ABSTRACT: The human T-cell lymphotropic virus type 1 is a retrovirus that may cause severe diseases such as infective dermatitis associated with HTLV-1 (IDH) and adult T-cell leukemia/lymphoma (ATL). IDH is a chronic relapsing infected eczema of childhood, and ATL is a distinct type of peripheral T-cell leukemia/lymphoma, which is classified into the following types: smoldering, primary cutaneous tumoral, chronic, lymphoma, and acute. Progression of IDH to ATL during the course of IDH has been previously reported in 3 young patients, two of them from Bahia (Brazil). We present the case of a 22-year-old man who had IDH since childhood and developed ATL 18 months ago. The lymphoma lesions were superimposed on previously existing IDH lesions (forehead, axillae, umbilical area, and neck) or in areas generally affected by IDH (external genitalia, hypogastrium, groin, and eyelid). Cutaneous lesions in ATL are very frequent, but in this patient, besides infiltrated plaques and papules presented vesicles on the skin corresponding histologically to dilated Pautrier abscesses. Vesicular ATL is a rare condition. This case constitutes a very demonstrative example of the close correlation between IDH and ATL.

Histoplasmosis is a systemic fungal disease that occurs worldwide, causing symptomatic infection mostly in immunocompromised hosts. Etiological agent is the dimorphic fungus, Histoplasma capsulatum, which occurs in soil contaminated with bird or bat droppings. Major limitation in recognition of H. capsulatum infections is the low awareness, since other diseases may have similar symptomatology. The molecular methods have gained importance because of unambiguous diagnostic ability and efficiency. The aim of this study was to develop and evaluate a padlock probe in view of rolling circle amplification (RCA) detection method which targets ITS (Internal Transcribed Spacer) rDNA of H. capsulatum enabling rapid and specific detection of the fungus in clinical samples. Two padlock probes were designed and one of these (HcPL2) allowed specific amplification of H. capsulatum DNA while no cross-reactivity was observed with fungi used as negative controls. This method proved to be effective for H. capsulatum specific identification and demonstrated to be faster than the traditional method of microbiological identification.

On the moderately complex terrain covered by dense tropical Amazon Rainforest (Reserva Biologica do Cuieiras--ZF2--02°36'17.1'' S, 60°12'24.4'' W), subcanopy horizontal and vertical gradients of the air temperature, CO(2) concentration and wind field were measured for the dry and wet periods in 2006. We tested the hypothesis that horizontal drainage flow over this study area is significant and can affect the interpretation of the high carbon uptake rates reported by previous works at this site. A similar experimental design as the one by Tóta et al. (2008) was used with a network of wind, air temperature, and CO(2) sensors above and below the forest canopy. A persistent and systematic subcanopy nighttime upslope (positive buoyancy) and daytime downslope (negative buoyancy) flow pattern on a moderately inclined slope (12%) was observed. The microcirculations observed above the canopy (38 m) over the sloping area during nighttime presents a downward motion indicating vertical convergence and correspondent horizontal divergence toward the valley area. During the daytime an inverse pattern was observed. The micro-circulations above the canopy were driven mainly by buoyancy balancing the pressure gradient forces. In the subcanopy space the microcirculations were also driven by the same physical mechanisms but probably with the stress forcing contribution. The results also indicated that the horizontal and vertical scalar gradients (e.g., CO(2)) were modulated by these micro-circulations above and below the canopy, suggesting that estimates of advection using previous experimental approaches are not appropriate due to the tridimensional nature of the vertical and horizontal transport locally. This work also indicates that carbon budget from tower-based measurement is not enough to close the system, and one needs to include horizontal and vertical advection transport of CO(2) into those estimates.

We present a new version of the Brazilian developments on the Regional Atmospheric Modeling System where different previous versions for weather, chemistry and carbon cycle were unified in a single integrated software system. The new version also has a new set of state-of-the-art physical parameterizations and greater computational parallel and memory usage efficiency. Together with the description of the main features are examples of the quality of the transport scheme for scalars, radiative fluxes on surface and model simulation of rainfall systems over South America in different spatial resolutions using a scale-aware convective parameterization. Besides, the simulation of the diurnal cycle of the convection and carbon dioxide concentration over the Amazon Basin, as well as carbon dioxide fluxes from biogenic processes over a large portion of South America are shown. Atmospheric chemistry examples present model performance in simulating near-surface carbon monoxide and ozone in Amazon Basin and Rio de Janeiro megacity. For tracer transport and dispersion, it is demonstrated the model capabilities to simulate the volcanic ash 3-d redistribution associated with the eruption of a Chilean volcano. Then, the gain of computational efficiency is described with some details. BRAMS has been applied for research and operational forecasting mainly in South America. Model results from the operational weather forecast of BRAMS on 5 km grid spacing in the Center for Weather Forecasting and Climate Studies, INPE/Brazil, since 2013 are used to quantify the model skill of near surface variables and rainfall. The scores show the reliability of BRAMS for the tropical and subtropical areas of South America. Requirements for keeping this modeling system competitive regarding on its functionalities and skills are discussed. At last, we highlight the relevant contribution of this work on the building up of a South American community of model developers.

Toxoplasmosis is a parasitic zoonosis caused by Toxoplasma gondii protozoan with a worldwide distribution. The aim of this study was to determine the frequency of IgG anti-T. gondii antibodies in bats from São Paulo city, Brazil. A total of 616 serum samples were collected from 22 species of bats. Anti-T. gondii antibodies were searched using the indirect fluorescent antibody test (IFAT ≥ 1:16) and IgG anti-bat antibodies produced in sheep on samples collected during 2006-2011; 32.62% (201/616) of bats had T. gondii antibodies. The modified agglutination test (MAT ≥ 1:25) was performed on samples collected during 2010-2011; 18.61% (35/188) were seropositive. The concordance between IFAT and MAT (serum samples from 2010 to 2011) by Kappa (95% CI) was 0.144, resulting in a low agreement between the techniques. The specificity and sensitivity of MAT and IFAT have not been evaluated for the diagnosis of toxoplasmosis in bats. Thus, it was verified that bats are exposed to T. gondii during their lifetime and they are also part of the toxoplasmosis epidemiology.

The genetic diversity of 47 Histoplasma capsulatum isolates from infected bats captured in Mexico, Brazil, and Argentina was studied, using sequence polymorphism of a 240-nucleotides (nt) fragment, which includes the (GA)(n) length microsatellite and its flanking regions within the HSP60 gene. Three human clinical strains were used as geographic references. Based on phylogenetic analyses of 240-nt fragments achieved, the relationships among H. capsulatum isolates were resolved using neighbour-joining and maximum parsimony methods. The tree topologies obtained by both methods were identical and highlighted two major clusters of isolates. Cluster I had three sub-clusters (Ia, Ib, and Ic), all of which contained Mexican H. capsulatum samples, while cluster II consisted of samples from Brazil and Argentina. Sub-cluster Ia included only fungal isolates from the migratory bat Tadarida brasiliensis. An average DNA mutation rate of 2.39 × 10(-9) substitutions per site per year was estimated for the 240-nt fragment for all H. capsulatum isolates. Nucleotide diversity analysis of the (GA)(n) and flanking regions from fungal isolates of each cluster and sub-cluster underscored the high similarity of cluster II (Brazil and Argentina), sub-clusters Ib, and Ic (Mexico). According to the genetic distances among isolates, a network of the 240-nt fragment was graphically represented by (GA)(n) length haplotype. This network showed an association between genetic variation and both the geographic distribution and the ecotype dispersion of H. capsulatum, which are related to the migratory behaviour of the infected bats studied.
