RESUMEN

Purpose: Corneal epithelial homeostasis is maintained by coordinated gene expression across distinct cell populations, but the gene regulatory programs underlying this cellular diversity remain to be characterized. Here we applied single-cell multi-omics analysis to delineate the gene regulatory profile of mouse corneal epithelial cells under normal homeostasis. Methods: Single cells isolated from the cornea epithelium (with marginal conjunctiva) of adult mice were subjected to scRNA-seq and scATAC-seq using the 10×Genomics platform. Cell types were clustered by the graph-based visualization method uniform manifold approximation and projection and unbiased computational informatics analysis. The scRNA-seq and scATAC-seq datasets were integrated following the integration pipeline described in ArchR and Seurat. Results: We characterized diverse corneal epithelial cell types based on gene expression signatures and chromatin accessibility. We found that cell type-specific accessibility regions were mainly located at distal regions, suggesting essential roles of distal regulatory elements in determining corneal epithelial cell diversity. Trajectory analyses revealed a continuum of cell state transition and higher coordination between transcription factor (TF) motif accessibility and gene expression during corneal epithelial cell differentiation. By integrating transcriptomic and chromatin accessibility analysis, we identified cell type-specific and shared gene regulation programs. We also uncovered critical TFs driving corneal epithelial cell differentiation, such as nuclear factor I (NFI) family members, Rarg, Elf3. We found that nuclear factor-κB (NF-κB) family members were positive TFs in limbal cells and some superficial cells, but they were involved in regulating distinct biological processes. Conclusions: Our study presents a comprehensive gene regulatory landscape of mouse cornea epithelial cells, and provides valuable foundations for future investigation of corneal epithelial homeostasis in the context of cornea pathologies and regenerative medicine.

Two new complexes [Cu(I)(3)(L1)I(3)](n) (1, L1 = 2,5-bis(4-pyridyl)-1,3,4-oxadiazole) and [Cu(I)(3)(L2)I(2)](n) (2, L2 = 2,5-bis(4-pyridyl)-1,2,4-triazolate) are controllably formed by using aqueous ammonia to regulate the pH value of the reaction involving CuI and L1. Interestingly, L2 of 2 is in situ generated from the ring transform of L1 when increase the pH value of the reaction. 1 exhibits 2-D layer, while 2 shows 3-D MOFs with a novel 3-nodal 4,4,5-connected net topology of an unprecedented Point (Schlafli) symbol: (4·5(2)·6(2)·7)(5(4)·8(2))(4(3)·5·6(6)). Although both 1 and 2 are built of CuI and similar ligands, different arrangements of CuI chains and ligands endow them with different physical properties. 1 displays a strong pure red luminescence emission, while 2 is nonluminescent and shows a broad absorption band covering the whole UV-vis-NIR spectrum range. The emissive excited states of 1 and the charge transitions of the optical absorption for 2 are solved by DFT calculations.

BACKGROUND: The resection rate of primary liver tumor in China is only about 20%. A lot of patients with moderate and advanced liver tumor may lose the chance of operation. The objective of present research was to study the efficacy of transcatheter arterial chemoembolization (TACE) combined with percutaneous injection of chemical agents and acetic acids in the treatment of patients with primary liver cancer (PLC). METHODS: Thirty-three patients with middle and advanced stage of PLC were divided into two groups: percutaneous injection of chemical agents and acetic acids (15 patients, group A) and TACE (18 patients, group B). RESULTS: Tumor diameter and serum AFP level reduced to 86.6% and 83.3% in group A, and 55.5% and 40% in group B, respectively. There was significant difference between the two groups (P<0.01). The 1, 2, 3, 4-year survival rates of group A were 96.7%, 86.6%, 51.3%, 33.3%, respectively and in group B were 66.7%, 44.4%, 16.7%, 0%, respectively (P<0.01). CONCLUSION: TACE combined with percutaneous injection of chemical agents and acetic acids is efficacious to increase the survival rate of patients with PLC.

Using a flexible methylenebis(3,5-dimethylpyrazole) ligand (H2MDP) with different aromatic carboxylate ligands, six new complexes {[Cd(H2MDP)(OPh)](H2O)}n (1), {[Cd(H2MDP)(MPh)](H2O)(1/10)}n (2), {[Cd(H2MDP)(PPh)](H2O)(3/2)}n (3), {[Co(H2MDP)(OPh)](H2O)2}n (4), {Co(H2MDP)(MPh)}n (5), {Co(H2MDP)(HBTC)}n (6) (H2OPh = phthalic acid, H2MPh = isophthalic acid, H2PPh = terephthalic acid, H3BTC = benzene-1,3,5-tricarboxylic acid) have been isolated under hydrothermal conditions. The crystal structure analyses reveal that complexes 1 and 4 feature two-dimensional undulated sheets with 4(4)·6(2) topology. Both complexes 2 and 3 contain M2L2-type metallocyclic motifs composed of two H2MDP ligands and two metal atoms, while complex 2 features a two-dimensional 4(4)·6(2) network constructed from binuclear cadmium units as four connected nodes, and complex 3 shows a two-dimensional network with 6(3)-hcb topology. Complex 5 affords a one-dimensional chain structure, and complex 6 shows a two-dimensional (4,4) network further connected into a 3D supramolecular structure connected by intermolecular hydrogen bonds and by π-π interactions. In addition, the luminescent properties of complexes 2 and 3 as well as the magnetic properties of 4 and 5 were studied.

Two polyoxometalate-pillared 3D compounds, {Cu(5)(2-ptz)(6)(H(2)O)(4)(SiW(12)O(40))}·4H(2)O 1 and {Cu(9)(2-ptz)(12)(H(2)O)(6)(PMo(12)O(40))(2)}·H(2)O 2 (2-ptz = 5-(2-pyridyl)tetrazole) have been constructed based on different polyoxometalate anions, and copper-organic coordination polymer sheets by a hydrothermal method. Magnetic investigations reveal that both 1 and 2 exhibit antiferromagnetic coupling between the Cu(II) ions. Structural studies show the compound 1 exhibits a typical pcu-type net with the Schälfli symbol of {4(12)·6(3)}, and that compound 2 is a (3,4,6)-connected framework with novel {4(4)·6(10)·10}{6(3)}(2){6(5)·8} topology which has not been reported to date.
