RESUMO

The impact of dietary phosphorus on chronic renal disease in cats, humans and other species is receiving increasing attention. As Ca and P metabolism are linked, the ratio of Ca:P is an important factor for consideration when formulating diets for cats and other animals. Here, we describe a fully randomised crossover study including twenty-four healthy, neutered adult cats, investigating postprandial responses in plasma P, ionised Ca and parathyroid hormone (PTH) following one meal (50 % of individual metabolic energy requirement) of each of six experimental diets. Diets were formulated to provide P at either 0·75 or 1·5 g/1000 kcal (4184 kJ) from the soluble phosphorus salt sodium tripolyphosphate (STPP, Na5P3O10), variable levels of organic Ca and P sources, and an intended total Ca:P of about 1·0, 1·5 or 2·0. For each experimental diet, baseline fasted blood samples were collected prior to the meal, and serial blood samples collected hourly for 6 h thereafter. For all diets, a significant increase from baseline was observed at 120 min in plasma PTH (P < 0·001). The diet containing the highest STPP inclusion level and lowest Ca:P induced the highest peaks in postprandial plasma P and PTH levels (1·8 mmol/l and 27·2 pg/ml, respectively), and the longest duration of concentrations raised above baseline were observed at 3 h for P and 6 h for PTH. Data indicate that Ca:P modulates postprandial plasma P and PTH. Therefore, when formulating diets containing soluble P salts for cats, increasing the Ca:P ratio should be considered.

Early responses in healthy adult dogs fed grain-free diets with high inclusion of split peas (20%) and lentils (40%) that may lead to canine diet-induced dilated cardiomyopathy (DCM) were investigated. To help understand the clinical relevance of the findings, a survey of electronic health records (EHR) was conducted of dogs with and without suspected DCM for comparison. Control and Test diets were fed to Labrador retriever dogs for 30 days (n = 5 and 6, respectively). Blood and urine samples collected at baseline and days 3, 14 and 28/30 were analyzed for hematology, clinical biochemistry and taurine concentrations. The EHRs of dogs at Banfield® Pet Hospitals in the 2-year period 2018-2019 were surveyed, revealing 420 dogs diagnosed with DCM, which were compared with 420 breed, gender and age-matched healthy control dogs. Compared to baseline values, feeding the Test diet for 28 days caused progressive, significant (p < 0.001) decreases in red blood cell counts (RBC), hematocrit and total hemoglobin by 7.7, 8.3 and 6.3%, respectively, and a 41.8% increase in plasma inorganic phosphate. Commonalities in these parameters were observed in clinical DCM cases. Regarding taurine status, Test dogs transiently increased whole-blood (23.4%) and plasma (47.7%) concentrations on day 14, while taurine:creatinine ratio in fresh urine and taurine in pooled urine were reduced by 77 and 78%, respectively, on day 28/30. Thus grain-free, legume-rich Test diets caused reduced RBC and hyperphosphatemia, findings also indicated in dogs with suspected DCM. Changes in taurine metabolism were indicated. The data will aid in generating hypotheses for future studies.

High dietary phosphorus (P), particularly soluble salts, may contribute to chronic kidney disease development in cats. The aim of the present study was to assess the safety of P supplied at 1 g/1000 kcal (4184kJ) from a highly soluble P salt in P-rich dry format feline diets. Seventy-five healthy adult cats (n 25/group) were fed either a low P control (1·4 g/1000 kcal [4184kJ]; Ca:P ratio 0·97) or one of two test diets with 4 g/1000 kcal (4184 kJ); Ca:P 1·04 or 5 g/1000 kcal (4184kJ); Ca:P 1·27, both incorporating 1 g/1000 kcal (4184 kJ) sodium tripolyphosphate (STPP) - for a period of 30 weeks in a randomised parallel-group study. Health markers in blood and urine, glomerular filtration rate, renal ultrasound and bone density were assessed at baseline and at regular time points. At the end of the test period, responses following transition to a commercial diet (total P - 2·34 g/1000 kcal [4184kJ], Ca:P 1·3) for a 4-week washout period were also assessed. No adverse effects on general, kidney or bone (skeletal) function and health were observed. P and Ca balance, some serum biochemistry parameters and regulatory hormones were increased in cats fed test diets from week 2 onwards (P ≤ 0·05). Data from the washout period suggest that increased serum creatinine and urea values observed in the two test diet groups were influenced by dietary differences during the test period, and not indicative of changes in renal function. The present data suggest no observed adverse effect level for feline diets containing 1 g P/1000 kcal (4184 kJ) from STPP and total P level of up to 5 g/1000 kcal (4184 kJ) when fed for 30 weeks.

Phosphorus is present in diets as naturally occurring P from raw materials or added as an inorganic salt. However, little is known about postprandial kinetics of P absorption in cats. Here, we describe several studies quantifying postprandial kinetics following the ingestion of diets of varying composition. Briefly, cats were fed a meal consisting of 50 % of their metabolic energy requirement in a randomised crossover design. A pre-meal baseline blood sample was taken via cephalic catheter and repeated measurements taken regularly up to 6 h post-meal to assess the whole blood ionised Ca, plasma P and parathyroid hormone concentrations. A diet containing 4·8 g total P/4184 kJ (1000 kcal), 3·5 g P from sodium dihydrogen phosphate (NaH2PO4)/4184 kJ (1000 kcal) and Ca:P 0·6 caused a marked increase in plasma P from baseline to a peak of 1·976 (95% CI 1·724, 2·266) mmol/l (P <0·001), whereas a diet containing 3·38 g total P/4184 kJ (1000 kcal), no added inorganic P and Ca:P 1·55 resulted in a postprandial decrease in plasma P (P = 0·008). Subsequent data indicate that added inorganic P salts in the diet above 0·5 g P/4184 kJ (1000 kcal) cause an increase in plasma P in cats, while diets below this do not. The data presented here demonstrate that sources of added inorganic P salts cause a temporary postprandial increase in plasma P in a dose-dependent manner, prolonged in diets with Ca:P <1·0. Dietary P derived from natural food ingredients (e.g. meat or vegetable matter) does not appear to have any effect on postprandial plasma P.

Renal disease has a high incidence in cats, and some evidence implicates dietary P as well. To investigate this further, two studies in healthy adult cats were conducted. Study 1 (36 weeks) included forty-eight cats, stratified to control or test diets providing 1·2 or 4·8 g/1000 kcal (4184 kJ) P (0 or approximately 3·6 g/1000 kcal (4184 kJ) inorganic P, Ca:P 1·2, 0·6). Study 2 (29 weeks) included fifty cats, stratified to control or test diets, providing 1·3 or 3·6 g/1000 kcal (4184 kJ) P (0 or approximately 1·5 g/1000 kcal (4184 kJ) inorganic P, Ca:P 1·2, 0·9). Health markers, glomerular filtration rate (GFR) and mineral balance were measured regularly, with abdominal ultrasound. Study 1 was halted after 4 weeks as the test group GFR reduced by 0·4 (95 % CI 0·3, 0·5) ml/min per kg, and ultrasound revealed changes in renal echogenicity. In study 2, at week 28, no change in mean GFR was observed (P >0·05); however, altered renal echogenicity was detected in 36 % of test cats. In agreement with previous studies, feeding a diet with Ca:P <1·0, a high total and inorganic P inclusion resulted in loss of renal function and changes in echogenicity suggestive of renal pathology. Feeding a diet containing lower total and inorganic P with Ca:P close to 1·0 led to more subtle structural changes in a third of test cats; however, nephrolithiasis occurred in both diet groups, complicating data interpretation. We conclude that the no observed adverse effects level for total dietary P in adult cats is lower than 3·6 g/1000 kcal (4184 kJ), however the effect of inorganic P sources and Ca:P require further investigation.

RESUMO

In pets, as in humans, there is increasing interest in interventions that promote 'health and well-being' into later life and extend these beyond their current limits. The purpose of this review was to assess the relevance of current knowledge of ageing in humans, described in a companion paper, as well as reviewing recent research on ageing in pet populations. The role of diet and other factors that influence the ageing process and ultimately lifespan in pets are highlighted in this review; in addition, future opportunities and challenges to further our understanding of the ageing process in pets are identified. Advancing knowledge of the fundamental biology of ageing will be key for the development and evaluation of strategies that extend both the quality and the quantity of lifespan in human and pet populations.

The first aim of these studies was to compare growth patterns of healthy kittens neutered during growth with growth standards created for sexually-intact kittens. A second aim was to clarify the impact of neutering in kittens on body composition and body shape. Study 1 was a retrospective observational study comparing median growth trajectories of healthy, client-owned domestic shorthair (DSH) kittens in optimal body condition and neutered at different ages, with previously-created growth standards from a similar, sexually-intact, population. The neuter groups contained between 3.0k and 9.3k cats. For all neuter groups in both sexes, the median growth trajectory inclined upwards after the procedure, with this being more marked in female than in male kittens. This upwards inclination was less marked for kittens neutered later during growth in both sexes, with the effect being least in kittens neutered after 28-29 weeks. Study 2 was an analysis of new body composition and zoometric data from a previously-published randomised study, comparing growth-related measures between 11 pairs of sexually-intact and neutered (at 19 weeks age) female DSH cats in a research population. Before neutering, the growth pattern in neutered kittens and sexually-intact kittens was similar, but neutered kittens were heavier by 52 weeks (mean difference in fold change vs. 10 weeks 1.34, 95-CI: 1.07-1.72), had a greater fat mass (mean difference in fold change vs. 10 weeks 1.91, 95-CI 1.09-3.21) and greater lean mass (mean difference in fold change vs. 10 weeks 1.23, 95-CI: 1.03-1.48). Abdominal girth (mean difference in fold change vs. 10 weeks 1.20, 95-CI: 1.04-1.39) and rib cage length (mean difference in fold change vs. 10 weeks 1.18, 95-CI: 1.02-1.36) were also greater, but there were no differences in other zoometric measurements. Veterinarians should consider the potential impact that neutering has on gain of adipose tissue, especially early neutering in female kittens. Bodyweight should be monitored closely during growth and especially after neutering to prevent inappropriate weight gain.

The optimal growth of domesticated cats has not yet been well defined. This study first aimed to create evidence-based growth standards for healthy pet domestic shorthair (DSH) kittens, and then compare the pattern of growth curves depicted by the standards with growth patterns in other healthy DSH kittens and those with abnormal body condition. Data were derived from the clinical records of the BANFIELD® Pet Hospital (BANFIELD) network in the USA and from a research population in the UK (Waltham Petcare Science Institute, UK). A 'modelling' dataset was first created from the BANFIELD records, comprising bodyweight data from immature client-owned DSH cats that had remained healthy and in ideal body condition within the first 2.5y of life. This was used to construct growth centile curves for male and female kittens, covering the age range 8 to 78 weeks, using Generalised Additive Models for Location, Shape and Scale. Growth patterns depicted by the centile curves were compared with the growth patterns of healthy DSH kittens from both research colonies and kittens attending BANFIELD that were classified as overweight or underweight. Overall, there was a broad agreement to the growth standards with approximately half of the research population (206/507, 49%) staying within 2 centile lines of their starting centile, and upward and downward movements outside this range being roughly equally distributed. Compared with the growth standards, the 122 overweight BANFIELD kittens were heavier on average at the start of monitoring and subsequently grew more quickly with 63 (52%) crossing at least 2 standard centile lines upwards. Only 4 underweight DSH kittens were available in the BANFIELD database; compared with the standards, there was a marked initial dip in growth followed by subsequent catch-up growth and 2/4 kittens crossed 2 or more centile lines downwards at some point. Evidence-based growth standards are developed here for male and female sexually-intact DSH kittens. Crossing centiles in an upwards and downwards direction is associated with cats becoming overweight or underweight by early adulthood, respectively. Further work is required to determine whether the clinical use of these growth standards will improve the health and wellbeing of pet cats.

To understand the effects of neutering on food intake, body weight (BW) and body composition in kittens, data from an unrelated study were subjected to post hoc analysis. A total of twelve pairs of 11-week-old female littermates were randomly assigned to either a neutered group (neutered at 19 weeks old) or an entire group (kept entire) and offered free access to a dry diet until the age of 1 year. Neutered kittens exhibited increased food intake and increased BW after neutering (both P < 0.00 001). Food intake (per kg BW) peaked 10 weeks after neutering; the mean intake of neutered kittens was 17 (95 % CI 8, 27) % more than entire littermates (P = 0.00 014). The intake was then reduced until there was no significant difference between the groups 18 weeks post-neutering. By 52 weeks of age, the neutered kittens were 24 (95 % CI 11, 39) % heavier than entire littermates (P < 0.0001) with a body condition score (BCS) 16.6 (95 % CI 0.9, 34.8) % higher (P = 0.0028). Neutered kittens continued to grow significantly fatter after neutering (all P < 0.0014), while entire kittens showed no significant change after 18 weeks of age. As neutered kittens consumed similar amounts of energy to their entire littermates from 18 weeks post-neutering, while their BW, BCS and percentage fat continued to increase, we suggest that neutered kittens have a reduced metabolisable energy requirement, and should therefore be fed to maintain an ideal BCS rather than ad libitum. Moreover, to maintain an ideal BCS, entire kittens consumed 93 (95 % CI 87, 100) % of their theoretical intake at 26 weeks of age, and 79 (95 % CI 72, 87) % at 52 weeks of age, suggesting that the current energy recommendation is inappropriate for these kittens.

RESUMO

The purpose of the present review is to highlight some of the challenges and issues in developing nutritional guidelines for companion animals, and to provide some insights that may influence their future direction. For this purpose, we have chosen to provide a brief historical review of the development of dog and cat nutrient guidelines, and an analysis of current recommendations and of key institutions and bodies (notably the National Research Council) that are influential in defining nutrient guidelines for companion animals. In addition, we have also included a review of current approaches for defining nutritional guidelines for humans and farm animal livestock, as they provide differing perspectives and insights that may be instructive for the future development of nutritional guidelines for companion animals.

Data from intravenous (i.v.) glucose tolerance tests suggest that glucose clearance from the blood is slower in cats than in dogs. Since different physiological pathways are activated following oral administration compared with i.v. administration, we investigated the profiles of plasma glucose and insulin in cats and dogs following ingestion of a test meal with or without glucose. Adult male and female cats and dogs were fed either a high-protein (HP) test meal (15 g/kg body weight; ten cats and eleven dogs) or a HP + glucose test meal (13 g/kg body-weight HP diet + 2 g/kg body-weight D-glucose; seven cats and thirteen dogs) following a 24 h fast. Marked differences in plasma glucose and insulin profiles were observed in cats and dogs following ingestion of the glucose-loaded meal. In cats, mean plasma glucose concentration reached a peak at 120 min (10.2, 95 % CI 9.7, 10.8 mmol/l) and returned to baseline by 240 min, but no statistically significant change in plasma insulin concentration was observed. In dogs, mean plasma glucose concentration reached a peak at 60 min (6.3, 95 % CI 5.9, 6.7 mmol/l) and returned to baseline by 90 min, while plasma insulin concentration was significantly higher than pre-meal values from 30 to 120 min following the glucose-loaded meal. These results indicate that cats are not as efficient as dogs at rapidly decreasing high blood glucose levels and are consistent with a known metabolic adaptation of cats, namely a lack of glucokinase, which is important for both insulin secretion and glucose uptake from the blood.

A charge made against feeding dry foods to cats is that the high carbohydrate (i.e. starch) content results in high blood glucose levels which over time may have detrimental health effects. The present study determined the post-meal concentrations of plasma glucose and insulin in adult cats (seven males and four females) and dogs (Labrador retrievers; four males and five females) fed dry diets with low-starch (LS), moderate-starch (MS) or high-starch (HS) levels. In a cross-over design with at least 7 d between the test meals, plasma glucose and insulin concentrations were measured following a single meal of a LS, MS and HS diet (209 kJ/kg bodyweight). Only the HS diet resulted in significant post-meal increases in plasma glucose concentration in cats and dogs although the time-course profiles were different between the species. In cats, plasma glucose concentration was significantly increased above the pre-meal concentration from 11 h until 19 h after the meal, while in dogs, a significant increase above baseline was seen only at the 7 h time point. Plasma insulin was significantly elevated in dogs 4-8 h following the MS diet and 2-8 h after the HS diet. In cats, plasma insulin was significantly greater than baseline from 3-7 and 11-17 h after the HS diet. The time lag (approximately 11 h) between eating the HS diet and the subsequent prolonged elevation of plasma glucose concentration seen in cats may reflect metabolic adaptations that result in a slower digestive and absorptive capacity for complex carbohydrate.

In dogs, optimal growth is critical for future health and wellbeing. Recently, a series of evidence-based growth standards, based on bodyweight, were developed for male and female dogs across 5 different size categories. The aim of the current study was to compare growth curves depicted by the standards with patterns of growth in dogs that were either healthy, had abnormal body condition, or had various diseases with the potential to affect growth. The data came from 2 research colonies in Europe (France and UK), and a large corporate network of primary care veterinary hospitals across the USA. Age and bodyweight data were used to model growth in healthy dogs, in dogs that became overweight or underweight by 3 years of age, and in dogs with diseases associated with altered growth. Centile line crossing during the growth phase was uncommon in healthy dogs, with <5% of dogs crossing >2 centile lines. In contrast, centile line crossing was more frequent in dogs with abnormal growth patterns or abnormal body condition. Dogs that developed obesity by 3 years grew faster than the growth standards predicted, and 68% crossed ≥2 centile lines in an upwards direction. Dogs with conditions associated with accelerated growth also grew faster than expected, and 54% crossed ≥2 centile lines. In contrast dogs that became underweight by 3 years gained weight slower than expected, and 49% crossed ≥2 centile lines in a downwards direction. These results suggest that the growth standards are useful for monitoring healthy growth in dogs. Prospective studies are now required to confirm these findings and to determine whether early intervention can prevent the development of diseases.

Elevated concentrations of serum phosphate are linked with progression and increased case fatality rate in animals and humans with chronic kidney disease. Elevated concentrations of serum phosphate can be a risk factor for development of renal and cardiovascular diseases or osteoporosis in previously healthy people. In rodents, an excess intake of dietary phosphorus combined with an inverse dietary calcium : phosphorus ratio (<1 : 1) contributes to renal calcification. Renal injury also has occured in cats fed experimental diets supplemented with highly soluble phosphate salts, especially in diets with inverse calcium : phosphorus ratios. However, not all phosphorus sources contribute similarly to this effect. This review, which focuses on cats, summarizes the published evidence regarding phosphorus metabolism and homeostasis, including the relative impact of different dietary phosphorus sources, and their impact on the kidneys. No data currently shows that commercial cat foods induce renal injury. However, some diets contain high amounts of phosphorus relative to recommendations and some have inverse Ca : P ratios and so could increase the risk for development of kidney disease. While limiting the use of highly soluble phosphates appears to be important, there are insufficient data to support a specific upper limit for phosphate intake. This review also proposes areas where additional research is needed in order to strengthen conclusions and recommendations regarding dietary phosphorus for cats.

The dog chew studied here is a starch-based, twin-screw cooker extruded dog care and treat (C&T) product with oral health benefits. The manufacturing process and nutrient profile of such products are markedly different from those of main meal pet foods. Predicted metabolisable energy (PME) in pet food is calculated using equations derived from main meal feeding studies so it is unclear whether these equations can be applied to C&T products. The present study aimed to directly measure metabolisable energy (ME) content of the dog chew in dogs and compare with calculated PME. A batch of dog chews was manufactured and the product rendered micronutrient complete to allow solus feeding. Following a 3 d standard diet pre-feed phase, the test product was fed solus to a panel of seven adult dogs for a period of 8 d. Dietary intake was recorded daily and faecal matter collected for the last 5 d. Test product and pooled faecal samples were analysed for proximate nutrients, and digestibility coefficients were calculated as the difference in intake and faecal excretion (7-11 d). Digestible energy was converted to ME by correcting for energy losses in urine. PME was calculated using proximate analysis and modified Atwater factors according to National Research Council 2006. The results showed close agreement between actual ME (1272 (sd 12·1) kJ/100 g) and calculated PME (1268 (sd 12·6) kJ/100 g), indicating transferability of the NRC 2006 PME equations to the dog chew tested here.

Limited information is available on what constitutes optimal growth in dogs. The primary aim of this study was to develop evidence-based growth standards for dogs, using retrospective analysis of bodyweight and age data from >6 million young dogs attending a large corporate network of primary care veterinary hospitals across the USA. Electronic medical records were used to generate bodyweight data from immature client-owned dogs, that were healthy and had remained in ideal body condition throughout the first 3 years of life. Growth centile curves were constructed using Generalised Additive Models for Location, Shape and Scale. Curves were displayed graphically as centile charts covering the age range 12 weeks to 2 years. Over 100 growth charts were modelled, specific to different combinations of breed, sex and neuter status. Neutering before 37 weeks was associated with a slight upward shift in growth trajectory, whilst neutering after 37 weeks was associated with a slight downward shift in growth trajectory. However, these shifts were small in comparison to inter-individual variability amongst dogs, suggesting that separate curves for neutered dogs were not needed. Five bodyweight categories were created to cover breeds up to 40kg, using both visual assessment and hierarchical cluster analysis of breed-specific growth curves. For 20/24 of the individual breed centile curves, agreement with curves for the corresponding bodyweight categories was good. For the remaining 4 breed curves, occasional deviation across centile lines was observed, but overall agreement was acceptable. This suggested that growth could be described using size categories rather than requiring curves for specific breeds. In the current study, a series of evidence-based growth standards have been developed to facilitate charting of bodyweight in healthy dogs. Additional studies are required to validate these standards and create a clinical tool for growth monitoring in pet dogs.

OBJECTIVE: To evaluate the effect of dietary supplementation with the probiotic strain Lactobacillus acidophilus DSM13241 in healthy adult cats. ANIMALS: 15 adult cats. PROCEDURES: Cats were fed a nutritionally complete dry food for 5 weeks. Fecal character was assessed daily, and a single fecal sample and 3-mL blood sample were collected for bacterial enumeration and hematologic analysis, respectively. Cats were then fed the same diet supplemented with L acidophilus DSM13241 (2 x 10(8) CFU/d) for 4.5 weeks. Repeat fecal and hematologic measurements were taken prior to the return to control diet for a 4-week period. RESULTS: The probiotic species was recovered from feces, demonstrating survival through the feline gastrointestinal tract. Probiotic supplementation was associated with increased numbers of beneficial Lactobacillus and L acidophilus groups in feces and decreased numbers of Clostridium spp and Enterococcus faecalis, indicating an altered bacterial balance in the gastrointestinal tract microflora. Fecal pH was also decreased suggesting a colonic environment selective for the beneficial lactic acid bacterial population. Systemic and immunomodulatory effects were associated with administration of L acidophilus DSM13241 including altered cell numbers within WBC subsets and enhanced phagocytic capacity in the peripheral granulocyte population. In addition, plasma endotoxin concentrations were decreased during probiotic feeding, and RBCs had a decreased susceptibility to osmotic pressure. CONCLUSIONS AND CLINICAL RELEVANCE: Probiotic strain L acidophilus DSM13241 fed at 2 x 10(8) CFU/d can alter the balance of gastrointestinal microflora in healthy cats. Furthermore, administration of this probiotic results in beneficial systemic and immunomodulatory effects in cats.

OBJECTIVE: To determine whether mild restriction of food intake affects clinicopathologic variables, body composition, and performance of dogs undertaking intense sprint exercise. ANIMALS: 9 trained healthy adult Greyhounds. PROCEDURE: Dogs were offered food free choice once daily for 9 weeks until body weight and food intake stabilized. Dogs were then randomly assigned to be fed either 85% or 100% of this quantity of food in a crossover study (duration of each diet treatment period, 9 weeks). Dogs raced a distance of 500 m twice weekly. Clinicopathologic variables were assessed before and 5 minutes after racing; food intake, weight, body composition, body condition score, and race times were compared at the end of each diet period. RESULTS: Compared with values associated with unrestricted access to food, there were significant decreases in mean body weight (by 6%) and median body condition score (from 3.75 to 3.5 on a 9-point scale) and the mean speed of the dogs was significantly faster (by 0.7 km/h) when food intake was restricted. Body composition and most clinicopathologic variables were unaffected by diet treatment, but dogs given restricted access to food had slightly fewer neutrophils, compared with values determined when food intake was unrestricted. CONCLUSIONS AND CLINICAL RELEVANCE: Results indicate that the common practice among Greyhound trainers of mildly restricting food intake of racing dogs to reduce body weight does improve sprint performance. A body condition score of approximately 3.5 on a 9-point scale is normal for a trained Greyhound in racing condition.

OBJECTIVE: To evaluate viability of a probiotic strain of Lactobacillus acidophilus in a dry dog food, determine its ability to survive transit through the gastrointestinal tract and populate the colon, and assess its effects on intestinal and systemic parameters. ANIMALS: 15 adult dogs. PROCEDURE: Dogs were sequentially fed a dry control food for 2 weeks, the same food supplemented with > 10(9) L. acidophilus for 4 weeks, and the control food again for 2 weeks. Fecal score was assessed daily, and fecal and blood samples were collected for enumeration of bacterial populations and measurement of hematologic variables. RESULTS: Recovery of L. acidophilus from the supplemented food was 71% and 63% at the start and end of the study, respectively, indicating that the bacteria were able to survive manufacture and storage. The probiotic bacterium was detected in feces via ribotyping and RNA gene sequencing during the probiotic administration phase but not 2 weeks after cessation of administration. Administration of the probiotic-supplemented food was associated with increased numbers of fecal lactobacilli and decreased numbers of clostridial organisms. There were significant increases in RBCs, Hct, hemoglobin concentration, neutrophils, monocytes, and serum immunoglobin G concentration and reductions in RBC fragility and serum NO concentration. CONCLUSIONS AND CLINICAL RELEVANCE: These data indicate that L. acidophilus can be successfully incorporated into a dry dog food, survive transit through the canine gastrointestinal tract, and populate the colon and are associated with local and systemic changes. This probiotic bacterium may have the potential to enhance intestinal health and improve immune function in dogs.
