The brown trout (Salmo trutta) is a salmonid residing in riverine and coastal waters throughout the Northern Hemisphere, whose various populations evolved into distinct ecological morphs, differing in their migratory tendencies and preferred habitats. Unfortunately, due to progressing degradation of natural environment, the conservation of these populations is of growing importance and is undoubtedly a challenging task. Therefore, various means to refine the preparatory protocols for restocking using hatchery-reared fish are being pursued, some of which involve the administration of immunity-boosting substances. The current study assessed the effects of two dietary immunostimulants: Bioimmuno (4% inosine pranobex and 96% ß-glucan) and Focus Plus (commercial preparation by Biomar, Denmark) on two morphs of the brown trout - the river trout (S. trutta morpha fario) and the sea trout (S. trutta morpha trutta). Tissue samples were obtained from ∼75 to 100g fish after 0, 2 and 4 weeks of experimental feeding. Multi-factorial analysis of conducted histological measurements of melanomacrophage centers (MMCs) revealed no changes of their parameters within spleens, but showed a decrease of the occupied tissue area and MMC counts in the livers, progressing with time regardless of the applied diet. Immunohistochemical analysis of CD3+ T cells showed their increased recruitment into mucosal folds of pyloric caeca in the 2-week sampling of trouts fed with the diet with 2% Bioimmuno addition, but this effect was not present in the 4-week sampling. When studying all groups jointly within each morph, there was a significant difference in terms of maintained CD3+ T cells levels, as sea trouts showed significantly higher tissue areas occupied by these cells than river trouts, both in the pyloric caeca and hepatic parenchyma. The study revealed that feeding with a diet enriched with Bioimmuno for 2 weeks may be a favorable enhancement of rearing protocols of brown trout stocks prior to their release, but more studies need to be conducted to test the possibility of an even shorter feeding period.

BACKGROUND: The balance of the intestinal commensal microbiome of fish and other animals plays an important role in the physiological processes of healthy animals, contributes to the defense against pathogens, stimulates the immune system and facilitates nutrient metabolism. In the last decade, the interest in the application of the insects in fish nutrition increased, although little is known regarding the effects of insect meals on the gastrointenstinal tract microbiome of the sea trout fingerlings. The aim of this study was to evaluate the effect of two diets containing mealworm (MW) and superworm (SW) on the microbiome of the digesta of sea trout fingerlings and the relative abundances of different taxa among communities under controlled conditions. RESULTS: The insect meals produced a similar weight gain and survival rate to sea trout fed fishmeal. The most abundant bacterial phylum in all the treatment groups was Firmicutes followed by Proteobacteria and Actinobacteria, and significant differences in the amount of Cyanobacteria were observed in the SW group. CONCLUSIONS: The insect meals did not produce differences in the three most abundant phyla in the sea trout digesta. However, the effect of each type of meal on the lower taxonomic levels was evident, particularly in the case of the superworm meal. These microbiome differences indicated that mealworm meal was more related to fishmeal than superworm meal. Our results highlight the potential effects of insect meals, such as mealworm and superworm meals, on the microbiota of sea trout.

Proliferative kidney disease caused by the myxozoan parasite Tetracapsuloides bryosalmonae has been actively studied in juvenile salmonids for decades. However, very little is known about parasite prevalence and its geographical and intra-host distribution at older life stages. We screened T. bryosalmonae among adult sea trout (Salmo trutta) (n = 295) collected along the Estonian Baltic Sea coastline together with juvenile trout from 33 coastal rivers (n = 1752) to assess spatial infection patterns of the adult and juvenile fish. The parasite was detected among 38.6% of adult sea trout with the prevalence increasing from west to east, and south to north, along the coastline. A similar pattern was observed in juvenile trout. Infected sea trout were also older than uninfected fish and the parasite was detected in sea trout up to the age of 6 years. Analysis of intra-host distribution of the parasite and strontium to calcium ratios from the otoliths revealed that (re)infection through freshwater migration may occur among adult sea trout. The results of this study indicate that T. bryosalmonae can persist in a brackish water environment for several years and that returning sea trout spawners most likely contribute to the parasite life cycle by transmitting infective spores.

The rivers of southern England and northern France which drain into the English Channel contain several genetically unique groups of trout (Salmo trutta L.) that have suffered dramatic declines in numbers over the past 40 years. Knowledge of levels and patterns of genetic diversity is essential for effective management of these vulnerable populations. Using restriction site-associated DNA sequencing (RADseq) data, we describe the development and characterisation of a panel of 95 single nucleotide polymorphism (SNP) loci for trout from this region and investigate their applicability and variability in both target (i.e., southern English) and non-target trout populations from northern Britain and Ireland. In addition, we present three case studies which demonstrate the utility and resolution of these genetic markers at three levels of spatial separation:(a) between closely related populations in nearby rivers, (b) within a catchment and (c) when determining parentage and familial relationships between fish sampled from a single site, using both empirical and simulated data. The SNP loci will be useful for population genetic and assignment studies on brown trout within the UK and beyond.

Brown trout (Salmo trutta L. 1758) and Arctic charr [Salvelinus alpinus (L. 1758)] tagged with acoustic transmitters migrated from fresh water to the sea mainly in May and June, but with large individual variation in migration timing. For S. trutta, large individuals (42-86 cm total length) migrated earlier in the season than small individuals (18-27 cm). For S. alpinus, no such pattern was found, likely because of the small size range of tagged fish (28-41 cm). S. trutta stayed longer at sea than S. alpinus (average 2 vs. 1 month). Early migrants of S. trutta stayed for a shorter period at sea than late migrants, whereas no such pattern was observed for S. alpinus. Large S. trutta moved quickly away from the river and spent average 3 days to reach a receiver line 20 km from the river mouth, whereas small S. trutta and S. alpinus migrating that far spent 2-3 weeks on the same distance. S. trutta utilized the entire fjord system and had a greater proportion of long-distance migrants (>20 km, 78% and 59% of large and small S. trutta, respectively) than S. alpinus (29%). S. alpinus mostly stayed in the inner fjord areas, and none were recorded in the outermost part of the fjord. The difference in the use of marine areas may be caused by variation in prey choice and spatial distribution of the preferred prey groups. Stable isotope analysis showed that S. trutta had been feeding at a higher trophic level than S. alpinus. S. trutta had mainly fed on marine fish and shrimps, whereas S. alpinus had large proportions of freshwater invertebrates in the diet, suggesting that the estuary with benthos and amphipods drifting from the river was an important feeding habitat for S. alpinus. In conclusion, major differences in habitat use, migration patterns and feeding strategies were found between sympatric anadromous S. trutta and S. alpinus while at sea.

We tested for reproductive isolation between Salmo trutta abanticus, S.t. labrax, S.t. caspius and S.t. fario by conducting crosses to produce F1 and F2 offspring. We also estimated the extent of genetic divergence between all three entities by examining sequence variation across the coI, d-loop and cytb mitochondrial genes. All of the F1 cross-types were successfully produced. After 2 years of culturing, F2 generation were produced as well. Fertilization, hatching and survival rates and hatching performance of F1 and F2 generations were evaluated. F2 generation had similar performance to their parent. Fertilization, hatching, larval survival rate and hatchery performance of F1 and F2 generation were similar except pure bred F2 S.t. abanticus. Purebred F1 individuals shared similar coloration patterns and spots with their parents but direction of the hybridization appeared to be decisive on morphology of hybrids. Some of the hybrids exhibit different morphological characters than their parents. Based on partial alignments of the three genes, phylogenetic analysis showed that these S. trutta are gathering within the same clade and appeared as monophyletic group. We found that there were some morphologic and genetic variation among S. trutta subspecies but the degree of variation does not warrant species level recognition. These findings indicate that the four subspecies constitute a single biological entity, corresponding to different morphs of the Danubian lineage. We therefore recommend that S. trutta belonging to Danubian lineage in Turkey be referred to as Salmo trutta and that strains be named according to location, such as Abant, Caspian, Black Sea and Anatolian.

Study of anadromous brown trout Salmo trutta in Orkney, U.K., burns (small streams) with a common-garden sea in Scapa Flow supports the key role of nutrient availability in fresh water, independent of day length, as a determinant of smolt age, with a systematic increase in mean smolt age from 1 to 3 years related inversely to productivity. Whole catchment (8 km2 ) population budgets indicated annual smolt production of around 650 individuals from approximately 100 spawners. Egg-to-smolt survival was 0·65%, while marine survival was estimated from mark-recapture to be between 3·5 and 10%. The question of B-type growth (accelerated growth immediately prior to or during smolt migration) was also addressed, with a strong negative correlation between B-type growth and size at end of winter suggesting that this represents a freshwater compensatory growth response. The data obtained indicate the potential importance of small catchments for supporting anadromous Salmo trutta populations and suggest that small runs of spawners (<100 individuals) are adequate to maintain stocks in such situations. Furthermore, they support the key role of freshwater productivity in determining life-history characteristics over small spatial scales, with Orkney providing a useful natural laboratory for future research into metapopulation genetic structuring and environmental factors at a tractable scale.

A total of 52 sea trouts, Salmo trutta trutta, were studied for parasites, originating from German freshwater streams and coastal waters of the Baltic Sea. While 35 specimens were caught mainly close to the shoreline in the Baltic Sea, 17 were sampled during their spawning migration in Warnow River and other neighboring rivers in Mecklenburg-Western Pomerania. A total of 12 different metazoan parasite species were found in sea trout originating from the Baltic Sea, including five digeneans, two cestodes, three nematodes, and two acanthocephalans. Marine and freshwater species were found. In the migratory trout, seven different parasite species were recorded (one digenean, two cestodes, one nematode, and one acanthocephalan), demonstrating lower parasite diversity and load during the spawning migration compared with the fish from the Baltic Sea. The anisakid nematodes Anisakis simplex (sensu stricto), Contraceacum rudolphii, and Hysterothylacium aduncum were identified by molecular analyses of the ITS-1, 5.8S, ITS-2, and flanking sequences of the rDNA. Together with the digenean Derogenes varicus, Hemiurus communis and H. luehei, and the cestode Diphyllobothrium dendriticum seven new host records for sea trout from the German part of the Baltic Sea are made.

The aim of this study was to test the hypothesis that hatchery brown trout Salmo trutta smolts, with 50% reduced or no feeding over the last 5 months before release, were more likely to migrate to the sea than individuals with standard feeding ratios. The juvenile fish were divided into three groups 176 days before release: (A) with no feeding, (B) with 50% and (C) with 100% feeding. To study their seaward migration, 40 fish from each feeding group were tagged with acoustic transmitters and tracked by automatic listening stations in the River Nidelva, Trondheim, Norway, its estuary and in the nearest marine environment. At the time of release, mean condition factor was significantly lower in group A and the fish from groups A and B had higher levels of Na+, K+-ATPase. Significantly more fish from group A migrated to the sea, but the rate of downstream progression from release to the estuary did not differ between the three groups. In conclusion, the S. trutta smolts with no access to food in the last 176 day before release were more likely to migrate to the sea. Fish from all three feeding groups, however, appeared to smoltify and had the same rate of downstream progression to the estuary. This indicates that differences in migratory behaviour between individuals from the three feeding groups begin from the time when the fish reach saline waters. It is suggested that feeding in hatcheries has to be greatly reduced (by 50% or more) over several months to have a pronounced effect on the migratory behaviour in S. trutta.

To study migration performance and return rates of hatchery brown trout Salmo trutta smolts the first 5 months after release, 50 fish in each year (fork length, LF , 158-288 mm) were in two subsequent years tagged with acoustic transmitters and recorded by automatic listening stations in the River Nidelva (central Norway), its estuary and in the marine environment. More than half of the smolts became anadromous migrants (52% in 2011 and 70% in 2012). The fish spent longer time in the estuary than in the marine environment and the results suggest that migratory behaviour of S. trutta smolts is not only restricted to be resident or anadrome-lacustrine, but that there is also an intermediary strategy of estuarine feeding. There were no differences in LF or mass between groups of smolts with different migration patterns. Return rates from the sea within the first 5 months after release were in both years 16%. Median progression rate in the river was 0·090 LF s(-1) but decreased significantly as the smolts entered the estuary (0·015 LF s(-1) ). The long residential time in the estuary may increase the risk of negative effects of anthropogenic activities in estuaries, such as harbours and industrial development, and special attention should be given to evaluate effects of such activities.

Several rivers that are tributaries of the Oder estuary are inhabited by Salmo trutta L, the most important of which are Ina, Gowienica, and Wolczenica. Both forms of the species, sea trout and resident brown trout, are present. All rivers are traditionally stocked with either sea trout from the neighboring Pomeranian river Rega basin or resident brown trout from various locations. To examine populations in these rivers in terms of genetic structure, genetic diversity, and origin, they were analyzed using 13 microsatellite loci. Relatedness was also assessed for fish stocked in the same year. The obtained genotypes were compared with breeding stocks used for stocking in Poland. The analyses revealed a significant genetic distance between adult individuals from Ina and Rega Rivers and fish caught during electrofishing. Strong kinship relationships were identified in the sampled areas, with high proportions of fish originating from stocking and their dominance in numbers over wild juveniles, primarily in smaller tributaries. Additionally, clear separation in the origin of stocked individuals was observed. Adult trout from Ina and Rega are genetically closer to northern brown trout lineages, providing crucial information for the management and biodiversity conservation of Polish Salmo trutta populations.

The longest Baltic River, the Vistula, historically hosted numerous sea trout populations. However, dam construction in the twentieth century drastically reduced the spawning migration rate. Reduced natural reproduction has resulted in a population collapse and, consequentially, a substantial reduction in catches. In response, like other Baltic countries, Poland has initiated an intensive stocking program, mainly involving smolt. Initially, stocking was conducted primarily with offspring of sea trout caught during spawning migration. Currently, due to difficulties in obtaining fish, most stocking involves fish from breeding stocks. Therefore, determining the proportion of fish derived from stocking has become an important issue. Experiments based on traditional tagging did not provide sufficient material for analysis; hence, we decided to use genetic methods based on analysis of relatedness. In this study, we performed parentage-based tagging and an analysis of the origins of parent animals used for artificial spawning in 2013, and offspring returning to the Vistula in subsequent years. We based the analysis on three different algorithms and compared the results, showing that the presented methods were effective for estimating mass stocking success. The study also indicated that a certain level of natural reproduction in the Vistula continues to occur. The proportion of sea trout from spawning in 2013 in Vistula sea trout catches from 2017 to 2018 was approximately 30%.

In November 2016, infectious pancreatic necrosis virus (IPNV) was isolated from a broodstock female of landlocked sea trout (Salmo trutta) in Lake Vänern in Sweden. VP2 gene sequencing placed the IPNV isolate in genogroup 6, for which pathogenicity is largely unknown. Lake Vänern hosts landlocked sea trout and salmon populations that are endangered, and thus the introduction of new pathogens poses a major threat. In this study we characterized the novel isolate by conducting an infection trial on three salmonid species present in Lake Vänern, whole genome sequencing of the isolate, and prevalence studies in the wild sea trout and salmon in Lake Vänern. During the infection trial, the pathogenicity of the Swedish isolate was compared to that of a pathogenic genogroup 5 isolate. Dead or moribund fish were collected, pooled, and analyzed by cell culture to identify infected individuals. In the trial, the Swedish isolate was detected in fewer sample pools in all three species compared to the genogroup 5 isolate. In addition, the prevalence studies showed a low prevalence (0.2-0.5%) of the virus in the feral salmonids in Lake Vänern. Together the data suggest that the novel Swedish IPNV genogroup 6 isolate is only mildly pathogenic to salmonids.

The aims of this study were to compare male and female sea trout (Salmo trutta) with respect to their hypo-osmoregulatory ability over a simulated migration to seawater and their clinical and physiological response to salmon louse (Lepeophtheirus salmonis) infection in seawater and over a simulated pre-mature return to freshwater. For this purpose, 2-year-old hatchery-reared male and female brown trout (F1 offspring of wild caught anadromous fish) were infected with salmon lice and measured for changes in plasma ions, glucose, lactate and osmolality and relative heart, liver and gonad sizes during a simulated seawater migration and thereafter a premature return to freshwater after 4 weeks in seawater (pre-adult louse). Un-infected trout served as control. Male trout used longer time to develop full hypo-osmoregulatory ability in seawater and showed a stronger response in plasma glucose and lactate following simulated premature return to freshwater, compared to female trout. Response to salmon louse was stronger in female trout, shown by stronger osmotic stress by chalimus (plasma Cl-) and pre-adult louse (plasma osmolality) and elevated relative liver size (hepatosomatic index) by pre-adult louse in female compared to male trout. Moreover, high plasma cortisol in infected female and low plasma cortisol in infected male trout produced a significant treatment-sex interaction on plasma cortisol. Lice infection intensity was initially higher in male (0.18 lice g-1) compared to female trout (0.11 lice g-1) at the chalimus stage, but equal between sexes at the pre-adult stage (male 0.15 and female 0.17 lice g-1). This study showed that female trout were better adapted for changes in water salinity, while male trout were more robust against salmon louse infection. These results suggests that the elevated salmon louse infection pressure generated by salmon farming have strong and unexplored negative effects on wild sea trout populations. Further research on this topic is vital for the conservation of wild sea trout populations.

Fish absorb dioxins from the environment through water and contact with sediments but the main source is food. These contaminants also enter the human body with food, including through the consumption of fish. This study presents the dioxin and polychlorinated biphenyl (PCB) concentrations in 145 samples of four fish species (herring, sprat, sea trout, salmon) caught in the southern Baltic Sea. The study results permit assessing whether the efforts made in recent years to reduce dioxin and PCB emissions into the environment have translated into decreases in concentrations of these pollutants in Baltic Sea fishes and whether these raw fish materials meet food law requirements. The safest fish species with regard to dioxins were designated, as were those of which consumption should be limited. The influence of fish age (length) and weight on dioxin and dioxin-like polychlorinated biphenyl (dl-PCB) concentrations was investigated. The correlation between fat content and dioxin concentration was determined. The Baltic Sea region and fishing ports from which the least contaminated fish come were also determined. The studies indicated that dioxin and PCB concentrationsin fishes from Polish fishing areas are currently lower than they were twenty years ago.

The impacts and interactions between hatchery-bred fish and wild fish populations has been a topic of active investigation in recent decades. In some instances, the benefits of stocking can be overshadowed by negative effects such as genetic introgression with natural populations, loss of genetic diversity, and dilution of local adaptations. Methods that facilitate the identification of stocked fish enable us to estimate not only the effectiveness of stocking but also the level of natural reproduction and the degree of hybridization. The longest Baltic river, the Vistula, also has the second highest discharge. Historically, it hosted numerous populations of the anadromous form of brown trout (sea trout); however, dam construction has since interfered with and reduced spawning migration to a rate that is much lower than before. Reduced spawning has resulted in a population collapse and a negative flow-on effect on commercial catches. In response, Poland (along with many other Baltic countries) initiated an intensive stocking program which continues today and which sees the average annual release of 700,000 smolts. As a consequence, today's main-river and inshore catches come from stock-enhanced populations. High-throughput single-nucleotide polymorphism (SNP) genotyping was performed on samples of sea trout from southern Baltic populations; results suggest that a significant portion of the sea trout catches in the Vistula mouth region have direct hatchery origin and indicate the presence of Pomeranian specimens. SNP loci identified as outliers indicate a potential selection pressure that may be related with effects of hatchery breeding and mixing with natural populations. The brown trout SNP array applied in this study showed high effectiveness not only for population differentiation, but more importantly, it emerged as a sensitive tool to provide evidence of detection selection.

Nanoproducts are being increasingly used in various industrial products, leading to a greater risk of water pollution through their discharge into environment as production byproducts. Increased levels of environmental pollution with nanoproducts pose a threat to all living organisms. Nanopollutants may have toxic effects on gametes and fertilization process in species with external fertilization, thereby reducing effectiveness of reproduction or greatly impairing it. The objective of the present study was to determine the effect of copper nanoparticles (Cu and CuO) and copper ions (CuSO4·5H2O) on the spermatozoa motility of sea trout and compare their harmful effects. Copper nanoparticles (NPs) of primary particle size <100 nm and CuO NPs of particle size <50 nm as well as a solution of CuSO4·5H2O were used for the study. Concentration of the products tested (CuNPs, CuONPs, and CuSO4·5H2O) was expressed as mg Cu L-1 in the media regardless of dissolution and aggregation. A suspension with concentration up to 500 mg L-1 in an aquatic environment was tested. Spermatozoa motility under direct contact with copper at 10 s post activation was evaluated using an automated system (CASA). Copper products in an aqueous environment primarily cause a reduction of spermatozoa velocity (VCL). A significant reduction of velocity in CuSO4·5H2O salt solution occurred at concentration starting from 8 mg L-1, whereas in solutions containing CuNPs and CuONPs this process is observed from 50 mg L-1. Reduction of percentage of motile spermatozoa occurred in CuNPs from a concentration of 125 mg L-1, while in CuONPs and CuSO4·5H2O, the effect began at concentration from 250 mg L-1. In a solution of CuSO4·5H2O at a concentration of 500 mg L-1, strong agglutination of sperm cells occurred and movement ceased (immobilization IC100). Exposure to CuONPs decreased the motility rate to below IC50, while for CuNPs, the motility rate reached 80%. Changes in linearity (LIN) and amplitude of head displacement (ALH) differed according to Cu products. Cu as CuSO4·5H2O and CuONPs impaired the LIN, while CuNPs lowered ALH. Duration of motility increased at low CuSO4·5H2O concentration. The effect of Cu ion on sea trout spermatozoa motility was more harmful than that of copper nanoparticles. The various copper products used affected motility parameters differently. Contamination of aqueous environment with copper primarily caused reduction in male gamete velocity, which may impair reproduction. Various copper products had different effects on LIN, ALH, and motility duration, which can also affect fertilization.

Using a stage- and sex-based multivariate significance tests on the sea trout Salmo trutta m. trutta L. model, we show dependencies in the balance between lipid peroxidation processes, levels of carbonyl derivatives, and activity of antioxidant enzymes (superoxide dismutase SOD, catalase CAT, glutathione reductase GR, and peroxidase GPx) in the processes of antioxidant profile formation during the fish growing process. The study was aimed at examination of the relationships between the biomarkers of oxidative stress estimated by the total antioxidant status as well as the dependencies between the sex (male, female) and developmental stage of the wild sea trout from the Slupia River and its catchment area rivers. Functioning of the pro/antioxidant balance of the liver tissue reflected the course of the individual developmental stages of the trout and was associated with significant intensification of lipoperoxidation, oxidative modification of proteins, and reduction of the total antioxidant capacity of fish along with age. Formation of a holistic model for the analysis of the involvement of all parameters of antioxidant protection in all stages of development and sex allowed us to obtain the following rank order for the level of lipoperoxidation processes, modified proteins, and antioxidant enzyme complex: CAT > SOD > GPx > GR and TBARS > OMP KD > TAC > OMP AD.

The extent to which no-take marine reserves can benefit anadromous species requires examination. Here, we used acoustic telemetry to investigate the spatial behavior of anadromous brown trout (sea trout, Salmo trutta) in relation to a small marine reserve (~1.5 km2) located inside a fjord on the Norwegian Skagerrak coast. On average, sea trout spent 42.3 % (±5.0% SE) of their time in the fjord within the reserve, a proportion similar to the area of the reserve relative to that of the fjord. On average, sea trout tagged inside the reserve received the most protection, although the level of protection decreased marginally with increasing home range size. Furthermore, individuals tagged outside the reserve received more protection with increasing home range size, potentially opposing selection toward smaller home range sizes inflicted on fish residing within reserves, or through selective fishing methods like angling. Monthly sea trout home ranges in the marine environment were on average smaller than the reserve, with a mean of 0.430 (±0.0265 SE) km2. Hence, the reserve is large enough to protect the full home range of some individuals residing in the reserve. Synthesis and applications: In general, the reserve protects sea trout to a varying degree depending on their individual behavior. These findings highlight evolutionary implications of spatial protection and can guide managers in the design of marine reserves and networks that preserve variation in target species' home range size and movement behavior.

The aim of this study was to confirm the taxonomic status of Piscicola pojmanskae Bielecki, 1994 found on Salmonidae fish. The fish leech was identified based on a diligent morphological analysis as well as COI gene sequence (DNA barcoding). The phylogenetic relationship with other piscicolid leeches was analyzed as well. Piscicola pojmanskae was found on various fins of both graylings and the resident form of trouts. The prevalence of infection was 1.63%. In this paper, probable causes of the lack of relation between the number of leeches on the fins and the fish body length as well as the host-searching strategy used by P. pojmanskae are discussed.
