RESUMEN

Porcine rotavirus A (PoRVA) is an enteric pathogen capable of causing severe diarrhea in suckling piglets. Investigating the prevalence and molecular characteristics of PoRVA in the world, including China, is of significance for disease prevention. In 2022, a total of 25,768 samples were collected from 230 farms across China, undergoing porcine RVA positivity testing. The results showed that 86.52% of the pig farms tested positive for porcine RVA, with an overall positive rate of 51.15%. Through the genetic evolution analysis of VP7, VP4 and VP6 genes, it was revealed that G9 is the predominant genotype within the VP7 segment, constituting 56.55%. VP4 genotypes were identified as P[13] (42.22%), P[23] (25.56%) and P[7] (22.22%). VP6 exhibited only two genotypes, namely I5 (88.81%) and I1 (11.19%). The prevailing genotype combination for RVA was determined as G9P[23]I5. Additionally, some RVA strains demonstrated significant homology between VP7, VP4 and VP6 genes and human RV strains, indicating the potential for human RV infection in pigs. Based on complete genome sequencing analysis, a special PoRVA strain, CHN/SD/LYXH2/2022/G4P[6]I1, had high homology with human RV strains, revealing genetic reassortment between human and porcine RV strains in vivo. Our data indicate the high prevalence, major genotypes, and cross-species transmission of porcine RVA in China. Therefore, the continuous monitoring of porcine RVA prevalence is essential, providing valuable insights for virus prevention and control, and supporting the development of candidate vaccines against porcine RVA.

Introduction: Pseudorabies virus (PRV) is a linear DNA virus with a double-stranded structure, capable of infecting a diverse array of animal species, including humans. This study sought to ascertain the seroprevalence of Pseudorabies Virus (PRV) in China by conducting a comprehensive collection of blood samples from 16 provinces over the course of 2022. Methods: The presence of PRV gE antibodies was detected through the utilization of an enzyme-linked immunosorbent assay (ELISA) technique. Logistic regression analysis was conducted to identify potential related factors associated with the serologic status of PRV gE at the animal level. Additionally, the SaTScan 10.1 software was used to analyze the spatial and temporal clusters of PRV gE seroprevalence. Results: A comprehensive collection of 161,880 samples was conducted, encompassing 556 swine farms throughout the country. The analysis revealed that the seroprevalence of PRV gE antibodies was 12.36% (95% confidence interval [CI], 12.20% to 12.52%) at the individual animal level. However, at the swine farm level, the seroprevalence was considerably higher, reaching 46.22% (95% CI, 42.08% to 50.37%). Related factors for PRV infection at the farm level included the geographic distribution of farms and seasonal variables. Moreover, five distinct high seroprevalence clusters of PRV gE were identified across China, with the peak prevalence observed during the months of April through June 2022. Conclusion: Our findings serve as a valuable addition to existing research on the seroprevalence, related factors, and temporal clustering of PRV gE in China. Furthermore, our study provides a reference point for the development of effective strategies for the prevention and control of pseudorabies and wild virus outbreaks.

Tibial dyschondroplasia (TD) is a skeletal deformity disease in broilers that occurs when vascularization in the growth plate (GP) is below normal. Although, blood vessels have been reported to contribute significantly in bone formation. Therefore, in the current study, we have examined the mRNA expression of angiogenesis-related genes in erythrocytes of thiram induced TD chickens by qRT-PCR and performed histopathological analysis to determine regulatory effect of recombinant Glutathione-S-Transferase A3 (rGSTA3) protein in response to the destructive effect of thiram following the injection of rGSTA3 protein. Histopathology results suggested that, blood vessels of GPs were damaged in thiram induced TD chicken group (D), it also affected the area and density of blood vessels. In the 20 and 50 µg·kg-1 of rGSTA3 protein-administered groups, E and F vessels appeared to be normal and improved on day 6 and 15. Furthermore, qRT-PCR results showed that rGSTA3 protein significantly (P < .05) up-regulated the expression of the most important angiogenesis-related integrin family genes ITGA2, ITGA5, ITGB2, ITGB3, ITGAV. The expression level of other genes including TBXA2R, FYN, IQGAP2, IL1R1, GIT1, RAP1B, RPL17, RAC2, MAML3, PTPN11, VAV1, PTCH1, NCOR2, CLU and ITGB3 up-regulated on dosage of rGSTA3 protein. In conclusion, angiogenesis is destroyed in thiram induced TD broilers, and rGSTA3 protein injection improved the vascularization of GPs by upregulating the angiogenesis related genes most importantly integrin family genes ITGAV, ITGA2, ITGB2, ITGB3, ITGA5.
