RESUMO

Ainuovirine (ANV), a novel non-nucleoside reverse-transcriptase inhibitor (NNRTI), was approved in China in 2021. In a previous randomized phase 3 trial, ANV demonstrated non-inferior efficacy relative to efavirenz (EFV) and was associated with lower rates of dyslipidemia. In this study, we aimed to explore lipid changes in treatment-experienced people with human immunodeficiency virus (HIV)-1 (PWH) switching to ANV from EFV in real world. At week 24, 96.65% of patients in the ANV group and 93.25% in the EFV group had HIV-1 RNA levels below the limit of quantification (LOQ). Median changes from baseline in CD4 +T cell counts (37.0 vs 36.0 cells/µL, P = 0.886) and CD4+/CD8 +ratio (0.03 vs 0.10, P = 0.360) were similar between the two groups. The ANV group was superior to the EFV group in mean changes in total cholesterol (TC, -0.06 vs 0.26 mmol/L, P = 0.006), triglyceride (TG, -0.6 vs 0.14 mmol/L, P < 0.001), high-density lipoprotein cholesterol (HDL-C, 0.09 vs 0.08 mmol/L, P = 0.006), and low-density lipoprotein cholesterol (LDL-C, -0.18 vs 0.29 mmol/L, P < 0.001) at week 24. We also observed that a higher proportion of patients demonstrated improved TC (13.55% vs 4.45%, P = 0.015) or LDL-C (12.93% vs 6.89%, P = 0.017), and a lower proportion of patients showed worsened LDL-C (5.57% vs 13.52%, P = 0.017) with ANV than with EFV at week 24. In conclusion, we observed good efficacy and favorable changes in lipids in switching to ANV from EFV in treatment-experienced PWH in real world, indicating a promising switching option for PWH who may be more prone to metabolic or cardiovascular diseases.

BACKGROUND: The effectiveness of post-exposure prophylaxis (PEP) depends on participants adherence, making it crucial to assess and compare regimen options to enhance human immunodeficiency virus (HIV) prophylaxis strategies. However, no prospective study in China has shown that the completion rate and adherence of single-tablet regimens in HIV PEP are higher than those of multi-tablet preparations. Therefore, this study aimed to assess the completion rate and adherence of two HIV PEP regimens. METHODS: In this single-center, prospective, open-label cohort study, we included 179 participants from May 2022 to March 2023 and analyzed the differences in the 28-day medication completion rate, adherence, safety, tolerance, and effectiveness of bictegravir/emtricitabine/tenofovir alafenamide (BIC/FTC/TAF) and tenofovir disoproxil fumarate, emtricitabine, and dolutegravir (TDF/FTC + DTG). RESULTS: The PEP completion rate and adherence were higher in the BIC/FTC/TAF group than in the TDF/FTC + DTG group (completion rate: 97.8% vs. 82.6%, P = 0.009; adherence: 99.6 ± 2.82% vs. 90.2 ± 25.29%, P = 0.003). The incidence of adverse reactions in the BIC/FTC/TAF and TDF/FTC + DTG groups was 15.2% and 10.3% (P = 0.33), respectively. In the TDF/FTC + DTG group, one participant stopped PEP owing to adverse reactions (1.1%). No other participants stopped PEP due to adverse events. CONCLUSIONS: BIC/FTC/TAF and TDF/FTC + DTG have good safety and tolerance as PEP regimens. BIC/FTC/TAF has a higher completion rate and increased adherence, thus, is recommended as a PEP regimen. These findings emphasize the importance of regimen choice in optimizing PEP outcomes. Intensive anthropogenic activities have led to drastic changes in land use/land cover (LULC) and impacted the carbon storage in high-groundwater coal basins. In this paper, we conduct a case study on the Yanzhou Coalfield in Shandong Province of China. We further classify waterbodies by using the Google Earth Engine (GEE) to better investigate the process of LULC transformation and the forces driving it in four periods from 1985 to 2020 (i.e., 1985-1995, 1995-2005, 2005-2015, and 2015-2020). We modeled the spatiotemporal dynamics of carbon storage by using InVEST based on the transformation in LULC and its drivers, including mining (M), reclamation (R), urbanization and village relocation (U), and ecological restoration (E). The results indicate that carbon storage had depleted by 19.69 % (321099.06 Mg) owing to intensive transformations in LULC. The area of cropland shrank with the expansion of built-up land and waterbodies, and 56.31 % of the study area underwent transitions in land use in the study period. U was the primary driver of carbon loss while E was the leading driver of carbon gain. While the direct impact of M on carbon loss accounted for only 5.23 % of the total, it affected urbanization and led to village relocation. R led to the recovery of cropland and the reclamation of water for aquaculture, which in turn improved the efficiency of land use. However, it contributed only 2.09 % to the total increase in carbon storage. Numerous complicated and intertwined processes (211) drove the changes in carbon storage in the study area. The work here provides valuable information for decision-makers as well as people involved in reclamation and ecological restoration to better understand the link between carbon storage and the forces influencing it. The results can be used to integrate the goals of carbon sequestration into measures for land management.

Loxoprofen sodium is a chiral drug with two chiral centers. In our previous work, we found that the elimination of its four isomers showed stereospecificity in rats, while how the stereospecific behavior occurred in vivo was unclear. To clarify this issue, each single isomer of loxoprofen sodium was prepared by a chiral semi-preparative high-performance liquid chromatography (HPLC) and then administered to rats. By analysis of each isomer in rat plasma utilizing an analytical chiral HPLC, it was discovered that the chiral inversion occurred only to its (2R)-isomers, one from (1'S,2R)- to (1'S,2S)-isomer and the other from (1'R,2R)- to (1'R,2S)-isomer. The reduction of α-substituted cyclopentanone occurred only to its (1'R)-isomers, with (1'R,2R)-isomer reduced to (2'S,1'R,2R)-trans-alcohol and (1'R,2S)- to (2'S,1'R,2S)-trans-alcohol. Interestingly, both the inversion and the reduction reaction occurred to its (1'R,2R)-isomer due to the special stereo-structure with both (2R)- and (1'R)-configuration, and conversely, neither of them occurred to its (1'S,2S)-isomer, which caused the significantly different elimination rate in vivo. These new findings were meaningful for evaluation of the safety and efficacy of chiral drugs.

ZnAl2O4 with a typical spinel structure is highly expected to be a novel rare-earth-free ion-activated oxide phosphor with red emission, which holds high actual meaning for advancing phosphor-converted light-emitting diode (pc-LED) lighting. Among the rare-earth-free activators, Mn4+ ions have emerged as one of the most promising activators. Considering the price advantage of MnCO3 generating Mn2+ ions and the charge compensation effect potentially obtaining Mn4+ ions from Mn2+ ions, this research delves into a collection of ZnAl2O4:Mn2+(Mn4+), x Li+ (x = 0%-40%) phosphors with Li+ as co-dopant and MnCO3 as Mn2+ dopant source prepared by a high temperature solid-state reaction method. The lattice structure was investigated using X-ray diffraction (XRD), photoluminescence (PL), and photoluminescence excitation (PLE) spectroscopy. Results suggest a relatively high probability of Li+ ions occupying Zn2+ lattice sites. Furthermore, Li+ ion doping was assuredly found to facilitate the oxidization of Mn2+ to Mn4+, leading to a shift of luminescence peak from 516 to 656 nm. An intriguing phenomenon that the emission color changed with the Li+ doping content was also observed. Meanwhile, the luminescence intensity and quantum yield (QY) at different temperatures, as well as the relevant thermal quenching mechanism, were determined and elucidated detailedly.

BACKGROUND: Diabetes mellitus (DM) is a prevalent chronic disease marked by significant metabolic dysfunctions. Understanding its molecular mechanisms is vital for early diagnosis and treatment strategies. METHODS: We used datasets GSE7014, GSE25724, and GSE156248 from the GEO database to build a diagnostic model for DM using Random Forest (RF) and LASSO regression models. GSE20966 served as a validation cohort. DM patients were classified into two subtypes for functional enrichment analysis. Expression levels of key diagnostic genes were validated using quantitative real-time PCR (qRT-PCR) on Peripheral Blood Mononuclear Cells (PBMCs) from DM patients and healthy controls, focusing on CXCL12 and PPP1R12B with GAPDH as the internal control. RESULTS: After de-batching the datasets, we identified 131 differentially expressed genes (DEGs) between DM and control groups, with 70 up-regulated and 61 down-regulated. Enrichment analysis revealed significant down-regulation in the IL-12 signaling pathway, JAK signaling post-IL-12 stimulation, and the ferroptosis pathway in DM. Five genes (CXCL12, MXRA5, UCHL1, PPP1R12B, and C7) were identified as having diagnostic value. The diagnostic model showed high accuracy in both the training and validation cohorts. The gene set also enabled the subclassification of DM patients into groups with distinct functional traits. qRT-PCR results confirmed the bioinformatics findings, particularly the up-regulation of CXCL12 and PPP1R12B in DM patients. CONCLUSION: Our study pinpointed seven energy metabolism-related genes differentially expressed in DM and controls, with five holding diagnostic value. Our model accurately diagnosed DM and facilitated patient subclassification, offering new insights into DM pathogenesis.

In this work, the pKa values of 69 polar alkanes (YH2) in acetonitrile were computed using the method developed by Luo and Zhang in 2020, and representative 69 thermodynamic network cards on 22 elementary steps of YH2 and related polar alkenes (Y) releasing or accepting H2 were naturally established. Potential electron reductants (YH-), hydride reductants (YH-), antioxidants (YH2 and YH-), and hydrogen molecule reductants (YH2) are unexpectedly discovered according to thermodynamic network cards. It is also found that there are great differences between YH2 and common hydrogen molecule reductants (XH2), such as Hantzsch ester (HEH2), benzothiazoline (BTH2), and dihydro-phenanthridine (PH2), releasing two hydrogen ions to unsaturated compounds. During the hydrogenation process, XH2 release hydrides first, then the oxidation state XH+ release protons. However, in the case of YH2, YH2 release protons first, then YH- release hydrides. It is the differences on acidic properties of YH2 and XH2 that result in the behavioral and thermodynamic differences on YH2 and XH2 releasing two hydrogen ions (H--H+). The redox mechanisms and behaviors of Y, YH-, and YH2 as electron, hydrogen atom, hydride, and hydrogen molecule donors or acceptors in the chemical reaction are reasonably investigated and discussed in this paper using thermodynamics.

Step-feature lines are one of the important geometrical elements for drawing the status quo maps of open-pit mines, and the efficient and accurate automatic extraction and updating of step-feature lines is of great significance for open-pit-mine stripping planning and analysis. In this study, an automatic extraction method of step-feature lines in an open-pit mine based on unmanned-aerial-vehicle (UAV) point-cloud data is proposed. The method is mainly used to solve the key problems, such as low accuracy, local-feature-line loss, and the discontinuity of the step-feature-line extraction method. The method first performs the regular raster resampling of the open-pit-mine cloud based on the MLS algorithm, then extracts the step-feature point set by detecting the elevation-gradient change in the resampled point cloud, further traces the step-feature control nodes by the seed-growth tracking algorithm, and finally generates smooth step-feature lines by fitting the space curve to the step-feature control nodes. The results show that the method effectively improves the accuracy of step-feature-line extraction and solves the problems of local-feature-line loss and discontinuity.

The evolution of land-use spatial conflict is a comprehensive reflection of regional development and a common phenomenon in urbanization. The research on land-use spatial conflicts can provide theoretical basis and practical support for spatial control. The present study employs Tianjin as the research object, and ecological risk assessment model was improved based on landscape pattern analysis by selecting appropriate indicators that may bring conflict risk problems. The spatial conflict measurement model of land-use based on the coordination of human-land-landscape elements was preliminarily constructed. The spatial and temporal evolution of land-use spatial conflict degree, and spatial autocorrelation from 2000 to 2020 was preliminarily analyzed. Results showed that (1) the conflict measurement level of the model was verified by Google Earth remote sensing image platform and field survey, and the results were relatively consistent with the actual situation, which proves that this method was feasible. (2) The areas with low intensity of land-use conflict were mainly distributed in remote mountainous areas with less human activities, while the areas with high comprehensive conflict index were mainly distributed in the central urban area with more active human activities and urbanization, the urban fringe area, the water-land ecotone, and some landscape transition areas. (3) There was a close relationship between land use spatial conflict and land-use change. Areas with severe and frequent land-use changes were often areas with noticeable land-use conflict changes. In the period of high land-use change intensity and frequent land-use mode conversion, it was also a period of a large area where regional land-use conflict was enhanced. (4) The overall level of conflict showed an average trend year by year, and the high conflict fluctuates greatly with first rising and then falling, but the change range was small. Moderate conflict mainly showed a decreasing trend, with an extensive change range. The early stage of the mild conflict was relatively stable, and the later stage changed significantly.

Soil nutrient is one of the most important properties for improving farmland quality and product. Imaging spectrometry has the potential for rapid acquisition and real-time monitoring of soil characteristics. This study aims to explore the preprocessing and modeling methods of hyperspectral images obtained from an unmanned aerial vehicle (UAV) platform for estimating the soil organic matter (SOM) and soil total nitrogen (STN) in farmland. The results showed that: (1) Multiplicative Scattering Correction (MSC) performed better in reducing image scattering noise than Standard Normal Variate (SNV) transformation or spectral derivatives, and it yielded a result with higher correlation and lower signal-to-noise ratio; (2) The proposed feature selection method combining Successive Projections Algorithm (SPA) and Competitive Adaptive Reweighted Sampling algorithm (CARS), could provide selective preference for hyperspectral bands. Exploiting this method, 24 and 22 feature bands were selected for SOM and STN estimation, respectively; (3) The particle swarm optimization (PSO) algorithm was employed to obtain optimized input weights and bias values of the extreme learning machine (ELM) model for more accurate prediction of SOM and STN. The improved PSO-ELM model based on the selected preference bands achieved higher prediction accuracy (R2 of 0.73 and RPD of 1.91 for SOM, R2 of 0.63, and RPD of 1.53 for STN) than support vector machine (SVM), partial least squares regression (PLSR), and the ELM model. This study provides an important guideline for monitoring soil nutrient for precision agriculture with imaging spectrometry.

Underground coal mining leads to land subsidence, and the situation is particularly serious in the Coal-Grain Complex in eastern China, causing the crop production to be reduced or to be taken out. Backfilling with Yellow River sediment is one of the effective methods to solve the land subsidence in this area, but a key issue is how to select the optimal soil reconstruction profile so that the crop yield after backfilling and reclamation is unaffected. The main purpose of this study is to verify the feasibility of selecting the optimal soil reconstruction profile by rapid monitoring of crop growth and judging soil quality with the aid of unmanned aerial vehicle systems (UAVs). A control treatment and 13 experimental treatments were established for the study area. The control treatment consisted of using 30 cm topsoil and 90 cm subsoil and the topsoil is a proxy for native (undisturbed) soil from the study sites. All other treatments consisted of using varying combinations of subsoil and sediment overlain by 30 cm of topsoil. The vegetation indices from the UAV multispectral images, and the plant height and vegetation coverage from the UAV RGB images were used for estimation of the winter wheat biomass in a random forest regression. The results showed that the random forest regression model yielded accurate estimation of the aboveground biomass. Furthermore, knowledge of plant height and vegetation coverage improved the accuracy of prediction such that crop growth was well characterized. The optimal soil profile consisted of 0.3 m topsoil + 0.2 m subsoil + 0.2 m sediment + 0.2 m subsoil + 0.3 m sediment. A fast and effective airborne monitoring method for soil quality was established, thus providing greatly improved monitoring efficiency.

In this work, thermodynamic driving forces on 20 possible elementary steps of Hantzsch ester (HEH2), benzothiazoline (BTH2), and dihydrophenanthridine (PDH2) releasing two hydrogen atoms or ions were measured or derived from the related thermodynamic data using Hess' law in acetonitrile. Furthermore, thermodynamic network cards of HEH2, BTH2, and PDH2 releasing two hydrogen atoms or ions on 20 elementary steps were first established. Based on the thermodynamic network cards, hydride-donating, hydrogen-atom-donating, and electron-donating abilities of XH2 and XH-, and two hydrogen-atom(ion)-donating abilities of XH2 are discussed in detail. Obviously, the thermodynamic network cards of HEH2, BTH2, and PDH2 not only offer rational data guidance for organic synthetic chemists to properly choose an appropriate reducer among the three reducing agents to hydrogenate various unsaturated compounds but also strongly promote elucidatation of the detailed hydrogenation mechanisms.

Copper is an important national resource, which is widely used in various sectors of the national economy. The traditional detection of copper content in copper ore has the disadvantages of being time-consuming and high cost. Due to the many drawbacks of traditional detection methods, this paper proposes a new method for detecting copper content in copper ore, that is, through the spectral information of copper ore content detection method. First of all, we use chemical methods to analyze the copper content in a batch of copper ores, and accurately obtain the copper content in those ores. Then we do spectrometric tests on this batch of copper ore, and get accurate spectral data of copper ore. Based on the data obtained, we propose a new two hidden layer extreme learning machine algorithm with variable hidden layer nodes and use the regularization standard to constrain the extreme learning machine. Finally, the prediction model of copper content in copper ore is established by using the algorithm. Experiments show that this method of detecting copper ore content using spectral information is completely feasible, and the algorithm proposed in this paper can detect the copper content in copper ores faster and more accurately.

In this work, kinetic isotope effect (KIEself) values of 68 hydride self-exchange reactions, XH(D) + X(+) → X(+) + XH(D), in acetonitrile at 298 K were determined using a new experimental method. KIE values of 4556 hydride cross transfer reactions, XH(D) + Y(+) → X(+) + YH(D), in acetonitrile were estimated from the 68 determined KIEself values of hydride self-exchange reactions using a new KIE relation formula derived from Zhu's kinetic equation and the reliability of the estimations was verified using different experimental methods. A new KIE kinetic model to explain and predict KIE values was developed according to Zhu's kinetic model using two different Morse free energy curves instead of one Morse free energy curve in the traditional KIE theories to describe the free energy changes of X-H bond and X-D bond dissociation in chemical reactions. The most significant contribution of this paper to KIE theory is to build a new KIE kinetic model, which can be used to not only uniformly explain the various (normal, enormous and inverse) KIE values but also safely prodict KIE values of various chemical reactions.

A series of analogues of indazolium alkaloids were designed and synthesized. The thermodynamic driving forces of the 6 elemental steps for the analogues of indazolium alkaloids to obtain hydride in acetonitrile were determined using an isothermal titration calorimeter (ITC) and electrochemical methods, respectively. The effects of molecular structure and substituents on the thermodynamic driving forces of the 6 steps were examined. Meanwhile, the oxidation mechanism of NADH coenzyme by indazolium alkaloids was examined using the chemical mimic method. The result shows that the oxidation of NADH coenzyme by indazolium alkaloids in vivo takes place by one-step concerted hydride transfer mechanism.

RESUMO

Based on the whole life cycle theory, the carbon effect of three different sizes of high-standard basic farmland construction projects is measured and analysed. The results show that the carbon emissions generated during the construction of high-standard basic farmland projects and the carbon absorption capacity at the later stage are different for projects of different sizes. The carbon emissions of different scales of high-standard basic farmland projects will increase during the construction stage. The results of carbon effect changes in the later operation and management stage show that the high-standard basic farmland construction projects will help reduce the carbon emissions of the field ecosystem where the farmland is located and increase its carbon sink capacity after the completion of construction, which is more obvious in larger projects. The emission reduction and carbon sequestration capacity of the farmland after remediation are improved to different degrees, which is more conducive to the ecological development of agricultural production and ecological environmental protection in the relevant areas. The study contributes to the green development of farmland, which is of some significance for the sustainable development of agriculture in Tianjin and the whole country.

In China, non-nucleoside reverse transcriptase inhibitors (NNRTIs) are integral to the antiretroviral therapy (ART) regimen for persons living with HIV (PWH), comprising over 80% of such treatments. To broaden treatment options and improve therapeutic effectiveness, Ainuovirine (ANV), a new NNRTI, was authorized for ART in 2021. Nevertheless, the clinical efficacy of ANV and its impact on blood biochemical markers remain somewhat underexplored. This study seeks to evaluate ANV's clinical performance in ART and its influence on relevant treatment parameters. A retrospective analysis was performed on 208 patients treated with an ANV-based regimen from July 2021 to July 2023, monitoring indicator changes from baseline to week 24. The primary endpoint was the proportion of participants achieving HIV-1 RNA levels of less than 50 copies/mL by week 24. Secondary endpoints involved assessing variations in CD4+ T cell counts and blood biochemical markers from baseline. These outcomes were also compared with data from 241 patients treated with an Efavirenz (EFV)-based regimen in the same time frame. The findings suggest that the ANV-based regimen is as effective as the EFV-based regimen in viral suppression (p > .05) and offers superior improvements in lipid profiles, liver function, and immune system indicators, alongside fewer adverse reactions. These results affirm ANV's efficacy and safety as an antiretroviral therapy option, offering Acquired Immune Deficiency Syndrome patients a wider array of treatment possibilities and the potential for better treatment outcomes.

Toxic epidermal necrolysis (TEN) is a type of drug eruption in dermatology emergencies that is rare in clinical practice but has a high mortality rate. The main causes are drug and viral infections. Unfortunately, no expert consensus on treating this disease exists, and a standard therapy is absent. Up to now, glucocorticoids combined with gamma globulin are commonly used in clinical practice, but their efficacy is highly controversial. This study reports on a 7-year-old girl with TEN who did not respond to traditional therapy, such as methylprednisolone combined with gamma globulin, but was finally cured with an additional low-dose etanercept. The results showed that etanercept therapy in paediatric TEN is safe, reliable and worth recommending.

BACKGROUND: This study aimed to evaluate the therapeutic effect and tolerance of bictegravir/emtricitabine/tenofovir alafenamide fumarate (BIC/FTC/TAF) use for 24 weeks in anti-retroviral therapy (ART)-naïve patients in China. METHODS: This single-center retrospective cohort study included ART-naïve patients who received BIC/FTC/TAF from July 2021 to April 2022. The proportion of patients with HIV RNA < 50 copies/mL at the end point of 24 weeks (virological suppression rate) was the primary outcome, and the changes in CD4 cell count, CD4/CD8 ratio, weight, blood lipid, and safety were secondary outcomes. RESULTS: A total of 80 ART-naïve patients were enrolled. The virological suppression rate was 86.3% at 24 weeks. The median CD4 cell count increased from 212 cells/µL (interquartile range [IQR]: 90.3-398.3) at baseline to 348 cells/µL (IQR: 219.8-541.0) at 24 weeks. The median CD4/CD8 ratio increased from 0.25 (IQR: 0.13-0.37) at baseline to 0.40 (IQR: 0.26-0.66) at 24 weeks. During the follow-up of 80 ART-naïve patients using BIC/FTC/TAF, 16 participants had adverse events; however, these events did not lead to drug withdrawal. CONCLUSION: This real-world cohort study showed that BIC/FTC/TAF could achieve good immunological and virological responses in ART-naïve patients. In addition, this study also shows good safety.

INTRODUCTION: The efficacy and safety of ainuovirine+lamivudine+tenofovir (ANV+3TC+TDF) and efavirenz+lamivudine+tenofovir (EFV+3TC+TDF) have been confirmed in previous clinical trials; however, there are no related studies on patient-reported outcomes. This study aimed to evaluate the effectiveness and safety of these 2 antiretroviral therapy regimens and to understand the patient's symptom experience and subjective experience of sleep quality through patient-reported outcomes. METHODS: This is a single-center prospective cohort study with 243 patients evaluated from October 1, 2021 to June 30, 2022. Virological effectiveness and patient-reported outcomes results were analyzed. The primary endpoint was the proportion of HIV viral load <50 copies/mL (virological suppression rate) at 48 weeks and the changes in the HIV symptom index and Pittsburgh sleep quality index. RESULTS: The virological suppression rates in the ANV+3TC+TDF and EFV+3TC+TDF groups were 83.6% (102/122) and 87.6% (106/121), respectively, at 48 weeks. In the ANV+3TC+TDF group, the scores of HIV symptom index and pittsburgh sleep quality index in the 48th week were lower than the baseline level (p < 0.05). Logistic regression results showed that the baseline regimen EFV+3TC+TDF was a risk factor for dizziness/lightheadedness (odds ratio = 3.153, 95% confidence interval: 1.473-6.748, p = 0.003), sadness/depression odds ratio = 2.404, 95% confidence interval:1.188-4.871, p = 0.015), and difficulty sleeping (odds ratio = 2.802, 95% confidence interval: 1.437-5.463, p = 0.002) at 48 weeks. CONCLUSIONS: Both regimens showed good virological effectiveness; however, compared with ANV+3TC+TDF, the EFV+3TC+TDF regimen reduced the prevalence of HIV-related symptoms.
