BACKGROUND: Neurological complications due to reactivation of varicella-zoster virus (VZV) are very uncommon in immunocompetent patients. Generally a vesicular rash is present on one or more dermatomes, preceding or following the main manifestation. Few cases are reported in the international literature, but they concern mainly adult or elderly patients. CASE PRESENTATION: A 12-year-old girl was referred to our hospital for persisting headache, cough and rhinitis for six days. After first examination, diagnosis of anterior sinusitis was made by nasal endoscopy. The day after, the girl developed psychotic symptoms and altered mental status. Computed tomography (CT) scan was immediately performed but was unremarkable; lumbar puncture revealed leukocytosis with lymphocytic predominance and cerebrospinal fluid polymerase chain reaction (PCR) detected varicella-zoster virus DNA. The diagnosis of acute VZV encephalitis was made. The patient was promptly treated with acyclovir infused intravenously and her clinical conditions rapidly improved. Tests made did not show any condition of immunosuppression. CONCLUSIONS: Although if rare, reactivation of VZV can occur in immunocompetent children and its complications can involve central nervous system. Among these complications, meningitis is more common, but cerebral parenchyma can also be involved leading to a severe medical condition that is defined meningoencephalitis. In rare cases vesicular rash may be absent; therefore high level of suspicion is required even in those patients in which suggestive clinical features are not present to guide the diagnosis. Intravenous acyclovir represents the treatment of choice to obtain a fast clinical response and to prevent the onset of late-term complications.

To validate the accuracy of 2 commercially available activity loggers in determining lying, standing, walking, and number of steps in dairy cows, 30 cows were fitted with the CowScout Leg (GEA Farm Technologies, Bönen, Germany) system and the IceTag (IceRobotics Ltd., Edinburgh, Scotland) system. The CowScout Leg logger reports standing and lying in 15-min periods, whereas the IceTag logger reports standing and lying every second. To make data comparable, the IceTag data were therefore also summarized over 15-min periods corresponding to the paired CowScout Leg sensor. These data from the 2 systems were then analyzed (more than 1,000 cow days in total). Video recordings of a total of 29.5 h were used for labeling the behaviors of the selected cows (n = 10) and these labels were used as a gold standard to determine the accuracy with which these 2 loggers can record behavioral states lying, standing, walking, and the behavioral event number of steps. A concordance correlation coefficient analysis showed that both the standing and lying durations obtained with the 2 systems were almost perfectly correlated with the video labeling (IceTag: ρc = 0.999 and 0.999, respectively; CowScout Leg: ρc = 0.995 and 0.996, respectively). However, both loggers performed poorly regarding number of steps (classified as an event; IceTag: ρc = 0.629; CowScout Leg: ρc = 0.678) and CowScout Leg did not detect walking (classified as a state) very accurately (ρc = 0.860). The IceTag system does not measure walking behavior. When comparing the 2 loggers, the correlation between them for standing and lying was substantial (ρc = 0.953 and ρc = 0.953, respectively). The number of steps poorly correlated between the 2 loggers (ρc = 0.593), which might be due to the CowScout Leg logger being attached to the front leg and the IceTag logger being attached to the hind leg. We conclude that both the IceTag and the CowScout Leg logger are able to record standing and lying almost perfectly, but the step counting by both loggers and the walking recording by the CowScout Leg logger are not very accurate.

Bovine respiratory disease (BRD) complex in calves impairs health and welfare and causes severe economic losses for the Stockperson. Early recognition of BRD should lead to earlier veterinary (antibiotic/anti-inflammatory) treatment interventions thereby reducing the severity of the disease and associated costs. Coughing is one of the clinical manifestations of BRD. It is believed that by automatically and continuously monitoring the sounds within calf houses, and analysing the coughing frequency, early recognition of BRD in calves is possible. Therefore, the objective of the present study was to develop an automated calf cough monitor and examine its potential as an early warning system for BRD in artificially reared dairy calves. The coughing sounds of 62 calves were continuously recorded by a microphone over a three-month period. A sound analysis algorithm was developed to distinguish calf coughs from other sounds (e.g. mechanical sounds). During the sound recording period the health of the calves was assessed and scored periodically per week by a trained human observer. Calves presenting with BRD received antibiotic and/or anti-inflammatory treatment and the dates of treatment were recorded. This treatment date reference served as a comparison for the investigation of whether an increase in coughing frequency could be related to calves developing BRD. The calf cough detection algorithm achieved 50.3% sensitivity, 99.2% specificity and 87.5% precision. Four out of five periods, where coughing frequency was observed to be increased, coincided with the development of BRD in more than one calf. This period of increased coughing frequency was always observed before the calves were treated. Therefore, the calf cough monitor has the potential to identify early onset of BRD in calves.

The pattern of body weight gain during the commercial growing of broiler chickens is important to understand growth and feed conversion ratio of each flock.The application of sound analysis techniques has been widely studied to measure and analyze the amplitude and frequency of animal sounds. Previous studies have shown a significant correlation (P ≤ 0.001) between the frequency of vocalization and the age and weight of broilers. Therefore, the aim of this study was to identify and validate a model that describes the growth rate of broiler chickens based on the peak frequency of their vocalizations and to explore the possibility to develop a tool capable of automatically detecting the growth of the chickens based on the frequency of their vocalizations during the production cycle. It is part of an overall goal to develop a Precision Livestock Farming tool that assists farmers in monitoring the growth of broiler chickens during the production cycle. In the present study, sounds and body weight were continuously recorded in an intensive broiler farm during 5 production cycles. For each cycle the peak frequencies of the chicken vocalizations were used to estimate the weight and then they were compared with the observed weight of the birds automatically measured using on farm automated weighing devices. No significant difference is shown between expected and observed weights along the entire production cycles; this trend was confirmed by the correlation coefficient between expected and observed weights (r = 96%, P value ≤ 0.001).The identified model used to predict the weight as a function of the peak frequency confirmed that bird weight might be predicted by the frequency analysis of the sounds emitted at farm level. Even if the precision of the weighing method based on sounds investigated in this study has to be improved, it gives a reasonable indication regarding the growth of broilers opening a new scenario in monitoring systems in broiler houses.

BACKGROUND & AIMS: Ergothioneine (EGT) is a natural occurring compound, synthesized by soil bacteria in fungal substrates, exhibiting antioxidant functions in many cell models. The aim of this study was to assess the effect of EGT in the prevention of H2O2-dependent cell death and oxidative damage on a model of neural cell derived from rat pheocromocytoma, the PC12. METHODS: The ability of EGT was tested by the 3 (4,5-dimethylthiazol-2-yl) 2, 5-diphenyltetrazolium bromide (MTT) assay and Comet assay. H2O2 insult was challenged with increasing concentration of antioxidant using two different incubation periods: 1 and 23 h of EGT pre-treatment followed by 23 and 1 h of H2O2, respectively, for both the MTT and the Comet assay data. CONCLUSION: The pre-treatment for 23 h with EGT, 250 microM and 1mM, followed by 1h of H2O2 incubation at the concentration of 250 and 500 microM, resulted in increased cell viability (P < 0.001) compared to the H2O2 cell batch. This correlated with a decrease in DNA damage as visualized by the Comet assay. Moreover, protein analysis reveals that in the presence of 250 microM of H2O2, EGT acted as a p38-MAPK and Akt specific inhibitor. EGT may play a protective role in rescuing cells from stress-induced apoptosis, likely by activating an intracellular antioxidant pathway involving p38 MAPK genes cascade.

The involvement of oxidative and nitrosative stress mechanisms in several biological and pathological processes including aging, cancer, cardiovascular and neurodegenerative diseases has continued to fuel suggestions that processes can potentially be modulated by treatment with free-radical scavengers and antioxidant. The fermented papaya preparation (FPP) derived from Carica papaya Linn was investigated for its ability to modulate oxidative DNA damage due to H2O2 in rat pheochromocytoma (PC12) cells and protection of brain oxidative damage in hypertensive rats. Cells pre-treated with FPP (50 microg/ml) prior to incubation with H2O2 had significantly increased viability and sustenance of morphology and shape. The human hepatoma (HepG2) cells exposed to H2O2 (50 microM) showed an olive tail moment of 10.56 +/- 1.44 compared to 1.37 +/- 0.29 of the solvent control. A significant reduction (P < or = 0.05) of DNA damage was observed at concentrations > or = 10 microg/ml FPP, with 50 microg/ml FPP reducing the genotoxic effect of H2O2 by about 1.5-fold compared to only H2O2 exposed cells.

We conducted a longitudinal study of 30 preschool children with autism spectrum disorders (ASDs) to evaluate the potential benefits of the Treatment and Education of Autistic and related Communication Handicapped Children (TEACCH). Fifteen children following a low intensity TEACCH program were assessed four times for autism severity, adaptive functioning, language skills, maladaptive behaviors and parental stress and compared with a control group of 15 children following a non-specific approach. Findings suggest that a low intensity home and school TEACCH program may provide benefits for children with ASD by reducing autistic symptoms and maladaptive behaviors. Furthermore, a decrease in parental stress indicates that parents' involvement in the rehabilitation program is a crucial factor and contributes greatly to treatment efficacy.

Maternal impairments in folate metabolism and elevated homocysteinemia are known risk factors for having a child with Down syndrome (DS) at a young age. The 80G>A polymorphism of the reduced folate carrier gene (RFC-1) has been recently demonstrated to affect plasma folate and homocysteine levels, alone or in combination with the 677C>T polymorphism in the methylenetetrahydrofolate reductase (MTHFR) gene. We performed the present study on 80 Italian mothers of DS individuals, aged less than 35 at conception, and 111 Italian control mothers, to study the role of the RFC-1 80G>A, MTHFR 677C>T, and MTHFR 1298A>C genotypes to the risk of a DS offspring at a young maternal age. When polymorphisms were considered alone, both allele and genotype frequencies did not significantly differ between DS mothers and control mothers. However, the combined MTHFR677TT/RFC-1 80GG genotype was borderline associated with an increased risk (OR 6 (CI 95%: 1.0-35.9), P = 0.05), and to be MTHF1298AA/RFC-1 80(GA or AA) was inversely associated with the risk (OR 0.36 (CI 95%: 0.14-0.96), P = 0.04). Present results seem to indicate that none of the RFC-1 80G>A, MTHFR 677C>T, and MTHFR 1298A>C polymorphisms is an independent risk factor for a DS offspring at a young maternal age; however, a role for the combined MTHFR/RFC-1 genotypes in the risk of DS pregnancies among young Italian women cannot be excluded.
