This study aimed to investigate the central involvement of 5-HT1A receptors in the nociceptive behavior of mice submitted to the chronic constriction injury (CCI) of sciatic nerve and the subsequent application of photobiomodulation (PBM). Male mice (Swiss-albino) were submitted to CCI and subsequently received an infusion of WAY100635 (5-HT1A receptor antagonist) or intracerebroventricular saline (ICV), followed by infrared laser irradiation (808 nm), in continuous mode, with the power of 100 mW and a dose of 0 J/cm2 (control group) or 50 J/cm2. The thermal hyperalgesia was evaluated by hot plate test, while mechanical allodynia was evaluated by von Frey filaments. After CCI, animals showed a reduction in the nociceptive threshold (p<0.001) when compared to the sham group. In von Frey test, the CCI + saline + PBM 50 J/cm2 group showed an increase in nociceptive threshold (p<0.001) in all measurement moments in comparison with groups CCI + SALINE + PBM 0 J/cm2, CCI + WAY100635 + PBM 50 J/cm2, and CCI + WAY100635 + PBM 0 J/cm2. Similarly, in hot plate test, CCI + SALINE + PBM 50 J/cm2 group showed an increase in nociceptive threshold after application of PBM at 120 and 180 min. Because of the results found, it can be suggested the involvement of 5-HT1A receptors in the central nervous system, since WAY100635 was able to reverse the antinociceptive effect provided by PBM in animals submitted to CCI.

BACKGROUND: Haloperidol (HAL) is an antipsychotic used in the treatment of schizophrenia. However, adverse effects are observed in the extrapyramidal tracts due to its systemic action. Natural compounds are among the treatment alternatives widely available in Brazilian biodiversity. Mygalin (MY), a polyamine that was synthesized from a natural molecule present in the hemolymph of the Acanthoscurria gomesian spider, may present an interesting approach. AIMS: This study aimed to evaluate the effect of MY in mice subjected to HAL-induced catalepsy. METHODS: Male Swiss mice were used. Catalepsy was induced by intraperitoneal administration of HAL (0.5 mg/kg - 1 mL/Kg) diluted in physiological saline. To assess the MY effects on catalepsy, mice were assigned to 4 groups: (1) physiological saline (NaCl 0.9 %); (2) MY at 0.002 mg/Kg; (3) MY at 0.02 mg/Kg; (4) MY at 0.2 mg/Kg. MY or saline was administered intraperitoneally (IP) 10 min b HAL before saline. Catalepsy was evaluated using the bar test at 15, 30, 60, 90, and 120 min after the IP administration of HAL. RESULTS: The latency time in the bar test 15, 30, 60, and 90 min increased (p < 0.05) after IP administration of HAL compared to the control group. Catalepsy was attenuated 15, 30, 90, and 120 min (p < 0.05) after the IP-administration of MY at 0.2 mg/Kg; while MY at 0.02 mg/Kg attenuated catalepsy 15 min after the HAL treatment. Our findings showed that MY attenuates the HAL-induced cataleptic state in mice.

Microglia are glial cells centrally related to pathophysiology and neuroimmunological regulation of pain through microglia-neuron crosstalk mechanisms. In contrast, anti-inflammatory mechanisms guided by immunological effectors such as IL-10 trigger the secretion of analgesic substances, culminating in the differential expression of genes encoding endogenous opioid peptides, especially ß-endorphin. Thus, when ß-endorphin binds to the µ-opioid receptor, it generates neuronal hyperpolarization, inhibiting nociceptive stimuli. This review aimed to summarize the recent advances in understanding the mechanism by which IL-10/ß-endorphin can reduce pain. For this, databases were searched for articles from their inception up until November 2022. Two independent reviewers extracted the data and assessed the methodological quality of the included studies, and seventeen studies were considered eligible for this review. Several studies have demonstrated the impact of IL-10/ß-endorphin in reducing pain, where IL-10 can stimulate GLP-1R, GRP40, and α7nAChR receptors, as well as intracellular signaling pathways, such as STAT3, resulting in increased ß-endorphin expression and secretion. In addition, molecules such as gabapentinoids, thalidomide, cynandione A, morroniside, lemairamin, and cinobufagin, as well as non-pharmacological treatments such as electroacupuncture, reduce pain through IL-10 mediated mechanisms, reflecting a microglia-dependent ß-endorphin differential increase. This process represents a cornerstone in pain neuroimmunology knowledge, and the results obtained by different studies about the theme are presented in this review.

ABSTRACT BACKGROUND AND OBJECTIVES: Studies have demonstrated that the cannabinoid CB1 receptor is involved in the modulation of pain, mainly by activating the descending pain control pathway. However, the role of photobiomodulation in this process is not well elucidated. Thus, the present study aimed to investigate the involvement of the CB1 receptor in the supraspinal photobiomodulation-induced antinociception. METHODS: Male albino swiss mice were submitted to chronic constriction injury and treated with photobiomodulation. To evaluate the supraspinal involvement of the CB1 receptor in the photobiomodulation-induced antinociception, the cannabinoid CB1 receptor antagonist AM251 (0.1µg/vol 0.2µL) was injected 5 minutes before the photobiomodulation treatment. The photobiomodulation treatment was performed on the fifth day after the stereotactic surgery and chronic constriction injury at a dose of 50J/cm2 in acute condition. The hot plate and von Frey monofilaments tests were performed to evaluate the thermal and mechanical pain sensitivity, respectively. RESULTS: The thermal and mechanical nociceptive threshold was higher in mice with chronic constriction injury, injected with saline and treated with photobiomodulation at the dose of 50J/cm2 in both the hot plate (p<0.001) and von Frey (p>0.001) tests. These antinociceptive effects were not detected in mice with chronic constriction injury pre-treated with AM251. CONCLUSION: The present study suggests that CB1 receptors located in Supraspinal structures, participate in the control of neuropathic pain following photobiomodulation treatment in animals undergoing chronic constriction injury.


RESUMO JUSTIFICATIVA E OBJETIVOS: Estudos demonstraram que o receptor canabinóide CB1 está envolvido na modulação da dor, principalmente pela ativação da via descendente de controle da dor, porém o papel da fotobiomodulação nesse processo não é bem elucidado. Assim, o presente estudo teve como objetivo investigar o envolvimento do receptor CB1 na antinocicepção induzida pela fotobiomodulação a nível supraespinhal. MÉTODOS: Camundongos machos suíço albinos foram submetidos à lesão por constrição crônica e tratados com fotobiomodulação. Para avaliar o envolvimento supraespinhal do receptor CB1 na antinocicepção induzida por fotobiomodulação foi injetado o antagonista do receptor canabinóide CB1, AM251 (0,1µg/vol 0,2µL) 5 minutos antes do tratamento com fotobiomodulação. O tratamento de fotobiomodulação foi realizado no quinto dia após cirurgia estereotática e lesão por constrição crônica, na dose de 50J/cm2 em estado agudo. Os testes de placa quente e monofilamentos de von Frey foram realizados para avaliar a sensibilidade térmica e mecânica à dor, respectivamente. RESULTADOS: O limiar térmico e mecânico nociceptivo foi maior nos camundongos com lesão por constrição crônica, injetados com solução salina e tratados com fotobiomodulação na dose de 50J/cm2 nos testes de placa quente (p<0,001) e von Frey (p>0,001). Esses efeitos antinociceptivos não foram detectados em camundongos com lesão por constrição crônica tratados com AM251. CONCLUSÃO: O presente estudo sugere que os receptores CB1 localizados nas estruturas supraespinhais participam do controle da dor neuropática, após tratamento com fotobiomodulação em animais submetidos à lesão por constrição crônica.

Alterações no equilíbrio são observadas em hemiparéticos após um acidente vascular encefálico (AVE), gerando impactos físicos, sociais e econômicos significativos; assim, uma avaliação foi desenvolvida para facilitar os planos de tratamento para os pacientes. A goniometria é atualmente usada como ferramenta de avaliação da amplitude de movimento (ADM); no entanto, o desenvolvimento de exergames trouxe uma nova perspectiva para a avaliação, que utiliza um mecanismo dinâmico e tem pouca subjetividade. Objetivo: Este estudo avaliou a confiabilidade inter e intraexaminadores da medida da ADM da abdução do ombro em pacientes pós-AVE usando o exergame PhysioPlay™. Método: Treze voluntários com AVE crônico, com idade de 58,23 ± 9,96 anos (homens e mulheres), participaram deste estudo. Dois fisioterapeutas avaliaram a abdução do ombro usando goniometria e o exergame PhysioPlay™. Um reteste foi realizado uma semana depois. Resultados: Os resultados das análises utilizando o coeficiente de correlação intraclasse (CCI) mostraram excelente nível de confiabilidade inter e intraexaminadores (r> 0,90; p <0,05). A correlação de Pearson entre as medidas máximas obtidas na goniometria e o software PhysioPlay™ apresentou alta correlação (r> 0,90, p = 0,001). Conclusão: O Kinect associado ao exergame PhysioPlay™ apresentou excelente confiabilidade na captura da medida da ADM em comparação à goniometria.


Changes in balance are observed in some hemiparetics following a stroke, generating significant physical, social, and economic impacts; thus, an assessment was developed to facilitate treatment plans for patients. Goniometry is currently used as an evaluation tool for range of motion (ROM); however, the development of exergames has brought a new perspective to the assessment, which uses a dynamic mechanism and has little subjectivity. Objective: This study evaluates the inter-and intra-rater reliability of ROM measurement of the shoulder abduction in post-stroke patients using the exergame PhysioPlay™. Method: Thirteen volunteer chronic stroke survivors, aged 58.23 ± 9.96 years (men and women), participated in this study. Two physiotherapists evaluated the abduction of the shoulder using goniometry and the exergame PhysioPlay™. A retest was performed one week later. Clinical trial registry number ­ RBR-55smwr. Results: The results of the analyses using intraclass correlation coeficient (ICC) showed an excellent inter- and intra-rater reliability level (r > 0.90; p < .05). The Pearson correlation between the maximum measures obtained in the goniometry and the software PhysioPlay™ showed a high correlation (r > 0.90, p= .001). Conclusion: The Kinect associated with the exergame PhysioPlay™ presented excellent reliability in capturing the ROM measure compared to the conventional goniometry.

Introduction: This study aims to analyze the influence of quick massage and pilatespilates on bank employees. The project was sent to the Research Ethics Committee and obtained a favorable opinion (1,045,371 / 2015). Methods: It is a clinical, controlled, randomized, and blind study. The study had had initially involved 70 employeesparticipants, but only 50 were included. They were divided into 3 groups: quick massage group (n = 20), ppilates group (n = 20) and control group (n = 10). They were evaluated with experimental times of 3, 6 and 9 months and the protocol was applied 3 times per week. Results: The results of the present study were presented in standard deviation and mean of all groups, then a one-way analysis of the variance was performed, obtaining a value of P <0.0198, being considered significant. However, it is noted that when presenting the detailed data of each group (mean and standard deviation) it is observed that the group which received the quick massage had a small advantage.: (6 months) 4.03 ± 0.82, (12 months) 3.43 ± 1.15 and pilatespilates group (3 months) 6.21 ± 1.98016, (6 months) 4.26 ± 0.57873 (9 months) 4.14 ± 0.63. Conclusions: The quick massage techniques and the pilatespilates protocol have shown great potential in reducing anxiety, depression and fatigue. (AU)
