RESUMO

The evaluation of ecosystem quality is important for land-management and land-use planning. Evaluation is unavoidably subjective, and robust metrics must be based on consensus and the structured use of observations. We devised a transparent and repeatable process for building and testing ecosystem metrics based on expert data. We gathered quantitative evaluation data on the quality of hypothetical grassy woodland sites from experts. We used these data to train a model (an ensemble of 30 bagged regression trees) capable of predicting the perceived quality of similar hypothetical woodlands based on a set of 13 site variables as inputs (e.g., cover of shrubs, richness of native forbs). These variables can be measured at any site and the model implemented in a spreadsheet as a metric of woodland quality. We also investigated the number of experts required to produce an opinion data set sufficient for the construction of a metric. The model produced evaluations similar to those provided by experts, as shown by assessing the model's quality scores of expert-evaluated test sites not used to train the model. We applied the metric to 13 woodland conservation reserves and asked managers of these sites to independently evaluate their quality. To assess metric performance, we compared the model's evaluation of site quality with the managers' evaluations through multidimensional scaling. The metric performed relatively well, plotting close to the center of the space defined by the evaluators. Given the method provides data-driven consensus and repeatability, which no single human evaluator can provide, we suggest it is a valuable tool for evaluating ecosystem quality in real-world contexts. We believe our approach is applicable to any ecosystem.

Actinomycetes are the most important microorganisms for the industrial production of secondary metabolites with antimicrobial and anticancer properties. However, they have not been implicated in biorefineries. Here, we study the ability of the ε-poly-L-lysine producing Streptomyces albulus BCRC 11814 to utilize biodiesel-derived crude glycerol. S. albulus was cultured in a mineral medium supplemented with up to 10% w/v sodium chloride or potassium chloride, and with crude glycerol as the sole carbohydrate source. Under these conditions, the strain produced 0.1 g ε-poly-L-lysine per 1 g of biomass. RNA sequencing revealed upregulation of the ectoine biosynthetic pathway of S. albulus, which provides proof of halotolerance. S. albulus has several silent secondary metabolite biosynthetic clusters predicted within the genome. Based on the results, we conclude that S. albulus BCRC 11814 is a halotolerant microorganism capable of utilizing biodiesel-derived crude glycerol better than other actinomycetes included in the present study. S. albulus has the potential to be established as microbial platform production host for a range of high-value biological products.

AIM: Given the Government's commitment to reducing tobacco availability to minimal levels by 2025, finding ways to decrease the number of tobacco retailers is an important task. We assessed the perceived importance of selling tobacco among dairy owners and managers. METHOD: We conducted a face-to-face survey to explore retailers' views on selling tobacco products, tobacco retailer licensing and tobacco-free retailing. Descriptive statistics were used to analyse the data. RESULTS: Of the 112 individuals invited to participate, 62 (55.4%) completed the survey. Most respondents felt that selling tobacco was important for their business, and almost two-thirds had concerns about tobacco products being a security risk. Twice as many respondents thought tobacco retail outlet licensing was a potentially viable option as those who expressed caution. Almost one-third of respondents were potentially interested in becoming a tobacco-free retailer. CONCLUSION: Selling tobacco products is perceived as important for many dairies, and just over half were not interested in becoming a tobacco-free retailer. However, there is some support among dairy owners/managers for tobacco product licensing. These findings strengthen the case for regulatory measures to decrease tobacco availability, as voluntary adoption of tobacco-free retailing is unlikely to result in substantial reductions in outlet numbers.

RESUMO

During the last few decades, Streptomycetes have shown to be a very important and adaptable group of bacteria for the production of various beneficial secondary metabolites. These secondary metabolites have been of great interest in academia and the pharmaceutical industries. To date, a vast variety of techniques and tools for metabolic engineering of relevant structural biosynthetic gene clusters have been developed. The main aim of this review is to summarize and discuss the published literature on tools for metabolic engineering of Streptomyces over the last decade. These strategies involve precursor engineering, structural and regulatory gene engineering, and the up or downregulation of genes, as well as genome shuffling and the use of genome scale metabolic models, which can reconstruct bacterial metabolic pathways to predict phenotypic changes and hence rationalize engineering strategies. These tools are continuously being developed to simplify the engineering strategies for this vital group of bacteria.

RESUMO

Here, we report the draft genome sequence of Streptomyces albulus strain CCRC 11814, a soil-dwelling, Gram-positive bacterium. S. albulus produces ε-poly-l-lysine, which has diverse antimicrobial activity. The genome is 9.43 Mb in size, with a G+C content of 72.2%, and contains 9,177 protein-coding sequences.
