingly. Approximately, 54% of women participants displayed an HW phenotype. Regarding AIP and FRS values, 0.12 ±â€¯0.15 and 7.50 ±â€¯8.00 were found, respectively. Besides, strong and significant associations (p < 0.05) between UAs and AIP, ADMA, and FABP4 were distinguished. Also, after a multivariate analysis, the association between those variables persisted after adjustment for traditional risk factors of CVD. In conclusion, according to the results found in this research, the most sensible CVD biomarkers distinguished in this study were AIP, ADMA, and FABP4. Nevertheless, more studies are necessary to confirm the results found in this investigation.

Household air pollution has been associated as a risk factor for cardiovascular diseases (CVD). Therefore, the aim of this study was to assess the expression of vascular inflammation regulators miR-126 and miR-155 in plasma from women that cook with wood and women that cook with liquid petroleum gas (LPG). A cumulative index of exposure to smoke (CIES) was estimated, urinary 1-hydroxypyrene (1-OHP) levels were quantified and miRNAs expression levels were determined by quantitative real-time PCR (qRT-PCR). Biochemical clinical parameters were also evaluated. The average values for CIES and 1-OHP were 140 ± 86.8 hours-years (12.0-270 hours-years) and 0.52 ± 0.45 µmol/mol creatinine, respectively. miR-126 and miR-155 expression levels were significantly higher (p < 0.01) in the wood users compared to LPG users. Besides, we found a significant association (p < 0.01) between miR-126 and miR-155 expression levels and CIES and urinary 1-OHP concentrations. These results contribute to the current evidence about the cardiovascular risk related to biomass smoke exposure, from an epigenetic level.

Health complications have been associated with polycyclic aromatic hydrocarbons (PAHs) exposure, a widespread environmental pollutants family. Therefore, the objective of this investigation was to develop a probabilistic health risk evaluation (using Monte-Carlo simulation) in an infantile population living in areas with a high risk of pollution by PAHs (indoor wood combustion, brick kiln industry, municipal landfill, and low and high vehicular traffic) in Mexico. Urine samples were obtained from Mexican children (n = 135) and urinary 1-OHP concentrations (used as a PAHs biomarker) were quantified. Highest urinary 1-OHP concentrations were detected in children living in areas that use wood combustion as the principal indoor fuel (3.50 ± 0.95 µg/L). Nevertheless, estimated hazard quotients (HQ) lower than 1 were found in all assessed sites after Monte-Carlo analysis. Although HQ <1.0 (a toxic effect is not expected), more data are necessary to determine the real impact of PAHs exposure on children health status.

The purpose of this study was to evaluate lead exposure and its relationship with serum levels of predictive CVD biomarkers [asymmetric dimethylarginine (ADMA), adipocyte fatty acid-binding protein (FABP4), adiponectin, and chemerin] in women living in San Luis Potosi, Mexico. A mean blood lead level (BLL) of 11.5 ± 9.00 µg/dL (mean ± standard deviation) was found after all blood samples were analyzed. Regarding serum predictive CVD biomarkers, mean serum level of 0.68 ± 0.38 µmol/L, 20.5 ± 16.5 ng/mL, 12.5 ± 3.30 µg/mL, and 255 ± 130 ng/mL were found for ADMA, FABP4, adiponectin, and chemerin, respectively. Simple significant associations (Pearson´s correlations) between BLL and ADMA (r = 0.17; p = 0.04) and FABP4 (r = 0.23; p = 0.03) were found. Furthermore, a multivariate linear regression model showed that BLL was a significant predictor of serum ADMA (ß = 0.06; p = 0.001) and FABP4 (ß = 1.75; p = 0.0004) concentrations after adjusting by confounders. For serum chemerin and adiponectin levels, no associations were found with BLL. In conclusion, high serum ADMA and FABP4 (predictive CVD biomarkers) levels were found in women exposed to lead. Consequently, this research can be used as a point of departure for the prevention of CVD events in populations living in sites environmentally impacted with lead.

The aim of this study was to perform a polybrominated diphenyl ethers (PBDEs) exposure assessment using blood samples collected from children living in the metropolitan area of Guadalajara, Jalisco, Mexico (GDL). Five congeners of PBDEs were analyzed using a gas chromatography/mass spectrometry technique. The blood concentrations of total PBDEs ranged from 5.50 to 169 ng/g lipid (42.0 ± 18.0 ng/g lipid; mean ± standard deviation). Regarding BDE congeners, the main congener (highest blood levels) was BDE99 (14.5 ± 5.50 ng/g lipid), followed by BDE100 (9.80 ± 3.40 ng/g lipid) and BDE154 (9.80 ± 5.90 ng/g lipid), and finally BDE153 (5.80 ± 2.30 ng/g lipid) and BDE47 (2.20 ± 1.20 ng/g lipid). In conclusion, blood PBDEs concentrations of concern were detected in this study, as blood levels were similar to the ones found in North America (the highest worldwide).

AIM: This study aimed to evaluate the genetic effects of PON1 Q192R polymorphism on serum FABP4 levels in Mexican women. METHODS: PON1 Q192R polymorphism was genotyped using a TaqMan allelic discrimination assay and serum FABP4 concentration was measured using an enzyme-linked immunosorbent assay. RESULTS: The distribution of genotype frequencies in the assessed women (PON1 Q192R polymorphism) was QQ = 20%, QR = 48% and RR = 32%. Significantly higher serum FABP4 levels were found in women with genotype QR/RR (20.6 ± 2.20 ng/mL), when compared with the levels found in the QQ group (12.8 ± 1.70 ng/mL) (p = .004). After, the odds ratio (OR) was calculated by binomial logistic regression analysis and a significantly higher OR was found in the QR/RR group when compared with the QQ group (OR = 3.45; 95% CI = 1.80-16.50; p < .05). CONCLUSION: The results support an association between 192R-allele of the PON1 polymorphism (Q192R) and increased serum FABP4 levels (suggested as an early biomarker of CVDs risk) in assessed Mexican women.

Recent studies have documented environmental contamination by PCBs in soil from different areas in Mexico (industrial, mining, and urban sites). However, the real significance of that soil contamination has not been established. Therefore, the aim of this study was to perform a human health risk assessment (Monte Carlos simulation) to evaluate the probable toxic effects of soils contaminated with PCBs on children in four sites in Mexico. A high non-carcinogenic risk (total nHQ = 1.1E+01; if nHQ ≥1, hazardous health effects cannot be ruled out) was found in Alpuyeca, Morelos, Mexico. Moreover, the total CR (cancer risk) found in Alpuyeca, Morelos is of concern (total CR = 5.1E-03), being that a cut-point of 1.0E-06 has been suggested as a safe level for cancer risk. Taking into consideration the data shown in this research, we conclude that a strategy to protect human health is necessary for the assessed sites.

Human biomonitoring (HBM) is an appreciated tool used to evaluate human exposure to environmental, occupational or lifestyle chemicals. Therefore, the aim of this study was to evaluate the exposure levels for environmental chemicals in urine and blood samples of children from San Luis Potosí, Mexico (SLP). This study identifies environmental chemicals of concern such as: arsenic (45.0 ± 15.0 µg/g creatinine), lead (5.40 ± 2.80 µg/dL), t,t-muconic acid (266 ± 220 µg/g creatinine), 1-hydroxypyrene (0.25 ± 0.15 µmol/mol creatinine), PBDEs (28.0 ± 15.0 ng/g lipid), and PCBs (33.0 ± 16.0 ng/g lipid). On the other hand, low mercury (1.25 ± 1.00 µg/L), hippuric acid (0.38 ± 0.15 µg/g creatinine) and total DDT (130 ± 35 ng/g lipid) exposure levels were found. This preliminary study showed the tool's utility, as the general findings revealed chemicals of concern. Moreover, this screening exhibited the need for HBM in the general population of SLP.

It has been demonstrated that the human biomonitoring of susceptible populations is a valuable method for the identification of critical contaminants. Therefore, the purpose of this study was to assess the exposure profile for arsenic (As), lead (Pb), mercury (Hg), 1-hydroxypyrene (1-OHP), 1,1-bis(p-chlorophenyl)-2,2,2-trichloroethane (DDT), 1,1-dichloro-2,2-bis(p-chlorophenyl) ethylene (DDE), polybrominated diphenyl ethers (PBDEs), and polychlorinated biphenyls (PCBs) in children living in Ciudad Juarez, Chihuahua, Mexico (a major manufacturing center in Mexico). In 2012, we evaluated a total of 135 healthy children living in Ciudad Juarez since birth. The total PBDEs levels ranged from nondetectable (< LOD) to 215 ng/g lipid, with a mean total PBDEs level of 29.5 ± 53.0 ng/g lipid (geometric mean ± standard deviation). The mean total PCBs level in the study participants was 29.0 ± 10.5 ng/g lipid (range 4.50-50.0 ng/g lipid). The purpose of this study was to assess the levels of polybrominated diphenyl ethers (PBDEs), polychlorinated biphenyls (PCBs), dichlorodiphenyltrichloroethane (DDT), dichlorodiphenyldichloroethane (DDE), and four heavy metals (arsenic, cadmium, and lead) in outdoor surface soils (50 samples) collected from the metropolitan area of Monterrey in Mexico. Total PBDEs levels ranged from 1.80 to 127 µg/kg, with mean total PBDEs level of 14.2 ± 21.5 µg/kg (geometric mean ± standard deviation). For PCBs, the mean total level in the studied soils was 23.5 ± 20.2 µg/kg (range 4.0-65.5 µg/kg). An important finding in our study was that all soil samples (100%) had detectable levels of the metabolite p,p'-DDE. Moreover, the mean total DDT level (∑p'p-DDT and p'p-DDE) was approximately 132 ± 175 µg/kg. The mean levels for arsenic, cadmium, and lead in soil were 5.30 ± 1.35 (range 1.55-7.85) mg/kg, 2.20 ± 1.20 (range 0.65-6.40) mg/kg, and 455 ± 204 (range 224-1230) mg/kg, respectively. Our study has several limitations, the most notable of which is the small sample of soils evaluated. However, this screening study provided concentration data for the occurrence of POPs and four heavy metals in soil from the metropolitan area of Monterrey, Nuevo Leon, Mexico, and taking into consideration that soil is an important pathway of exposure for people, a biomonitoring program for the surveillance of the general population in the metropolitan area of Monterrey, Nuevo Leon is deemed necessary.

The aim of this study was to assess the levels of polybrominated diphenyl ethers (PBDEs) in soils from the city of San Luis Potosi in Mexico and perform an ecological and human health risk characterization. In order to confirm the presence of PBDEs, outdoor surface soil samples were collected and the concentrations of PBDEs in urban, industrial, agricultural, and brick kiln industry areas were determined. The mean total PBDEs levels obtained in the study sites were 25.0 ± 39.5 µg/kg (geometric mean ± standard deviation) in the brick kiln industry zone; 34.5 ± 36.0 µg/kg in the urban zone; 8.00 ± 7.10 µg/kg in the industrial zone and 16.6 ± 15.3 µg/kg in the agricultural zone. The ecological and human health risk characterization showed relatively low-hazard quotient values. However, the moderately high PBDEs levels found in soils highlight the necessity to establish a systematic monitoring process for PBDEs in environmental and biological samples.

The antimicrobial properties of silver nanoparticles (AgNPs) have made these particles one of the most used nanomaterials in consumer products. Therefore, an understanding of the interactions (unwanted toxicity) between nanoparticles and human cells is of significant interest. The aim of this study was to assess the in vitro cytotoxicity effects of silver nanoclusters (AgNC, < 2 nm diameter) on peripheral blood mononuclear cells (PBMC). Using flow cytometry and comet assay methods, we demonstrate that exposure of PBMC to AgNC induced intracellular reactive oxygen species (ROS) generation, DNA damage and apoptosis at 3, 6 and 12 h, with a dose-dependent response (0.1, 1, 3, 5 and 30 µg ml(-1)). Advanced electron microscopy imaging of complete and ultrathin-sections of PBMC confirmed the cytotoxic effects and cell damage caused by AgNC. The present study showed that AgNC produced without coating agents induced significant cytotoxic effects on PBMC owing to their high aspect ratio and active surface area, even at much lower concentrations (<1 µg ml(-1)) than those applied in previous studies, resembling what would occur under real exposure conditions to nanosilver-functionalized consumer products.

CONTEXT: The use of solid fuels for cooking and heating is likely to be the largest source of indoor air pollution on a global scale. OBJECTIVE: The aim of this study was to investigate the urinary excretion of 1-hydroxypyrene (1-OHP) in women living in a rural community, where biomass combustion is used as main energy source during a working day. MATERIALS AND METHODS: The study was performed on urine samples collected in 2012, of 30 healthy women who were residents of a rural community in San Luis Potosi, Mexico. Urine was collected from each woman at three time points (morning, post-lunch and evening) during a working day. The analysis of urinary 1-OHP was performed using high-performance liquid chromatography (HPLC). Also, a health-risk assessment was conducted. RESULTS: The highest levels of 1-OHP in this study were found in the samples taken in the evening (geometric mean ± SD; 0.36 ± 0.13 µg/g creatinine). However, no significant differences among 1-OHP concentrations in the evening and post-lunch samples (0.27 ± 0.10 µg/g creatinine; 0.58 ± 0.67 µg/L) were observed. But, the 1-OHP levels (0.17 ± 0.13 µg/g creatinine; 0.19 ± 0.21 µg/L) found in samples collected in the morning were significantly lower than the 1-OHP levels found in samples collected during post-lunch and evening time. DISCUSSION AND CONCLUSION: The data shown in this study demonstrated an increment in the exposure levels to PAHs in women across the shift. However, no health risk was found in this study.

The aim of this study was to assess the levels of polybrominated diphenyl ethers (PBDEs), polychlorinated biphenyls (PCBs), p,p'-dichlorodiphenyltrichloroethane (DDT), p,p'-dichlorodiphenyldichloroethylene (DDE), and four heavy metals (arsenic, cadmium, lead, and mercury) in soil from the city of San Luis Potosí in Mexico. In order to confirm the presence of the previously mentioned compounds, outdoor surface soil samples were collected and analyzed by gas chromatography/mass spectrometer for PBDEs, PCBs, DDT, and DDE. Meanwhile, heavy metals were quantified using the atomic absorption spectrophotometry technique. The total PBDEs levels ranged from 5.0 to 134 µg/kg dry weight (dw), with a total mean PBDEs level of 22.0 ± 32.5 µg/kg dw (geometric mean ± standard deviation). Biomass combustion indoors has been associated with generation of various pollutants, such as carbon monoxide, formaldehyde, respirable particles, toluene, benzene and polycyclic aromatic hydrocarbons (PAHs), among others. In this context, the aim of this study was to evaluate the urinary 1-hydroxypyrene (as a biomarker exposure to PAHs) levels in children living in three states in Mexico. We evaluated children living in communities that use biomass fuels to cook and to heat homes in the next states: Chiapas, Oaxaca and Quintana Roo. We found similar levels of 1-OHP in urine of children living in the three studied states, with mean levels of 3.5 ± 1.0; 4.5 ± 2.8 and 4.4 ± 2.5 µg/L (geometric mean ± standard deviation), respectively for Chiapas, Oaxaca and Quintana Roo. In conclusion, our data indicate high exposure levels to PAHs in children living in the states studied in this work.

In Mexico, polychlorinated biphenyls (PCBs) were principally used as heat transfer chemicals in electric transformers and capacitors as well as hydraulic fluids and lubricants in heavy electrical equipment since the early 1940s. However, although PCBs have been banned in Mexico, their past and present improper disposal has resulted in environmental contamination. Therefore, the aim of this study was to evaluate the PCBs levels in soil samples in the immediate area of an electric capacitor manufacturing industry, which was established several years ago in Alpuyeca, Morelos, Mexico. To confirm the presence of PCBs, surface soil samples (1-5 cm in depth) were collected from the vicinity of the industry. We determined the concentrations of 40 PCB congeners in soil samples using gas chromatography/mass spectrometry. The total PCBs levels in the soil samples ranged from 6.2 to 108460.6 µg kg(-1). Moreover, when we analyzed the results of the congeners (non-dioxin-like PCBs and dioxin-like PCBs), the levels of non-dioxin-like PCB congeners ranged from 5.7 to 103469 µg kg(-1) and the levels of dioxin-like PCB congeners ranged from 0.5 to 4992 µg kg(-1). Considering that soil is an important pathway of exposure in humans, analysis of PCBs levels in blood (as a biomarker of exposure) is necessary in individuals living in Alpuyeca, Morelos.
