RESUMO

Urban sanitation infrastructure is inadequate in many low-income countries, leading to the presence of highly concentrated, uncontained fecal waste streams in densely populated areas. Combined with mechanisms of aerosolization, airborne transport of enteric microbes and their genetic material is possible in such settings but remains poorly characterized. We detected and quantified enteric pathogen-associated gene targets in aerosol samples near open wastewater canals (OWCs) or impacted (receiving sewage or wastewater) surface waters and control sites in La Paz, Bolivia; Kanpur, India; and Atlanta, USA, via multiplex reverse-transcription qPCR (37 targets) and ddPCR (13 targets). We detected a wide range of enteric targets, some not previously reported in extramural urban aerosols, with more frequent detections of all enteric targets at higher densities in La Paz and Kanpur near OWCs. We report density estimates ranging up to 4.7 × 102 gc per mair3 across all targets including heat-stable enterotoxigenic Escherichia coli, Campylobacter jejuni, enteroinvasive E. coli/Shigella spp., Salmonella spp., norovirus, and Cryptosporidium spp. Estimated 25, 76, and 0% of samples containing positive pathogen detects were accompanied by culturable E. coli in La Paz, Kanpur, and Atlanta, respectively, suggesting potential for viability of enteric microbes at the point of sampling. Airborne transmission of enteric pathogens merits further investigation in cities with poor sanitation.

Antibiotic resistance poses a major global health threat. Understanding emergence and dissemination of antibiotic resistance in environmental media is critical to the design of control strategies. Because antibiotic resistance genes (ARGs) may be aerosolized from contaminated point sources and disseminated more widely in localized environments, we assessed ARGs in aerosols in urban La Paz, Bolivia, where wastewater flows in engineered surface water channels through the densely populated urban core. We quantified key ARGs and a mobile integron (MI) via ddPCR and E. coli spp. as a fecal indicator by culture over two years during both the rainy and dry seasons in sites near wastewater flows. ARG targets represented major antibiotic groups-tetracyclines (tetA), fluoroquinolines (qnrB), and beta-lactams (blaTEM)-and an MI (intI1) represented the potential for mobility of genetic material. Most air samples (82%) had detectable targets above the experimentally determined LOD: most commonly blaTEM and intI1 (68% and 47% respectively) followed by tetA and qnrB (17% and 11% respectively). ARG and MI densities in positive air samples ranged from 1.3 × 101 to 6.6 × 104 gene copies/m3 air. Additionally, we detected culturable E. coli in the air (52% of samples <1 km from impacted surface waters) with an average density of 11 CFU/m3 in positive samples. We observed decreasing density of blaTEM with increasing distance up to 150 m from impacted surface waters. To our knowledge this is the first study conducting absolute quantification and a spatial analysis of ARGs and MIs in ambient urban air of a city with contaminated surface waters. Environments in close proximity to urban wastewater flows in this setting may experience locally elevated concentrations of ARGs, a possible concern for the emergence and dissemination of antimicrobial resistance in cities with poor sanitation.

The health effects associated with human exposure to airborne fine particulate matter (PM2.5) have been linked to the ability of PM2.5 to facilitate the production of excess cellular reactive oxygen species (oxidative potential). Concern about the adverse human health impacts of PM2.5 has led to the increased use of indoor air cleaners to improve indoor air quality, which can be an important environment for PM2.5 exposure. However, the degree to which the oxidative potential of indoor and personal PM2.5 can be influenced by an indoor air cleaner remains unclear. In this study we enrolled 43 children with physician diagnosed asthma in suburban Shanghai, China and collected two paired-sets of 48-h indoor, outdoor, and personal PM2.5 exposure samples. One set of samples was collected under "real filtration" during which a functioning air cleaner was installed in the child's bedroom, and the other ("false filtration") with an air cleaner without internal filters. The PM2.5 samples were characterized by inductively coupled plasma mass spectroscopy for elements, and by an alveolar macrophage assay for oxidative potential. The sources of metals contributing to our samples were determined by the EPA Positive Matrix Factorization model. The oxidative potential was lower under real filtration compared to sham for indoor (median real/sham ratio: 0.260) and personal exposure (0.813) samples. Additionally, the sources of elements in PM2.5 that were reduced indoors and personal exposure samples by the air cleaner (e.g. regional aerosol and roadway emissions) were found by univariate multiple regression models to be among those contributing to the oxidative potential of the samples. An IQR increase in the regional aerosol and roadway emissions sources was associated with a 107% (95% CI: 80.1-138%) and 38.1% (17.6-62.1%) increase in measured oxidative potential respectively. Our results indicate that indoor air cleaners can reduce the oxidative potential of indoor and personal exposure to PM2.5, which may lead to improved human health.

The urban heat island effect causes increased heat stress in urban areas. Cool roofs and urban greening have been promoted as mitigation strategies to reduce this effect. However, evaluating their efficacy remains a challenge, as potential temperature reductions depend on local characteristics. Existing methods to characterize their efficacy, such as computational fluid dynamics and urban canopy models, are computationally burdensome and require a high degree of expertise to employ. We propose a data-driven approach to overcome these hurdles, inspired by recent innovations in spatial causal inference. This approach allows for estimates of hypothetical interventions to reduce the urban heat island effect. We demonstrate this approach by modeling evening temperature in Durham, North Carolina, using readily retrieved air temperature, land cover, and satellite data. Hypothetical interventions such as lining streets with trees, cool roofs, and changing parking lots to green space are estimated to decrease evening temperatures by a maximum of 0.7-0.9   [Formula: see text], with reduced effects on temperature as a function of distance from the intervention. Because of the ease of data access, this approach may be applied to other cities in the U.S. to help them come up with city-specific solutions for reducing urban heat stress.

Railyards have the potential to influence localfine particulate matter (aerodynamic diameter < or = 2.5 microm; PM2.5) concentrations through emissions from diesel locomotives and supporting activities. This is of concern in urban regions where railyards are in proximity to residential areas. Northwest of Atlanta, Georgia, Inman and Tilford railyards are located beside residential neighborhoods, industries, and schools. The PM2.5 concentrations near the railyards is the highest measured amongst the state-run monitoring sites (Georgia Environmental Protection Division, 2012; http://www.georgiaair.org/amp/report.php). The authors estimated fuel-based black carbon (BC) and PM2.5 emission factors for these railyards in order to help determine the impact of railyard activities on PM2.5 concentrations, and for assessing the potential benefits of replacing current locomotive engines with cleaner technologies. High-time-resolution measurements of BC, PM2.5, CO2, and wind speed and direction were made at two locations, north and south of the railyards. Emissions factors (i.e., the mass of BC or PM2.5 per gallon of fuel burned) were estimated by using the downwind/upwind difference in concentrations, wavelet analysis, and an event-based approach. By the authors' estimates, diesel-electric engines used in the railyards have average emission factors of 2.8 +/- 0.2 g of BC and 6.0 +/- 0.5 g of PM2.5 per gallon of diesel fuel burned. A broader mix of railyard supporting activities appear to lead to average emission factors of 0.7 +/- 0.03 g of BC and 1.5 +/- 0.1 g of PM2.5 per gallon of diesel fuel burned. Railyard emissions appear to lead to average enhancements of approximately 1.7 +/- 0.1 microg/m3 of PM2.5 and approximately 0.8 +/- 0.01 microg/m3 of BC in neighboring areas on an annual average basis. Uncertainty not quantified in these results could arise mainly from variability in downwind/upwind differences, differences in emissions of the diverse zones within the railyards, and the influence of on-road mobile source emissions.

Surgery of the middle ear is a delicate process that requires the surgeon to manipulate the ossicles, the smallest bones in the body. Excessive force applied to the ossicles can easily be transmitted through to the inner ear which may cause a permanent sensorineural hearing loss. An instrument was required to measure the forces applied to cadaveric temporal bone ossicles with the vision of measuring forces in vivo at a later stage. A feasibility study was conducted to investigate a method of measuring force and torque applied to the ossicles of the middle ear. Information from research papers was gathered to determine the expected amplitudes. The study looked at commercially available transducers as well as constructing an instrument using individual axis transducers coupled together. A prototype surgical instrument was constructed using the ATI industrial automation Nano17 six axis transducer. The Nano17 allows for the measurement of force and torque in the X, Y and Z axis to a resolution of 1/320 N. The use of the Nano17 enabled rapid development of the surgical instrument. It meets the requirements for its use on cadaveric models and has the potential to be a useful data collection tool in vivo.

Increased public awareness of the health impacts of atmospheric fine particulate matter (PM2.5) has led to increased demand and deployment of indoor air cleaners. Yet, questions still remain about the effectiveness of indoor air cleaners on indoor PM2.5 concentrations and personal exposure to potentially hazardous components of PM2.5. Metals in PM2.5 have been associated with adverse health outcomes, so knowledge of their sources in urban indoor and outdoor areas and how exposures are influenced by indoor air cleaners would be beneficial for public health interventions. We collected 48-h indoor, outdoor, and personal PM2.5 exposure samples for 43 homes with asthmatic children in suburban Shanghai, China during the spring months. Two sets of samples were collected for each household, one set with a functioning air filter placed in the bedroom ("true filtration") and the other with a non-functioning ("sham") air cleaner. PM2.5 samples were analyzed for elements, elemental carbon, and organic carbon. The major sources of metals in PM2.5 were determined by Positive Matrix Factorization (PMF) to be regional aerosol, resuspended dust, residual oil combustion, roadway emissions, alloy steel abrasion, and a lanthanum (La) and cerium (Ce) source. Under true filtration, the median indoor to outdoor percent removal across all elements increased from 31% to 78% and from 46% to 88% across all sources. Our findings suggest that indoor air cleaners are an effective strategy for reducing indoor concentrations of PM2.5 metals from most sources, which could translate into improved health outcomes for some populations.

In many developing regions with poor air quality, the use of air filtration devices to clean indoor air is growing rapidly. In this study, we collected indoor, outdoor and personal exposure filter-based samples of fine particulate matter (PM2.5) with both properly operating, and sham air cleaners in six Beijing residences from July 24th to August 17th, 2016. Mass concentrations of PM2.5 and several health relevant components of PM2.5 including organic carbon, elemental carbon, sulfate, nitrate, ammonium, and 21 selected metals, were analyzed to evaluate the effectiveness of air cleaners. The effect of air purification on PM2.5 reactive oxygen species (ROS) activity, a metric of the oxidative potential of the aerosol, was also evaluated. The average indoor PM2.5 concentration during true filtration was 8.47µg/m3, compared to 49.0µg/m3 during sham filtration; thus, air cleaners can significantly reduce the indoor PM2.5 concentration to well below WHO guideline levels and significantly lower all major components of PM2.5. However, the utility of air cleaners in reducing overall personal exposure to PM2.5 and its components was marginal in this study: the average personal exposure PM2.5 concentration was 67.8 and 51.1µg/m3 during true and sham filtration respectively, and it is likely due to the activity patterns of the subjects. Short-term exposure contributions from environments with high PM2.5 concentrations, including exposure to traffic related emissions as well as uncharacterized indoor microenvironments, likely add substantially to the total PM2.5 exposure burden. The toxicity assay indicates that the air cleaners can also significantly reduce ROS activity in the indoor environment; however, this decrease did not translate to a reduction in personal exposure. Elemental carbon, lead, and arsenic were well-correlated with the ROS activity, thus adding to the knowledge base of drivers for ROS activity.

An enzyme-based monitoring system provides the basis for continuous sampling of organophosphate contamination in air. The enzymes butyrylcholinesterase (BuChE) and organophosphate hydrolase (OPH) are stabilized by encapsulation in biomimetic silica nanoparticles, entrained within a packed bed column. The resulting immobilized enzyme reactors (IMERs) were integrated with an impinger-based aerosol sampling system for collection of chemical contaminants in air. The sampling system was operated continuously and organophosphate detection was performed in real-time by single wavelength analysis of enzyme hydrolysis products. The resulting sensor system detects organophosphates based on either enzyme inhibition (of BuChE) or substrate hydrolysis (by OPH). The detection limits of the IMERs for specific organophosphates are presented and discussed. The system proved suitable for detection of a range of organophosphates including paraoxon, demeton-S and malathion.

An investigation of the potential environmental and health impacts in the immediate aftermath of one of the largest coal ash spills in U.S. history at the Tennessee Valley Authority (TVA) Kingston coal-burning power plant has revealed three major findings. First the surface release of coal ash with high levels of toxic elements (As = 75 mg/kg; Hg = 150 microg/kg) and radioactivity (226Ra + 228Ra = 8 pCi/g) to the environment has the potential to generate resuspended ambient fine particles (< 10 microm) containing these toxics into the atmosphere that may pose a health risk to local communities. Second, leaching of contaminants from the coal ash caused contamination of surface waters in areas of restricted water exchange, but only trace levels were found in the downstream Emory and Clinch Rivers due to river dilution. Third, the accumulation of Hg- and As-rich coal ash in river sediments has the potential to have an impact on the ecological system in the downstream rivers by fish poisoning and methylmercury formation in anaerobic river sediments.
