RESUMO

The performance of various measures of rectal mucosal proliferation has been evaluated in the literature, but the performance of the forceps used to obtain the tissue has received little attention. We used data from two large studies of proliferation at a single institution to compare reusable and disposable endoscopic forceps. Endoscopic pinch biopsies were taken 10 cm from the anal verge using either reusable or disposable, oval-cupped, sheathed forceps. The specimens were fixed, embedded, and sectioned, taking care to orient the specimens longitudinally. Five sections were placed on each slide. We determined how many slides did not contain eight scorable crypts (inadequate) and how many sections were necessary to identify eight complete crypts. There were 395 subjects who had biopsies taken with reusable forceps and 185 subjects who had biopsies taken with disposable forceps. The specimens were inadequate in 27.6% of the reusable forceps specimens versus 2.7% of the disposable forceps (P < 0.0001). The mean number of tissue sections necessary to identify eight scorable crypts for the reusable forceps was 3.82 (SD, 0.87) compared with 3.17 (SD, 0.83) for disposable forceps (P = 0.0001). The specimens taken with the disposable forceps were better, probably because the forceps were sharper. We believe that the better quality of the specimens and the sterility justify the higher cost of disposable forceps. We would urge investigators in proliferation studies to evaluate the biopsy equipment as carefully as they evaluate other aspects of their methods.

Modified Nash and Snyder's medium (MNSM) has been used to study soil populations of the fungus Fusarium solani f. sp. glycines, the causal agent of sudden death syndrome (SDS) of soybean. However, no studies have been conducted to confirm the accuracy of this technique in enumerating the SDS pathogen. To determine what portion of the colonies enumerated on MNSM are the SDS pathogen, 282 isolates resembling F. solani f. sp. glycines were collected from soil of two fields with a history of SDS in Arkansas using a soil dilution method with MNSM. The colony morphology of these isolates was compared on MNSM and on potato dextrose agar (PDA). Of these isolates, 112 were tested for pathogenicity on soybean in the greenhouse. In addition to fungi that produced colonies typical of F. solani f. sp. glycines on MNSM, 5 isolates each of four common colony types on MNSM were collected from each field for a total of 40 isolates. These isolates were compared to F. solani f. sp. glycines for colony morphology on PDA and for pathogenicity. Isolates that had colonies on MNSM that resembled F. solani f. sp. glycines had colony morphologies on PDA similar to this pathogen and produced SDS-like foliar symptoms in greenhouse inoculations of soybean. There was a significant quadratic relationship between foliar symptom severity and plant fresh weight. None of the other isolates tested resembled F. solani f. sp. glycines on either MNSM or PDA or produced SDS-foliar symptoms in greenhouse inoculations. These non-SDS isolates were F. solani, F. merismoides, a Pythium sp., and a Paecilomyces sp. All of these isolates produced some root rot, but significantly less than the F. solani f. sp. glycines reference isolate. The results of this study established that soil populations of F. solani f. sp. glycines can be accurately enumerated using MNSM. It should be noted that, on both MNSM and PDA, F. solani f. sp. glycines can resemble some isolates of F. solani f. sp. phaseoli, so pathogenicity tests of random isolates may be required to confirm the forma specialis especially in fields that have a history of both Glycines max and Phaseolus vulgaris.

This study compared the effectiveness of electromyographic (EMG) biofeedback and relaxation treatment in producing flexibility increases at the hip joint. Retention of flexibility gains and changes in sprinting performance were also examined. Fifteen male and 15 female athletes, predominantly sprinters, were matched according to age, sex, and flexibility and were randomly assigned to one of three groups. The control group received only the testing, the biofeedback group received visual and auditory EMG training, and the relaxation group received modified PMR and imagery exercises. Subjects in the two experimental groups were given eight twice-weekly 10-minute sessions and then were retested for flexibility. Retention of flexibility was reassessed 2 and 4 weeks later. Sprint performance improved for all groups. EMG biofeedback and relaxation treatment methods were no more effective than stretching exercises used by the control group for improving flexibility during the treatment period, but they did produce evidence of superior flexibility gains in the retention period. This may be important as flexibility has been reported to be related to the incidence of injuries.

The reference staff of the Washington University School of Medicine Library, in an attempt to consider an alternative way of providing the information contained in the printed Chemical Abstracts, designed a study to examine whether the on-line version of Chemical Abstracts could be substituted for the hard copy. For a thirteen-week period, all patrons using the printed index were offered a free computer search of Chemical Abstracts in exchange for evaluating the searches. Only 39.6% of those offered a free search chose to do so. Of these patrons, 62% still planned to refer to the abstracts later, which are only available in the printed index. The hypothesis that the on-line version could be substituted for the printed index was not confirmed.
