RESUMO

This research presents a comprehensive review of Salmonella presence in retail fresh fruits and vegetables from 2010 to 2023, utilizing data from recognized sources such as PubMed, Scopus, and Web of Science. The study incorporates a meta-analysis of prevalence, serovar distribution, antimicrobial susceptibility, and antimicrobial resistance genes (ARGs). Additionally, it scrutinizes the heterogeneous sources across various food categories and geographical regions The findings show a pooled prevalence of 2.90% (95% CI: 0.0180-0.0430), with an increase from 4.63% in 2010 to 5.32% in 2022. Dominant serovars include S. Typhimurium (29.14%, 95% CI: 0.0202-0.6571) and S. Enteritidis (21.06%, 95% CI: 0.0181-0.4872). High resistance rates were noted for antimicrobials like erythromycin (60.70%, 95% CI: 0.0000-1.0000) and amoxicillin (39.92%, 95% CI: 0.0589-0.8020). The most prevalent ARGs were blaTEM (80.23%, 95% CI: 0.5736-0.9692) and parC mutation (66.67%, 95% CI: 0.3213-0.9429). Factors such as pH, water activity, and nutrient content, along with external factors like the quality of irrigation water and prevailing climatic conditions, have significant implications on Salmonella contamination. Nonthermal sterilization technologies, encompassing chlorine dioxide, ozone, and ultraviolet light, are emphasized as efficacious measures to control Salmonella. This review stresses the imperative need to bolster prevention strategies and control measures against Salmonella in retail fresh fruits and vegetables to alleviate related food safety risks.

RESUMO

OBJECTIVE: To develop a polymerase chain reaction(PCR) method for rapid detection of Listeria monocytogenes in oysters without pre-enrichment. METHODS: The combination of ß-cyclodextrin and bentonite-coated activated carbon was used to remove PCR inhibitors from oyster samples, and the target gene inlB was used for the PCR subsequently. The specificity, sensitivity, and application of the developed method were verified, and the stability and application of the reagents stored under cryopreservation conditions were evaluated. RESULTS: The specificity of the developed PCR method was 100% for the detection of 130 target bacterial strains and 63 non-target bacterial strains. The method reduced the time required for Listeria monocytogenes detection to 4 h without pre-enrichment, and the detection limit was 10 CFU/25 g. The method was consistent with the conventional culture method on the detection rate and viable bacteria detection rate of Listeria monocytogenes in natural oyster samples(the coincidence rate was 100%). Additionally, the reagents could be used normally after storing at-20 ℃ for at least one year. CONCLUSION: The PCR method developed in this study has high specificity and sensitivity, and can be used for rapid, accurate detection of Listeria monocytogenes in oysters.

RESUMO

Multiple drug-resistant (MDR) Shigella isolates have been reported worldwide. Between May 2017 and September 2018, 55 Shigella flexneri 2a isolates were collected from 3322 stool samples of 0-10-year-old outpatients with diarrhea at the Children's Hospital of Urumqi, China. All isolates were characterized using serotyping, antimicrobial susceptibility testing, and whole-genome sequencing. A total of 54 of 55 (98.2%) isolates exhibited MDR phenotypes and had accumulated multiple resistance determinants, particularly of fluoroquinolones and cephalosporins preferred for shigellosis treatment: point mutations in quinolone resistance-determining regions (QRDRs) of topoisomerases (GyrA (S83L, D87N) and ParC (S80I) [n = 9]; GyrA (S83L) and ParC (S80I) [n = 45]) and acquisition of qnrS1 (n = 3) and blaCTX-M (n = 8). Over 70% of isolates acquired two point mutations of GyrA (S83L) and ParC (S80I) in QRDRs and 11 highly resistant isolates accumulated three point mutations in QRDRs or acquired qnrS1. Four S. flexneri 2a isolates from three single-nucleotide polymorphism clusters exhibited coresistance to ciprofloxacin, cefotaxime, or azithromycin (AZM), which are used as first- and second-line shigellosis treatment antimicrobials in clinics. Our data indicated that fluoroquinolones should be terminated in shigellosis treatment for outpatients in Urumqi. The transferable antimicrobial resistance determinants have been identified for third-generation cephalosporins and AZM. Novel strategies are urgently required for developing empirical medication to reduce the antimicrobial selective pressure and prevent dissemination of MDR S. flexneri 2a isolates.

RESUMO

Unsustainable diet is one of the main reasons for the nutrition-health-environment trilemma. However, information on environment-nutrition-obesity effects associated with food consumption is still limited. This study analyzes these diet-related impacts of different groups classified by various socio-economic attributes: location, gender, age, income, education, and occupation. We applied the samples in China Health and Nutrition Survey and divided them into advantaged group and dis-advantaged group according to the probability of access to more nutritious food. Results show that the advantaged groups had higher and more rapidly increasing dietary and nutrition quality than their counterpart during 1997-2011. On the contrary, the non-advantaged group' body mass index increased faster. Meanwhile, the high-income group as well as government and professional & technological workers have passed the criterion for overweight. The environmental footprints, i.e., nitrogen, phosphorus, carbon, and water footprints, of high-income group were higher 89%, 70%, 98%, and 41% than low-income group, respectively. Notably, food consumption sustainability of each group has declined, and the non-advantaged groups' is much more sustainable. We concluded that inequality existed and tends to expand in food consumption and its related impacts of different socio-economic groups. A reformed responsibility allocation system is needed during dietary transition for better environmental management. Strategies to improve dietary quality for advantaged group focus on improving the types of high-quality protein foods, such as milk and seafood, while the non-advantaged group can choose to increase the types of high-quality but relatively cheap foods like vegetables and fruits considering the availability and living cost.

RESUMO

Food waste (FW) has received increasing attention because of its immense production quantities and significance to resource and environmental impacts related to disposal approaches. We combined life cycle assessment (LCA) with society's willingness to pay (WTP) index to evaluate energy, water, and environmental impacts on three food waste-to-energy (FWTE) options in China. For anaerobic digestion (AD) mode, the results showed that 1140 MJ of energy consumption could be saved by power generation from methane, power transmission, and biodiesel production from per ton of FW; the cost of climate change for treating FW was 137.8 kg CO2e t-1 FW, failing to be climate-sound due to the end life of digestate in practice. The total impact to AD mode in the form of monetized value for WTP was 13.3 CNY t-1 FW, of which the collection and transportation, pretreatment, AD reaction, wastewater treatment, biodiesel production, and residue landfilling stages contributed by 10.5%, 6.5%, 19.3%, 27.6%, 4.7%, and 75.7%, respectively, while biogas utilization offset it by 43.9%. Notably, a considerable amount of water used in AD prevented it from showing an advantage compared to incineration (-5.1 CNY t-1 FW), which performed best overall attributing to the generated electricity compensated for primary energy demand, water, and terrestrial acidification to a great extent. Landfilling turned out to be an unappealing FW disposal method due to the low landfill gas capture ratio. Given that AD is touted for its environmental benefits, potential approaches-such as developing a reliable and supportive technology to facilitate digestate recycling into agriculture-were discussed to improve its competitiveness and attractiveness. Our study employed a way to accumulate and compare impact indicators to better interpret FW management impacts and advantages, considering energy recovery, resource recycling, and the environment.

RESUMO

Most outbreaks of Shiga toxin-producing Escherichia coli (STEC) are attributed to consumption of contaminated foodstuffs including beef and dairy products. In this study, we evaluated the prevalence of non-O157 STEC cultured from beef and dairy cattle and collected in Xinjiang Uygur Autonomous Region in China. Results identified 67 non-O157 STEC recovered from the 793 samples including beef cattle (10.28%, 43/418) and dairy cattle (6.40%, 24/375). A total of 67 non-O157 STEC was sequenced allowing for in silico analyses of their serotypes, virulence genes, and identification of the corresponding multilocus sequence types (STs). Twenty-one O serogroups and nine H serotypes were identified and the dominant serotype identified was O22:H8. One stx1 subtype (stx1a) and four stx2 subtypes (2a, 2b, 2c, and 2d) were found in the 67 non-O157 STEC isolates. The results revealed that stx1a+stx2a-positive STEC isolates were predominant (32.83%, 22/67), followed by stx1a+stx2d (29.85%, 20/67) and stx2a alone (17.91%, 12/67). Non-O157 STEC isolates carried virulence genes ehxA (98.51%), subA (53.73%), and cdtB (17.91%). Of the four adherence-associated genes tested, eaeA was absent, whereas lpfA and iha were present in 67 and 55 non-O157 STEC isolates, respectively. The STEC isolates were divided into 48 pulsed-field gel electrophoresis patterns and 10 STs, and ST446 (O22:H8) was the dominant clone (22.38%). Our results revealed that there was a high genetic diversity among non-O157 STEC isolated from beef and dairy cattle, some of which have potential to cause human diseases.

RESUMO

While Shiga toxin-producing Escherichia coli (STEC) is a major foodborne pathogen worldwide, data on the molecular and phylogenetic properties of STEC isolates from retail beef samples in China remain scant. Fresh retail beef samples (n = 1062) were collected from eight provinces, and STEC isolates were recovered and characterized. PCR data showed that more than 50% of the samples were stx positive, and 82 STEC isolates were recovered from 14.8% (79/535) stx-positive enriched broths. In contrast, all ciprofloxacin resistant isolates (n = 19) and 13 cefotaxime (CTX) resistant isolates were eae positive and belonged to three serotypes: O111:H8, O26:H11, or O157:H7. Point mutations in quinolone resistance-determining regions and plasmid-mediated quinolone resistance determinants were identified in 16 and 20 isolates, respectively. BlaCTX-M and a point mutation (C-42T) in ampC promoter were detected in 15 and 8 of the CTX resistant isolates, respectively. In addition, macrolide resistance gene mphA was identified in eight azithromycin resistant O111:H8 isolates and one O26:H11 isolate. Single nucleotide polymorphism analysis demonstrated that the O26 and O157 isolates had multiple origins, but the O111 isolates were closely related. Taken together, our data demonstrated that several sequence types associated with hemolytic uremic syndrome from the retail beef samples in China had developed into dangerous multidrug resistant pathogens. The resistant phenotype can facilitate their transmission among the farm animals and human beings when there is an antimicrobial selective pressure.

RESUMO

Food supply has been the central issue of human development for millennia and has become increasingly critical in an urbanizing world. However, the environmental footprints and associated mitigation strategies of food consumption have rarely been comprehensively characterized at urban or regional scales. Here, we analyze the water, carbon, reactive nitrogen, and phosphorus footprints of food consumption in Chinese urban regions and demonstrate how such information can help to formulate tailored mitigation strategies. The results show that in three of the largest urban regions of China, 44-93% of the four footprints are embodied in transboundary food supply. The size of the footprints and the effectiveness of mitigation measures in food supply chain vary across the environmental footprints and urban regions. However, targeting agriculture and food processing sectors in Hebei, Shandong, and Henan provinces can reduce these footprints by up to 47%. Our findings show that the analysis of the environmental footprints along the transboundary food supply chains could inform individualized and effective mitigation targets and strategies.

RESUMO

OBJECTIVE: To study the safety of eight nucleic acid dyes: EB, GelGreen, GoldView, GeneGreen, SYBRGreen I, GelRed, SYBRSafe and SYBRGold. METHODS: Salmonell atyphimurium TA97, TA98, TA100 and TA102 involved in the minimum inhibitory concentration(MIC) of the 8 nucleic acid dyes by the Broth dilution, which follows Clinical and Laboratory Standards Institute. The experiment tests the reverse mutation of bacteria with the 8 nucleic acid dyes, which follows GB 15193. 4-2014. RESULTS: EB and its metabolites show significant mutagenicity on TA97, TA98, TA100 and TA102. SYBRGold and its metabolites do not show mutagenicity on TA97, TA98, TA100 and TA102. The other six dyes show varying mutagenicity on TA97, TA98 and TA102, meanwhile show no mutagenicity on TA100. CONCLUSION: This research shows that, except EB, the other dyes show no mutagenicity under working concentration, but show varying mutagenicity under high concentration. SYBRSafe and SYBRGreenI are highly toxic, and the operators must have high precautions when making diluted solution of them.

RESUMO

The plasmid-mediated high-level tigecycline resistance gene, tet(X4), was detected in seven Escherichia coli isolates from pork in two Chinese provinces. Two isolates belonged to the epidemic spreading sequence type ST101. Tet(X4) was adjacent to ISVsa3 and concurrent with floR in all seven isolates. In addition to IncFIB, the replicon IncFII was found to be linked to tet(X4). This report follows a recent detection of tet(X3)/(X4) in E. coli from animals and humans in China.

RESUMO

Nitrogen flows in urban food systems are attracting increasing concern. However, characteristics of nitrogen flow and systematic measures to reduce reactive nitrogen losses in the food systems of consumption-oriented cities in developing countries have not been well understood, especially in a quantitative way. This study empirically investigates the transforming nitrogen flows of an urban food system in a food-sink city in China, with a nitrogen metabolism model. Three types of nitrogen loads transfer are identified: from production to consumption side, between different environmental media, and from areas within to areas beyond the city boundary. By integrating sensitivity analysis into the metabolism model, increases in the sewage treatment rate, the sewage nitrogen removal rate, and the ratio of animal excreta returned to field are found to contribute the most to the water nitrogen load reduction, and reducing food waste at the consumer level is the most influential measure for lowering soil nitrogen loads, under the existing nitrogen flow regime. Additionally, a three-tier template framework is proposed to streamline city strategies (prevention, abatement, recycling, regional cooperation, etc.) for reducing the N loads of urban food systems, providing references for sustainable nutrient management in urban ecosystems.

RESUMO

Soil carbon sequestration is being considered as a potential pathway to mitigate climate change. Cropland soils could provide a sink for carbon that can be modified by farming practices; however, they can also act as a source of greenhouse gases (GHG), including not only nitrous oxide (N2 O) and methane (CH4 ), but also the upstream carbon dioxide (CO2 ) emissions associated with agronomic management. These latter emissions are also sometimes termed "hidden" or "embedded" CO2 . In this paper, we estimated the net GHG balance for Chinese cropping systems by considering the balance of soil carbon sequestration, N2 O and CH4 emissions, and the upstream CO2 emissions of agronomic management from a life cycle perspective during 2000-2017. Results showed that although soil organic carbon (SOC) increased by 23.2 ± 8.6 Tg C per year, the soil N2 O and CH4 emissions plus upstream CO2 emissions arising from agronomic management added 269.5 ± 21.1 Tg C-eq per year to the atmosphere. These findings demonstrate that Chinese cropping systems are a net source of GHG emissions and that total GHG emissions are about 12 times larger than carbon uptake by soil sequestration. There were large variations between different cropping systems in the net GHG balance ranging from 328 to 7,567 kg C-eq ha-1  year-1 , but all systems act as a net GHG source to the atmosphere. The main sources of total GHG emissions are nitrogen fertilization (emissions during production and application), power use for irrigation, and soil N2 O and CH4 emissions. Optimizing agronomic management practices, especially fertilization, irrigation, plastic mulching, and crop residues to reduce total GHG emissions from the whole chain is urgently required in order to develop a low-carbon future for Chinese crop production.

RESUMO

We characterized antibiotic resistance profiles, antibiotic resistance-associated genes, and pulsed-field gel electrophoresis (PFGE) patterns of 145 Salmonella enterica serotype Typhimurium isolates from human infections and retail foods that were possibly responsible for salmonellosis outbreaks from 2008 to 2012 in Shanghai, China. Resistance to at least three antibiotics was found in 66.7% of chicken isolates, 76.5% of duck isolates, 77.8% of pork isolates, and 80.5% of human isolates. Seven antibiotic resistance phenotypes were detected in chicken isolates, 16 in pork isolates, 17 in duck isolates, and 50 in human isolates. No significant difference (p > 0.05) was found between Salmonella isolates derived from human salmonellosis and from retail foods in terms of the percent resistance of ampicillin, amoxicillin/clavulanic acid, ceftiofur, ceftriaxone, nalidixic acid, chloramphenicol, gentamicin, kanamycin, streptomycin, tetracycline, sulfisoxazole, and sulfamethoxazole/trimethoprim. PFGE using XbaI and BlnI showed that some Salmonella isolates recovered from human infections and retail foods had same or highly similar genetic profile. Same or similar antibiotic resistance profiles, antibiotic resistance associated genes (i.e., qnrA, qnrB, qnrS, aac(6')-Ib, and oqxAB), gene cassettes (i.e., aadA2, dfrA12-aadA2, and aadA1), and mutations were detected in those isolates that exhibited high genetic similarities. These findings highlighted the frequent presence of Salmonella Typhimurium in retail chicken, pork, duck, and humans.

RESUMO

One thousand four hundred ninety-one Salmonella isolates recovered from retail foods including chicken, beef, fish, pork, dumplings, and cold dishes in China in 2007, 2008, 2010, 2011, and 2012 were analyzed for distribution of serotype and antimicrobial susceptibility. A total of 129 Salmonella serotypes were detected among 1491 isolates. Salmonella Enteritidis (21.5%), Typhimurium (11.0%), Indiana (10.8%), Thompson (5.4%), Derby (5.1%), Agona (3.8%), and Shubra (3.0%) were the seven most important serotypes in 1491 isolates. For antibiotic susceptibility, except 16 (1.1%) isolates were susceptible to all tested antibiotics, 131 (8.8%) resisted 1-2 and 1344 (90.1%) resisted three or more antibiotics. One thousand forty-six (70.2%) of 1491 Salmonella isolates were identified as multidrug-resistant (MDR) isolates, which could resist three or more categories of antibiotics. Resistance to sulfisoxazole (78.1%) was most common among the tested Salmonella, followed by tetracycline (70.6%), trimethoprim/sulfamethoxazole (68.0%), and nalidixic acid (63.4%). Resistances to amikacin (20.0%), levofloxacin (18.7%), gatifloxacin (17.9%), ceftriaxone (17.7%), and cefoxitin (13.2%) were less frequently detected. Resistance to fluoroquinolones was most common among Salmonella Shubra and Indiana isolates, while resistance to cephalosporins was frequently detected among Salmonella Thompson isolates. The results highlighted the diversity of Salmonella serotypes and the high prevalence of Salmonella MDR isolates in China. Compared with Salmonella Enteritidis and Typhimurium isolates, the higher fluoroquinolones and cephalosporins resistance rates of some individual serotypes (Salmonella Shubra, Indiana, and Thompson) also provided more information for further study related to fluoroquinolones or cephalosporin-resistant Salmonella.

RESUMO

Salmonellosis is a major global foodborne infection, and strains that are resistant to a great variety of antibiotics have become a major public health concern. The aim of this study was to identify genes conferring resistance to fluoroquinolones and extended-spectrum ß-lactams in nontyphoidal Salmonella (NTS) from patients and food-producing animals in China. In total, 133 and 21 NTS isolates from animals and humans, respectively, exhibiting concurrent resistance to ciprofloxacin and cefotaxime were cultured independently from 2009 to ∼2013. All of the isolates were identified, serotyped, and subjected to antimicrobial susceptibility testing. Importantly, the isolates with concurrent resistance to ciprofloxacin and cefotaxime all were confirmed as S. enterica serovar Indiana. The presence of fluoroquinolone resistance genes and extended-spectrum ß-lactamases (ESBLs) was established by PCR and DNA sequencing. The occurrence and diversity of different genes conferring fluoroquinolone resistance [qepA, oqxAB, and aac(6')-Ib-cr] with mutations in topoisomerase-encoding genes (gyrA and parC) and several ESBLs (including CTX-M-65, CTX-M-27, CTX-M-15, CTX-M-14, and CTX-M-14/CTX-M-15) were noteworthy. Genes located on mobile genetic elements were identified by conjugation and transformation. Pulsed-field gel electrophoresis, used to determine the genetic relationships between these isolates, generated 91 pulsotypes from 133 chicken isolates and 17 pulsotypes from the 21 clinical isolates that showed considerable diversity. Analysis of the pulsotypes obtained with the isolates showed some clones appeared to have existed for several years and had been disseminating between humans and food-producing animals. This study highlights the emergence of ciprofloxacin- and cefotaxime-resistant S. enterica serovar Indiana, posing a threat to public health.

RESUMO

Rapid and adequate identification of anaerobic bacterial species still presents a challenge for most diagnostic laboratories, hindering the selection of appropriate therapy. In this study, the identification capacity of 16S rRNA sequence analysis, VITEK 2 (BioMérieux, Lyon, France) compact analysis and VITEK MS-mediated identification for anaerobic bacterial species was compared. Eighty-five anaerobic bacterial isolates from 11 provinces in China belonging to 14 genera were identified by these three methods. Differences in identification between these three methods were compared. Consistent identification results were obtained for 54 (54/85, 63.5%) isolates by all three methods, the most discordant results being concentrated in Clostridium XI (n = 8) and Bacteroides fragilis (n = 9) clusters. Using the VITEK MS system, 74 (74/90, 82.2%) isolates were identified as single species consistent with 16S rRNA sequence analysis, which was significantly better than the results obtained with VITEK 2 Compact (P < 0.01). Misidentifications by the Vitek 2 Compact and Vitek MS systems were mainly observed in the Clostridium XI (n = 8)and B. fragilis clusters (n = 9). VITEK MS identified anaerobic bacteria even after they had been exposed to oxygen for a week. Identification by the Vitek MS system was more consistent with 16S rRNA sequence analysis than identification by Vitek 2 Compact. Continuous expansion of the VITEK MS database with rare described anaerobic species is warranted to improve both the efficiency and accuracy of VITEK MS identification in routine diagnostic microbiology.

RESUMO

A global multiregional input-output (MRIO) model was built for eight Chinese cities to track their carbon flows. For in-depth understanding of urban carbon footprint from the perspectives of production, consumption, and trade balance, four kinds of footprints and four redefined measurement indicators were calculated. From the global supply chain, urban carbon inflows from Mainland China were larger than outflows, while the carbon outflows to European, principal North American countries and East Asia were much larger than inflows. With the rapid urbanization of China, Construction was the largest consumer and Utilities was the largest producer. Cities with higher consumption (such as Dalian, Tianjin, Shanghai, and Beijing) should change their consumption patterns, while cities with lower production efficiency (such as Dalian, Shanghai, Ningbo, and Chongqing) should improve their technology. The cities of net carbon consumption tended to transfer carbon emissions out of them by trading in carbon-intensive products, while the cities of net carbon production tended to produce carbon-intensive products for nonlocal consumers. Our results indicated that urban carbon abatement requires not only rational consumption and industrial symbiosis at the city level, but also tighter collaboration along all stages of the global supply chain.

RESUMO

Human mobilization and use of reactive nitrogen (Nr) has been one of the major aspects of global change over the past century. Nowhere has that change been more dramatic than in China, where annual net Nr creation increased from 9.2 to 56 Tg from 1910 to 2010. Since 1956, anthropogenic Nr creation exceeded natural Nr creation, contributing over 80% of total Nr until 2010. There is great interest and uncertainty in the fate and effects of this Nr in China. Here, a comprehensive inventory of Nr in China shows that Nr (including recycled Nr) has continuously and increasingly accumulated on land (from 17 to 45 Tg), accompanied by increasing transfers to the atmosphere (before deposition; from 7.6 to 20 Tg), inland waters (from 2.7 to 9.6 Tg), and coastal waters (from 4.5 to 7.7 Tg) over the past 30 y. If current trends continue, Nr creation from human activities will increase to 63 Tg by 2050, raising concerns about deleterious environmental consequences for land, air, and water at regional and global scales. Tremendous amounts of Nr have accumulated in plants, soils, and waters in China over the past 30 y, but the retention capacity of the terrestrial landscape seems to be declining. There is a possibility that the negative environmental effects of excessive Nr may accelerate in coming decades, increasing the urgency to alter the trajectory of increasing Nr imbalance. Here, a conceptual framework of the relationships between human drivers and Nr cycling in China is oriented and well-targeted to Chinese abatement strategies for Nr environmental impact.

RESUMO

ß-Lactamases and extended-spectrum ß-lactamases (ESBLs) producing pathogenic bacteria were widely studied previously in China, but were seldom focused on foodborne Salmonella. In this study, an investigation concerning ß-lactamases and ESBLs producing Salmonella recovered from retail raw chickens was performed. Sixty of 699 foodborne Salmonella isolates were detected as ß-lactamases and ESBLs-producing ones that covered 12 Salmonella serotypes and exhibited different pulsed-field gel electrophoresis genotypes. Forty-four of 60 ß-lactamases and ESBLs-producing strains were simultaneously resistant to ampicillin, amoxicillin/clavulanic acid, ceftiofur, ceftriaxone, and cefoxitin. The most commonly detected ß-lactamases and ESBLs-encoding gene was bla(TEM-1) (n = 44), followed by bla(OXA-1) (n = 38), bla(CMY-2) (n = 29), bla(PSE-1-like) (n = 1), bla(CTX-M-3) (n = 16), and bla(CTX-M-15) (n = 1), respectively. Fourteen, 24, 21, and 1 isolates were detected simultaneously positive for 1, 2, 3, and 4 of the detected ß-lactamases and ESBLs-encoding genes, respectively. A Salmonella strain simultaneously co-carrying bla(TEM-1), bla(OXA-1), bla(CMY-2), and bla(CTX-M-3) was first reported in the present study. Amino acid substitution of Trp244Cys/His247Leu was detected in PSE-1, Val218Asp in CMY-2, and Asp242Gly in CTX-M-15 enzymes, respectively. A difference was found among the amino acid sequences of the detected OXA-1, CMY-2, CTX-M, PSE-1, and TEM-1. The results demonstrated that ß-lactamases and ESBLs were emerging and prevalent in foodborne Salmonella.

RESUMO

OBJECTIVE: To elucidate the epidemic condition and molecular subtyping of ciprofloxacin and cefotaxime co-resistant Salmonella Indiana (S. Indiana) isolated from retail chicken carcasses in six provinces of China. METHODS: A total of 2 647 Salmonella strains isolated from retail chicken carcasses collected from six provinces of China were subjected to antimicrobial susceptibility testing. All Salmonella isolates co-resistant to ciprofloxacin and cefotaxime were further characterized by serotyping, extended-spectrum beta-lactamases (ESBLs) producing strains screening and pulsed field gel electrophoresis (PFGE) typing. RESULTS: Among 2 629 Salmonella isolates tested, 227 (8.52%) isolates were co-resistant to ciprofloxacin and ceftazidime/cefotaxime (Beijing: 11.67% (99/874), Jilin: 8.20% (60/726), Guangdong: 1.39% (7/502), Jiangsu: 15.61% (42/260), Shaanxi: 8.56% (16/186), Inner Mongolia: 0 (0/81)), and 224 of them were identified as S. Indiana. 213 (95.10%) isolates of S. Indiana were ESBLs producing strains. All ciprofloxacin and cefotaxime co-resistant S. Indiana isolates developed a multi-drug resistant profile and 17.86% (40/224) of them were resistant to all antibiotics tested except carbapenems, and 50.89% (114/224) of them resistant to 9 antibiotics, additionally, 25.45% (57/224) of them showed multi-drug resistance to 8 antibiotics. All ciprofloxacin and cefotaxime co-resistant S. Indiana isolates were divided into 32 PFGE clusters and 150 PFGE patterns. Strains of S. Indiana from same or different sampling site and time seemed to either share the same PFGE patterns or be differential to each other in different regions. CONCLUSION: The results indicated that chicken carcasses collected from parts of China were heavily contaminated by ciprofloxacin and cefotaxime co-resistant S. Indiana and could serve as an important reservoir of ciprofloxacin and cefotaxime co-resistant Salmonella. Molecular subtyping results indicated that cross contamination or common pollution source might be in these strains.
