RESUMEN

Incomplete milking (IM) is one way of mitigating the negative energy balance (NEB) that is characteristic for early lactation and may increase the risk for disease. Our objectives were to test the effects of IM in early lactation on energy balance (EB), metabolic status, udder health, and subsequent performance. To facilitate the practical application, an automated system was used to remove the milking clusters once a predefined amount of milk is withdrawn. Forty-six Holstein cows were equally allocated to either the treatment (TRT, starting on 8 d in milk) or the control group (CON; conventional cluster removal at milk flow rate <0.3 kg/min). Milk removal in the TRT group was limited to the individual cow's milk yield 1 d before IM started and held constant for 14 d. Thereafter, all cows were conventionally milked and records related to EB, performance, and udder health were continued up to 15 wk of lactation. During the 14 d of IM, on average 11.1% less milk was obtained from the TRT cows than from the CON cows. Thereafter, milk yield increased in the TRT group, eliminating the group difference throughout the remaining observation period until wk 15 of lactation. The TRT cows tended to have less dry matter intake and also water intake than the CON cows. The extent of the NEB and the circulating concentrations of fatty acids, ß-hydroxybutyrate, insulin-like growth factor-1, and leptin mostly did not differ between the groups. The IM did not affect body condition. Udder health was maintained over the entire observation period in all cows. Our results demonstrate the applicability of the automated cluster removal for limiting milk withdrawal to a defined amount in early lactation. However, it remains to be determined whether the absent effect on energy metabolism was due to the relatively stable energy status of the cows or to the relatively mild IM setting used herein.

The interrelationships between silage preference of ruminants and compounds that may affect forage choice are not yet fully enlightened. Analysis of the forage metabolome in addition to conventional chemical analysis and preference trials can provide new insights. Six silage treatments each of alfalfa (AL) and red clover (RC), with different dry matter concentrations (222-391 g/kg), silage additives, and intended addition of soil, were produced in quadruplicate to obtain a range of qualities. After 120 d of ensiling, silages were sampled for chemical analysis, vacuum-packed, and refrigerated for subsequent preference trials with goats. Within 21 d, each possible combination of 2 silages and an AL hay that served as control (n = 21) was presented to goats (Saanen-type wethers, n = 8, body weight 105 ± 2.7 kg) for 3 h for ad libitum intake. Comparisons among means for 3-h dry matter intake (DMI) for forages offered in choice situations were made using variance analysis, including terms for treatment and animal and the Waller-Duncan k-ratio t-test to separate means. The most preferred and avoided treatments of AL and RC silage amounted to 863, 858, 226, and 282 g DMI/3 h, respectively. To further explore relations between silage composition and preference, a metabolome analysis of the most preferred and most avoided AL and RC treatments were conducted. Metabolites (all low molecular weight molecules) were analyzed by a nontargeted metabolite profiling in the range of 50-1,700 Da. Metabolites showing the most distinct difference between preferred and avoided silages were identified by partial least squares discriminant analysis. In the 2 selected treatments of each plant species (those that were most different in forage preference), more than 6,400 compounds were detected and 2,010 were identified. Between preferred and avoided treatments, 934 of the detected compounds differed in RC and 1,860 in AL, of which 475 were altered in both plant species (251 were reduced and 186 were increased; only 38 behaved contrarily, meaning that they were increased in one substrate and decreased in the other). The database provides a useful foundation for the approach of explaining silage preference by ruminants.

Information about the individual herbage DMI (HDMI) of grazing dairy cows is important for an efficient use of pasture herbage as an animal feed with a range of benefits. Estimating HDMI, with its multifaceted influencing variables, is difficult but may be attempted using animal, performance, behavior, and feed variables. In our study, 2 types of approaches were explored: 1 for HDMI estimation under a global approach (GA), where all variables measured in the 4 underlying experiments were used for model development, and 1 for HDMI estimation in an approach without information about the amount of supplements fed in the barn (WSB). The accuracy of these models was assessed. The underlying data set was developed from 4 experiments with 52 GA and 50 WSB variables and one hundred thirty 7-d measurements. The experiments differed in pasture size, herbage allowance, pregrazing herbage mass, supplements fed in the barn, and sward composition. In all the experiments, cow behavioral characteristics were recorded using the RumiWatch system (Itin and Hoch GmbH, Liestal, Switzerland). Herbage intake was estimated by applying the n-alkane method. Finally, HDMI estimation models with a minimal relative prediction error of 11.1% for use under GA and 13.2% for use under WSB were developed. The variables retained for the GA model with the highest accuracy, determined through various selection steps, were herbage crude protein, chopped whole-plant corn silage intake in the barn, protein supplement or concentrate intake in the barn, body weight, milk yield, milk protein, milk lactose, lactation number, postgrazing herbage mass, and bite rate performed at pasture. Instead of the omitted amounts of feed intake in the barn and, due to the statistical procedure for model reduction, the unconsidered variables postgrazing herbage mass and bite rate performed at pasture, the WSB model with the highest accuracy retained additional variables. The additional variables were total eating chews performed at pasture and in the barn, total eating time performed at pasture, number of total prehension bites, number of prehension bites performed at pasture, and herbage ash concentration. Even though behavioral characteristics alone did not allow a sufficiently accurate individual HDMI estimation, their inclusion under WSB improved estimation accuracy and represented the most valid variables for the HDMI estimation under WSB. Under GA, the inclusion of behavioral characteristics in the HDMI estimation models did not reduce the root mean squared prediction error. Finally, further adaptation, as well as validation on a more comprehensive data set and the inclusion of variables excluded in this study such as body condition score or gestation, should be considered in the development of HDMI estimation models.

Observation of ingestive and rumination behaviors of dairy cows may assist in detecting diseases, controlling reproductive status, and estimating intake. However, direct observation of cows on pasture is time consuming and can be difficult to realize. Consequently, different systems have been developed to automatically record behavioral characteristics; among them is the RumiWatch System (RWS; Itin and Hoch GmbH, Liestal, Switzerland). Until now, the RWS has not been thoroughly validated under grazing conditions. The aim of the current study was to validate the RWS, against direct observation, in measuring ingestive and rumination behaviors of dairy cows during grazing and supplementation in the barn. A further objective was to examine whether it is possible to refine the algorithm used by the evaluation software RumiWatch Converter 0.7.3.2 to improve the accuracy of the RWS. The data were collected from an experiment carried out with 18 lactating Holstein cows in a crossover block design including 3 treatments and 3 measuring periods. All cows grazed night and day, 19 h/d, and were either unsupplemented or supplemented, with chopped whole-plant corn silage, or chopped whole-plant corn silage mixed with a protein concentrate. During the measuring periods, cows were equipped with the RumiWatch Halter, and their ingestive and rumination behaviors were recorded concurrently by the RumiWatch Halter and by direct observation (690 × 10 min). Comparison of concurrently measured data shows that the RWS detected jaw movements reliably, but classification errors occurred. A low relative prediction error of ≤0.10 for the number of rumination boluses, rumination chews, and total eating chews was found. A high relative prediction error of >0.10 was found for the number of prehension bites and time spent in prehension and eating. Both converter versions performed equally well in differentiating ingestive and rumination behaviors when cows were supplemented in the barn or when grazing and supplementation activities were combined. For grazing cows, with no supplementation, more reliable results for the total number of eating chews, rumination chews, prehension bites, and time spent in these activities were obtained, by using the RumiWatch Converter 0.7.3.11. In light of these findings, further research is warranted to improve the accuracy of the RWS and to allow a differentiation between mastication chews and prehension bites while eating.

Rumen health is of vital importance in ensuring healthy and efficient dairy cattle production. Current feeding programs for cattle recommend concentrate-rich diets to meet the high nutritional needs of cows during lactation and enhance cost-efficiency. These diets, however, can impair rumen health. The term "subacute ruminal acidosis" (SARA) is often used as a synonym for poor rumen health. In this review, we first describe the physiological demands of cattle for dietary physically effective fiber. We also provide background information on the importance of enhancing salivary secretions and short-chain fatty acid absorption across the stratified squamous epithelium of the rumen; thus, preventing the disruption of the ruminal acid-base balance, a process that paves the way for acidification of the rumen. On-farm evaluation of dietary fiber adequacy is challenging for both nutritionists and veterinarians; therefore, this review provides practical recommendations on how to evaluate the physical effectiveness of the diet based on differences in particle size distribution, fiber content, and the type of concentrate fed, both when the latter is part of total mixed ration and when it is supplemented in partial mixed rations. Besides considering the absolute amount of physically effective fiber and starch types in the diet, we highlight the role of several feeding management factors that affect rumen health and should be considered to control and mitigate SARA. Most importantly, transitional feeding to ensure gradual adaptation of the ruminal epithelium and microbiota; monitoring and careful management of particle size distribution; controlling feed sorting, meal size, and meal frequency; and paying special attention to primiparous cows are some of the feeding management tools that can help in sustaining rumen health in high-producing dairy herds. Supplementation of feed additives including yeast products, phytogenic compounds, and buffers may help attenuate SARA, especially during stress periods when the risk of a deficiency of physically effective fiber in the diet is high, such as during early lactation. However, the usage of feed additives cannot fully compensate for suboptimal feeding management.

The nutrition of captive giraffe (Giraffa camelopardalis), a browsing ruminant, is challenging because browse availability is limited in zoos and rations need to be composed of compensatory feeds. In this study, ration composition for giraffes in 12 German zoos was documented and linked to animal variables that indicate suitability of nutrition. Rations differed in proportion of ration items and chemical composition resulting in various grades of accordance with feeding recommendations. An estimated daily metabolisable energy (ME) intake (MEI; mean ± SD) of 0.61 MJ ME/kg0.75 body weight (BW; ±0.1) was sufficient to cover estimated energy requirements. Daily dry matter (DM) intake (DMI) was 61 g DM/kg0.75 BW (±10) and correlated negatively to dietary ME content (p = .009; r = -.596). Apparently, feed intake was regulated by energetic satiety and not by physical properties of forage. A negative correlation between produce proportion and DMI (p = .002; r = -.676) led to the assumption of a low ruminal pH in giraffes fed high proportions of produce. Increasing dietary forage proportions led to an increasing duration of feed intake (p = .045; r = .477) and decreasing occurrence of oral stereotypies (p = .047; r = -.474). The weighted average faecal particle size was larger than reported for free-ranging giraffes, but no relation to ration characteristics among the facilities existed. The abrasiveness of rations was not excessive, as contents of silicate in faeces were similar to values from the wild. Body condition was generally acceptable, but there was no evident relation to ration characteristics. The capacity to self-regulate DM and ME intakes with lucerne hay may work at higher forage proportions than often assumed for captive giraffes. Rations with less energetic density can result in a greater DMI, including maximisation of forage intake and reduction of oral stereotypies.

The prediction of renal energy excretion is crucial in a metabolizable energy system for horses. Phenolic acids from forage cell walls may affect renal energy losses by increasing hippuric acid excretion. Therefore, the relationships were investigated between renal energy, nitrogen (N) and hippuric acid excretion of four adult ponies (230-384 kg body weight (BW)) consuming diets based on fresh grass, grass silage, grass cobs (heat-dried, finely chopped, pressed grass), alfalfa hay, straw, extruded straw and soybean meal. Feed intake was measured; urine and faeces were quantitatively collected for three days. Feed was analysed for crude nutrients, gross energy, amino acids and neutral-detergent-insoluble crude protein (CP); faeces were analysed for crude nutrients and cross energy; urine was analysed for N, hippuric acid, creatinine and gross energy. Renal energy excretion (y; kJ/kg BW0.75 ) correlated with renal N excretion (x1 ; g/kg BW0.75 ) and renal hippuric acid excretion (x2 ; g/kg BW0.75 ): y = 14.4 + 30.2x1 +20.7x2 (r = .95; n = 30; p < .05). Renal hippuric acid excretion was highest after intake of fresh grass and lowest after intake of soybean meal. The ratio of hippuric acid to creatinine in urine and the excretion of hippuric acid per gram of dry matter intake was significantly higher for fresh grass than for all other rations. There was no relationship between aromatic amino acid intake and renal hippuric acid excretion. The results of the present study and literature data suggest that feed can be categorized into four groups with regard to the energy losses per gram CP intake: (i) protein supplements (e.g., soybean meal): 4.2-4.9 kJ/g CP intake (ii) alfalfa hay, grains, dried sugar beet pulp: 6.4 kJ/g CP intake, (iii) hay, preserved grass products, straw: 5.2-12.3 kJ/g CP intake (mean 8) and (iv) fresh grass. For group (iii) a negative relationship was observed between renal energy losses per gram of CP and the content of CP or neutral-detergent-insoluble CP in dry matter.

Lactating ruminants require an adequate supply of absorbable amino acids for the synthesis of milk protein from two sources, that is crude protein (CP) synthesized microbially in the rumen and ruminally undegraded CP (RUP) from feed which can both be digested in the small intestine. Several chemical and physical methods have been identified as being effective in increasing the proportion of RUP of total CP of a feedstuff, yet there is a continuing need for developing and establishing methods which protect feed protein from ruminal degradation with acceptable expenditure of labour and other costs. The objective of this study was to identify and quantify effects of and interactions between chlorogenic acid and protein in solvent-extracted sunflower meal (SFM) as induced by alkali treatment. Response surface methodology was employed to investigate the influence of pH, reaction time and drying temperature on the resulting SFM and, subsequently, its protein value for ruminants estimated from laboratory values. For this purpose, alkali-treated SFM was subjected to a fractionation of feed CP according to the Cornell net carbohydrate and protein system as a basis for estimating RUP at different assumed ruminal passage rates (Kp ). To estimate the intestinal digestibility of the treated SFM and its RUP, a three-step enzymatic in vitro procedure was applied. Alkaline treatment of SFM increased RUP values with factors ranging from approximately 3 (Kp =.08/hr) to 12 (Kp =.02/hr). Furthermore, the intestinal digestibility of the alkali-treated SFM was enhanced by approximately 10% compared to untreated SFM. Increasing pH and reaction time led to both increasing RUP values and intestinal digestibility. In conclusion, a targeted alkaline treatment of naturally occurring compounds in feedstuffs might be a promising approach to provide high-RUP feeds for ruminants which, at the same time, have improved intestinal digestibility values.

The objective of this study was to test whether diet selection of dairy cows under grazing conditions could be estimated using plant wax markers. Furthermore, differences between 2 cow strains and the effect of concentrate supplementation on plant species selection were investigated. The experiment was a study with a crossover design performed on an organic farm with 12 Swiss Holstein cows and 12 New Zealand Holstein cows. Both experimental periods consisted of a 21-d adaptation and a 7-d measurement period. All cows grazed full time in a rotational stocking system and received either no concentrate or 6 kg/d of a commercial cereal-grain mix. Representative herbage samples of each grazed paddock were taken and botanical composition of subsamples was manually determined. The average proportions of the plant species were 27.8% Lolium perenne, 6.1% Dactylis glomerata, 10.4% Trifolium repens, and 9.0% Taraxacum officinale. Other grass species were merged as "other grass" (38.2%) and other forb species as "other forbs" (8.5%). n-Alkanes, long-chain fatty acids, and long-chain alcohols (LCOH) were analyzed in the samples of plant species, concentrate, and feces from each cow. A linear discriminant analysis indicated that diet components were differentiated best with LCOH (96%) and worst with the combination of all marker groups together (12%). For each marker, the fecal marker recovery (FR) relative to dosed ytterbium was determined in 2 ways. Estimation of diet composition was performed with the software "EatWhat," and results were compared with botanical composition with the Aitchison distance. The results indicate that the diet composition of grazing dairy cows can be estimated using plant wax markers. Additionally, the calculation of FR led to mostly reliable results, yet this approach needs further validation. The most accurate estimation was achieved with the marker combination of n-alkanes and LCOH with a correction for FR. Less accurate estimations were achieved with long-chain fatty acids alone or in combination with n-alkanes. No difference relating to diet selection between the 2 cow strains was recorded, but supplemented cows apparently ingested higher proportions of T. repens than nonsupplemented cows. Awareness that supplementation influences selection behavior of grazing dairy cows may lead to adaptations in botanical composition of the pasture according to the demand of the animals.

Carnivores do not vary markedly in their digestive efficiency for protein and fat, but whether they resemble other trophic guilds (omnivores and herbivores) in this respect has not been evaluated. We collated data on apparent crude protein (CP) and crude fat (ether extracts, EE) digestibility in 157 mammal species, applying the Lucas principle of regressing digestible nutrient content against nutrient content, where the slope of the regression equation represents the true digestibility and the intercept the metabolic losses per unit dry matter intake. The data collection is marked by the evident uneven distribution of dietary nutrient contents across trophic guilds and differences in the nutrient range by which different species have been evaluated, making statistical interpretation difficult. Results indicate a lower true digestibility of CP in herbivores compared to carnivores, most likely due to a lower digestibility of fibre-bound protein in herbivore diets. Metabolic CP losses did not appear to differ between trophic guilds, but herbivores had higher metabolic EE losses, compatible with the hypothesis that a higher proportion of metabolic CP losses were bound in microbes that also contain lipids in herbivores. Among herbivores, no clear pattern was evident that would indicate a difference in metabolic losses associated with microbes between digestive strategies (coprophagy, foregut/hindgut fermentation). Foregut fermenters had a lower true EE digestibility, possibly linked to the hydrogenation of lipids in their forestomach prior to digestion. The results do not demonstrate clear differences in digestive efficiency and metabolic losses for protein and fat between mammalian trophic guilds and digestive strategies, leading to the hypothesis that the process of CP and EE digestion is not physiologically challenging and hence does not lead to a noticeable differentiation between species or species groups.

Growth, apparent nutrient digestibility, ileal digesta viscosity, and energy metabolizability of growing turkeys fed diets containing malted sorghum sprouts (MSP) supplemented with enzyme or yeast were investigated using 120, 28-day-old male turkeys. Six treatments were laid out in a 3 × 2 factorial arrangement of treatments with three dietary inclusion levels of MSP (0, 50, and 100 g/kg) and supplemented with 200 mg/kg yeast (Saccharomyces cerevisiae) or 200 mg/kg of a commercial enzyme. The experiment lasted for the starter (day 28-56) and grower phases (day 57-84) of the birds. Each treatment group consisted of 20 turkeys replicated four times with five birds each. Data were analysed using analysis of variance while polynomial contrast was used to determine the trends (linear and quadratic) of MSP inclusion levels. Irrespective of dietary supplementation with enzyme or yeast, final body weight (BW), total BW gain, and feed intake for turkey poults from day 29-56 was reduced (p < 0.05) with increasing inclusion level of MSP. Dietary supplementation with yeast resulted in increased (p < 0.05) feed intake while enzyme supplementation improved (p < 0.05) feed conversion ratio of the poults. Turkeys fed enzyme-supplemented MSP diets had higher (p < 0.05) BW gain than their counterparts fed yeast-supplemented MSP diets. Apparent ash digestibility reduced linearly (p < 0.05) with increasing inclusion levels of MSP. Apparent metabolizable energy (AME) did not vary significantly (p > 0.05) with MSP inclusion levels. Enzyme supplementation reduced (p < 0.05) ileal viscosity but had no effect (p > 0.05) on AME. Inclusion of MSP resulted in poor growth performance. This confirms earlier studies that utilization of MSP by poultry is rather poor. Supplementation with enzyme or yeast did not lead to any appreciable improvement in performance of turkeys in this study.

Energy balance in dairy cows changes during the course of lactation due to alterations in voluntary feed intake and energy required for milk synthesis. To adapt to the demands of lactation, energy metabolism needs to be regulated and coordinated in key organs such as adipose tissue (AT), liver, and mammary gland. Mitochondria are the main sites of energy production in mammalian cells and their number varies depending on age, organ, and physiological condition. The copy number of the mitochondrial genome, the mitochondrial DNA (mtDNA), reflects the abundance of mitochondria within a cell and is regulated by transcriptional and translational factors. Environmental, physiological, and energetic conditions change during lactation and we thus hypothesized that these changes may influence the mtDNA copy number and the abundance of genes regulating mitochondrial biogenesis. Therefore, we aimed to provide an overview of mitochondrial biogenesis in liver, subcutaneous (sc)AT, mammary gland, and peripheral blood cells during early and late lactation in dairy cows. German Holstein cows (n=21) were fed according to their requirements, and biopsies from scAT, liver, mammary gland, and blood were collected in early and late lactation and assayed for relative mtDNA copy numbers and the mRNA abundance of genes regulating mitochondrial biogenesis, such as nuclear-respiratory factor 1 and 2 (NRF-1, NRF-2), mitochondrial transcription factor A (TFAM), and peroxisome proliferator-activated receptor-gamma coactivator 1-α (PGC-1α). The number of mtDNA copies increased from early to late lactation in all tissues, whereas that in peripheral blood cells was greater in early compared with late lactation. Moreover, mitochondrial activity enzymes (i.e., citrate synthase and cytochrome c oxidase) increased from early to late lactation in scAT. Comparing the number of mtDNA copies between tissues and blood in dairy cows, the highest mtDNA content was observed in liver. The mRNA abundance of genes related to mitochondrial biogenesis changed in a tissue-specific manner when comparing early versus late lactation. The mtDNA copy number was associated with transcriptional factors only in AT, suggesting nontranscriptional regulation of mtDNA in the other tissues. We detected strong correlations between peripheral blood mtDNA and tissue mtDNA content in early lactation. Peripheral blood forms an appropriate medium to display the cellular content of mtDNA copy numbers and consequently the cellular energy status of tissues during early lactation.

Telomeres create a protective cap on the ends of chromosomes that shorten with cell division and are influenced by stressful conditions. With the onset of lactation, high-yielding dairy cows are exposed to metabolic stress. In the present study, we aimed to analyze telomere length (TL) in key metabolic organs, such as liver, subcutaneous (sc) adipose tissue (AT), and mammary gland, as well as in peripheral blood cells during early and late lactation in German Holstein cows (n=21). Animals were fed according to their requirement, and biopsies from scAT, liver, and mammary gland as well as blood cells were collected in early and late lactation. The relative quantity of telomere products (qT), which is proportional to the average TL, was determined in genomic DNA by multiplex quantitative PCR. In this study, relative qT varied widely in the investigated tissues and blood. In late lactation, slowly proliferating tissues, such as liver and scAT, had the highest qT, whereas peripheral blood cells and in the mammary gland had the lowest qT. Comparing early with late lactation, relative qT attrition was limited to blood and mammary gland. Relationships between relative qT in blood, mammary gland, scAT, and liver suggest that blood qT might serve as a surrogate marker for tissue-specific qT. Cows with high initial qT in tissues and blood in early lactation had greater qT attrition during the course of lactation than cows with lower qT. The determination of qT could be included when phenotyping dairy cattle to test for associations with performance and fitness traits.

Feed efficiency is an important trait of dairy production. However, assessing feed efficiency is constrained by the associated cost and difficulty in measuring individual feed intake, especially on pastures. The objective of this study was to investigate short-term feed efficiency traits of herbage-fed dairy cows and screening of potential biomarkers (n = 238). Derived feed efficiency traits were ratio-based (i.e., feed conversion ratio (FCR) and N use efficiency (NUE)) or residual-based (i.e., residual feed intake (RFI), residual energy intake (REI), and residual N intake (RNI)). Thirty-eight Holstein and 16 Swiss Fleckvieh dairy cows underwent a 7-d measurement period during mid- and/or late-lactation. The experimental data (n = 100 measurement points) covered different lactational and herbage-fed system situations: mid-lactation grazing (n = 56), late-lactation grazing (n = 28), and late-lactation barn feeding (n = 16). During each measuring period, the individual herbage intake of each cow was estimated using the n-alkane marker technique. For each cow, biomarkers representing milk constituents (n = 109), animal characteristics (n = 13), behaviour, and activity (n = 46), breath emissions (n = 3), blood constituents (n = 35), surface, and rectal temperature (n = 29), hair cortisol (n = 1), and near-infrared (NIR) spectra of faeces and milk (n = 2) were obtained. The relationships between biomarkers and efficiency traits were statistically analysed with univariate linear regression and for NIR spectra using partial least squares regression with feed efficiency traits. The feed efficiency traits were interrelated with each other (r: -0.57 to -0.86 and 0.49-0.81). The biomarkers showed varying R2 values in explaining the variability of feed efficiency traits (FCR: 0.00-0.66, NUE: 0.00-0.74, RFI: 0.00-0.56, REI: 0.00-0.69, RNI: 0.00-0.89). Overall, the feed efficiency traits were best explained by NIR spectral characteristics of milk and faeces (R2: 0.25-0.89). Biomarkers show potential for predicting feed efficiency in herbage-fed dairy cows. NIR spectra data analysis of milk and faeces presents a promising method for estimating individual feed efficiency upon further validation of prediction models. Future applications will depend on the ability to improve the robustness of biomarkers to predict feed efficiency in a greater variety of environments (locations), managing conditions, feeding systems, production intensities, and other aspects.

Despite the fact that llamas have become increasingly popular as companion and farm animals in both Europe and North America, scientific knowledge on their nutrient requirements is scarce. Compared with other livestock species, relatively little is known especially about the nutrient and energy requirements for lactating llamas. Therefore, we aimed to measure milk output in llama dams using an isotope dilution technique and relate it to energy intakes at different stages of lactation. We also validated the dilution technique by measuring total water turnover (TWT) directly and comparing it with values estimated by the isotope dilution technique. Our study involved 5 lactating llama dams and their suckling young. Milk output and TWT were measured at 4 stages of lactation (wk 3, 10, 18, and 26 postpartum). The method involved the application of the stable hydrogen isotope deuterium ((2)H) to the lactating dam. Drinking water intake and TWT decreased significantly with lactation stage, whether estimated by the isotope dilution technique or calculated from drinking water and water ingested from feeds. In contrast, lactation stage had no effect on dry matter intake, metabolizable energy (ME) intake, or the milk water fraction (i.e., the ratio between milk water excreted and TWT). The ratios between TWT measured and TWT estimated (by isotope dilution) did not differ with lactation stage and were close to 100% in all measurement weeks, indicating that the D(2)O dilution technique estimated TWT with high accuracy and only small variations. Calculating the required ME intakes for lactation from milk output data and gross energy content of milk revealed that, with increasing lactation stage, ME requirements per day for lactation decreased but remained constant per kilogram of milk output. Total measured ME intakes at different stages of lactation were similar to calculated ME intakes from published recommendation models for llamas.

The objective of this study was to improve knowledge regarding the amino acid profile of the insoluble portion of ingested forage escaping rumen degradation. Six forage categories were analyzed. Categories varied in botanical composition and each contained 2 samples. Samples within categories were derived from the same parent material but differed in harvest, maturity, or conservation type. The rumen-undegradable protein of all forages was measured by incubation for 16h in the rumen of 3 nonlactating cows. All residues were corrected for microbial colonization. The AA profile of the residue was different to the original profile. Degradation trends of individual AA, in terms of increase or decrease relative to the original concentration, were similar between all forages. The AA profiles of forage residues, both within and between categories, were more similar to each other than to their respective original profile. This information may aid in improving the accuracy of estimating postruminal AA supply from forages while decreasing the number of samples required to be analyzed.

The present study aimed to investigate whether phagocytic immune cells infiltrate into bovine adipose tissue (AT) and to study the effects of lactation and conjugated linoleic acid (CLA) supplementation on the invasion of phagocytic immune cells into different s.c. and visceral (v.c.) fat depots of primiparous dairy cows during the first 105 d in milk (DIM). German Holstein-Friesian cows (HF; n = 25) with a mean body condition score of 3.0 were divided into a control (CON) and a CLA group. From 1 DIM until sample collection, CLA cows were fed 100 g of CLA supplement/d (about 6% of c9,t11 and t10,c12 isomers each), whereas the CON cows received 100 g/d of a fatty acid mixture instead of CLA. The CON cows (n = 5 each) were slaughtered at 1, 42, and 105 DIM, and the CLA cows (n = 5 each) were slaughtered at 42 and 105 DIM. Adipose tissues (n = 150) from 3s.c. (tailhead, withers, and sternum) and 3 v.c. (omental, mesenteric, and retroperitoneal) depots were sampled. In addition, s.c. tailhead biopsies were collected by repeated surgical biopsies (3 samplings within 7 wk; n = 36) from 12 nonpregnant, nonlactating Simmental heifers (SM; mean body condition score = 5.0) fed diets of varying energy density to compare the changes in phagocytic immune cell infiltration with early lactating cows. Immunohistochemical analyses of different fat depots revealed a low incidence of phagocytic immune cell infiltration in early lactating cows. The portion of infiltrating macrophages (CD68+) in a few positive AT samples of HF cows was slightly lower in s.c. than v.c. fat and was positively correlated with both empty body weight and adipocyte size. However, no differences with regard to DIM and CLA supplementation were observed in HF cows. Increased accumulation of phagocytic immune cells, albeit at low cell numbers, in nonpregnant, over-conditioned SM heifers might be related to larger adipocytes secreting higher amounts of chemoattractant adipokines compared with the early lactating cows. In conclusion, the extent of fatness in HF cows may not be high enough to stimulate significant infiltration of phagocytic cells in AT and, therefore, these immune cells might have no major role in the immunologic and metabolic adaptations of AT during early lactation.

The present study was conducted to investigate the effect of dietary Rhodobacter capsulatus on lipid fractions and egg-yolk fatty acid composition in laying hens. Thirty-six laying hens (30 weeks old) were randomly assigned into two dietary groups fed diets with (0.04%) or without (control) R. capsulatus for a 60-day feeding trial. Dietary R. capsulatus decreased (p < 0.05) serum and hepatic cholesterol and increased (p < 0.05) the excreta cholesterol, and resultant lower (p < 0.05) cholesterol contents in egg yolk. The concentration of polyunsaturated fatty acids (PUFA) and ratio to saturated fatty acids in egg yolk was improved (p < 0.05) by dietary R. capsulatus. The concentration of hepatic bile acid was increased (p < 0.05) and excreta bile acid was decreased (p < 0.01) in the laying hens fed R. capsulatus diet. The incorporation of 1-(14) C-palmitic acid into hepatic lipids and lipid fractions was increased (p < 0.05) in laying hens fed R. capsulatus diet. Moreover, dietary R. capsulatus did not appear to cause any adverse effects on laying hen performances. Therefore, dietary supplementation of R. capsulatus in layer diets may be a feasible means of producing eggs with lower cholesterol and higher PUFA contents for health conscious consumers.

Seasonal chemical composition and ruminal organic matter (OM) and crude protein (CP) degradabilities were determined in four tropical multi-purpose tree species (MPTS) namely; Pterocarpus santalinoides, Grewia pubescens, Enterolobium cyclocarpum and Leucaena leucocephala. Three West African dwarf (WAD) rams fitted with permanent rumen cannula were used for the degradability trials. Foliage samples were collected four times to represent seasonal variations as follows: January--mid dry; April--late dry; July--mid rainy and October--late rainy seasons. Leaf samples were randomly collected from the trees for estimation of dry matter (DM) and chemical composition. Ruminal in sacco OM and CP degradabilities were estimated from residues in nylon bags. All samples had high CP (161-259 g/kg DM) and moderate fibre concentrations [neutral detergent fibre (without residual ash], 300-501 g/kg DM; acid detergent fibre (without residual ash), 225-409 g/kg DM and acid detergent lignin, 87-179 g/kg DM across seasons. Interaction effects of species and season on chemical composition were highly significant (p = 0.001) except for trypsin inhibitor (p = 0.614). The MPTS recorded more than 60% OM and CP degradability at 24 h, which implied that they were all highly degradable in the rumen. Their incorporation into ruminant feeding systems as dry season forage supplements is therefore recommended.
