RESUMEN

Heterochromatin is critical for maintaining genome stability, especially in flowering plants, where it relies on a feedback loop involving the H3K9 methyltransferase, KRYPTONITE (KYP), and the DNA methyltransferase CHROMOMETHYLASE3 (CMT3). The H3K9 demethylase INCREASED IN BONSAI METHYLATION 1 (IBM1) counteracts the detrimental consequences of KYP-CMT3 activity in transcribed genes. IBM1 expression in Arabidopsis is uniquely regulated by methylation of the 7th intron, allowing it to monitor global H3K9me2 levels. We show the methylated intron is prevalent across flowering plants and its underlying sequence exhibits dynamic evolution. We also find extensive genetic and expression variations in KYP, CMT3, and IBM1 across flowering plants. We identify Arabidopsis accessions resembling weak ibm1 mutants and Brassicaceae species with reduced IBM1 expression or deletions. Evolution towards reduced IBM1 activity in some flowering plants could explain the frequent natural occurrence of diminished or lost CMT3 activity and loss of gene body DNA methylation, as cmt3 mutants in A. thaliana mitigate the deleterious effects of IBM1.

Dust loading in West and South Asia has been a major environmental issue due to its negative effects on air quality, food security, energy supply and public health, as well as on regional and global weather and climate. Yet a robust understanding of its recent changes and future projection remains unclear. On the basis of several high-quality remote sensing products, we detect a consistently decreasing trend of dust loading in West and South Asia over the last two decades. In contrast to previous studies emphasizing the role of local land use changes, here, we attribute the regional dust decline to the continuous intensification of Arctic amplification driven by anthropogenic global warming. Arctic amplification results in anomalous mid-latitude atmospheric circulation, particularly a deepened trough stretching from West Siberia to Northeast India, which inhibits both dust emissions and their downstream transports. Large ensemble climate model simulations further support the dominant role of greenhouse gases induced Arctic amplification in modulating dust loading over West and South Asia. Future projections under different emission scenarios imply potential adverse effects of carbon neutrality in leading to higher regional dust loading and thus highlight the importance of stronger anti-desertification counter-actions such as reforestation and irrigation management.

Heat waves and air pollution extremes exert compounding effects on human health and food security and may worsen under future climate change. On the basis of reconstructed daily O3 levels in China and meteorological reanalysis, we found that the interannual variability of the frequency of summertime co-occurrence of heat wave and O3 pollution in China is regulated mainly by a combination of springtime warming in the western Pacific Ocean, western Indian Ocean, and Ross Sea. These sea surface temperature anomalies impose influences on precipitation, radiation, etc., to modulate the co-occurrence, which were also confirmed with coupled chemistry-climate numerical experiments. We thus built a multivariable regression model to predict co-occurrence a season in advance, and correlation coefficient could reach 0.81 (P < 0.01) for the North China Plain. Our results provide useful information for the government to take actions in advance to mitigate damage from these synergistic costressors.

The ongoing and projected impacts from human-induced climate change highlight the need for mitigation approaches to limit warming in both the near term (<2050) and the long term (>2050). We clarify the role of non-CO2 greenhouse gases and aerosols in the context of near-term and long-term climate mitigation, as well as the net effect of decarbonization strategies targeting fossil fuel (FF) phaseout by 2050. Relying on Intergovernmental Panel on Climate Change radiative forcing, we show that the net historical (2019 to 1750) radiative forcing effect of CO2 and non-CO2 climate forcers emitted by FF sources plus the CO2 emitted by land-use changes is comparable to the net from non-CO2 climate forcers emitted by non-FF sources. We find that mitigation measures that target only decarbonization are essential for strong long-term cooling but can result in weak near-term warming (due to unmasking the cooling effect of coemitted aerosols) and lead to temperatures exceeding 2 °C before 2050. In contrast, pairing decarbonization with additional mitigation measures targeting short-lived climate pollutants and N2O, slows the rate of warming a decade or two earlier than decarbonization alone and avoids the 2 °C threshold altogether. These non-CO2 targeted measures when combined with decarbonization can provide net cooling by 2030 and reduce the rate of warming from 2030 to 2050 by about 50%, roughly half of which comes from methane, significantly larger than decarbonization alone over this time frame. Our analysis demonstrates the need for a comprehensive CO2 and targeted non-CO2 mitigation approach to address both the near-term and long-term impacts of climate disruption.

Microglial polarization and associated inflammatory activity are the key mediators of depression pathogenesis. The natural Smilax glabra rhizomilax derivative engeletin has been reported to exhibit robust anti-inflammatory activity, but no studies to date have examined the mechanisms through which it can treat depressive symptoms. We showed that treatment for 21 days with engeletin significantly alleviated depressive-like behaviours in chronic stress social defeat stress (CSDS) model mice. T1-weighted imaging (T1WI), T2-weighted imaging (T2WI) imaging revealed no significant differences between groups, but the bilateral prefrontal cortex of CSDS mice exhibited significant increases in apparent diffusion coefficient and T2 values relative to normal control mice, with a corresponding reduction in fractional anisotropy, while engeletin reversed all of these changes. CSDS resulted in higher levels of IL-1ß, IL-6, and TNF-a production, enhanced microglial activation, and greater M1 polarization with a concomitant decrease in M2 polarization in the mPFC, whereas engeletin treatment effectively abrogated these CSDS-related pathological changes. Engeletin was further found to suppress the LCN2/C-X-C motif chemokine ligand 10 (CXCL10) signalling axis such that adeno-associated virus-induced LCN2 overexpression ablated the antidepressant effects of engeletin and reversed its beneficial effects on the M1/M2 polarization of microglia. In conclusion, engeletin can alleviate CSDS-induced depressive-like behaviours by regulating the LCN2/CXCL10 pathway and thereby altering the polarization of microglia. These data suggest that the antidepressant effects of engeletin are correlated with the polarization of microglia, highlighting a potential avenue for future design of antidepressant strategies that specifically target the microglia.

Long noncoding RNAs (lncRNAs) have emerged as important molecules and potential new targets for human cancers. This study investigates the function of lncRNA CTBP1 antisense RNA (CTBP1-AS) in prostate cancer (PCa) and explores the entailed molecular mechanism. Aberrantly expressed genes potentially correlated with PCa progression were probed using integrated bioinformatics analyses. A cohort of 68 patients with PCa was included, and their tumor and para-cancerous tissues were collected. CTBP1-AS was highly expressed in PCa tissues and cells and associated with poor patient prognosis. By contrast, tumor protein p63 (TP63) and S100 calcium binding protein A14 (S100A14) were poorly expressed in the PCa tissues and cells. CTBP1-AS did not affect TP63 expression; however it blocked the TP63-mediated transcriptional activation of S100A14, thereby reducing its expression. CTBP1-AS silencing suppressed proliferation, apoptosis resistance, migration, invasion, and tumorigenicity of PCa cell lines, while its overexpression led to inverse results. The malignant phenotype of cells was further weakened by TP63 overexpression but restored following artificial S100A14 silencing. In conclusion, this study demonstrates that CTBP1-AS plays an oncogenic role in PCa by blocking TP63-mediated transcriptional activation of S100A14. This may provide insight into the management of PCa.

RESUMEN

We have developed a miniature two-photon microscope equipped with an axial scanning mechanism and a long-working-distance miniature objective to enable multi-plane imaging over a volume of 420 × 420 × 180 µm3 at a lateral resolution of ~1 µm. Together with the detachable design that permits long-term recurring imaging, our miniature two-photon microscope can help decipher neuronal mechanisms in freely behaving animals.

Small-molecular-induced intermolecular hydrogen bonding (inter-HB) interactions were reported to increase the glass transition temperature (Tg) while decrease the dynamic fragility (m) of polymers. Herein, enthalpy relaxation parameters heat capacity jump (ΔCp) at Tg and enthalpy hysteresis (ΔHR) were investigated to help clarify the effect of macromolecular-induced inter-HB on Tg and m using amorphous polyamides as model polymers. The inter-HB strength was weakened by random copolymerization with varied chain rigidity, but was enhanced by decreasing steric hindrance. It was found that Tg and m increased after copolymerization due to the increased chain rigidity. Nevertheless, increasing steric hindrance leads to an increased Tg while anomalously reduced m. Further results found that m can be well correlated to Tg·ΔCp/ΔHR. ΔCp increases more significantly than ΔHR in co-polyamides, and thus the entropy change dominates the activation free energy of cooperative rearrangement. By contrast, ΔHR increases more significantly than ΔCp with increasing steric hindrance, and thus it is reasonable that Tg increases while m decreases. Most importantly, ΔCp and ΔHR decrease with increasing inter-HB strength regardless of the variation of Tg. These results indicate that the inter-HB strength may be very strong and insensitive to temperature in polyamides, thus behaving like physical cross-linking.

Poly(ethylene glycol) (PEG) or oligo (ethylene glycol) (OEG) grafted anion exchange membranes (AEMs) exhibit improved ionic conductivity, high alkaline stability, and subsequent boosted AEM fuel cell performance, but too much PEG/OEG side chains may can result in a reduction in the ion exchange capacity (IEC), which can have adverse effects on ion transport. Here, a series of partially PEG-grafted poly(terphenyl piperidinium) with different side chain length are synthesized using simple postpolymerization modification to produce AEMs with balanced properties. The polar and flexible PEG side chains are responsible for the controlled water uptake and swelling, superior hydroxide conductivity (122 mS cm-1 at 80 °C with an IEC of 1.99 mmol g-1), and enhanced alkaline stability compared to the reference sample without PEG grafts (PTP). More importantly, the performance of AEM fuel cell (AEMFC) with the membrane containing partial PEG side chains surpasses that with PTP membrane, demonstrating a highest peak power density of 1110 mW cm-2 at 80 °C under optimized conditions. This work provides a novel approach to the fabrication of high-performance AEM materials with balanced properties for alkaline fuel cell application.

Major depressive disorder (MDD) is a severe mental disorder associated with high rates of morbidity and mortality. Current first-line pharmacotherapies for MDD are based on enhancement of monoaminergic neurotransmission, but these antidepressants are still insufficient and produce significant side-effects. Consequently, the development of novel antidepressants and therapeutic targets is desired. Engeletin, a natural Smilax glabra rhizomilax derivative, is a compound with proven efficacy in treating ischemic stroke, yet its therapeutic effects and mechanisms for depression remain unexplored. The effects of engeletin were assessed in the forced swimming test (FST) and tail suspension test (TST) in mice. Engeletin was also investigated in the chronic restraint stress (CRS) mouse model of depression with fluoxetine (FLX) as the positive control. Changes in prefrontal cortex (PFC) spine density, synaptic plasticity-linked protein expressions and the brain-derived neurotrophic factor (BDNF)-tyrosine kinase B (TrkB)- mammalian target of rapamycin complex 1 (mTORC1) signalling pathway after chronic stress and engeletin treatment were then investigated. The TrkB and mTORC1 selective inhibitors, ANA-12 and rapamycin, respectively, were utilized to assess the engeletin's antidepressive mechanisms. Our data shows that engeletin exhibited antidepressant-like activity in the FST and TST in mice without affecting locomotor activity. Furthermore, it exhibited efficiency against the depression of CRS model. Moreover, it enhanced the BDNF-TrkB-mTORC1 pathway in the PFC during CRS and altered the reduction in dendritic spine density and levels of synaptic plasticity-linked protein induced by CRS. In conclusion, engeletin has antidepressant activity via activation of the BDNF-TrkB-mTORC1 signalling pathway and upregulation of PFC synaptic plasticity.

High-mobility group box1 (HMGB1) induces inflammatory injury, and emerging reports suggest that it is critical for brain ischemia reperfusion. Engeletin, a natural Smilax glabra rhizomilax derivative, is reported to possess anti-inflammatory activity. Herein, we examined the mechanism of engeletin-mediated neuroprotection in rats having transient middle cerebral artery occlusion (tMCAO) against cerebral ischemia reperfusion injury. Male SD rats were induced using a 1.5 h tMCAO, following by reperfusion for 22.5 h. Engeletin (15, 30 or 60 mg/kg) was intravenously administered immediately following 0.5 h of ischemia. Based on our results, engeletin, in a dose-dependent fashion, reduced neurological deficits, infarct size, histopathological alterations, brain edema and inflammatory factors, namely, circulating IL-1ß, TNF-α, IL-6 and IFN-γ. Furthermore, engeletin treatment markedly reduced neuronal apoptosis, which, in turn, elevated Bcl-2 protein levels, while suppressing Bax and Cleaved Caspase-3 protein levels. Meanwhile, engeletin significantly reduces overall expressions of HMGB1, TLR4, and NF-κB and attenuated nuclear transfer of nuclear factor kappa B (NF-κB) p65 in ischemic cortical tissue. In conclusion, engeletin strongly prevents focal cerebral ischemia via suppression of the HMGB1/TLR4/NF-κB inflammatory network.

A dielectric/Ag-coated hollow fiber (HF) temperature sensor based on long-range surface plasmon resonance (LRSPR) is proposed and experimentally demonstrated. The structural parameters, including the dielectric material and layer thicknesses, are optimized through comprehensive theoretical analysis to achieve the best performance. By filling it with a high refractive index (RI) thermosensitive liquid, the GK570/Ag-coated HF temperature sensor with optimal structural parameters is fabricated. Due to the high sensitivity of the LRSPR sensor and the optimized design, the fabricated sensor achieves a temperature sensitivity of 3.6∼20.5 nm/°C, which is almost the highest among the optical fiber temperature sensors based on surface plasmon resonance reported experimentally. Moreover, the detection range of the proposed sensor can be easily tuned up to 170°C by varying the RI of the filled thermosensitive liquid, and the sensor performance remains stable. Considering that most temperature sensors using polydimethylsiloxane have a fixed detection range, this is an outstanding advantage that could expand the application field of the optical fiber temperature sensor.

BACKGROUND: Pancreatic cancer is a growing public health concern in China, and depicting it from different perspectives would provide a comprehensive understanding of its epidemiological characteristics. METHODS: Data from the National Mortality Surveillance System (NMSS) in China was used to estimate the number of deaths, years of life lost (YLL), age-standardized mortality rate (ASMR) and age-standardized YLL rate in China, its provinces and urban-rural areas from 2005 to 2020. Joinpoint regression analysis was employed to explore the temporal trends of ASMR and age-standardized YLL rate. Decomposition analysis was conducted to assess the contribution of population growth, population aging and cause-specific mortality rate to the increment of pancreatic cancer deaths. RESULTS: A total of 100,427 pancreatic cancer deaths and 2,166,355 pancreatic cancer related YLL were estimated in China in 2020. The overall ASMR significantly increased from 6.6/100 000 in 2005 to 7.4/100 000 in 2020, and was higher in men than that in women. Age-standardized YLL rate showed a similar trend. The mortality rates of pancreatic cancer were generally higher in northeast China than in southwest China. The highest ASMRs were found in Jilin, Zhejiang, Inner Mongolia and Anhui, and the lowest ones in Guangxi, Yunnan, Tibet, and Hainan. The disease burden due to pancreatic cancer presented a significant upward trend in rural areas and a downward trend in urban areas. CONCLUSIONS: The burden associated with pancreatic cancer had been increasing in China from 2005 to 2020. The escalating disease burden of pancreatic cancer in rural areas necessitates the implementation of effective control and prevention measures. Relevant provinces should pay greater attention to the prevailing of pancreatic cancer, particularly those exhibiting higher mortality rates.

OBJECTIVE: To develop a 3D scoring system of tumor anatomy and intrarenal relationship for assessing surgical complexity and outcomes of robot-assisted partial nephrectomy (RAPN). METHODS: We prospectively enrolled patients with a renal tumor who had a 3D model and underwent RAPN between Mar 2019 and Mar 2022. The ADDD nephrometry consisted of the contact surface area between tumor and parenchyma (A), the depth of tumor invasion into the renal parenchyma (D1), the distance from tumor to the main intrarenal artery (D2), and to the collecting system (D3). The primary outcomes included perioperative complication rate and trifecta outcome (WIT ≤ 25 min, negative surgical margins, and no major complications). RESULTS: We enrolled a total of 301 patients. The mean tumor size was 2.93 ± 1.44 cm. There were 104 (34.6%) patients, 119 (39.5%) patients, and 78 (25.9%) patients in the low-, intermediate-, and high-risk groups, respectively. Each point increase in the ADDD score increased the risk of complications [hazard ratio (HR) 1.501]. A lower grade indicated a lower risk of failed trifecta (HR low group 15.103, intermediate group 9.258) and renal function damage (HR low risk 8.320, intermediate risk 3.165) compared to the high-risk group. The AUC of ADDD score and grade were 0.738 and 0.645 for predicting major complications, 0.766 and 0.714 for predicting trifecta outcome, and 0.746 and 0.730 for predicting postoperative renal function reservation. CONCLUSION: The 3D-ADDD scoring system shows the tumor anatomy and its intraparenchymal relationships and has better efficacy in predicting surgical outcomes of RAPN.

BACKGROUND: Ovarian epithelial tumors are common in adults, and their peak incidence of onset is over 40 years of age. In children, most ovarian tumors are germ cell-derived, whereas epithelial tumors are rare and mostly benign. CASE PRESENTATION: This report describes a case of a 14-year-old Chinese girl with ovarian mucinous cystadenocarcinoma. She was admitted with a small amount of bloody vaginal discharge during the past month. Magnetic resonance imaging of the abdomen and pelvis showed a large solid cystic mass lesion in the left ovary. Tumor marker levels were within normal limits ( CA-125: 22.3 U/mL, HE4: 28.5 pmol/L, HCG: < 1.20 mIU/ml, AFP: 3.3 ng/ml, CEA: 2.2 ng/ml, CA19-9: < 2.0 U/mL). Laparoscopic exploration revealed a large left ovarian tumor. The patient underwent left salpingo-oophorectomy, and showed no significant issues during follow-up, as well as no evidence of recurrence or metastasis. CONCLUSIONS: We report the first pediatric case of ovarian mucinous cystadenocarcinoma in China. Given the scarcity of reports addressing the clinical management of this condition, the present study provides a useful contribution to its further understanding in light of developing future treatment strategies.

RESUMEN

The application of spinal cord stimulation (SCS) and deep brain stimulation (DBS) for disorders of consciousness (DoC) has been increasingly reported. However, there is no sufficient evidence to determine how effective and safe SCS and DBS are for DoC owing to various methodological limitations. We conducted a systematic review to elucidate the safety and efficacy of SCS and DBS for DoC by systematically reviewing related literature by searching PubMed, EMBASE, Medline, and Cochrane Library. Twenty eligible studies with 608 patients were included in this study. Ten studies with 508 patients reported the efficacy of SCS for DoC, and the estimated overall effectiveness rate was 37%. Five studies with 343 patients reported the efficacy of SCS for VS, and the estimated effectiveness rate was 30%. Three studies with 53 patients reported the efficacy of SCS for MCS, and the estimated effectiveness rate was 63%. Five studies with 92 patients reported the efficacy of DBS for DoC, and the estimated overall effectiveness rate was 40%. Four studies with 63 patients reported the efficacy of DBS for VS, and the estimated effectiveness rate was 26%. Three studies with 19 patients reported the efficacy of DBS for MCS, and the estimated effectiveness rate was 74%. The adverse event rate of DoC was 8.1% and 18.2% after SCS and DBS, respectively. These results suggest that SCS and DBS can be considered reasonable treatments for DoC with considerable efficacy and safety.

In this paper, the silver-coated large-core suspended-core fiber (LSCF) probe was fabricated by the dynamic chemical liquid phase deposition method for surface-enhanced Raman scattering (SERS) sensing. The 4-mercaptophenylboronic acid (4-MPBA) monolayer was assembled in the LSCF as the recognition monolayer. Taking advantage of the appropriate core size of the LSCF, a custom-made Y-type optical fiber patch cable was utilized to connect the semiconductor laser, Raman spectrometer, and the proposed fiber SERS probe. The SERS signal is propagated in the silver-coated air channels, which can effectively reduce the Raman and fluorescence background of the silica core. Experiments were performed to measure the Raman scattering spectra of the 4-MPBA in the silver-coated LSCF in a non-enhanced and enhanced case. The experiment results showed that the Raman signal strength was enhanced more than 6 times by the surface plasmon resonance compared with the non-enhanced case. The proposed LSCF for SERS sensing technology provides huge research value for the fiber SERS probes in biomedicine and environmental science. The combination of SERS and microstructured optical fibers offers a potential approach for SERS detection.

A novel surface plasmon resonance (SPR) temperature sensor based on a silver-coated multi-hole optical fiber (SMHOF) is presented. The central and surrounding air holes of the SMHOF are filled with two kinds of thermosensitive liquid with high and low refractive index (RI), respectively. Two separated resonance dips, which are related to the high and low RI filled liquid respectively, are observed at different wavelength in the transmission spectrum. Advantageously, the two dips move towards opposite direction with the temperature variation. The interval between the two SPR dips is measured under different environmental temperature and exhibits a good linearity. The proposed sensor with different detection range is fabricated by changing the RIs of the filled thermosensitive liquids. The temperature sensitivity of 7.72 nm/°C and -7.81 nm/°C is obtained in the range of 20-60 °C and -20-20 °C, respectively. Owing to the high temperature sensitivity and tunable detection range, the proposed sensor is expected to find potential applications in biomedicine, health care and environmental monitoring.

A reasonable design of samples with efficient spatial separation for photoinduced electron-hole pairs toward the photocatalytic hydrogen evolution reaction (HER) has gained significant attention. Herein, a new C/MoS2@ZnIn2S4/Co3O4 composite with a core-shell structure is designed toward photocatalytic hydrogen production on C/MoS2 and Co3O4 dual electron collectors. Co3O4 nanoparticles as the co-catalyst would form a Schottky junction with ZnIn2S4 nanosheets while the C/MoS2 hollow core would form the step-scheme (S-scheme) heterojunction with ZnIn2S4 sheets, which provides a dual photogenerated electron transfer pathway during the light irradiation process. In addition, the unique core-shell architecture offers large contact interfaces favoring the exposure of rich active sites, which facilitated the separation and the transfer of charges. Consequently, all these factors endowed the C/MoS2@ZnIn2S4/Co3O4 composite with enhanced light absorption ability and an increased hydrogen evolution rate of 6.7 mmol·g-1·h-1 under 420 nm light irradiation, which is about 23.4- and 4.5-fold that of ZnIn2S4 and CMZ, respectively. This work offers a guideline for designing efficient composite photocatalysts toward the photocatalytic HER.

In both organic and polymer synthesis, photochemistry of charge transfer complexes (CTCs) is considered as a powerful approach to expand visible-light-driven radical chemistry reaction. One reports herein on the development of a class of useful CTCs using pyridinium salts as efficient electron acceptors (combined with N, N, 3,5-tetramethylaniline, TMA) to achieve a multiwavelength (375-560 nm) metal-free LED photopolymerization process under mild conditions (open to air, without monomer purification and inhibitor removal). The UV-vis absorption spectra and molecular modeling simultaneously verify its potential blue-green absorbing wavelength range. Also, their good thermal initiation behavior at relatively low temperatures makes it easier to achieve thick samples and/or polymerization in the shadow region in practice. More importantly, with excellent photoinitiating capability, the formulation is successfully applied to direct laser write (DLW) and high-resolution 3D printing, yielding a series of objects with well-defined structures, such as letters, ring, solid squares, and chess pieces. These new pyridinium salt acceptors further extend the applicability to visible photopolymerizable resins and additive-containing formulations for efficient surface and deep curing.
