RESUMEN

Immune checkpoint blockade (ICB) treatment has demonstrated excellent medical effects in oncology, and it is one of the most sought after immunotherapies for tumors. However, there are several issues with ICB therapy, including low response rates and a lack of effective efficacy predictors. Gasdermin-mediated pyroptosis is a typical inflammatory death mode. We discovered that increased expression of gasdermin protein was linked to a favorable tumor immune microenvironment and prognosis in head and neck squamous cell carcinoma (HNSCC). We used the mouse HNSCC cell lines 4MOSC1 (responsive to CTLA-4 blockade) and 4MOSC2 (resistant to CTLA-4 blockade) orthotopic models and demonstrated that CTLA-4 blockade treatment induced gasdermin-mediated pyroptosis of tumor cells, and gasdermin expression positively correlated to the effectiveness of CTLA-4 blockade treatment. We found that CTLA-4 blockade activated CD8+ T cells and increased the levels of interferon γ (IFN-γ) and tumor necrosis factor α (TNF-α) cytokines in the tumor microenvironment. These cytokines synergistically activated the STAT1/IRF1 axis to trigger tumor cell pyroptosis and the release of large amounts of inflammatory substances and chemokines. Collectively, our findings revealed that CTLA-4 blockade triggered tumor cells pyroptosis via the release of IFN-γ and TNF-α from activated CD8+ T cells, providing a new perspective of ICB.

RESUMEN

Tomato zonate spot virus (TZSV, Orthotospovirus tomatozonae, genus Orthotospovirus, family Tospoviridae) was first reported to infect tomato (Solanum lycopersicum) in China in 2008 (Dong et al. 2008). Belamcanda chinensis (L.) Redouté is a perennial herbaceous medicinal plant of the family Iridaceae, which is widely distributed in China. Its rhizome contains abundant active components, mainly including flavonoids, and has antibacterial, anticancer, and antioxidative effects. In July 2023, four B. chinensis plants with virus-like symptoms were collected in Fuyuan County, Yunnan Province in China. The diseased leaves showed chlorosis and ringspots (Fig. S1). Spherical virus particles with a diameter of 80-100 nm were observed in the saps of diseased leaves under a transmission electron microscope (Fig. S2). The presence of an orthotospovirus was confirmed by the previously reported method to amplify the partial sequence (312 nt) of L segment (Huang et al. 2018) (Fig. S3). BLASTn analysis showed that the obtained 312-nt sequence was 95.62% nucleotide identity with TZSV tomato-YN isolate (accession no. NC_010491.1). To obtain the complete genome of this isolate, total RNA from symptomatic leaves of two single diseased B. chinensis were extracted using Hipure Universal RNA Mini Kit (Magen Biotech) and subjected to high-throughput sequencing with a NovaPE150 (Illumina, USA) at MAGIGENE (Shenzhen, China). A total of 41,144,571 clean reads were obtained after removing low quality reads. Quality-controlled, qualified reads were assembled into contigs using Megahit v1.1.2 software. Thirteen contigs shared nucleotide identity ranging 86.94%-97.73% with the L, S, and M segments of TZSV using BLASTn searches online (https://blast.ncbi.nlm.nih.gov/Blast.cgi). In addition, no contigs were mapped to other viral (taxid:10239) and viroidal (taxid:12884) sequences in GenBank Databases. The full-length L, M, and S RNA segments of TZSV-Bc isolate was determined tbe 8917 nt (PP314222), 4718 nt (PP314223) and 3213 nt (PP314224), respectively. These segments were validated by RT-PCR, and Sanger sequencing. They shared nucleotide sequence identities of 95.9%, 97.2%, and 93.1% of the L (NC_010491.1), M (NC_010490.1), and S (NC_010489.1) segments, of the TZSV tomato-YN isolate, respectively. Compared to the TZSV tomato-YN isolate, there exists a missing segment with 113 nt in the intergenic region of S RNA and a segment with 199 nt in M RNA. To further confirm the TZSV infection on B. chinensis, three primers pairs Tosp10/ Tosp11, Tosp5/Tosp6, and NSs-F/NSs-R were tested by RT-PCR for TZSV based on the previous report (Dong et al, 2008). The sequences of amplicons shared >99% nucleotide identity with the corresponding TZSV-Bc isolate sequences. Total of 14 B. chinensis samples were detected with the primer pair N-F/N-R (5'-ATGTCTAACGTCCGGAGTTTAACA-3'/ 5'-AAAAGACAGATCATTGCTGCTCTT-3') by One Step RT-PCR, 6 samples (42.85%) showed the positive results. The mechanical inoculation and RT-PCR detection confirmed TZSV-Bc isolate can infect N. bethamiana. So far, tomato zonate spot virus has been detected in different plants including tobacco (N. tabacum) (Huang et al. 2017), sticktight (Bidens pilosa) (Xu et al. 2022), pepper (Capsicum annuum) (Li et al. 2023) in China. To our knowledge, it is the first report of TZSV naturally infecting B. chinensis plants, which enriches information on the host range of TZSV and will be helpful for disease management.

RESUMEN

Covalent organic frameworks (COFs) have emerged as a promising class of crystalline porous materials for cancer phototherapy, due to their exceptional characteristics, including light absorption, biocompatibility, and photostability. However, the aggregation-caused quenching effect and apoptosis resistance often limit their therapeutic efficacy. Herein, we demonstrated for the first time that linking luminogens with aggregation-induced emission effect (AIEgens) into COF networks via vinyl linkages was an effective strategy to construct nonmetallic pyroptosis inducers for boosting antitumor immunity. Mechanistic investigations revealed that the formation of the vinyl linkage in the AIE COF endowed it with not only high brightness but also strong light absorption ability, long lifetime, and high quantum yield to favor the generation of reactive oxygen species for eliciting pyroptosis. In addition, the synergized system of the AIE COF and αPD-1 not only effectively eradicated primary and distant tumors but also inhibited tumor recurrence and metastasis in a bilateral 4T1 tumor model.

RESUMEN

We report spectroscopic measurements of the local pH and pKa at an electrode/electrolyte interface using surface enhanced Raman scattering (SERS) spectroscopy of 4-mercaptobenzoic acid (4-MBA). In acidic and basic solutions, the protonated and deprotonated carboxyl functional groups at the electrode surface exist in the solution as -COOH and -COO-, which have different Raman active vibrational features at around 1697 and 1414 cm-1, respectively. In pH neutral water, as the applied electrochemical potential is varied from negative to positive, the acidic form of the 4-MBA (i.e., -COOH) decreases in Raman intensity and the basic form (i.e., -COO-) increases in Raman intensity. The change in local ion concentration is due to the application of electrochemical potentials and the accumulation of ions near the electrode surface. Under various applied potentials, the ratio of 1697 and 1587 cm-1 (pH-independent) peak areas spans the range between 0.7 and 0, and the ratio of the 1414 and 1587 cm-1 peak areas ranges from 0 to 0.3. By fitting these data to a normalized sigmoid function, we obtain the percentage of surface protonation/deprotonation, which can be related to the pKa and pH of the system. Thus, we can measure the local pKa at the electrode surface using the surface enhanced Raman signal of the 4-MBA.

RESUMEN

The complete genome sequence of a putative novel potyvirus, tentatively named "polygonatum mosaic-associated virus 1" (PMaV1), was sequenced from naturally infected Polygonatum cyrtonema Hua in China. PMaV1 has a typical genome organization of potyviruses with a single large open reading frame (nt 119-9448) that encodes a 3109-aa polyprotein that is predicted to be cleaved into 10 mature proteins by virus-encoded proteases. Pairwise comparisons revealed that PMaV1 shares 71.50% complete genome sequence identity with Polygonatum kingianum virus 4 and 80.00% amino acid sequence identity with Polygonatum kingianum virus 3 of the genus Potyvirus. Phylogenetic analysis indicated that PMaV1 clustered with other potyviruses and that it was most closely related to Polygonatum kingianum virus 3 and Polygonatum kingianum virus 4. These results suggest that PMaV1 is a new member of the genus Potyvirus of the family Potyviridae (Nucleotide sequence data reported are available in the GenBank databases under the accession number OP380926).

RESUMEN

The complete genome sequence of a new potyvirus from Paris polyphylla var. yunnanensis was determined. Its genomic RNA consists of 9571 nucleotides (nt), excluding the 3'-terminal poly(A) tail, containing the typical open reading frame (ORF) of potyviruses and encoding a putative large polyprotein of 3061 amino acids. The virus shares 54.20%-59.60% nt sequence identity and 51.80%-57.90% amino acid sequence identity with other potyviruses. Proteolytic cleavage sites and conserved motifs of potyviruses were identified in the polyprotein and within individual proteins. Phylogenetic analysis indicated that the virus was most closely related to lily yellow mosaic virus. The results suggest that the virus should be classified as a member of a novel species within the genus Potyvirus, and we have tentatively named this virus "Paris yunnanensis mosaic chlorotic virus" (PyMCV).

RESUMEN

Energy-restricted diet is a specific dietary regimen, including the continuous energy-restricted diet and the intermittent energy-restricted diet. It has been proven effective not only to reduce weight and extend the lifespan in animal models, but also to regulate the development and progression of various neurological diseases such as epilepsy, cerebrovascular diseases (stroke), neurodegenerative disorders (Alzheimer's disease and Parkinson's disease) and autoimmune diseases (multiple sclerosis). However, the mechanism in this field is still not clear and a systematic neurological summary is still missing. In this review, we first give a brief summary of the definition and mainstream strategies of energy restrictions. We then review evidence about the effects of energy-restricted diet from both animal models and human trials, and update the current understanding of mechanisms underlying the biological role of energy-restricted diet in the fight against neurological diseases. Our review thus contributes to the modification of dietary regimen and the search for special diet mimics.

RESUMEN

Understanding the relaxation and injection dynamics of hot electrons is crucial to utilizing them in photocatalytic applications. While most studies have focused on hot carrier dynamics at metal/semiconductor interfaces, we study the in situ dynamics of direct hot electron injection from metal to adsorbates. Here, we report a hot electron-driven hydrogen evolution reaction (HER) by exciting the localized surface plasmon resonance (LSPR) in Au grating photoelectrodes. In situ ultrafast transient absorption (TA) measurements show a depletion peak resulting from hot electrons. When the sample is immersed in solution under -1 V applied potential, the extracted electron-phonon interaction time decreases from 0.94 to 0.67 ps because of additional energy dissipation channels. The LSPR TA signal is redshifted with delay time because of charge transfer and subsequent change in the dielectric constant of nearby solution. Plateau-like photocurrent peaks appear when exciting a 266 nm linewidth grating with p-polarized (on resonance) light, accompanied by a similar profile in the measured absorptance. Double peaks in the photocurrent measurement are observed when irradiating a 300 nm linewidth grating. The enhancement factor (i.e., reaction rate) is 15.6× between p-polarized and s-polarized light for the 300 nm linewidth grating and 4.4× for the 266 nm linewidth grating. Finite-difference time domain (FDTD) simulations show two resonant modes for both grating structures, corresponding to dipolar LSPR modes at the metal/fused silica and metal/water interfaces. To our knowledge, this is the first work in which LSPR-induced hot electron-driven photochemistry and in situ photoexcited carrier dynamics are studied on the same plasmon resonance structure with and without adsorbates.

RESUMEN

The Shank family proteins are enriched at the postsynaptic density (PSD) of excitatory glutamatergic synapses. They serve as synaptic scaffolding proteins and appear to play a critical role in the formation, maintenance and functioning of synapse. Increasing evidence from genetic association and animal model studies indicates a connection of SHANK genes defects with the development of neuropsychiatric disorders. In this review, we first update the current understanding of the SHANK family genes and their encoded protein products. We then denote the literature relating their alterations to the risk of neuropsychiatric diseases. We further review evidence from animal models that provided molecular insights into the biological as well as pathogenic roles of Shank proteins in synapses, and the potential relationship to the development of abnormal neurobehavioral phenotypes.

RESUMEN

Thin layer chromatography in tandem with surface-enhanced Raman scattering (TLC-SERS) has demonstrated tremendous potentials as a new analytical chemistry tool to detect a wide range of substances from real-world samples. However, it still faces significant challenges of multiplex sensing from complex mixtures due to the imperfect separation by TLC and the resulting interference of SERS detection. In this article, we propose a multiplex sensing method of complex mixtures by machine vision analysis of the scanning image of the TLC-SERS results. Briefly, various pure substances in solution and the complex mixture solution are separated by TLC followed by one-dimensional SERS scanning of the entire TLC plate, which generates TLC-SERS images of all target substances along the chromatography path. After that, a machine vision method is employed to extract the template images from the TLC-SERS images of pure substance solutions. Finally, we apply a feature point matching strategy based on the Winner-take-all principle, which matches the template image of each pure substance with the mixture image to confirm the existence and derive the position of each target substance in the TLC plate, respectively. Our experimental results based on the mixture solution of five different substances show that the proposed machine vision analysis is highly selective, sensitive and does not require artificial analysis of the SERS spectra. Therefore, we envision that the proposed machine vision analysis of the TLC-SERS imaging is an objective, accurate, and efficient method for multiplex sensing of trace level of target substances from complex mixtures.

RESUMEN

The effect of resolution bandwidth (RBW) and video bandwidth (VBW) of a radio-frequency spectrum analyzer on the characteristics of a power spectrum for chaos generated by a semiconductor laser with external optical feedback is investigated experimentally and numerically. We describe the spectral characteristics with effective bandwidth and time-delay signature (TDS) quantificationally. The experimental results demonstrate that the ratio of VBW to RBW has a significant impact on the smoothness of the power spectrum and effective bandwidth of chaos. Meanwhile, the RBW affects the resolution of periodical peaks of the power spectrum and the TDS of chaos, which is obtained by the power spectrum. The incorrect characterization of chaos can be avoided by setting the RBW to no more than 0.1 times as much as the resonance frequency of the external cavity of chaos and setting the VBW/RBW to no more than 0.01. The simulation results qualitatively agree with the analysis of the experiment.

RESUMEN

Frequent monitoring of sea food, especially shellfish samples, for the presence of biotoxins serves not only as a valuable strategy to mitigate adulteration associated health risks, but could also be used to develop predictive models to understand algal explosion and toxin trends. Periodic toxin assessment is often restricted due to poor sensitivity, multifarious cleaning/extraction protocols and high operational costs of conventional detection methods. Through this work, a simplistic approach to quantitatively assess the presence of a representative marine neurotoxin, Domoic acid (DA), from spiked water and crab meat samples is presented. DA sensing was performed based on surface-enhanced Raman scattering (SERS) using silver nanoparticle enriched diatomaceous eartha biological photonic crystal material in nature. Distinctive optical features of the quasi-ordered pore patterns in diatom skeleton with sporadic yet uniform functionalization of silver nanoparticles act as excellent SERS substrates with improved DA signals. Different concentrations of DA were tested on the substrates with the lowest detectable concentration being 1 ppm that falls well below the regulatory DA levels in seafood (>20 ppm). All the measurements were rapid and were performed within a measurement time of 1 min. Utilizing the measurement results, a standard calibration curve between SERS signal intensity and DA concentration was developed. The calibration curve was later utilized to predict the DA concentration from spiked Dungeness crab meat samples. SERS based quantitative assessment was further complemented with principal component analysis and partial least square regression studies. The tested methodology aims to bring forth a sensitive yet simple, economical and an extraction free routine to assess biotoxin presence in sea food samples onsite.

RESUMEN

Disinfection is a common practice to inhibit pathogens, yet success is limited by microbial adaptation and our poor knowledge of viral transmission, notably in the current COVID-19 pandemic. There is a need for alternative disinfection strategies and techniques that are adapted to the actual behavior of humans living in densely populated mega-cities. Here, high public circulation in shared passenger vehicles such as taxis, buses and personal cars represents a major risk of viral transmission due to confined space and commonly touched surfaces. Actual regulatory guidelines are not fully successful because they rely both on passengers' willingness to wear face masks and on drivers' willingness to disinfect cars after each shift or each ride with symptomatic individuals. Here we propose that passive solar heating, a sustainable technique that has been used in agronomy to kill weeds and soil pathogens, could inactivate the virus in vehicles during warm-to-hot weather within few minutes to half an hour at 50-60 °C. We measured temperatures in a white compact-size sedan left in a parking lot under direct sunlight. Air temperatures increased from 30 to 42-49 °C after 30 min and then reached a plateau at 52-57 °C after 90 min. Temperatures were about 3 °C higher in front versus back of the car and about 5 °C higher at face height compared to knee height. Since COVID-19 is inactivated in 30 min at 56 °C, our findings confirm that hot air generated passively by solar heating in enclosed spaces is a promising strategy of disinfection with benefits of no added costs, chemicals or worktime. Though this technique appears limited to hot climate, possible heating systems that work during parking time might be developed by vehicle makers to extend the technique to cold climates.

RESUMEN

Multiple myeloma (MM) is characterised by the proliferation and accumulation of malignant plasma cells in the bone marrow. Despite the progress in treatment over the last few years, MM remains incurable and the majority of patients relapse. MM stem-like cells (MMSCs) have been considered as the main reason for drug resistance and eventual relapse. Currently, therapeutic agents are not enough to eradicate MMSCs, and finding effective strategies to eradicate MMSCs may improve the outcome of patients. Here we showed that lycorine, a natural compound from the Amaryllidaceae species, effectively inhibits the proliferation of myeloma cells from cell lines or patients, mainly through decreasing ALDH1+ cells. Mechanistically, lycorine decreases the MMSC population through inhibition of the Wnt/ß-catenin pathway by reducing the ß-catenin protein level. Moreover, lycorine could overcome the increasing proportion of ALDH1+ cells caused by bortezomib (BTZ) treatment, and a combination BTZ and lycorine have a synergistic effect on anti-myeloma cells. Furthermore, we found a similar reduction of MMSC characteristics by lycorine in BTZ-resistant MM cells and primary CD138+ plasma cells. Collectively, our findings indicate lycorine as a promising agent to target MMSCs to overcome the drug resistance of BTZ, and that, alone or in combination with BTZ, lycorine is a potential therapeutic strategy for MM treatments.

RESUMEN

The release of metal ions may play an important role in toxicity of metal-based nanoparticles. In this report, a life cycle study is carried out in a greenhouse, to compare the effects of ceria nanoparticles (NPs) and Ce3+ ions at 0, 50, 100, and 200 mg Ce kg-1 on plant growth, biological and physiological parameters, and nutritional value of soil-grown common bean plants. Ceria NPs have a tendency to negatively affect photosynthesis, but the effect is not statistically significant. Ce3+ ionic treatments at 50, 100, and 200 mg Ce kg-1 result in increases of 1.25-, 0.66-, and 1.20-fold in stomatal conductance, respectively, relative to control plants. Both ceria NPs and Ce3+ ions disturb the homeostasis of antioxidant defense system in the plants, but only 200 mg Ce kg-1 ceria NPs significantly induce lipid peroxidation in the roots. Ceria NP treatments tend to reduced fresh weight and to increase mineral contents of the green pods, but have no effect on the organic nutrient contents. On the contrary, Ce3+ ion treatments modify the organic compositions and thus alter the nutritional quality and flavor of the green pods. These results suggest that the two Ce forms may have different mechanisms on common bean plants.

RESUMEN

Recent evidence suggests that long noncoding RNAs (lncRNAs) are essential regulators of many cancer-related processes, including cancer cell proliferation, invasion, and migration. There is thus a reason to believe that the detection of lncRNAs may be useful as a diagnostic and prognostic strategy for cancer detection, however, at present no effective genome-wide tests are available for clinical use, constraining the use of such a strategy. In this study, we performed a comprehensive assessment of lncRNAs expressed in samples in the head and neck squamous cell carcinoma (HNSCC) cohort available in The Cancer Genome Atlas database. A risk score (RS) model was constructed based on the expression data of these 15 lncRNAs in the validation data set of HNSCC patients and was subsequently validated in validation data set and the entire data set. We were able to stratify patients into high- and low-risk categories, using our lncRNA expression panel to determine an RS, with significant differences in overall survival (OS) between these two groups in our test set (median survival, 1.863 vs. 5.484 years; log-rank test, p < 0.001). We were able to confirm the predictive value of our 15-lncRNA signature using both a validation data set and a full data set, finding our signature to be reproducible and effective as a means of predicting HNSCC patient OS. Through the multivariate Cox regression and stratified analyses, we were further able to confirm that the predictive value of this RS was independent of other predictive factors such as clinicopathological parameters. The Gene set enrichment analysis revealed potential functional roles for these 15 lncRNAs in tumor progression. Our findings indicate that an RS established based on a panel of lncRNA expression signatures can effectively predict OS and facilitate patient stratification in HNSCC.

RESUMEN

BACKGROUND: Transcription factors (TFs) are responsible for the regulation of various activities related to cancer like cell proliferation, invasion, and migration. It is thought that, the measurement of TFs levels could assist in developing strategies for diagnosis and prognosis of cancer detection. However, due to lack of effective genome-wide tests, this cannot be carried out in clinical settings. METHODS: A complete assessment of RNA-seq data in samples of a head and neck squamous cell carcinoma (HNSCC) cohort in The Cancer Genome Atlas (TCGA) database was carried out. From the expression data of six TFs, a risk score model was developed and further validated in the GSE41613 and GSE65858 series. Potential functional roles were identified for the six TFs via gene set enrichment analysis. RESULTS: Based on our multi-TF signature, patients are stratified into high- and low-risk groups with significant variations in overall survival (OS) (median survival 2.416 vs. 5.934 years, log-rank test P < 0.001). The sensitivity and specificity evaluation of our multi-TF for 3-year OS in TCGA, GSE41613 and GSE65858 was 0.707, 0.679 and 0.605, respectively, demonstrating good reproducibility and robustness for predicting overall survival of HNSCC patients. Through multivariate Cox regression analyses (MCRA) and stratified analyses, we confirmed that the predictive capability of this risk score (RS) was not dependent on any of other factors like clinicopathological parameters. CONCLUSIONS: With the help of a RS obtained from a panel of TFs expression signatures, effective OS prediction and stratification of HNSCC patients can be carried out.

RESUMEN

We report electrochemical measurements using in situ Raman spectroscopy at graphene/D2O interfaces under extremely low applied potentials. Here, the hydrophobic and catalytically inert nature of graphene and the insulating nature of the deionized (DI) water enables potentials as low as Vapplied = -7 V vs Ag/AgCl to be applied without exceeding 200 µA/cm2 of current density. At higher currents, bubble formation (i.e., hydrogen evolution reaction) prohibits reliable spectra from being obtained from the electrode surface. Using CaF2 as the supporting substrate enables significantly lower reducing potentials to be reached compared to glass substrates, likely due to trapped charge and impurities in the glass substrate. G band Raman spectra taken under various applied electrochemical potentials exhibit a linear relationship between the G band shift (ΔωG) and the applied potential, with blueshifts as high as ΔωG = 18 cm-1. These large Raman shifts indicate a large change in the Fermi level of ΔEF = -0.43 eV for graphene electrodes in contact with water, favoring reduction half-reactions. Based on the solution resistance measurement, there is a VIR = 3.1 V voltage drop across the solution for D2O (when the applied potential was Vapplied = -7 V vs Ag/AgCl) and the effective reducing potential on the working electrode is Veffective = -3.9 V vs Ag/AgCl. We have also tested these graphene electrodes in ionic liquids [DEME][TFSI], which are limited to applied potentials above Vapplied = -2.7 V vs Ag/AgCl and a corresponding shift in the Fermi level ΔEF = -0.32 eV, indicating that pure water can provide a more robust electrolyte for reaching low reducing potentials than ionic liquids.

RESUMEN

The hybrid energy storage system of wind power involves the deep coupling of heterogeneous energy such as electricity and heat. Exergy as a dual physical quantity that takes into account both 'quantity' and 'quality, plays an important guiding role in the unification of heterogeneous energy. In this paper, the operation characteristics of the system are related to the energy quality, and the operation strategy of the wind power hybrid energy storage system is proposed based on the exergoeconomics. First, the mathematical model of wind power hybrid energy storage system is established based on exergoeconomics. Then, wind power experiments of three forms of thermal-electric hybrid energy storage are carried out, and RSM is used to analyze the power quality and exergoeconomic characteristics of the system, and the optimal working conditions of the experiment are obtained. Finally, an optimization strategy is proposed by combining experiment and simulation. The system efficiency, unit exergy cost and current harmonic distortion rate are multi-objective optimization functions. The three algorithms evaluate the optimal solution based on standard deviation. The results show that the exergoeconomics can effectively judge the production-storage-use characteristics of the new system of ' wind power + energy storage'. The thermal-electric hybrid energy storage system can absorb the internal exergy loss of the battery, increase the exergy efficiency by 10%, reduce the unit exergy cost by 0.03 yuan/KJ, and reduce the current harmonic distortion rate by 8%. It provides guidance for improving the power quality of wind power system, improving the exergy efficiency of thermal-electric hybrid energy storage wind power system and reducing the unit cost.

RESUMEN

Because of the high dielectric strength of water, it is extremely difficult to discharge plasma in a controllable way in the aqueous phase. By using lithographically defined electrodes and metal/dielectric nanoparticles, we create electric field enhancement that enables plasma discharge in liquid electrolytes at significantly reduced applied voltages. Here, we use high voltage (10-30 kV) nanosecond pulse (20 ns) discharges to generate a transient plasma in the aqueous phase. An electrode geometry with a radius of curvature of approximately 10 µm, a gap distance of 300 µm, and an estimated field strength of 5 × 106 V/cm resulted in a reduction in the plasma discharge threshold from 28 to 23 kV. A second structure had a radius of curvature of around 5 µm and a gap distance of 100 µm had an estimated field strength of 9 × 106 V/cm but did not perform as well as the larger gap electrodes. Adding gold nanoparticles (20 nm diameter) in solution further reduced the threshold for plasma discharge to 17 kV due to the electric field enhancement at the water/gold interface, with an estimated E-field enhancement of 4×. Adding alumina nanoparticles decorated with Pt reduced the plasma discharge threshold to 14 kV. In this scenario, the emergence of a triple point at the juncture of alumina, Pt, and water results in the coexistence of three distinct dielectric constants at a singular location. This leads to a notable concentration of electric field, effectively aiding in the initiation of plasma discharge at a reduced voltage. To gain a more comprehensive and detailed understanding of the electric field enhancement mechanism, we performed rigorous numerical simulations. These simulations provide valuable insights into the intricate interplay between the lithographically defined electrodes, the nanoparticles, and the resulting electric field distribution, enabling us to extract crucial information and optimize the design parameters for enhanced performance.
