RESUMO

Patterns of late Palaeogene mammalian evolution appear to be very different between Eurasia and North America. Around the Eocene-Oligocene (EO) transition global temperatures in the Northern Hemisphere plummet: following this, European mammal faunas undergo a profound extinction event (the Grande Coupure), while in North America they appear to pass through this temperature event unscathed. Here, we investigate the role of surface uplift to environmental change and mammalian evolution through the Palaeogene (66-23 Ma). Palaeogene regional surface uplift in North America caused large-scale reorganization of precipitation patterns, particularly in the continental interior, in accord with our combined stable isotope and ecometric data. Changes in mammalian faunas reflect that these were dry and high-elevation palaeoenvironments. The scenario of Middle to Late Eocene (50-37 Ma) surface uplift, together with decreasing precipitation in higher-altitude regions of western North America, explains the enigma of the apparent lack of the large-scale mammal faunal change around the EO transition that characterized western Europe. We suggest that North American mammalian faunas were already pre-adapted to cooler and drier conditions preceding the EO boundary, resulting from the effects of a protracted history of surface uplift.

Trace elements are found in most drugs as a result of the drug formulation and drug production methods. An inductively coupled plasma-mass spectrometry method for the determination of 24 trace elements (Mg, Ti, V, Cr, Mn, Cu, Fe, Co, Ni, Zn, As, Se, Mo, Ru, Rh, Pd, Ag, Cd, Sb, Ba, Ir, Pt, Au, and Pb) in solid ibuprofen tablets was established in relation to the ICH Q3D(R1) guideline, to evaluate the possibility of linking trace elemental profiles to drug formulation strategies, and to differentiate between drug products based on the trace elemental profiles. Ten European ibuprofen drug products were evaluated (n=3). The sample preparation was performed by microwave-assisted acid digestion using only 10 mg of homogenized sample and 900 µL of a mix of 65% HNO3, 37% HCl, and 30% H2O2. Solid residuals primarily composed of insoluble SiO2 excipients were removed by centrifugation. Only concentrations of Mg, Fe, Ti, Mn, Cr, and Ni were detected above the limits of detection and did not exceed the ICH Q3D(R1) guideline permitted daily exposure limits. The trace elemental profiles were evaluated through principal component analysis. Three principal components describing 96% of the variance were useful in grouping the ibuprofen drug products, and the detected trace elemental remnants could be related to drug formulation and drug production strategies. An in-house quality control material was used in lack of certified reference materials and was in combination with spike recoveries used for method validation. Good spike recoveries (94-119%) were obtained for all measured trace elements except Mg. Mg showed acceptable spike recoveries (75-155%) for mid and high-spike concentrations, but poor recoveries (30-223%) were detected with low spike concentrations in spike matrices containing high amounts of Mg. Overall, the method is suggested applicable for solid drugs containing insoluble SiO2 excipients and drugs comparable to ibuprofen.

This study examines detrimental effects of acid mine drainage (AMD) on stream invertebrate communities and tests for a direct relationship between trophic position and accumulation of three metals (Fe, Cu, Zn) by stream invertebrates in situ. On two dates in each of seven stream sites, we measured food chain length, mean trophic level, taxa richness, and trophic position of stream macroinvertebrates comprising the food webs using stable nitrogen isotope ratios. Metals in tissue of representatives of 35 taxa were measured by ICP-OES. Our results are the first direct comparison of uptake of these metals in stream invertebrate taxa according to trophic position as identified by delta15N. As predicted, metal concentrations were generally greater in water and insects from sites adjacent to mining activity and invertebrate taxa richness was significantly lower. Taxa richness increased with distance away from contaminated headwaters. Despite reductions in diversity at sites nearest AMD, food chain length and mean trophic level did not differ between streams. The relationship between trophic position and metal accumulation differed considerably among metals. Specifically, Fe declined (biodilution) and Zn increased (biomagnification) with trophic level, but trophic position had no effect on Cu levels in these insects. Our results highlight fundamental differences in trophic transfer of specific metals through aquatic food webs and identify ecologically important impacts of AMD on stream invertebrates.

Cadmium (Cd) and lead (Pb) concentrations were analysed in 96 samples of produce from seven urban farms, three suburban farms and three grocery stores in the San Francisco Bay Area in 2011-2012. Cd concentrations were highest in urban chard (0.043 mg kg(-1)) and lowest in urban, suburban and grocery squash (0.003 mg kg(-1)). Pb concentrations were highest in urban kale (0.080 mg kg(-1)) and lowest in grocery squash (0.008 mg kg(-1)). The mean heavy metal concentrations for Cd and Pb in all produce types were well below the maximum limits as set by the Food and Agriculture Organization (FAO) and the World Health Organization (WHO). Individual concentrations of Cd and Pb were below the limits of detection in 26 of 192 analyses. Cd and Pb concentrations in produce from urban farms were not significantly different from produce grown in suburban farms or grocery stores. It was concluded that produce from urban community farms in San Francisco, at least for the farms studied, is safe for human consumption.

We determine paleoelevation of the Sierra Nevada, California, by tracking the effect of topography on precipitation, as recorded in hydrogen isotopes of kaolinite exposed in gold-bearing river deposits from the Eocene Yuba River. The data, compared with the modern isotopic composition of precipitation, show that about 40 to 50 million years ago the Sierra Nevada stood tall (>/=2200 meters), a result in conflict with proposed young surface uplift by tectonic and climatic forcing but consistent with the Sierra Nevada representing the edge of a pre-Eocene continental plateau.

The endangered California Condor (Gymnogyps californianus) was reduced to a total population of 22 birds by the end of 1982. Their captive-bred descendants are now being released back into the wild in California, Arizona, and Baja California, where monitoring indicates they may accumulate lead to toxic levels. Fragments of ammunition in the carcasses of game animals such as deer, elk, and feral pigs not retrieved by hunters or in gut piles left in the field have been considered a plausible source of the lead, though little direct evidence is available to support this hypothesis. Here, we measured lead concentrations and isotope ratios in blood from 18 condors living in the wild in central California, in 8 pre-release birds, and in diet and ammunition samples to determine the importance of ammunition as a source of exposure. Blood lead levels in pre-release condors were low (average 27.7 ng/mL, SD 4.9 ng/ mL) and isotopically similar to dietary and background environmental lead in California. In contrast, blood lead levels in free-flying condors were substantially higher (average 246 ng/mL, SD 229 ng/mL) with lead isotopic compositions that approached or matched those of the lead ammunition. A two-endmember mixing model defined by the background 207Pb/206Pb ratio of representative condor diet samples (0.8346) and the upper 207Pb/206Pb ratio of the ammunition samples (0.8184) was able to account for the blood lead isotopic compositions in 20 out of the 26 live condors sampled in this study (i.e., 77%). Finally, lead in tissues and in a serially sampled growing feather recovered postmortem from a lead-poisoned condor in Arizona evidence acute exposure from an isotopically distinct lead source. Together, these data indicate that incidental ingestion of ammunition in carcasses of animals killed by hunters is the principal source of elevated lead exposure that threatens the recovery in the wild of this endangered species.

The geochemical composition of the Earth's upper mantle is thought to reflect 4.5 billion years of melt extraction, as well as the recycling of crustal materials. The fractionation of rhenium and osmium during partial melting in the upper mantle makes the Re-Os isotopic system well suited for tracing the extraction of melt and recycling of the resulting mid-ocean-ridge basalt. Here we report osmium isotope compositions of more than 700 osmium-rich platinum-group element alloys derived from the upper mantle. The osmium isotopic data form a wide, essentially gaussian distribution, demonstrating that, with respect to Re-Os isotope systematics, the upper mantle is extremely heterogeneous. As depleted and enriched domains can apparently remain unequilibrated on a timescale of billions of years, effective equilibration seems to require high degrees of partial melting, such as occur under mid-ocean ridges or in back-arc settings, where percolating melts enhance the mobility of both osmium and rhenium. We infer that the gaussian shape of the osmium isotope distribution is the signature of a random mixing process between depleted and enriched domains, resulting from a 'plum pudding' distribution in the upper mantle, rather than from individual melt depletion events.
