RESUMO

Squalene synthase (SQS) is a key enzyme in plant terpenoid biosynthetic pathway. This study focused on cloning and analysis of Huperzia serrata SQS (HsSQS1) gene. After searching the transcriptome dataset of H serrata, one unique sequence encoding SQS was discovered. The primers were designed according to the transcript sequence of HsSQS1 from the H. serrata transcriptome dataset. The open reading frame of HsSQS1 was cloned using RT-PCR strategy. The bioinformatic analysis of this gene and its corresponding protein were performed. The cDNA (named as HsSQS1) contains a 1263 bp open reading frame and encodes a predicted protein of 420 amino acids. The GenBank accession number for this gene is JQ004938. HsSQS1 contains two transmembrane regions, without signal peptide. The conserved domain of squalene synthase was presented in HsSQS1. HsSQS1 was more abundant in H. serrata root than in leaf and stem. This study cloned and analyzed squalene synthase gene from H. serrata for the first time. The result will provide a foundation for exploring the mechanism ofterpenoid biosynthesis in H. serrata plants.

RESUMO

OBJECTIVE: To analyze the volatile aromatic substances in Chaenomeles speciosa fruit produced in Chongqing in order to provide the characteristic data for it's resources development and flavors chemistry research. METHODS: The volatile aromatic substances were extracted by steam distillation from Chaenomeles speciosa fresh fruit and seperated and identified by GC-MS. RESULTS: 106 volatile aromatic substances were seperated and identified, mainly including esters, alcohols, carboxylic acids, alkanes and alkenes, ketones, which made a great contribution to flavor of Chaenomeles speciosa fresh fruit. CONCLUSION: This study elucidated the composition of volatile aromatic substances in Chaenomeles speciosa fruit produced in Chongqing, which could provide basic information for exploitation and utilization of it's flavor substances.

RESUMO

OBJECTIVE: To evaluate the quality of Fructus Toosendan. METHODS: HPLC method was used to determine the content of rutin, isoquercitrin and quercetin in Fructus Toosendan from different origins, and the HPLC fingerprints were established correspondingly. RESULTS: There were significant differences among the HPLC fingerprints, and the contents of rutin, isoquercitrin and quercetin were different in Fructus Toosendan from different origins. CONCLUSION: Quantitative analysis of effective component and integral analysis of HPLC fingerprints can be used for the quality evaluation of Fructus Toosendan.

RESUMO

OBJECTIVE: To study the chemical constituents in the rhizome of Elaeagnus bockii. METHOD: Compounds were isolated by silica gel and Sephadex LH-20 chromatography. Their structures were elucidated by means of spectral analysis. RESULT: Seven compounds were isolated from the rhizomes of E. bockii, and their structures were identified as angelicin (1), psoralen (2), vanillic acid (3), bakuchiol (4), oleanolic acid (5), ursolic acid (6) and beta-sitosterol (7 ). CONCLUSION: All these compounds were obtained from E. bockii for the first time.
