RESUMO

Screen-printed carbon nanofiber electrodes (SPCNFEs) represent an alternative with great acceptance due to their results, as well as their low impact on the environment. In order to improve their performance, in the present work they were modified with silver nanoparticles (Ag-NPs) and electrochemically characterized by using anodic stripping voltammetry. From the Ag-NP synthesis, silver seeds (Ag-NS) and silver nanoprisms (Ag-NPr) were obtained. The Ag-NP formation was confirmed by micrographs, where Ag-NPs with diameters of 12.20 ± 0.04 nm for Ag-NS and 20.40 ± 0.09 nm for Ag-NPr were observed. The electrodes were modified by using three different deposition methods-drop-casting, spin-coating, and in situ approaches-that offer different nanoparticle distribution and electrode modification times. It was observed that the last methodology showed a low amount of Ag-NS deposited on the electrode surface and deep alteration of this surface. Those facts suggest that the in situ synthesis methodology was not appropriate for the determination of heavy metals, and it was discarded. The incorporation of the nanoparticles by spin-coating and drop-casting strategies showed different spatial distribution on the electrode surface, as proved by scanning electron microscopy. The electrodes modified by these strategies were evaluated for the cadmium(II) and lead(II) detection using differential pulse anodic stripping voltammetry, obtaining detection limit values of 2.1 and 2.8 µg·L-1, respectively. The overall results showed that the incorporation route does not directly change the electrocatalytic effect of the nanoparticles, but the shape of these nanoparticles (spherical for seeds and triangular for prisms) has preferential electrocatalytic enhancement over Cd(II) or Pb(II).

The dissolution rates of unirradiated UO2 and unirradiated UO2 doped with Gd2O3 were determined as a function of pH using flow-through experiments in the presence of O2(g) and bicarbonate. The dissolution rate of non-doped UO2 was very low under hyperalkaline conditions (pH 12-13) whereas it increased drastically as the pH decreased to 9. The dissolution of non-doped UO2 in the pH range of 9-13 was consistent with the oxidative dissolution mechanism already described for UO2 dissolution in the presence of bicarbonate and oxygen. XPS analysis performed on the solid after dissolution experiments at pH 10 and 13 supported the bicarbonate effect to complex UO22+ and accelerate dissolution. Moreover, UO2 doped with Gd2O3 (5 wt% and 10 wt%) showed dissolution rates as low as non-doped UO2 under hyperalkaline conditions, which were maintained throughout the pH range studied (9-13). No substantial differences in the dissolution rates between these two doping levels were found. XPS analysis evidenced a similar surface composition both at pH 10 and 13, with U(V) being the dominant oxidation state. The low dissolution rates were assumed to be a consequence of the gadolinium capacity to retard the oxidation of U(V) to U(VI). The slight increase in dissolution rates observed in the hyperalkaline region was attributed to a shift in the oxidative dissolution mechanism, in which the presence of OH- promotes the formation of soluble uranyl hydroxo complexes.

NK1 is a tachykinin receptor highly relevant to tumorigenesis and metastasis development in breast cancer and other carcinomas. Despite the substantial efforts done to develop potent NK1 receptor antagonists, none of these antagonists had shown good antitumor activity in clinical trials. Now, we have tested the effect of inhibition of the neuropeptide Substance P (SP), a NK1 ligand, as a potential therapeutic approach in cancer. We found that the inhibition of SP with antibodies strongly inhibit cell growth and induce apoptosis in breast, colon, and prostate cancer cell lines. These effects were accompained by a decrease in the mitogen-activated kinase singaling pathway. Interestingly, in some cell lines SP abrogation decreased the steady state of Her2 and EGFR, suggesting that SP-mediated signaling is important for the basal activity of these ErbB receptors. In consequence, we observed a blockade of the cell cycle progression and the inhibition of several cell cycle-related proteins including mTOR. SP inhibition also induced cell death in cell lines resistant to Lapatinib and Trastuzumab that have increased levels of active Her2, suggesting that this therapeutic approach could be also effective for those cancers resistant to current anti-ErbB therapies. Thus, we propose a new therapeutic strategy for those cancers that express NK1 receptor and/or other tachykinin receptors, based in the immuno-blockade of the neuropeptide SP.

This work determines the capacity of uranophane, one of the long-term uranyl secondary solid phases formed on the spent nuclear fuel (SNF), to retain radionuclides (cesium and strontium) released during the dissolution of the SNF. Sorption was fast in both cases, and uranophane had a high sorption capacity for both radionuclides (maximum sorption capacities of 1.53·10-5 mol m-2 for cesium and 3.45·10-3 mol m-2 for strontium). The high sorption capacity of uranophane highlights the importance of the formation of uranyl silicates as secondary phases during the SNF dissolution, especially in retaining the release of radionuclides not retarded by other mechanisms such as precipitation.

Grape stalk wastes generated in the wine production process were used for the removal of nickel (II) from aqueous solution. The experimental breakthrough curves were obtained in fixed-bed columns. Experiments we carry out in order to evaluate the influence of inlet metal concentration (30 and 70 mg L(-1)) and the regeneration process in a double sorption cycle. The CXTFIT code was used to fit the experimental data and to determine the transport and sorption parameters of the convective-dispersive equation (CDE) and the two-site deterministic non-equilibrium (TSM/CDE) model by adjusting the models to the experimental breakthrough curves (BTC). The results showed that bed capacity as well as transport and sorption parameters were affected by the initial metal concentration, at the highest Ni(II) concentration the grape stalks column saturated quickly leading to earlier breakthrough. The sorption capacity of the sorbent was slightly reduced in a double sorption cycle, while the recovery of the metal in the desorption step was ranging between 80% and 85% in both cycles.

Strontium-90 is considered the most important radioactive isotope in the environment and one of the most frequently occurring radionuclides in groundwaters at nuclear facilities. The uranyl peroxide studtite (UO2O2 . 4H2O) has been observed to be formed in spent nuclear fuel leaching experiments and seems to have a relatively high sorption capacity for some radionuclides. In this work, the sorption of strontium onto studtite is studied as a function of time, strontium concentration in solution and pH. The main results obtained are (a) sorption is relatively fast although slower than for cesium; (b) strontium seems to be sorbed via a monolayer coverage of the studtite surface, (c) sorption has a strong dependence on ionic strength, is negligible at acidic pH, and increases at neutral to alkaline pH (almost 100% of the strontium in solution is sorbed above pH 10). These results point to uranium secondary solid phase formation on the spent nuclear fuel as an important mechanism for strontium retention in a high-level nuclear waste repository (HLNW).

BACKGROUND: The efficacy of oxaliplatin in cancer chemotherapy is limited by the development of drug resistance. MMP7 has been related to the loss of tumor cell response to cytotoxic agents although the exact mechanism is not fully understood. Moreover, MMP7 is an independent prognosis factor for survival in patients with colorectal cancer. The aim of the present study was to analyze the role of MMP7 and its cross-talk with the Fas/FasL system during the acquisition of oxaliplatin resistance in colon cancer cells. PRINCIPAL FINDINGS: For this purpose we have developed three different oxaliplatin-resistant cell lines (RHT29, RHCT116 p53(+/+), RHCT116 p53(-/-)) from the parental HT29, HCT116 p53(+/+) and HCT116 p53(-/-) colon cancer cells. MMP7 basal expression was higher in the resistant compared to the parental cell lines. MMP7 was also upregulated by oxaliplatin in both HT29 (p53 mutant) and RHCT116 p53(-/-) but not in the RHCT116 p53(+/+). Inhibition of MMP by 1,10-phenantroline monohydrate or siRNA of MMP7 restores cell sensitivity to oxaliplatin-induced apoptosis in both HT29 and RHCT116 p53(-/-) but not in the RHCT116 p53(+/+). Some of these effects are caused by alterations in Fas receptor. Fas is upregulated by oxaliplatin in colon cancer cells, however the RHT29 cells treated with oxaliplatin showed a 3.8-fold lower Fas expression at the cell surface than the HT29 cells. Decrease of Fas at the plasma membrane seems to be caused by MMP7 since its inhibition restores Fas levels. Moreover, functional analysis of Fas demonstrates that this receptor was less potent in inducing apoptosis in RHT29 cells and that its activation induces MAPK signaling in resistant cells. CONCLUSIONS: Taking together, these results suggest that MMP7 is related to the acquisition of oxaliplatin-resistance and that its inhibition restores drug sensitivity by increasing Fas receptor. Furthermore, Fas undergoes a change in its functionality in oxaliplatin-resistant cells inducing survival pathways instead of apoptotic signals.

The processes that affect the mobility of uranium and other radionuclides in the environment have been largely studied at both the laboratory and the field scales. The natural reactors found at the Oklo uranium mine in Gabon constitute a unique investigation setting as spontaneous fission reactions occurred two billion years ago. Oklo uraninites contain a large amount of other radionuclides as a result of the fission process. We have investigated the dissolution behavior of four uraninite samples from Oklo as a function of temperature (25 and 60 degrees C) and bicarbonate concentration (2.7-30 mmol/L). We have also investigated the dissolution behavior of minor components of the uraninites (i.e., Nd, Cs, Mo, Yb, and Sb) in relation to the dissolution of uranium. The results of the reported work are in good agreement with the kinetic rate laws derived from other uranium(IV) dioxide studies. Some of the minor components are found to be congruently released from the uraninite phase, while it is postulated that dissolution from segregated phases might affect the final concentrations of some of the rare earth elements, i.e., Nd and Yb. In addition, we have performed dissolution studies at 60 degrees C with two uraninites representative of different geochemical environments at Oklo, to study the uranium dissolution rates as a function of the temperature. This has allowed derivation of apparent activation energies for the bicarbonate-promoted oxidative dissolution of the Oklo uraninites. The dissolution behavior of the minor components of the uraninites at 60 degrees C was found to closely follow the behavior observed at 25 degrees C. This indicates that similar codissolution mechanisms operate in the temperature range studied. The implications for the mobility of uranium and other radionuclides in natural and anthropogenic environments are discussed.
