RESUMEN

Hexavalent chromium exposure via inhalation, ingestion, or both has been proven to adversely affect internal organs, induce toxic effects, cause allergies, and contribute to the development of cancer. It requires a substantial and challenging effort to detect several heavy metal ions conveniently, sensitively, and reliably by using materials that are easy to synthesize and have a high yield. The impact of light on the electrocatalytic oxidation/reduction process proves an environmentally friendly methodology with numerous applications in pollution control. The extensive use of photoactive materials in photoelectrochemical (PEC) sensors necessitates the development of stable and highly effective photoactive materials. Hence, the solvothermal synthesis of the organic-inorganic hybrid nanocomposite ß-Bi2O3-Bi2WO6/H2TPP with varying weight percentages of meso-tetraphenylporphyrin (H2TPP) resulted in a selective electrode for electrocatalytic and photoelectrocatalytic reduction of Cr6+ on fluorine-doped tin oxide (FTO) by an adsorption-reduction mechanism. H2TPP increases the active site density and provides an effective surface area for efficient adsorption by providing both pyridinic- and pyrrolic-N atoms to ß-Bi2O3-Bi2WO6/H2TPP. H2TPP could effectively adsorb Cr6+ in the ß-Bi2O3-Bi2WO6/H2TPP composite system through electrostatic interaction, and the adsorbed Cr6+ ions were reduced to trivalent chromium Cr3+, resulting in promising Cr6+ sensing. The projected density of states and Bader charge calculations result in the electrostatic attraction among the N-2p orbital of H2TPP and the 3d and 4s orbitals of the Cr atom, resulting in the adsorption of the hexavalent Cr atom onto the active center of H2TPP. Moreover, the addition of H2TPP results in the development of a mesoporous surface that offers strong electrical conductivity, a substantial surface area, improved charge-mass transport, intimate contact between the electrolyte and catalyst, an extended fluorescence lifetime, and increased stability. The role of pH values was thoroughly investigated. All electrochemical and photoelectrochemical studies were carried out on 5 wt % H2TPP-ornated ß-Bi2O3-Bi2WO6. Nanocomposite ß-Bi2O3-Bi2WO6/5 wt % H2TPP demonstrated reliable cyclic stability, reproducibility, good sensitivity (8.005 µA mM cm-2), and a low limit of detection (LOD) (8.0 nM) toward photoelectrocatalytic reduction of Cr6+. The interference study in the presence of a few inorganic entities exhibited excellent selectivity. This tale amplification approach for developing a ß-Bi2O3-Bi2WO6/5 wt % H2TPP nanocomposite system suggests a deeper understanding of the application of photoelectrocatalytic reduction of Cr6+ in environmental remediation with real samples under light irradiation.

A selective electrode for oxygen reduction reaction (ORR) and electrocatalytic reduction of 4-nitrophenol (p-NP) was fabricated on a glassy carbon electrode using organic-inorganic Bi2MoO6/H2TPP nanocomposites with different weight percentages of tetraphenylporphyrin, synthesized by the solvothermal process. Materials thus synthesized were characterized through UV-Vis diffuse reflectance spectroscopy, X-ray photoelectron spectroscopy (XPS), field emission scanning electron microscopy (FE-SEM), energy-dispersive X-ray spectroscopy (EDX), high-resolution transmission electron microscopy (HR-TEM), Fourier transform infrared spectroscopy (FT-IR), and X-ray diffraction (XRD) analysis. The electrocatalytic performance of the modified electrode toward ORR in the 0.1 M KOH solution, the onset potential Eonset (0.942 V), E1/2 (0.704 V) vs RHE, Jd (-5.545 mA cm-2), and n = 4 physicochemical parameters were well appreciable. It exhibits good catalytic activity toward ORR through a four-electron pathway with excellent stability and high active site density, and thus, the in situ Porphy-decorated metal oxide system facilitates the electron transport process. High selectivity and efficacy for the oxygen reduction reaction (ORR) are a significant measure for several energy-converting applications. The decorated electrode, glassy carbon electrode (GCE)/Bi2MoO6/3 wt % Porphy, serves as an electrochemical sensor that exhibited good sensitivity (0.4683 µAµM-1 cm-2), good reproducibility, a low detection limit (0.0940 µM), and long-term stability in the aqueous phase without any appreciable effect in the presence of some common organic and inorganic interferences for the detection of p-NP in a linear concentration range of 0.5-350 µM. Therefore, the material performs as an effective electrode for both the ORR and the electrocatalytic reduction of p-NP with real matrix samples at room conditions.

BACKGROUND: Grass pea (Lathyrus sativus) is an underutilised crop with high tolerance to drought and flooding stress and potential for maintaining food and nutritional security in the face of climate change. The presence of the neurotoxin ß-L-oxalyl-2,3-diaminopropionic acid (ß-L-ODAP) in tissues of the plant has limited its adoption as a staple crop. To assist in the detection of material with very low neurotoxin toxin levels, we have developed two novel methods to assay ODAP. The first, a version of a widely used spectrophotometric assay, modified for increased throughput, permits rapid screening of large populations of germplasm for low toxin lines and the second is a novel, mass spectrometric procedure to detect very small quantities of ODAP for research purposes and characterisation of new varieties. RESULTS: A plate assay, based on an established spectrophotometric method enabling high-throughput ODAP measurements, is described. In addition, we describe a novel liquid chromatography mass spectrometry (LCMS)-based method for ß-L-ODAP-quantification. This method utilises an internal standard (di-13C-labelled ß-L-ODAP) allowing accurate quantification of ß-L-ODAP in grass pea tissue samples. The synthesis of this standard is also described. The two methods are compared; the spectrophotometric assay lacked sensitivity and detected ODAP-like absorbance in chickpea and pea whereas the LCMS method did not detect any ß-L-ODAP in these species. The LCMS method was also used to quantify ß-L-ODAP accurately in different tissues of grass pea. CONCLUSIONS: The plate-based spectrophotometric assay allows quantification of total ODAP in large numbers of samples, but its low sensitivity and inability to differentiate α- and ß-L-ODAP limit its usefulness for accurate quantification in low-ODAP samples. Coupled to the use of a stable isotope internal standard with LCMS that allows accurate quantification of ß-L-ODAP in grass pea samples with high sensitivity, these methods permit the identification and characterisation of grass pea lines with a very low ODAP content. The LCMS method is offered as a new 'gold standard' for ß-L-ODAP quantification, especially for the validation of existing and novel low- and/or zero-ß-L-ODAP genotypes.

Here, iron chalcogenide thin films were developed for the first time by using the less hazardous electrodeposition technique at optimized conditions on an FTO glass substrate. The chalcogenides have different surface, morphological, structural, and optical properties, as well as an enzyme-free sensing behavior toward urea. Numerous small crystallites of about ∼20 to 25 nm for FeSe, ∼18 to 25 nm for FeTe, and ∼18 to 22 nm in diameter for FeSeTe are observed with partial agglomeration under an electron microscope, having a mixed phase of tetragonal and orthorhombic structures of FeSe, FeTe, and, FeSeTe, respectively. Profilometry, XRD, FE-SEM, HR-TEM, XPS, EDX, UV-vis spectroscopy, and FT-IR spectroscopy were used for the analysis of binary and ternary composite semiconductors, FeSe, FeTe, and FeSeTe, respectively. Electrochemical experiments were conducted with the chalcogenide thin films and urea as the analyte in phosphate-buffered media at a pH of ∼ 7.4 in the concentration range of 3-413 µM. Cyclic voltammetry was performed to determine the sensitivity of the prepared electrode at an optimized scan rate of 50 mV s-1. The electrodeposited chalcogenide films appeared with a low detection limit and satisfactory sensitivity, of which the ternary chalcogenide film has the lowest LOD of 1.16 µM and the maximum sensitivity of 74.22 µA µM-1 cm-2. The transition metal electrode has a very wide range of detection limit of 1.25-2400 µM with a short response time of 4 s. This fabricated biosensor is capable of exhibiting almost 75% of its starting activity after 2 weeks of storage in the freezer at 4 °C. Simple methods of preparation, a cost-effective process, and adequate electrochemical sensing of urea confirm that the prepared sensor is suitable as an enzyme-free urea sensor and can be utilized for future studies.

A new Mannich base (2-(4-(2-hydroxy-3-methoxy-5-methylbenzyl)-piperazin-1-yl)methyl)-6-methoxy-4-ethylphenol (H2L) and its tetranuclear NiII complex [Ni4L2(µ1,1-Cl)2(H2O)4]Cl2 (compound 1) are characterised using single-crystal X-ray diffraction measurements. Compound 1 contains four different NiII centres in a rhombus-like structure. Two Ni atoms (Ni1 and Ni2) have a NiN2O4 coordination sphere, while the other two (Ni3 and Ni4) have a NiO4Cl2 coordination environment and Ni-Cl-Ni bridges connect them. Ni1 and Ni2 are linked to Ni3 and Ni4 by phenoxide bridges. Variable temperature magnetic susceptibility measurements of 1 indicate the presence of alternating antiferromagnetic coupling (J1 = -6.6(1) cm-1) through the phenoxide bridge along the sides of the rhombus and ferromagnetic coupling (J2 = 8.4(1) cm-1) through the double Cl bridge along the short diagonal of the rhombus with a zero-field splitting of |D| = 2.7(1) cm-1. Compound 1 shows oxidase (catecholase-like and phenoxazinone synthase-like) activity. The oxidation of 3,5-di-tert-butylcatechol (3,5-DTBC) gives 3,5-di-tert-butylquinone (3,5-DTBQ) and H2O2 and the oxidation of o-aminophenol (OAP) produces 2-aminophenoxazin-3-one (APX) and H2O with turnover numbers of 28.32 and 17.52 h-1, respectively, under aerobic conditions. A mechanism for the oxidase activity catalysed by compound 1 is proposed in line with ESI-mass spectrometry, EPR spectroscopy, and electrochemical data. The reaction involves the cleavage of the tetranuclear Ni4-core to form a mononuclear NiII complex in the presence of the substrate (3,5-DTBC/OAP). This NiII complex is reduced to NiI with the concomitant oxidation of the substrate (3,5-DTBQ/APX). Formation of a radical intermediate is confirmed using EPR. In the catecholase-like activity, O2 is reduced to H2O2 while in the phenoxazinone synthase-like activity O2 produces H2O. Compound 1 participated in oxygen evolution reaction (OER) and hydrogen evolution reaction (HER) in a strongly basic medium with an onset potential of 418 mV and a Tafel slope of 121 mV dec-1 for OER and an onset potential of 477 mV and Tafel slope of 146 mV dec-1 for HER.

Grass pea (Lathyrus sativus L.) is a rich source of protein cultivated as an insurance crop in Ethiopia, Eritrea, India, Bangladesh, and Nepal. Its resilience to both drought and flooding makes it a promising crop for ensuring food security in a changing climate. The lack of genetic resources and the crop's association with the disease neurolathyrism have limited the cultivation of grass pea. Here, we present an annotated, long read-based assembly of the 6.5 Gbp L. sativus genome. Using this genome sequence, we have elucidated the biosynthetic pathway leading to the formation of the neurotoxin, ß-L-oxalyl-2,3-diaminopropionic acid (ß-L-ODAP). The final reaction of the pathway depends on an interaction between L. sativus acyl-activating enzyme 3 (LsAAE3) and a BAHD-acyltransferase (LsBOS) that form a metabolon activated by CoA to produce ß-L-ODAP. This provides valuable insight into the best approaches for developing varieties which produce substantially less toxin.

The Australian sheep blowfly Lucilia cuprina is a major pest for the Australian and New Zealand sheep industries. With the long-term aim of making a strain of L. cuprina suitable for a genetic control program, we previously developed a tetracycline-repressible female lethal genetic system in Drosophila. A key part of this system is a female-specific promoter from a yolk protein (yp) gene controlling expression of the tetracycline-dependent transactivator (tTA). Here we report the sequence of a 14.2 kb genomic clone from L. cuprina that contains a cluster of three complete yp genes and one partial yp gene. The Lcyp genes are specifically expressed in females that have received a protein meal. A bioinformatic analysis of the promoter of one of the yp genes (LcypA) identified several putative binding sites for DSX, a known regulator of yp gene expression in other Diptera. A transgenic strain of L. cuprina was made that contained the LcypA promoter driving the expression of the Escherichia coli lacZ reporter gene. Transgenic females express high levels of ß-galactosidase after a protein meal. Thus the LcypA promoter could be used to obtain female-specific expression of tTA in transgenic L. cuprina.

RESUMEN

'Speed breeding' (SB) shortens the breeding cycle and accelerates crop research through rapid generation advancement. SB can be carried out in numerous ways, one of which involves extending the duration of plants' daily exposure to light, combined with early seed harvest, to cycle quickly from seed to seed, thereby reducing the generation times for some long-day (LD) or day-neutral crops. In this protocol, we present glasshouse and growth chamber-based SB approaches with supporting data from experimentation with several crops. We describe the conditions that promote the rapid growth of bread wheat, durum wheat, barley, oat, various Brassica species, chickpea, pea, grass pea, quinoa and Brachypodium distachyon. Points of flexibility within the protocols are highlighted, including how plant density can be increased to efficiently scale up plant numbers for single-seed descent (SSD). In addition, instructions are provided on how to perform SB on a small scale in a benchtop growth cabinet, enabling optimization of parameters at a low cost.

BACKGROUND: Germ-line transformation of insects is now a widely used method for analyzing gene function and for the development of genetically modified strains suitable for pest control programs. The most widely used transposable element for the germ-line transformation of insects is piggyBac. The site of integration of the transgene can influence gene expression due to the effects of nearby transcription enhancers or silent heterochromatic regions. Position effects can be minimized by flanking a transgene with insulator elements. The scs/scs' and gypsy insulators from Drosophila melanogaster as well as the chicken beta-globin HS4 insulator function in both Drosophila and mammalian cells. RESULTS: To minimize position effects we have created a set of piggyBac transformation vectors that contain either the scs/scs', gypsy or chicken beta-globin HS4 insulators. The vectors contain either fluorescent protein or eye color marker genes and have been successfully used for germ-line transformation of Drosophila melanogaster. A set of the scs/scs' vectors contains the coral reef fluorescent protein marker genes AmCyan, ZsGreen and DsRed that have not been optimized for translation in human cells. These marker genes are controlled by a combined GMR-3xP3 enhancer/promoter that gives particularly strong expression in the eyes. This is also the first report of the use of the ZsGreen and AmCyan reef fluorescent proteins as transformation markers in insects. CONCLUSION: The insulated piggyBac vectors should protect transgenes against position effects and thus facilitate fine control of gene expression in a wide spectrum of insect species. These vectors may also be used for transgenesis in other invertebrate species.
