RESUMEN

Background and Objectives: Ultrasound (US) is a non-invasive tool for the in vivo detection of peripheral nerve alterations. Materials and Methods: In this study, we applied nerve US to assist the discrimination between the spectrum of amyotrophic lateral sclerosis (ALS, n = 11), chronic inflammatory demyelinating polyradiculoneuropathy (CIDP, n = 5), and genetically confirmed Charcot-Marie-Tooth disease (CMT, n = 5). All participants and n = 15 controls without neurological diseases underwent high-resolution US of the bilateral tibial nerve. The nerve cross-sectional area (CSA) and nerve microvascular blood flow were compared between the groups and related to cerebrospinal fluid (CSF) measures, clinical symptoms, and nerve conduction studies. The analyses are part of a larger multimodal study on the comparison between US and 7 Tesla (7T) magnetic resonance neurography (MRN). Results: The patients and controls were matched with respect to their demographical data. CMT had the longest disease duration, followed by CIDP and ALS. CSA was related to age, weight, and disease duration. CSA was larger in CMT and CIDP compared to ALS and controls. The blood flow was greatest in CIDP, and higher than in CMT, ALS, and controls. In ALS, greater CSA was correlated with greater CSF total protein and higher albumin quotient. The US measures did not correlate with clinical scores or nerve conduction studies in any of the subgroups. Conclusion: Our results point towards the feasibility of CSA and blood flow to discriminate between ALS, CIDP, and CMT, even in groups of small sample size. In ALS, larger CSA could indicate an inflammatory disease subtype characterized by reduced blood-nerve barrier integrity. Our upcoming analysis will focus on the additive value of 7T MRN in combination with US to disentangle the spectrum between more inflammatory or more degenerative disease variants among the disease groups.

Objective: To develop an efficacious and efficient method for treating chronic wounds using "nanosheet" that improves the survival and localization of transplanted cells without prior seeding to optimally derive the regenerative potentials of uncultured stromal vascular fraction (SVF) cells. Approach: We propose a method whereby the wound is covered by uncultured SVF cells using the nanosheet [porous poly(d, l,-lactic acid)] (PDLLA) films) designed to hold cells in a single-cell layer. A chronic wound model was created on 12-month-old db/db mice by inflecting a full-thickness skin excision on their dorsum and was subsequently given either no treatment or a treatment with SVF cells alone (with Tegaderm dressing), nanosheet alone, or nanosheet with SVF cells. Results: The placement of the nanosheet improved the grafted cell retention rate at day 10 timepoint by 5 folds, and the wound area was the smallest in the wounds treated with SVF cells plus nanosheet in comparison to the other groups. Collagen deposition and epidermal growth factor were significantly higher in the wound beds treated with SVF cells with the nanosheet, offering some mechanistic insights. Innovation: Porous poly(d, l,-lactic acid acid) (PDLLA) films or "nanosheet" printed on the nanoscale (1-100 nm in thickness) as a cellular scaffold for cytotherapy for the treatment of chronic wounds. Conclusion: The use of the nanosheet is an effective way to improve the transplanted SVF cell retention and accelerate the overall wound closure.

BACKGROUND: The process of cleaning and disinfection of animal transport vehicles after unloading animals at the abattoir is a critical control point regarding proper hygiene. It is an important step regarding the biosecurity. In the present study, a status quo analysis of the currently performed cleaning and disinfection measures of animal transport vehicles was carried out at the vehicle washing facilities of five different industrial abattoirs in Germany. For this purpose, a checklist was developed and validated to assess the washing procedure of transport vehicles in a standardised way. The evaluated phases of cleaning included the evaluation criteria "length of time per used floor", "visual cleaning success" and the "hygienic awareness of the driver". During disinfection, attention was paid to the internal and external surfaces of the transporter and to the methods used to disinfect them. In addition, the technical and structural equipment of the five different washing facilities were recorded using a questionnaire and compared to the legal regulations, respectively. At each location, approximately 150 vehicles of all delivery types (transport vehicles owned by the abattoir, external delivery companies and vehicles owned by the supplying farmers) were inspected so that in total a number of more than 750 vehicles were included in this study. The aim was to develop abattoir specific, as well as generally applicable intervention measures and to generate "standard-operation procedures" (SOP's) for the cleaning and disinfection of animal transporters. RESULTS: At two out of five locations vehicles have left the abattoir without cleaning and disinfection. In 31-97% of all vehicles, only a cleaning of the vehicle was carried out, a subsequent disinfection did not take place. A cleaning followed by disinfecting took place in only 3-59% of all vehicles. CONCLUSION: The results indicate a considerable need for improvement and standardisation in this relevant field of disease prevention.
