According to the French recommendations, the elimination of the hepatitis C virus by 2025 could be a realistic public health goal. Screening policies are being intensified, and access to treatment is promoted for patients who escape the usual care pathway. The 'Scanvir' program is an original strategy based on dedicated screening days, as part of the 'test, treat and cure HCV' event in addiction care centers in a French region, during which innovative screening technologies (RDTs, FibroScan® and point-of-care HCV RNA testing) are brought on site and access to a multidisciplinary team is offered. A total of 392 patients attended the 67 regional Scanvir sessions: 31.6% were HCV Ab-positive and 66% of them were HCV RNA-positive. Treatment was initiated in 79.3% of the patients. RDTs were accepted by 62% of the PWIDs (including those who already knew their status) and FibroScan® by 99.5% of the patients. 80% of the viremic patients started their treatment on site and are now cured or still under treatment. Advanced fibrosis evaluated by FibroScan® (LSM > 8 KPa) was suspected in 13.4% and 14.1% of the global and the HCV population, respectively. Scanvir is an efficient strategy for HCV elimination based on dedicated days aimed at increasing cost-effectiveness and offering a multidisciplinary service while saving human care resources. It is an exportable strategy that also offers comprehensive screening of associated chronic liver diseases via the elastometry device and interviews.

Nonpolar anabolic steroids are doping agents that typically do not provide strong signals by electrospray ionization-mass spectrometry (ESI-MS) owing especially to the low polarity of the functional groups present. We have investigated the addition of anions, in ammonium salt form, to anabolic steroid samples as ionization enhancers and have confirmed that lower instrumental limits of detection (as low as 10 ng/mL for fluoxymesterone-M) are obtained by fluoride anion attachment mass spectrometry, as compared to ESI(+)/(-) or atmospheric pressure photoionization (APPI)(+). Moreover, collision-induced decomposition (CID) spectra of precursor fluoride adducts of the bifunctional steroid "reduced pregnenolone" (containing two hydroxyl groups) and its d4-analogue provide evidence of regiospecific decompositions after attachment of fluoride anion to a specific hydroxyl group of the steroid. This type of charting of specific CID reaction pathways can offer value to selected reaction monitoring experiments (SRM) as it may result in a gain in selectivity in detection as well as in improvements in quantification.

RESUMO

The development of electrospray ionization mass spectrometry (ESI-MS) was a 20th century occurrence that underwent rapid acceleration especially in the 1990's. However, long prior to its coupling with mass spectrometry, the electrification of liquids had been studied in a variety of contexts. Although initial reports describing cone formation upon electrification of water drops came out of England, the first true experiments investigating the electrospray phenomenon were performed in the middle of the 18th century by Abbé Jean-Antoine Nollet. The current report, associated with the French Regional Issue of Mass Spectrometry Reviews, examines the contributions of Abbé Nollet to the earliest understanding of the electrospray phenomenon. A description of his accomplishments is placed in the context of the societal and scientific developments of the "Age of Enlightenment" out of which Jean-Antoine Nollet arose.

Volcanic activity can induce flank failure, sometimes generating large earthquakes and tsunamis. However, the failure structures have never been fully characterized and the failure mechanism is still debated. Magmatic activity is a possible trigger, either through fault slip, which might be induced by dyke intrusions, or through sill intrusions, which might be undergoing coeval normal displacements and slip. At the Piton de la Fournaise volcano, satellite imagery combined with inverse modeling highlights the pathways of 57 magmatic intrusions that took place between 1998 and 2020. We show that a major arcuate dyke intrusion zone is connected at depth to a sill intrusion zone, which becomes a fault zone towards the sea, forming a spoon-shaped structure. Some sills are affected by coeval normal displacement and seaward slip. Overall, the structure is characterized by a continuum of displacement from no slip, to sheared sills and finally pure slip. Repeated intrusions into this spoon-shaped structure could trigger catastrophic collapses.

Posttranslational modification by the small ubiquitin-related modifier (SUMO) is a highly regulated modification, which is often restricted to very specific cellular events. A number of analytical strategies for identification of SUMOylated proteins have been previously reported in the literature. A new screening method for SUMOylated peptides based on ion mobility mass spectrometry is presented. Using poly-SUMO2 as a model system, a two-enzyme trypsin/chymotrypsin digestion was performed to reduce the size of the isopeptide conjugated to the substrate lysine residue. Traveling wave ion mobility mass spectrometry was used to screen for peptides containing the QQQTGG isopeptide tag from SUMO, which increases the mass and size of the peptide by 618 Da. This increase in mass along with solution conditions to promote higher charge states allows the isopeptides to be separated from the typically smaller and lesser charged linear peptides. On the basis of these findings, this method can be used as a quick and easy screening method for identifying possible SUMO isopeptides.

A series of [Pd(NHC)(PR(3))] complexes quantitatively form [Pd(η(2)-O(2))(NHC)(PR(3))] products when submitted to an dioxygen atmosphere. Synthetic details and structural features of these Pd-peroxo complexes are described. The palladium compounds were found to be efficient catalysts in the oxidation of alcohols into corresponding carbonyl compounds using O(2) or air as oxidant. Reactions proceed at low catalyst loadings with a wide range of alcohols.
