OBJECTIVES: This study aimed to estimate five harmonized healthy aging indicators covering functional ability and intrinsic capacity among older women and men from Brazil and England and evaluate their association with loneliness. STUDY DESIGN: This was a cross-sectional study. METHODS: We used two nationally representative samples of men and women aged ≥60 years from the Brazilian Longitudinal Study of Aging (ELSI-Brazil) wave 2 (2019-2021; n = 6929) and the English Longitudinal Study of Aging wave 9 (2018-2019; n = 5902). Healthy aging included five separate indicators (getting dressed, taking medication, managing money, cognitive function, and handgrip strength). Loneliness was measured by the 3-item University of California Loneliness Scale. Logistic regression models stratified by sex and country were performed. RESULTS: Overall, age-adjusted healthy aging indicators were worse in Brazil compared with England for both men and women. Considering functional ability, loneliness was negatively associated with all indicators (ranging from odds ratio [OR] = 0.26, [95% confidence interval (CI) 0.13-0.52] in English men regarding the ability to take medication to OR = 0.49 [95% CI 0.27-0.89] in Brazilian women regarding the ability to manage money). Considering intrinsic capacity, loneliness was negatively associated with a higher cognitive function (OR = 0.72; 95% CI 0.55-0.95 in English women) and a higher handgrip strength (OR = 0.61; 95% CI 0.45-0.83 in Brazilian women). Lonely women demonstrated lower odds of a higher number of healthy aging indicators than men in both countries. CONCLUSIONS: Country-specific social environments should be targeted by public policies to decrease loneliness and promote healthy aging later in life.

Control of Chagas disease in endemic countries is primarily accomplished through insecticide spraying for triatomine vectors. In this context, pyrethroids are the first-choice insecticide, and the evolution of insect resistance to these insecticides may represent an important barrier to triatomine control. In insects, cytochrome P450s are enzymes involved in the metabolism of xenobiotics and endogenous chemicals that are encoded by genes divided into different families. In this work, we evaluated the role of three Rhodnius prolixus CYP4EM subfamily genes during blood meal and after deltamethrin exposure. CYP4 gene members were expressed in different insect organs (integument, salivary glands (SGs), midgut, fat body and malpighian tubules) at distinct transcriptional levels. CYP4EM1 gene was highly expressed in the SG and was clearly modulated after insect blood meal. Injection of CYP4EM1dsRNA promoted significant reduction in mRNA levels of both CYP4EM1 and CYP4EM2 genes and induced deleterious effects in R. prolixus nymphs subsequently exposed to sublethal doses of deltamethrin (3.4 or 3.8 ng/nymph treated). The higher dose reduced the survival over time and increased susceptibility of R. prolixus nymphs to deltamethrin. A better understanding of this mechanism can help in developing of more efficient strategies to reduce Trypanosoma cruzi vector transmission in Americas.

Following the appearance of several antimicrobial agents to control the spread of infections, two major challenges have emerged: (i) the occurrence and blowout of multiresistant bacteria and the increase of chronic diseases and (ii) difficult-to-eradicate infections. In this study, we tested five benzoylthiourea derivatives for their ability to inhibit and stop bacterial growth and evaluated the possible influence of 1,2,4-triazolyl-benzoylthiourea derivative 4 on the formation and eradication of Staphylococcus aureus biofilms. Benzoylthiourea derivatives 4, 6, 10, 11 and 13 were obtained in one or two steps with low cost and subjected to tests to identify their minimum inhibitory concentration (MIC) and minimum bactericidal concentration. In vitro tests were also performed to assess their effects on biofilm formation and in preformed biofilms and scanning electron microscopy was used to visualize the effects on biofilm formation. The 1,2,4-triazolyl-benzoylthiourea derivative 4 showed bacteriostatic activity against the S. aureus HU25 clinical strain with an MIC of 16 µg ml-1 , which is below the toxic concentration (at 2500 µg ml-1 , 62·25% of the cells remained viable). Compound 4 also effectively prevented biofilm formation at the three subinhibitory concentrations tested (1/2 MIC, 1/4 MIC and 1/8 MIC) as confirmed by scanning electron microscopy. For breakdown of formed biofilms, the main influence was at a subinhibitory concentration (1/2 MIC). These findings make compound 4 a strong candidate for studies on the development of new antimicrobial and antibiofilm agents.

Data about the genetic structure can help to understand the evolutionary process of natural populations as well as to drive strategies of conservation. Vriesea cacuminis, an endemic Brazilian Bromeliad, has been found in 2 areas of Minas Gerais State. One is a legal preservation unit (Ibitipoca State Park) and the other an unprotected area (Serra Negra). The 2 areas belong to the Mantiqueira Mountain Range Complex; both are characterized by steep relief with high altitudes and by heterogenic vegetation formed by a mosaic of rocky fields and forest fragments. According to International Union for Conservation of Nature criteria, V. cacuminis is designated as "vulnerable". We examined the genetic variability and population structure of 70 individuals (3 populations) of V. cacuminis, using 16 ISSR markers. Although V. cacuminis is considered a rare species, the estimated genetic diversity was found to be relatively high (Shannon index = 0.33; percentage of polymorphic bands = 87%). The populations were found not to be structured (AMOVA test, ΦST = 0.16), probably due to the cross-breeding. Based on Bayesian analysis, this species includes one cluster containing the populations from Ibitipoca State Park and another cluster including the population from Serra Negra. This information will help determine strategies to maintain the genetic variability of these populations.

Borderline personality disorder (BPD) is a severe psychiatric condition that affects up to 2.7% of the population and is highly linked to functional impairment and suicide. Despite its severity, there is a lack of knowledge about its pathophysiology. Studies show genetic influence and childhood violence as factors that may contribute to the development of BPD; however, the involvement of neuroinflammation in BPD remains poorly investigated. This article aimed to explore the pathophysiology of BPD according to the levels of brain-derived neurotrophic factor (BDNF), inflammatory cytokines, and oxidative stress substances that exacerbate neuronal damage. Few articles have been published on this theme. They show that patients with BPD have a lower level of BDNF and a higher level of tumor necrosis factor (TNF)-α and interleukin (IL)-6 in peripheral blood, associated with increased plasma levels of oxidative stress markers, such as malondialdehyde and 8-hydroxy-2-deoxyguanosine. Therefore, more research on the topic is needed, mainly with a pre-clinical and clinical focus.

Congenital malformations of the external and middle ear relatively frequent anomalies (one to five cases in 20,000 live births). They are part of genetic syndromes such as Treacher Collins and Goldenhar's syndromes, but most cases are isolated and sporadic. A few cases of familial incidence of isolated external and middle ear malformations with autosomal recessive, autosomal dominant, and sex-linked modes of transmission have been described. We report on two siblings with almost identical anomalies of the external and middle ear and no other congenital defects. Their father had similar malformations, but nobody else in his large sibship presented ear malformations. The defects were also absent in two previous generations. We discuss the possibility, not previously mentioned in the literature, that these congenital malformations could have appeared by spontaneous mutation in the father and transmitted themselves as an autosomal dominant trait to his children.

Upper urinary tract obstruction is very frequent. The authors present a review of the etiology and pathogenesis of upper urinary tract obstruction. The clinical aspects and diagnosis are described in detail and the most appropriate complementary exams are pointed out. Finally, the authors discuss the therapeutic options available and the prognosis.

Foram avaliados aspectos clínico-cirúrgicos, radiográficos e histológicos de discos intervertebrais cervicais caninos após quimionucleólise com quimiopapaína associada à espondilectomia ventral em 24 cães. Em todos foi realizada a espondilectomia ventral na região do disco intervertebral C2-C3 e quimionucleólises nos demais discos cervicais. Os cães foram sacrificados às 24 horas, e aos 8, 30, 60, 90 e 120 dias após aplicação da enzima para a obtenção de cortes sagitais dos discos intervertebrais tratados e das estruturas cartilaginosas e ósseas adjacentes. Nos mesmos períodos, foram realizadas radiografias para monitoração da espessura dos espaços intervertebrais. Todos toleraram bem o procedimento cirúrgico e não apresentaram alterações clínicas significativas. Na avaliação radiográfica, observou-se marcada redução dos espaços intervertebrais 24 horas após o tratamento. Do oitavo ao 90º dia após aplicação da enzima, foi verificada ausência total de espaço intervertebral na região dos discos tratados. Aos 120 dias, as espessuras dos espaços intervertebrais corresponderam, em média, a 59,1 por cento da espessura anterior ao procedimento. Na avaliação histológica, foram observadas digestão nuclear e redução da intensidade de coloração pela safranina-O 24 horas após a quimionucleólise. A partir do 60º dia, havia material amorfo no espaço nuclear, que, aos 120 dias, tinha aparência fibrocartilaginosa. A quimionucleólise associada à espondilectomia ventral determinou a lise dos discos intervertebrais e lesões nas estruturas cartilaginosas e ósseas adjacentes, com tendência à reparação por tecido fibrocartilaginoso ao longo do tempo.


The clinical-surgical, radiological, and histological aspects of chemonucleolysis with chymopapain in canine cervical intervertebral disks associated to ventral slot in 24 dogs were evaluated. The ventral slot was accomplished in the area of the intervertebral disks C2-C3 and chemonucleolysis in the other cervical disks. The dogs were clinically and radiographically monitored for up to 120 days. Each group of four animals was submitted to euthanasia with an anesthetic overdose at 24 hours, and at 8, 30, 60, 90, and 120 days after the enzyme injection for the acquisition of sagittal sections of the treated disks, as well as adjacent cartilaginous endplates and bony structures. In this study, all dogs tolerated the surgical procedure, without detectable postoperative pain or neurological deficit. In the radiographic evaluation, it was observed consistent disk space narrowing 24 hours after the procedure and total absence of space from the 8th to the 90th day of postoperative in the area of all treated disks. However, 120 days after treatment, there was an increase in disk height, corresponding on average to 59.1 percent of the preinjection value. In the histological evaluation, it was observed nuclear digestion and decrease of safranin-O staining intensity 24 hours after disk injection. At eight days, the nuclear content was still vacuolated, but more fibrillar. An irregularly defined mass was observed in the nuclear space on days 60th and 90th, but it seemed fibrocartilaginous tissue on day 120th. Chemonucleolysis with chymopapain associated to the ventral slot in the cervical column of dogs determined lysis of intervertebral disks, cervical instability, and lesions of cartilaginous and bone adjacent structures that tend to repair with fibrocartilaginous tissue along the time.

Foi avaliada a reparação óssea após implantação de proteínas morfogenéticas ósseas (BMP) em diferentes concentrações e períodos de observação, carreadas por auto-enxerto ósseo esponjoso (EOE), em falhas ósseas, produzidas na região fronto-parietal do crânio de 20 coelhas. A falha I não foi preenchida, a II foi completamente preenchida com 3mg de EOE e as falhas III, IV, V e VI foram preenchidas com EOE associado a 0,5; 1; 2 e 5mg de BMP, respectivamente. Nas avaliações mesoscópicas, post mortem, verificou-se que, independentemente do período de tratamento, o preenchimento ósseo iniciou-se a partir das bordas para o centro e do fundo para a superfície das falhas. Na falha I manifestou-se o menor preenchimento ósseo quando comparada com as demais falhas, em todos os períodos, e nas que receberam 2mg de BMP exibiu-se a melhor cobertura óssea. Microscopicamente, verificou-se que, aos sete dias, o preenchimento ósseo iniciou-se a partir das bordas e do fundo da lesão, com mobilização e diferenciação de células provenientes do periósteo e das meninges, respectivamente e, nas avaliações subseqüentes, a atividade osteoblástica originou-se, também, de "ilhas de ossificação" semelhantes a centros de ossificação, localizadas no centro da falha. A formação trabecular aumentou, proporcionalmente, com a concentração utilizada de BMP, e a aposição e organização óssea aumentaram com o tempo de observação. Verificou-se também a presença de tecido cartilaginoso. A BMP associada ao EOE contribuiu para a formação de novo tecido ósseo, promovendo maior mobilização, diferenciação e organização celular, e abreviou o tempo de formação óssea, sugerindo processo de ossificação endocondral. Os melhores resultados foram observados com a associação de 2mg de BMP a 3mg de enxerto, e a adição de BMP, mesmo em menor quantidade, determinou precocidade de formação óssea. A maior quantidade de BMP não determinou maior preenchimento ósseo


Aspects of bone repair were evaluated after implantation of bone morphogenetic proteins (BMP) in different concentrations. They were carried by autogenous bone graft in defects created on skulls of 20 adult, young female rabbits, randomizedly divided into five experimental groups and were observed at five times. After exposure of skull bones, six bone defects on the fronto-parietal region of each animal were performed. The defect I was not filled, the II was completed filled with 3mg of autogenous bone graft and the defects III, IV, V, and VI were filled with autogenous bone graft associated with 0.5; 1; 2 and 5mg of BMP, respectively. In the post-mortem mesoscopic evaluations, it was observed that, independently of the treatment period of the defects, the bony filling began from the borders to the center, and from the botton to the surface of the lessions. The bony filling of the defect I was the smallest when compared with the others defects, in all the observation moments. It was also verified that until 2mg the higher the concentration of BMP used, better was the bone cover. Microscopically, it was verified in the first evaluations, on the seventh day, that the bony growth started from the borders and from the bottom of the lesion, with mobilization and differentiation of cells deriving from the periosteum and the meninges, respectively. In the subsequent evaluations, the osteoblastic activity also derived from "ossification islands" to ossification centers, located in the center of the flaw. The trabecular formation increased proportionally with the concentration of BMP used, and the apposition and bony organization increased proportionally with the time of observation. The presence of cartilaginous tissue was verified in all the flaws. In conclusion, the use the higher concentration of BMP did not determinate the better new bone formation. Trinta e quatro ratos foram alocados em quatro grupos experimentais: sem imobilização (G1), com imobilização do joelho direito por 45 dias (G2), com imobilização e remobilização com atividade livre por cinco semanas (G3), imobilização e remobilização com atividade livre e natação por cinco semanas (G4). A imobilização interferiu negativamente na marcha e amplitude articular e o G4 apresentou melhor evolução na marcha nos cinco primeiros dias, em relação ao G3. Após esse período, a evolução foi similar. Os componentes do G2 apresentaram rigidez articular, não observada em G3 e G4. Histologicamente, a imobilização promoveu aumento da espessura da cápsula articular, evidenciada pela presença do tecido conjuntivo fibroso que substituiu o tecido adiposo no G2, mas em menor proporção em G3 e G4. A imobilização determinou perda de proteoglicanos da matriz cartilaginosa, aumento do número de condrócitos, dispostos de forma irregular, aumento da espessura da cartilagem calcificada, irregularidade da superfície articular, proliferação de tecido conjuntivo no espaço intra-articular e aumento da espessura do osso subcondral. O G3 apresentou maior número de alterações na cartilagem e osso subcondral, quando comparado com G4. A imobilização degenerou as células sinoviais, indicando diminuição da produção de fluido sinovial e do suprimento nutricional à cartilagem. Tanto a atividade livre quanto sua associação com a natação favoreceram o retorno das condições biomecânicas e da cápsula articular, anteriores à imobilização.


Thirty-four rats were randomly allocated into one of four experimental groups: without immobilization (G1), immobilization of the right knee joint for 45 days (G2), immobilization and remobilization with free activity for 5 weeks (G3), and immobilization and remobilization with free activity and swimming program for 5 weeks (G4). The immobilization was prejudicial to march and flexibility articular. Animals from G4 showed a better march evolution on the first five days as compared to those from G3. After that periods, march evolution were similar in both G3 and G4. Animals from G2 showed rigid joint while in those from G3 and G4 the articular movement was close to normal. The histological analysis pointed out that immobilization led to increase in articular capsule thickness evidenced by the presence of fibrous connective tissue replacing adipose tissue in G2, but proportionally less in G3 and G4. It was observed that immobilization determined loss of proteoglycans from the cartilaginous matrix, increase in the number of regularly arranged condrocytes, increase in calcified cartilage thickness, irregularity in the articular surface, proliferation of connective tissue in the intra-articular space and increase in subchondral bone thickness. The animals from G3 showed a greater number of alterations in both articular cartilage and subchondral bone, as compared to those from G4. Immobilization degenerated synovial cells indicating decreased synovial fluid production and reduced nutritional supplying to the cartilage. The free cage activity and its association with swimming influenced positively the return of biomechanics and articular capsule morphologic conditions to those before immobilization.

Na resseccao parcial do baco, a friabilidade de seu parenquima torna dificil a execucao de sutura hemostatica da superficie cruenta. O autor idealizou uma variante tecnica usando um reforco da sutura por meio de fitas de dacron, teflon, gelfoan, dura-mater ou do proprio epiploon do operado.Operou 50 caes nos quais foram praticadas esplenectomias parciais com a nova tecnica. Apos a ligadura dos vasos segmentares penetrantes, e praticada sutura transfixante no plano limitrofe do segmento a ressecar. Os pontos da sutura feitos com dexon ou catgut cromado tambem transfixam as duas fitas de reforco colocadas nas duas faces opostas do baco.A seccao do parenquima e realizada apos a sutura e em plano proximo a ela, na zona isquemiada. A peritonizacao da surpeficie cruenta e feita com o grande epiploon. Os animais foram sacrificados de 15 a 30 dias apos a intervencao. Os segmentos restantes do baco foram estudados macroscopicamente e microscopicamente, atestando o bom resultado da experimentacao
