A multicenter cross-sectional study with the participation of 356 healthy children aged 1 to 3 years from the cities of the Central Federal District: Moscow (n=106), Ivanovo (n=126), Yaroslavl (n=124) has been carried out. Questionnaire method, the method of 24-hour diet recall for 3 days (2 weekdays and 1 weekend) and the method of diary recording of food intake were used. The average daily volumes of food were consistent with the recommended age norms only in 31.2% of the surveyed children; after 1 year of life children actively started to transfer to nutrition from the family table and only 51.7% of children continued to receive individually prepared for baby food. The recommended dietary allowances were inadequate in macro- and micronutrients: 48.6% of all children received excessive amounts of protein and fat, and 74.1% were less in carbohydrates. The consumption of vitamins D and A and minerals (calcium, iron and zinc) did not correspond to existing recommendations in the majority of cases: only 10-30% of the examined children received physiological norms of vitamins from food and 13-34% - some minerals and trace elements. These results indicate that the nutrition of young children living in the Central Federal District is imbalanced on daily caloric, macro- and micronutrient composition, does not fully meet the requirements of the balanced diet and needs serious optimization.

Proteoglycans play a crucial role in bone tissue formation promoting consolidation of collagen fibers and bonds between them and mineral crystals. The composition of extracellular matrix proteoglycans was analyzed in early aging OXYS rats with signs of early osteoporosis versus Wistar rats at the age of 2, 4, 6, and 8 months. It was found that bone mineral density in two month OXYS rats was lower than in age-matched Wistar rats. It increased only till 6 month of age in OXYS rats, and during the whole observation period in Wistar rats. Formation of peak bone mass in Wistar rats was accompanied by change in composition of proteoglycan pools in bone tissue matrix: accumulation of macromolecular keratan sulfate-containing proteoglycans, and decrease of uronic acid and sulfated glycosaminoglycan content. OXYS rats were noted for accumulation of micro- and macromolecular proteoglycans: uronic acids and sulfated glycosaminoglycans. Chondroitin sulfate AC and dermatan sulfate were accumulated in OXYS rat bone tissue with the increase in mineral density difference. Keratin sulfate was completely absent here, in contrast to Wistar rats.

A model of articular cavity with smooth cover was produced by influence of shape-forming element on regenerating bone tissue. Morphological, ultrastructural, histochemical, and biochemical peculiarities of the tissue of the formed articular surface are described.

The mode of Scheuermann's disease inheritance and its phenotypic traits in probands and their relatives were studied in 90 pedigrees (90 probands and 385 relatives). The disorder was identified as a genetically related pathology inherited by autosomal dominant type, controlled by a mutant major gene, as a kyphotic deformity without signs of vertebral bodies' anomaly and torsion. Morphological and biochemical studies showed disturbance in the structure of vertebral growth plate anterior aspects at the level of deformity, defects in proliferation and differentiation of chondrocytes, and change in proteoglycan spectrum in cells and matrix. Twelve candidate genes were studied in chondrocytes isolated from vertebral growth plates of patients with Scheuermann's disease. The study results included disorder in the IHH gene expression and preservation of the expression of PAX1, two aggrecan isoforms, link protein, types I and II collagen, lumican, versican, growth hormone and growth factor receptor genes, and proliferation gene. Preservation of the SOX9 gene (transcription gene) probably indicates posttranscriptional genetic disorders. The study is under way.

The concentrations of keratan sulfates and unmodified keratan sulfates increased in the vertebral body growth plate in patients with idiopathic scoliosis. Sulfation and acetylation of total glycosaminoglycans decreased by 50 and 30%, respectively. These changes reflect the decrease in biological activity of molecules that modulate function of the growth plate.

In 24 hours after a single administration of melleril (30 mg/kg) an elevated injuriousness of "heavy" and "light" liver lysosomes of rats subjected to "in vitro" treatment with a pH 5.0 solution at 37 degrees was recorded. Introduction of melleril to rats with toxic CCl4-induced hepatitis (CCl4 dosage of 0.15 ml per 100 g body weight) depresses free activity of acid phosphatase, this fall, however, failing to reach the corresponding values in intact animals. An elevated injuriousness of "heavy" and "light" lysosomas undergoing treatment at pH 5.0 and 37degrees, revealed in all variants of the tests, is related to activation of lysosomal phospholipases with subsequent liberation of surfactant lipids and volumetric changes of the particles.

Subcellular distribution of hyaluronidase of rat liver was studied during spontaneous restoration during the 1st, 2nd, 3rd days, and 1, 2, 3, 4 weeks of acute and chronic CCl4-hepatitis. Redistribution of relative specific activity of hyaluronidase was seen in the fractions of heavy and light mitochondria, microsomes and supernatant fraction, after an acute hepatic injury. Four weeks after the injury subcellular distribution was not yet normal. In chronic hepatitis the relative specific hyaluronidase activity was almost identical in the five subcellular fractions. In the course of restoration the relative specific activity was increased in the fractions of heavy and light mitochondria and microsomes, but decreased in the nuclear and supernatant fractions.

Acute hypobaric hypoxia in rats (260 mm Hg., 90 min) was accompanied by a greater liver lysosomes osmotic susceptibility and by an increase in the relative content of the lysosomal enzymes (acid RNAase and acid phosphatase) in the nuclear fraction. This indicates an enrichment of the liver cell lysosomes with secondary lysosomes. No significant signs of labilization of the liver lysosomes were found. The adaptation of rats to hypoxia, as well as administration of the antihypoxant guthimin or of 1,4-bis-(3'-morpholinopropin-1'-yl-1') benzene hinder these changes to occur in the liver lysosomes during the organism reaction to hypoxia. But the effect of stabilization of the lysosomal membranes in a minor component in the antihypoxia action of the treatment.
