RESUMEN

We have developed a novel aromatic fluorine-induced one-pot synthesis of ring-perfluorinated trimethine cyanine dye without the use of a pyridine by reacting hexafluorobenzoindolenine with 5 equiv of methyl trifluoromethanesulfonate in mixed solvents of dimethylformamide and toluene. The thus-obtained ring-perfluorinated trimethine cyanine dye shows much better fluorescence properties, including intensity, quantum yield, and lifetime, than the nonfluorinated dye, not only in CH2Cl2 solution and the poly(methyl methacrylate) film but also in the powder state. Furthermore, ring-perfluorinated trimethine cyanine dye 2a shows better photostability toward white light-emitting diode irradiation than nonfluorinated dye trimethine cyanine dye 2b.

RESUMEN

The high density nucleation of α-Al2O3 nanocrystallites was observed by rapid heating of the aluminum formate hydroxide-based precursor powder at 1200 °C for 50 s. The nucleation of α-Al2O3 nanocrystallites with less 10 nm in size from high purity aluminum oxide matrix has not been observed to our knowledge. Based on the results of XRD and TEM, α-Al2O3 nanocrystallites nucleated from the amorphous phase which formed after thermal decomposition of the precursor powder. Subsequently, α-Al2O3 with hollow rod-like morphology formed through coalescence and growth of nanocrystallites after heating at 1200 °C for 1 min. The results obtained in this paper indicates a possible beneficial effect of the rapid heating and cooling of the aluminum formate hydroxide-based precursor powder on the precipitation of α-Al2O3 nanocrystallites.

RESUMEN

S-Equol is a metabolite of daidzein, a type of soy isoflavone, and three reductases are involved in the conversion of daidzein by specific intestinal bacteria. S-Equol is thought to prevent hormone-dependent diseases. We previously identified the equol producing gene cluster (eqlABC) of Eggerthella sp. YY7918. Daidzein reductase (DZNR), encoded by eqlA, catalyzes the reduction of daidzein to dihydrodaidzein (the first step of equol synthesis), which was confirmed using a recombinant enzyme produced in Escherichia coli. Here, we purified recombinant DZNR to homogeneity and analyzed its enzymological properties. DZNR contained FMN, FAD, and one 4Fe-4S cluster per 70-kDa subunit as enzymatic cofactors. DZNR reduced the CC bond between the C-2 and C-3 positions of daidzein, genistein, glycitein, and formononetin in the presence of NADPH. R-Dihydrodaidzein and R-dihydrogenistein were highly stereo-selectively produced from daidzein and genistein. The Km and kcat for daidzein were 11.9 µM and 6.7 s-1, and these values for genistein were 74.1 µM and 28.3 s-1, respectively. This enzyme showed similar kinetic parameters and wide substrate specificity for isoflavone molecules. Thus, this enzyme appears to be an isoflavone reductase. Gel filtration chromatography and chemical cross-linking analysis of the active form of DZNR suggested that the enzyme consists of an octameric subunit structure. We confirmed this by small-angle X-ray scattering and transmission electron microscopy at a magnification of ×200,000. DZNR formed a globular four-petal cloverleaf structure with a central vertical hole. The maximum particle size was 173 Å.

Zirconium oxy-salts were hydrolyzed to form positively charged polymer or cluster species in acidic solutions. The zirconium hydrolyzed polymer was found to react with a negatively charged polyelectrolyte, such as poly(vinyl sulfate), and to form a stoichiometric polyion complex. Thus, colloidal titration with poly(vinyl sulfate) was applied to measure the zirconium concentration in an acidic solution by using a Toluidine Blue selective plasticized poly(vinyl chloride) membrane electrode as a potentiometric end-point detecting device. The determination could be performed with 1% of the relative standard deviation. The colloidal titration stoichiometry at pH < or = 2 was one mol of zirconium per equivalent mol of poly(vinyl sulfate).
