RESUMEN

BACKGROUND: In humans, it is now well documented that rising paternal age is correlated with decreased sperm DNA integrity and embryonic developmental failures. On the other side of the coin, it is also reported that very young fathers such as teenagers carry an increased risk of adverse birth outcomes. These observations suggest that, at least in humans, there is an age window for optimal sperm DNA integrity. In bovine, little is known about sperm DNA quality in young bulls and how it evolves with age. This study aimed to fill in this gap as it may be of importance for the bovine industry to know when exactly a bull is an optimal performer for reproductive programs. METHODS: Forty Nellore bulls were divided into three age groups: 1.8 to 2 years - young bulls; 3.5 to 7 years - adult bulls; and 8 to 14.3 years - aged bulls. Three ejaculates were collected from each bull, cryopreserved and evaluated for various parameters including: computer-assisted sperm analysis (CASA), plasma membrane and acrosome integrity, mitochondrial potential, sperm nuclear protamination, DNA oxidative damage, and Sperm Chromatin Structure Assay (SCSA). RESULTS: We report here that young bulls presented superior values for motility, plasma and acrosomal membrane integrity, and high mitochondrial potential. However, they also presented higher values for sperm morphological abnormalities compared to adult and aged animal groups (p < 0.05). In addition, young bulls exhibited more defective protamination than older animals did. The oldest bulls showed more nuclear oxidative damage than the younger groups of bulls while both the young and aged groups were found more susceptible to DNA denaturation as revealed with the SCSA test (p < 0.05). CONCLUSION: These results indicate that young bulls spermatozoa best survived the freezing procedure, followed by adult and aged bulls. However, young and aged bulls were found to be more susceptible to DNA damage, respectively caused by protamine deficiency and oxidation. Therefore, although young bulls have correct semen parameters according to classical evaluation, our results indicate that they may show some structural nuclear immaturity.


CONTEXTE: Chez l'homme, de nombreuses données indiquent maintenant que l'avancée de l'âge du père est associée à une réduction de l'intégrité de l'ADN des spermatozoïdes et aux échecs de développement embryonnaire. D'un autre côté, il est aussi rapporté que les jeunes pères, tels les adolescents, sont porteurs d'un risque accru d'issue défavorable de la grossesse. Ces observations suggèrent que, au moins chez l'humain, il existe une tranche d'âge dans laquelle l'intégrité de l'ADN des spermatozoïdes est optimale. Chez les bovins, on dispose de peu de connaissances sur la qualité de l'ADN des spermatozoïdes des jeunes taureaux et sur son mode d'évolution avec l'âge. La présente étude a pour but de combler ce manque car il peut être important, pour l'industrie bovine, de savoir à quelle période précise un taureau est un reproducteur optimal pour les programmes de reproduction. MATÉRIEL ET MÉTHODES: Quarante taureaux Nellore ont été répartis en trois groupes d'âge: 1, 8 à 2 ans ­ jeunes taureaux; 3,5 à 7 ans ­ taureaux adultes; et 8 à 14,3 ans ­ taureaux âgés. Trois éjaculats ont été collectés par taureau, cryopréservés et évalués pour différents paramètres incluant l'analyse assistée du sperme par ordinateur (CASA), l'intégrité des membranes plasmique et acrosomique, le potentiel mitochondrial, la protamination du noyau, l'altération oxydative de l'ADN et l'évaluation de la structure de la chromatine du noyau du spermatozoïde (SCSA). RÉSULTATS: Nous rapportons ici que les jeunes taureaux présentent des valeurs supérieures de la mobilité des spermatozoïdes et de l'intégrité des membranes plasmique et acrosomique, ainsi qu'un potentiel mitochondrial élevé. Cependant, les jeunes taureaux présentent aussi des valeurs plus élevées d'anomalies morphologiques des spermatozoïdes que celles des groupes adulte et âgé (p < 0.05). De plus, les jeunes taureaux ont une protamination plus défectueuse que celle des taureaux plus âgés. Les taureaux les plus âgés présentent plus d'altérations oxydatives du noyau que les jeunes taureaux alors que les deux groupes -jeunes et âgés- sont plus susceptibles d'avoir une dénaturation de l'ADN nucléaire comme indiqué par le SCSA (p < 0.05). CONCLUSIONS: Ces résultats indiquent que les spermatozoïdes des jeunes taureaux survivent le mieux au processus de congélation, suivis par les adultes puis les âgés. Toutefois, les jeunes taureaux et les âgés sont plus susceptibles d'avoir une altération de l'ADN, causée respectivement par une protamination déficiente et une oxydation. Par conséquent, bien que les jeunes taureaux aient des paramètres spermatiques corrects à l'évaluation classique, nos résultats indiquent que leurs spermatozoïdes peuvent présenter un certain degré d'immaturité structurale nucléaire.

The effects of ambient temperature and humidity, month, age and genotype on sperm production and semen quality in AI bulls in Brazil were evaluated. Data from two consecutive years were analyzed separately. Seven Bos indicus and 11 Bos taurus bulls from one artificial insemination (AI) center were evaluated in Year 1 and 24 B. indicus and 16 B. taurus bulls from three AI centers were evaluated in Year 2. Ambient temperature and humidity did not significantly affect sperm production and semen quality, probably because there was little variation in these variables. Month accounted for less than 2% of the variation in sperm production and semen quality. Increased bull age was associated with decreased sperm motility (P<0.10) and increased minor sperm defects (P<0.001) in Year 1. B. indicus bulls had greater (P<0.005) sperm concentration than B. taurus bulls in both years (1.7 x 10(9)/ml versus 1.2 x 10(9)/ml in Year 1 and 1.6 x 10(9)/ml versus 1.2 x 10(9)/ml in Year 2, respectively). Ejaculate volume was not significantly affected by genotype in Year 1 (6.6 ml versus 6.9 ml in B. indicus and B. taurus bulls, respectively), but B. indicus bulls had greater (P<0.05) total (11.4x10(9) versus 8.2 x 10(9)) and viable (6.7 x 10(9) versus 4.9 x 10(9)) numbers of spermatozoa in the ejaculate than B. taurus bulls. In Year 2, B. taurus bulls had greater (P<0.05) ejaculate volume than B. indicus bulls (8.2ml versus 6.7 ml, respectively) and total and viable number of spermatozoa in the ejaculate were not significantly different between genotypes (10.3 x 10(9) versus 9.1 x 10(9) and 6.1 x 10(9) versus 5.4 x 10(9) in B. indicus and B. taurus bulls, respectively). Sperm motility was not significantly affected by genotype (mean, 59%). In Year 1, B. indicus bulls tended (P<0.10) to have more major sperm defects and had more (P<0.05) total sperm defects than B. taurus bulls (11.8% versus 8.7% and 13.6% versus 10.0%, respectively). In Year 2, B. indicus bulls tended (P<0.10) to have more total sperm defects than B. taurus bulls (16.2% versus 13.3%, respectively). In conclusion, neither ambient temperature and humidity nor month (season) significantly affected sperm production and semen quality. B. indicus bulls had significantly greater sperm concentration and B. taurus bulls had significantly fewer morphologically defective spermatozoa.

The objectives were to determine the effects of age and genetic group on characteristics of the scrotum, testes and testicular vascular cones (TVC), and on sperm production and semen quality in 107 Bos indicus, B. taurus and cross-bred bulls at three artificial insemination (AI) centers in Brazil. In addition, predictors of sperm production and semen quality were identified. In general, scrotal circumference (SC), scrotal shape score, scrotal neck perimeter, and testicular size (length, width and volume) increased (P < 0.05) with age. Although there were no significant differences among genetic groups for SC or testicular size, B. indicus bulls had the least pendulous scrotal shape, the shortest scrotal neck length, and the greatest scrotal neck perimeter (P < 0.05). Fat covering the TVC was thinner (P < 0.05) in bulls < or = 36 months of age and in B. taurus bulls than in older bulls and B. indicus bulls, respectively. Age and genetic group did not affect testicular ultrasonic echotexture. B. indicus bulls tended (P < 0.1) to have the lowest average scrotal surface temperature (SST). In general, ejaculate volume, total number of spermatozoa and number of viable spermatozoa increased (P < 0.05) with age. However, there was no significant effect of age on sperm concentration, motility, major and total defects. The proportion of spermatozoa with minor defects was highest (P < 0.05) in bulls 37-60 months of age. B. indicus bulls had higher (P < 0.01) sperm concentration, total number of spermatozoa and number of viable spermatozoa than B. taurus bulls, with intermediate values for cross-bred bulls. Increased sperm production was associated with increased testicular volume, SC, TVC fat cover, and SST top-to-bottom gradient. Decreased semen quality was associated with increased SC and bottom SST, and decreased scrotal shape, scrotal neck perimeter and vascular cone diameter. In summary, age and genetic group affected the characteristics of the scrotum, testes, and TVC, sperm production and semen quality. In addition, characteristics of the scrotum, testes and TVC were associated with sperm production and semen quality in bulls and could be assessed for breeding soundness evaluation.

RESUMEN

Anomalous origin of the left main coronary artery from the right anterior aortic sinus is a rare condition (0.02 to 0.07%) and its clinical significance is sometimes controversial. The authors present the case of a 44-year-old female with this anomaly and septal course with clinical, electrocardiographic, scintigraphic and cineangiographic evidences of myocardial infarction without coronary lesions. Literature is reviewed regarding diagnosis, clinical significance, pathophysiology and management. It is probably the first evidence of the potential danger of this anomaly with septal course.

RESUMEN

A 60-year-old female with unstable angina was submitted to coronary arteriography. A 90% obstruction of the left main coronary artery ostium was detected, and the patient developed ventricular fibrillation. Circulatory support was instituted with cardiopulmonary bypass. Percutaneous transluminal coronary angioplasty was then successfully carried on. Ventricular fibrillation could be controlled and the patient was submitted to coronary artery bypass graft surgery 14 days later.

PURPOSE: To show the initial experience of Institute of Cardiology of Rio Grande do Sul in the treatment of congenital valvular aortic stenosis with percutaneous balloon aortic valvuloplasty. METHODS: Twenty four patients were submitted to the procedure, 14 males and 10 females. The mean age 7 years (4 days-17 years). Four patients were aged below 30 days and three patients had previous surgical valvuloplasty. The percutaneous balloon aortic valvuloplasty were made the retrograde approach in all patients. RESULTS: The peak systolic pressure gradient was reduced from 65.96 +/- 22.68 to 27.08 +/- 18.74 mmHg. The procedure resulted in aortic regurgitation in seven patients and worsened aortic regurgitation in two patients. One patient had cardiac arrest that was reverted by cardiopulmonary resuscitation, this patient had hospital discharge without sequel. Five patients had acute femoral artery thrombosis, and hemorrhage in the site of puncture happened in one patient. CONCLUSION: The percutaneous balloon aortic valvuloplasty results in effective reduction of the peak systolic pressure gradient, it is a save and effective therapy in patients with congenital valvular aortic stenosis. Further evaluation of the long term results are necessary for definitive conclusions.

The routine semen evaluation assessing sperm concentration, motility and morphology, does not identify subtle defects in sperm chromatin architecture. Bulls appear to have stable chromatin, with low levels of DNA fragmentation. However, the nature of fragmentation and its impact on fertility remain unclear and there are no detailed reports characterizing the DNA organization and damage in this species. The intensive genetic selection, the use of artificial insemination and in vitro embryo production associated to the cryopreservation process can contribute to the chromatin damage and highlights the importance of sperm DNA integrity for the success of these technologies. Frozen-thawed semen samples from three ejaculates from a Nellore bull showed high levels of morphological sperm abnormalities (55.8±5.1%), and were selected for complementary tests. Damage of acrosomal (76.9±8.9%) and plasma membranes (75.7±9.3%) as well as sperm DNA strand breaks (13.8±9.5%) and protamination deficiency (3.7±0.6%) were significantly higher compared to the values measured in the semen of five Nellore bulls with normospermia (24.3±3.3%; 24.5±6.1%; 0.6±0.5%; 0.4±0.6% for acrosome, plasma membrane, DNA breaks and protamine deficiency, respectively) (P<0.05). Motility and percentage of spermatozoa with low mitochondrial potential showed no differences between groups. This study shows how routine semen analyses (in this case morphology) may point to the length and complexity of sperm cell damage emphasizing the importance of sperm function testing.(AU)


O exame de rotina de sêmen, o qual avalia a concentração de espermatozoides, a motilidade e a morfologia, pode não identificar defeitos sutis na arquitetura da cromatina de espermatozoides. Os touros parecem ter cromatina estável com baixos níveis de fragmentação do DNA. No entanto, a natureza da fragmentação e o seu impacto sobre a fertilidade ainda não estão claros e não há relatos que caracterizam a organização do DNA e os danos nessa espécie com mais detalhes. A seleção genética intensiva e o uso da inseminação artificial e da produção in vitro de embriões, além do processo de criopreservação, podem contribuir para o dano da cromatina, e sabe-se a importância da integridade do DNA espermático para o sucesso dessas tecnologias. Amostras de sêmen de três ejaculados de um touro Nelore com altos níveis de alterações morfológicas (55,8±5,1%) foram selecionadas para realização de exames complementares. Os danos de acrossoma (76,9±8,9%) e das membranas plasmáticas (75,7±9,3%), bem como quebras de fita de DNA de espermatozoides (13,8±9,5) e deficiência de protamina (3,7±0,6) foram significativamente maiores em comparação aos valores avaliados no sêmen de cinco touros Nelore com normospermia (24,3±3,3%; 24,5±6,1%; 0,6±0,5%; 0,4±0,6% para acrossoma, membrana plasmática, quebras de DNA e deficiência de protamina, respectivamente) (p<0,05). Motilidade e porcentagem de espermatozoides com baixo potencial mitocondrial não diferiram estatisticamente. Essas avaliações mostram que análises de sêmen de rotina (neste caso, morfologia) podem apontar para a extensão e a complexidade dos danos na célula espermática, o que indica que a deficiência de protamina e os danos no DNA podem ocorrer simultaneamente a defeitos morfológicos. Tal ocorrência enfatiza a importância das análises de sêmen clássicas e dos testes complementares.(AU)

Objetivou-se avaliar as características morfológica e funcional do sêmen bovino congelado comparando-se a eficácia de dois diferentes diluidores. O ejaculado de quatro touros foi dividido em duas partes iguais, uma submetida ao diluidor Tris e gema de ovo (A) e outra ao diluidor à base de lecitina de soja (Andromed®) (B). No experimento I, cinco palhetas dos diluidores A e B de cada touro foram descongeladas e avaliadas quanto à motilidade, vigor, concentração, morfologia espermática e teste de termor-resistência lento. Foram feitas, ainda, avaliação da integridade de membranas, por meio da associação das sondas iodeto de propídio, isotiocionato de fluoresceína - Pisum sativum e carbocianina catiônica lipofílica, e avaliação funcional da membrana plasmática com teste hiposmótico. A avaliação da integridade da cromatina foi realizada pelo método de coloração com laranja de acridina. No experimento II, o sêmen com os diferentes diluidores foi utilizado na fecundação in vitro, sendo observadas taxas de clivagem e desenvolvimento embrionário in vitro. Em relação aos resultados obtidos, apenas a porcentagem de espermatozoides no sêmen congelado foi discretamente maior com o diluidor A, concluindo-se que o diluidor composto por lecitina de soja pode substituir o composto por Tris e gema de ovo, respeitando-se as variações individuais de cada touro utilizado no presente experimento.


The goal of this study was to evaluate the protective effect of egg yolk compared with a soybean lecithin-based extender on morphologic and functional characteristics of frozen bovine semen. The ejaculate of four bulls was divided into two equal parts, one diluted with egg-yolk-Tris extender (A) and the other with a soy-lecithin based extender (Andromed®) (B). In experiment I, five straws of extender A and B from each bull were thawed and assessed regarding motility (subjective and computerized analysis), vigor, concentration, sperm morphology and slow thermoresistance (STR), evaluation of membrane integrity through association of propidium iodite probes (PI), fluorescein isotiocianate - Pisum sativum (FITC-PSA) and lipophilic cationic carbocyanine (JC-1) and functional evaluation of the plasmatic membrane through Hyposmotic Swelling Test (HOST). An evaluation of chromatin integrity was performed with the acridine orange staining procedure. In experiment II, the frozen semen with different extenders was used for in vitro fertilization (IVF), analyzing cleavage rates and in vitro embryo development. No statistical difference between extenders was identified, suggesting that the soy lecithin extender may substitute egg-yolk-Tris extender, considering the individual variations of each bull used in this experiment.

Foram utilizados dez animais doadores de sêmen em nível de Central de Inseminaçäo Artificial, da raça Gir, divididos em dois grupos, de acordo com o grau de congelabilidade do sêmen de cada animal. Os animais com sêmen de alta congelabilidade foram aqueles cuja porcentagem de ejaculados viáveis pós-descongelaçäo foi superior a 80 por cento. O grupo de baixa congelabilidade tinha animais com porcentagem menor que 50 por cento de ejaculados viáveis pós-descongelaçäo. Os critérios de avaliaçäo da viabilidade do sêmen e seleçäo dos animais foram definidos pelo controle de qualidade do Departamento de Produçäo da Central de Inseminaçäo Artificial. Foram feitas quatro coletas semanais consecutivas, sendo que obtiveram-se as amostras de plasma seminal por centrifugaçäo a 1.500 g por 15 a 20 minutos a 4ºC, momentos após a coleta do sêmen em vagina artificial. O plasma seminal foi dialisado em membrana de celulose, em tampäo Tris-Glicina pH-7,4 por 24 horas a 4ºC, em agitaçäo lenta e constante. As amostras foram padronizadas em 1,0 mg/ml de proteína total, por diluiçäo em tampäo Tris-HCl 62mM pH-6,2 mais 20 por cento de glicerol e 4 por cento de SDS. Através de eletroforese do tipo SDS-PAGE, foram feitas as corridas em gel a 13 por cento. A corrida foi feita com a constante de 25 mA, por um período de 5 horas. A coloraçäo do gel foi feita por Coomassie Brilliant Blue. Pelos resultados obtidos, verificou-se que existe uma banda no grupo de alta congelabilidade, cujo fragmento polipeptídico desta proteína tem Mr (mobilidade relativa) 20,3 e PM (peso molecular) aproximado de 61.800 Da. Esta banda nao foi detectada nas amostras do grupo de baixa congelabilidade, o que sugere ser um possível marcador bioquímico quanto ao potencial de criopreservaçäo do sêmen de bovinos

O presente estudo utilizou 16 animais Bos taurus indicus da raça Nelore doadores de sêmen. Estes animais foram divididos em grupos de acordo com a idade em que o sêmen congelou pela primeira vez. O grupo I, considerado precoce, apresentou animais com sêmen passível de congelaçäo com idade inferior a 20 meses. O grupo II, composto por animais que tiveram o sêmen congelado com idade entre 21 e 26 meses. E o grupo III, tido como tardio, composto por animais com sêmen congelável com idade superior a 27 meses. Para análise dos padröes eletroforéticos da transferrina e albumina, amostras de sangue foram colhidas em tubos heparinizados e submetidos a centrifugaçäo de 2.500 G por 15 minutos para separaçäo do plasma sangüíneo. As amostras de plasma sangüíneo foram processadas para que a corrida eletroforética em gel de poliacrilamida pudesse ser realizada. Para a coloraçäo do gel, usou-se Coomasie Brilliant Blue. Após análise dos padröes eletroforéticos da transferrina e albumina, observou-se que näo houve relaçäo detectável entre os fenótipos da albumina e precocidade sexual de touros doadores. Entretanto, em relaçäo à transferrina, foi possível sugerir uma associaçäo entre o alelo Tf elevado a D com touros portadores de sêmen congelável precocemente ou medianamente em termos de idade à congelaçäo
