RESUMO

On 1 October 2019, a locally-acquired Zika virus disease case was laboratory confirmed in Hyères, Var department. Active case finding identified two additional locally-acquired cases living within 90 m, with symptom onset 8 days before the index case. Extensive patient interviews did not yield information supporting transmission through sexual contact or substances of human origin. Vector-borne transmission by local Aedes albopictus mosquitoes is the most likely mode of transmission. Here we describe the public health response.

RESUMO

In August 2017, an autochthonous chikungunya case was reported in south-east France. By mid-September, eight additional autochthonous cases were found in the index case's neighbourhood, where the chikungunya virus vector Aedes albopictus was observed. Genomic characterisation identified an East-Central South African (ECSA) lineage strain, probably from the Central African region and carrying an adaptive mutation facilitating transmission by Ae. albopictus. The event confirms we need early case detection and response to contain chikungunya in Europe.

Fecal pollution in coastal areas is of a high concern since it affects bathing and shellfish harvesting activities. Wild waterbirds are non-negligible in the overall signal of the detectable pollution. Yet, studies on wild waterbirds' gut microbiota focus on migratory trajectories and feeding impact on their shape, rare studies address their comparison to other sources and develop quantitative PCR (qPCR)-based Microbial Source Tracking (MST) markers to detect such pollution. Thus, by using 16S rRNA amplicon high-throughput sequencing, the aims of this study were (i) to explore and compare fecal bacterial communities from wild waterbirds (i.e., six families and 15 species, n = 275 samples) to that of poultry, cattle, pigs, and influent/effluent of wastewater treatment plants (n = 150 samples) and (ii) to develop new MST markers for waterbirds. Significant differences were observed between wild waterbirds and the four other groups. We identified 7,349 Amplicon Sequence Variants (ASVs) from the hypervariable V3-V4 region. Firmicutes and Proteobacteria and, in a lesser extent, Actinobacteria and Bacteroidetes were ubiquitous while Fusobacteria and Epsilonbacteraeota were mainly present in wild waterbirds. The clustering of samples in non-metric multidimensional scaling (NMDS) ordination indicated a by-group clustering shape, with a high diversity within wild waterbirds. In addition, the structure of the bacterial communities was distinct according to bird and/or animal species and families (Adonis R 2 = 0.13, p = 10-4, Adonis R 2 = 0.11, p = 10-4, respectively). The Analysis of Composition of Microbiomes (ANCOM) showed that the wild waterbird group differed from the others by the significant presence of sequences from Fusobacteriaceae (W = 566) and Enterococcaceae (W = 565) families, corresponding to the Cetobacterium (W = 1427) and Catellicoccus (W = 1427) genera, respectively. Altogether, our results suggest that some waterbird members present distinct fecal microbiomes allowing the design of qPCR MST markers. For instance, a swan- and an oystercatcher-associated markers (named Swan_2 and Oyscab, respectively) have been developed. Moreover, bacterial genera harboring potential human pathogens associated to bird droppings were detected in our dataset, including enteric pathogens, i.e., Arcobacter, Clostridium, Helicobacter, and Campylobacter, and environmental pathogens, i.e., Burkholderia and Pseudomonas. Future studies involving other wildlife hosts may improve gut microbiome studies and MST marker development, helping mitigation of yet unknown fecal pollution sources.

BACKGROUND: Ultra-low volume (ULV) insecticidal aerosols dispensed from vehicle-mounted cold-foggers are widely considered the method of choice for control of Aedes aegypti and Ae. albopictus during outbreaks of dengue and chikungunya and, more recently, Zika. Nevertheless, their effectiveness has been poorly studied, particularly in Europe. Nearly all published studies of ULV efficacy are bio-assays based on the mortality of caged mosquitoes. In our study we preferred to monitor the direct impact of treatments on the wild mosquito populations. This study was undertaken to evaluate the efficiency of the two widely used space spraying methods to control Ae. albopictus and Ae. aegypti. METHODS: We determined the susceptibility of local Ae. albopictus to deltamethrin by two methods: topical application and the "WHO Tube Test". We used ovitraps baited with hay infusion and adult traps (B-G Sentinel) baited with a patented attractant to monitor the mosquitoes in four residential areas in Nice, southern France. The impact of deltamethrin applied from vehicle-mounted ULV fogging-machines was assessed by comparing trap results in treated vs untreated areas for 5 days before and 5 days after treatment. Four trials were conducted at the maximum permitted application rate (1 g.ha-1). We also made two small-scale tests of the impact of the same insecticide dispensed from a hand-held thermal fogger. RESULTS: Susceptibility to the insecticide was high but there was no discernable change in the oviposition rate or the catch of adult female mosquitoes, nor was there any change in the parous rate. In contrast, hand-held thermal foggers were highly effective, with more than 90% reduction of both laid eggs and females. CONCLUSIONS: We believe that direct monitoring of the wild mosquito populations gives a realistic assessment of the impact of treatments and suggest that the lack of efficacy is due to lack of interaction between the target mosquitoes and the ULV aerosol. We discuss the factors that influence the effectiveness of both methods of spraying in the context of epidemic situations.

Molecular sexing revealed an unexpectedly strong female bias in the sex ratio of pre-breeding European Storm Petrels (Hydrobates pelagicus), attracted to playback of conspecific calls during their northwards migration past SW Europe. This bias was consistent across seven years, ranging from 80.8% to 89.7% female (mean annual sex ratio ± SD = 85.5% female ±4.1%). The sex ratio did not differ significantly from unity (i.e., 50% female) among (i) Storm Petrel chicks at a breeding colony in NW France, (ii) adults found dead on beaches in Southern Portugal, (iii) breeding birds attending nest burrows in the UK, captured by hand, and (iv) adults captured near a breeding colony in the UK using copies of the same sound recordings as used in Southern Europe, indicating that females are not inherently more strongly attracted to playback calls than males. A morphological discriminant function analysis failed to provide a good separation of the sexes, showing the importance of molecular sexing for this species. We found no sex difference in the seasonal or nocturnal timing of migration past Southern Europe, but there was a significant tendency for birds to be caught in sex-specific aggregations. The preponderance of females captured in Southern Europe suggests that the sexes may differ in migration route or in their colony-prospecting behaviour during migration, at sites far away from their natal colonies. Such differences in migration behaviour between males and females are poorly understood but have implications for the vulnerability of seabirds to pollution and environmental change at sea during the non-breeding season.
