Since genome instability can drive cancer initiation and progression, cells have evolved highly effective and ubiquitous DNA damage response (DDR) programs. However, some cells (for example, in skin) are normally exposed to high levels of DNA-damaging agents. Whether such high-risk cells possess lineage-specific mechanisms that tailor DNA repair to the tissue remains largely unknown. Using melanoma as a model, we show here that the microphthalmia-associated transcription factor MITF, a lineage addition oncogene that coordinates many aspects of melanocyte and melanoma biology, plays a nontranscriptional role in shaping the DDR. On exposure to DNA-damaging agents, MITF is phosphorylated at S325, and its interactome is dramatically remodeled; most transcription cofactors dissociate, and instead MITF interacts with the MRE11-RAD50-NBS1 (MRN) complex. Consequently, cells with high MITF levels accumulate stalled replication forks and display defects in homologous recombination-mediated repair associated with impaired MRN recruitment to DNA damage. In agreement with this, high MITF levels are associated with increased single-nucleotide and copy number variant burdens in melanoma. Significantly, the SUMOylation-defective MITF-E318K melanoma predisposition mutation recapitulates the effects of DNA-PKcs-phosphorylated MITF. Our data suggest that a nontranscriptional function of a lineage-restricted transcription factor contributes to a tissue-specialized modulation of the DDR that can impact cancer initiation.

PURPOSE OF REVIEW: Despite available treatments, many bladder pain syndrome/interstitial cystitis (BPS/IC) patients continue to have poor quality of life. Thus, there is an urge for new therapies. Our manuscript aims to review papers about BPS/IC treatments published in the last 2 years. RECENT FINDINGS: During this period, several treatments were tested, most of them new and others combining treatments already used. Pentosan polysulfate, interleukin 1 antagonist, low energy shock wave, physical therapy, hypnosis, acupuncture, clorpactin, dimethyl sulfoxide and hyaluronic acid plus botulinum toxin-A showed positive results. ASP3652 and lidocaine-releasing intravesical systems failed to prove their efficacy. SUMMARY: Validation of these studies is arduous due to the broad spectre of BPS/IC phenotypes, small number of patients enrolled, distinct outcome measures and short-term follow-up. It is also important to highlight that some authors combined therapies, and others split central and peripheric phenotypes before treatment. Therefore, soon, phenotyping and combining therapies with a step-by-step approach will be needed in BPS/IC treatment.

Silica aerogel composites with recycled tire rubber have been synthesized and evaluated for their potential use for thermal protection in buildings. The present work describes for the first time the preparation of silica-based aerogel composites containing recycled rubber tires reinforced with polyvinyl butyral (PVB) by hot pressing. The developed composite was extensively characterized regarding its physical, morphological, thermal and mechanical features, and the results showed their properties were relevant, leading to composites with different properties/performances. The obtained bulk density values were satisfactory, down to 474 kg·m-3, and very good thermal properties were achieved, namely, thermal conductivity as low as 55 mW·m-1·K-1 for composites with silica aerogel, recycled tire rubber and PVB. The most promising composites were those based on low bulk density and thermal conductivity values, and they were thermally stable, indicating their suitability for thermal insulation applications.

In melanomas, therapy resistance can arise due to a combination of genetic, epigenetic and phenotypic mechanisms. Due to its crucial role in DNA supercoil relaxation, TOP1 is often considered an essential chemotherapeutic target in cancer. However, how TOP1 expression and activity might differ in therapy sensitive versus resistant cell types is unknown. Here we show that TOP1 expression is increased in metastatic melanoma and correlates with an invasive gene expression signature. More specifically, TOP1 expression is highest in cells with the lowest expression of MITF, a key regulator of melanoma biology. Notably, TOP1 and DNA Single-Strand Break Repair genes are downregulated in BRAFi- and BRAFi/MEKi-resistant cells and TOP1 inhibition decreases invasion markers only in BRAFi/MEKi-resistant cells. Thus, we show three different phenotypes related to TOP1 levels: i) non-malignant cells with low TOP1 levels; ii) metastatic cells with high TOP1 levels and high invasiveness; and iii) BRAFi- and BRAFi/MEKi-resistant cells with low TOP1 levels and high invasiveness. Together, these results highlight the potential role of TOP1 in melanoma progression and resistance.

Significantly, the SUMOylation-defective MITF-E318K melanoma predisposition mutation recapitulates the effects of ATM/DNA-PKcs-phosphorylated MITF. Our data suggest that a non-transcriptional function of a lineage-restricted transcription factor contributes to a tissue-specialised modulation of the DDR that can impact cancer initiation.

Highly efficient simultaneous removal of paracetamol and Cu2+ ions from aqueous solutions was accomplished by using bovine bone char (BC). The adsorption behaviour was determined by kinetic and equilibrium studies of both single and binary system solutions. BC is a predominantly mesoporous material with a surface area of 103 m2 g-1. The influence of the initial pH on Cu2+ removal was tested, suggesting that the optimal pH was 3.0. The removal of paracetamol from single and binary systems was 9.45 and 12.7%, respectively. On the other hand, the Cu2+ removal was 36.2% for a single system, suggesting a higher affinity for BC. Moreover, in the case of binary mixtures, the presence of paracetamol led to an enhanced affinity of Cu2+ due to a synergistic/cooperative mechanism, which led to a copper removal of 97.3%. The cooperative model was successfully adjusted to the equilibrium data of the binary systems. The modelling results indicated the formation of a first adsorption layer where paracetamol and copper are retained, and a second layer with a great affinity for copper ions after the formation of a Cu-paracetamol complex, leading to higher removal of Cu2+.

The coronavirus disease 2019 pandemic abruptly changed the dynamics of basic health care, with the consequent need for adjustments in essential services. The objective of this study was to evaluate the acceptance and impact of telemedicine at a Reference Center for Special Immunobiologicals (CRIE). Methods: Patients aged 18 years or older who had a medical referral to CRIE and agreed to have a telemedicine consultation were included. After the medical appointments, participants answered a satisfaction survey. Results: From April 2021 to February 2022, 702 telemedicine consultation were conducted. Over 3,380 vaccines were prescribed via telemedicine. Of all the participants who answered the satisfaction questionnaire, 99.8% stated that they would recommend the service to other people. Conclusions: Telemedicine proved to be promising tool for healthcare at CRIE and had good acceptance by users, potentially improving access and extending the reach of the National Immunization Program.

Fish and other marine animals have a unique and intimate interaction with their surrounding environment. Diet type or trophic level play significant roles in modulating species digestive physiology. However, little is known about how the trophic niche influences digestive activity and gut microbiota in scombrids species. The aim of the present study was to analyse and describe the digestive physiology of the skipjack tuna (Katsuwonus pelamis) and the Atlantic bonito (Sarda sarda) as bioindicator of the feeding ecology and trophic niche for both species in the Gulf of Cádiz (Spain). For that, fish proximate composition, pH of digestive organs and digestive enzymes activity levels were analysed in 10 individuals of each species to gain insight into the digestive physiological adaptations of the two species of scombrids. In addition, intestinal microbiota composition was determined for the skipjack tuna. The integration of the results suggested a clear trophic niche segregation between both species. Stomach pH was associated in both species with infrequent feeding events. Body proximate composition and digestive lipases activity patterns pointed to a high predominance of lipids in the Atlantic bonito diet, suggesting oily fish as main prey. On the other hand, results supported the skipjack tuna as a highly opportunistic feeder with a more varied diet, including fish but also crustaceans as preys. The gut microbial community in the latter species is dominated by Firmicutes and Tenericutes at phylum level, and by Mycoplasma, Blautia and Dorea at genus level. The present study contributes to advance the knowledge on the feeding behaviour and physiology diversity in fish species as a result of adaptation to a particular habitat.

Alzheimer's disease (AD) is the most prevalent neurodegenerative disease commonly diagnosed among the elderly population. AD is characterized by the loss of synaptic connections, neuronal death, and progressive cognitive impairment, attributed to the extracellular accumulation of senile plaques, composed by insoluble aggregates of amyloid-ß (Aß) peptides, and to the intraneuronal formation of neurofibrillary tangles shaped by hyperphosphorylated filaments of the microtubule-associated protein tau. However, evidence showed that chronic inflammatory responses, with long-lasting exacerbated release of proinflammatory cytokines by reactive glial cells, contribute to the pathophysiology of the disease. NLRP3 inflammasome (NLRP3), a cytosolic multiprotein complex sensor of a wide range of stimuli, was implicated in multiple neurological diseases, including AD. Herein, we review the most recent findings regarding the involvement of NLRP3 in the pathogenesis of AD. We address the mechanisms of NLRP3 priming and activation in glial cells by Aß species and the potential role of neurofibrillary tangles and extracellular vesicles in disease progression. Neuronal death by NLRP3-mediated pyroptosis, driven by the interneuronal tau propagation, is also discussed. We present considerable evidence to claim that NLRP3 inhibition, is undoubtfully a potential therapeutic strategy for AD.

Pyrolysis chars derived from rice wastes were chemically activated and used in W(VI) oxyanion adsorption assays in synthetic and mining wastewaters. For comparison purposes, a commercial activated carbon (CAC) was also used. Different experimental conditions were tested in the adsorption assays: solid/liquid ratio (S/L), initial pH, contact time, and initial W concentration. The porous carbon P2C+KOH presented the overall best performance in both media, due to its high surface area (2610 m2 g-1), mesopore volume (1.14 cm3 g-1), and neutral pHpzc (6.92). In the synthetic wastewater, the highest uptake capacity of P2C+KOH (854 mg g-1) was found in the assays with an S/L 0.1 g L-1, an initial pH 2, and an initial W concentration of 150 mg L-1, for 24 h. This value was almost 8 times higher than the one obtained for CAC (113 mg g-1). In the mining wastewater, P2C+KOH showed an even higher uptake capacity (1561 mg g-1) in the assay with the same experimental conditions, which was almost 3 times higher than for CAC (561 mg g-1). These results suggest that P2C+KOH seems to be an efficient alternative to CAC in the W(VI) adsorption from liquid effluents.

Cocos nucifera L. is a palm tree (Arecaceae) with a high economic value. The coconut husk fibers are nonedible, thick, and abrasion-resistant and correspond up to 85% of biomass discarded as solid waste residue. Therefore, the husk fibers are an underexploited byproduct with a high content of extractives of unreported nature. Two varieties of C. nucifera L. husk extracts were investigated to uncover bioactive metabolites and their possible application as a green corrosion inhibitor for carbon steel AISI 1020 under neutral pH conditions. The chemical analysis indicated 3% (w/w) of proanthocyanidins in the husk fibers with a high B-type procyanidin content. The husk fibers' crude extract showed promising results as an eco-friendly corrosion inhibitor for carbon steel AISI 1020 under neutral pH conditions. Although it formed a film on the metal surface in all tested concentrations (0.4, 0.8, 1.2, and 1.6 g L-1), the highest protective efficiency was shown at a concentration of 1.2 g L-1, determined by electrochemical techniques and mass loss. This was the first comprehensive report on coconut husk fibers' chemical composition, which was similar between the two varieties with potential for industrial application.

The aim of the present work was to assess the efficiency of biochars obtained from the co-gasification of blends of rice husk + corn cob (biochar 50CC) and rice husk + eucalyptus stumps (biochar 50ES), as potential renewable low-cost adsorbents for Cr(III) recovery from wastewaters. The two gasification biochars presented a weak porous structure (ABET = 63-144 m2 g-1), but a strong alkaline character, promoted by a high content of mineral matter (59.8% w/w of ashes for 50CC biochar and 81.9% w/w for 50ES biochar). The biochars were used for Cr(III) recovery from synthetic solutions by varying the initial pH value (3, 4, and 5), liquid/solid (L/S) ratio (100-500 mL g-1), contact time (1-120 h), and initial Cr(III) concentration (10-150 mg L-1). High Cr(III) removal percentages (around 100%) were obtained for both biochars, due to Cr precipitation, at low L/S ratios (100 and 200 mL g-1), for the initial pH 5 and initial Cr concentration of 50 mg L-1. Under the experimental conditions in which other removal mechanisms rather than precipitation occurred, a higher removal percentage (49.9%) and the highest uptake capacity (6.87 mg g-1) were registered for 50CC biochar. In the equilibrium, 50ES biochar presented a Cr(III) removal percentage of 27% with a maximum uptake capacity of 2.58 mg g-1. The better performance on Cr(III) recovery for the biochar 50CC was attributed to its better textural properties, as well as its higher cation exchange capacity.

Twelve biochars from forest and agri-food wastes (pruning of Quercus ilex, Eucalyptus grandis, Pinus pinaster, Quercus suber, Malus pumila, Prunus spinosa, Cydonia oblonga, Eriobotrya japonica, Juglans regia, Actinidia deliciosa, Citrus sinensis and Vitis vinifera) were investigated as potential low-cost and renewable adsorbents for removal of a commonly used pharmaceutical, fluoxetine. Preliminary adsorption experiments allowed to select the most promising adsorbents, Quercus ilex, Cydonia oblonga, Eucalyptus, Juglans regia and Vitis vinifera pruning material. They were characterized by proximate, elemental and mineral analysis, thermogravimetric analysis, Fourier transform infrared spectroscopy, determination of specific surface area and pH at the point of zero charge. Batch and equilibrium studies were performed, and the influence of pH was evaluated. The equilibrium was reached in less than 15 min in all systems. The maximum adsorption capacity obtained was 6.41 mg/g for the Eucalyptus biochar, which also demonstrated a good behavior in continuous mode (packed column).

RESUMO Objetivo: identificar a presença de sinais e sintomas de depressão em pessoas idosas hospitalizadas e sua autopercepção sobre este problema de saúde. Métodos: pesquisa de métodos mistos do desenho paralelo convergente e ênfase qualitativa. Doze idosos internados durante o período de 22 a 27 de outubro de 2021 em um hospital do Sul do Brasil, selecionados por conveniência, responderam à entrevista e ao Inventário de Depressão de Beck-II, além da extração documental de variáveis demográficas e clínicas. Os dados foram analisados de forma descritiva e apresentados em categorias temáticas, com articulação em joint display e integração interpretativa. Resultados: os resultados foram organizados em três categorias temáticas: Sinais e sintomas de depressão entre idosos na hospitalização; Desafios enfrentados por idosos hospitalizados que implicam na saúde mental; Elementos de aporte à saúde mental e concepção da depressão entre idosos hospitalizados. Conclusão/Considerações finais: o estudo identificou sinais e sintomas de depressão e os desafios que as pessoas idosas enfrentam, que incluem a própria hospitalização; a sua concepção sobre a depressão e os mecanismos de aporte à sua saúde mental, com destaque à fé. Oportunizou um espaço de fala e escuta ativa dentro do cenário hospitalar, lançando luz sobre a temática.

Twelve older adults hospitalized during the period from October 22 to 27, 2021, in a hospital in Southern Brazil were selected by convenience. The participants were interviewed and answered the Beck Depression Inventory-II, and demographic and clinical variables were extracted from medical records. Data were analyzed descriptively and presented in thematic categories, with a joint display format and interpretive integration. Results: results were organized into three thematic categories: Signs and symptoms of depression among older adults in hospitalization; Challenges faced by hospitalized older adults that affect mental health; Elements of mental health support and conception of depression among hospitalized older adults. Conclusion/Final Considerations: the study identified signs and symptoms of depression and the challenges faced by older adults, which include hospitalization, their conception about depression, and mental health support mechanisms, with emphasis on faith. It provided an opportunity for speaking and active listening within the hospital setting, shedding light on the topic.

This work is dedicated to study the potential application of char byproducts obtained in the gasification of rice husk (RG char) and rice husk blended with corn cob (RCG char) as removal agents of two emergent aquatic contaminants: tetracycline and caffeine. The chars presented high ash contents (59.5-81.5%), being their mineral content mainly composed of silicon (as silica) and potassium. The samples presented a strong basic character, which was related to its higher mineral oxides content. RCG char presented better textural properties with a higher apparent surface area (144 m2 g-1) and higher micropore content (V micro = 0.05 cm3 g-1). The alkaline character of both chars promoted high ecotoxicity levels on their aqueous eluates; however, the ecotoxic behaviour was eliminated after pH correction. Adsorption experiments showed that RG char presented higher uptake capacity for both tetracycline (12.9 mg g-1) and caffeine (8.0 mg g-1), indicating that textural properties did not play a major role in the adsorption process. For tetracycline, the underlying adsorption mechanism was complexation or ion exchange reactions with the mineral elements of chars. The higher affinity of RG char to caffeine was associated with the higher alkaline character presented by this char.

Plantar fibromatosis, Ledderhose's disease, or Morbus Ledderhose is an uncommon benign nodular hyperplasia of the plantar aponeurosis. The aim of this paper was to report the case of a 47-year-old male patient who had concomitant Dupuytren's disease and failed all conservative measures. He was treated surgically with prompt and complete relief of symptoms postoperatively, and he has had no recurrence at the 2-year follow-up. In this richly documented case, we discuss details of the surgical technique and anatomy, which was important for a successful outcome and preventing complications. The technique for subtotal fasciectomy is reviewed and the relevance of the adequate choice of skin incision to prevent painful scarring, skin necrosis, and difficulties with shoe wearing is highlighted.

The main aim of this work was to study the removal efficiency of Pb from synthetic and industrial wastewaters by using biomass fly ashes. The biomass fly ashes were produced in a biomass boiler of a pulp and paper industry. Three concentrations of Pb(2+) were tested in the synthetic wastewater (1, 10 and 1000 mg Pb/L). Moreover, two different wastewaters were collected in an industrial wastewater treatment plant (IWWTP) of an industry of lead-acid batteries: (i) wastewater of the equalization tank, and (ii) IWWTP effluent. All the wastewaters were submitted to coagulation-flocculation tests with a wide range of biomass fly ashes dosage (expressed as Solid/Liquid - S/L - ratios). All supernatants were characterized for chemical and ecotoxicological parameters. The use of biomass fly ashes has reduced significantly the Pb concentration in the synthetic wastewater and in the wastewaters collected in the IWWTP. For example, the definitive coagulation-flocculation assays performed over the IWWTP effluent presented a very low concentration of Pb (0.35 mg/L) for the S/L ratio of 1.23 g/L. Globally, the ecotoxicological characterization of the supernatants resulting from the coagulation-flocculation assays of all wastewaters has indicated an overall reduction on the ecotoxicity of the crude wastewaters, due to the removal of Pb.

O presente artigo relata a prática pedagógica de docentes de Educação Física Escolar que desenvolveram dois projetos relacionados ao circo, sendo um com discentes do Ensino Fundamental e, outro, com os do Ensino Médio. Objetiva-se, inicialmente, discorrer sobre a importância do compromisso ético-político-pedagógico dos professores frente às disparidades sociais, a fim de estabelecer uma ação didática situada com vistas à emancipação coletiva, na acepção freireana. Em seguida, apresentam-se as potencialidades artísticas, afetivas, estéticas, críticas e socioculturais da tematização do circo nas aulas de Educação Física através do relato dos projetos "O circo chegou -o fazer coletivo das acrobacias" e "Circo: entre o espetáculo e o cotidiano". Os resultados evidenciam um rompimento com a visão tecnicista e projetam a potencialidade de tematizar o circo na educação básica como forma de possibilitar trocas humanizadoras provenientes de relações que se constituem a partir de um diálogo com o outro, com o mundo.


This article reports the pedagogical practice of Physical Education teachers who developed two projects related to the circus, one with elementary school students and the other with high school students. The objective is, initially, to discuss the importance of the ethical-political-pedagogical commitment of teachers in face of social disparities, in order to establish a didactic action situated with a view to collective emancipation, in the Freirean perspective. Then, the artistic, affective, aesthetic, critical and sociocultural potentials of the circus theme in Physical Education classes are presented through the report of the projects "The circus has arrived -the collective doing of acrobatics" and "Circus: between the spectacle and the everyday". The results show a break with the technicist view and project the potential to thematize the circus in basic education to enable humanizing exchanges arising from relationships that are constituted from a dialogue with the other, with the world.


Este artículo relata la práctica pedagógica de docentes de Educación Física que desarrollaron dos proyectos relacionados con el circo, uno con el alumnado de la Educación Primaria y otro de la Educación Secundaria. Inicialmente se buscó discutir la importancia del compromiso ético-político-pedagógico de los docentes frente a las desigualdades sociales, para establecer una acción didáctica situada que posibilite la emancipación colectiva, en la perspectiva freireana. A continuación, se presentan las potencialidades artísticas, afectivas, estéticas, críticas y socioculturales de abordar el circo en las clases de Educación Física, a través del relato de los proyectos "El circo llegó -la realización colectiva de acrobacias" y "Circo: entre el espectáculo y la vida cotidiana". Los resultados muestran una ruptura con la visión tecnicista y proyectan la potencialidad de tematizar el circo en la educación básica para posibilitar intercambios humanizadores que surgen de relaciones que se constituyen a partir del diálogo con el otro, con el mundo.

O presente trabalho teve por objetivo descrever e caracterizar a inserção da temática Educação Física no Ensino Médio no âmbito dos periódicos nacionais de Educação Física e Esporte (período 2005-2010). Foram explorados 3313 artigos publicados em periódicos classificados nos extratos A e B do sistema Qualis/CAPES. Destes, 31 artigos (1,06%) trazem a temática da Educação Física no Ensino Médio. Baseando-se na proposição de ANTUNES et al.14, foi possível classificar estes 31 artigos nas seis categorias seguintes: Processo Ensino-Aprendizagem e Indefinido: oito artigos (25,80% cada); Finalidade: sete (22,6%); Caracterização: seis (19,35%); Epistemologia: dois (6,5%); e Formação de Professores: nenhum artigo (0%). Os dados revelam uma restrição na produção de conhecimento no campo da Educação Física e Educação Física no Ensino Médio, sugerindo a necessidade de desenvolver um entrecruzamento estratégico das dimensões epistemológicas da área com as pesquisas de caráter aplicado, perpassando pelas questões inerentes ao processo de formação docente...


The aim of this study was to describe and characterize the insertion of Physical Education in High School" as a scientific subject in national Physical Education and Sport Journals (period 2005-2010). We explored 3313 articles published in journals ranked in extracts A and B of the Qualis/CAPES System. Of these, 31 articles (1.06%) brought the theme of Physical Education in High School. Based on ANTUNES et al.14, it was possible to classify these 31 articles in the following 6 categories: Teaching-Learning Process and Indefinite: 8 articles (25.80% each) Purpose: 7 (22.6%), Characterization: 6 (19.35%), Epistemology: 2 (6.5%) and Teachers Training Process: no articles (0%). These data show that there is a restriction in knowledge production in the Physical Education and Physical Education in High School areas, suggesting that is needed to develop a strategic intersection between epistemologist dimension of the area, going through researches and teachers trainings process...
