Salmonellosis remains a major foodborne disease threat to public health worldwide. Swine are considered a reservoir for many Salmonella serotypes affecting humans; however, not all serotypes of concern in food animal products cause clinical signs of infection in swine. The objective of this study was to evaluate the presence and distribution of Salmonella spp. in finishing pigs at commercial farms across Kansas (USA). Five farms were selected and sampled when pigs weighed between 125 and 136 kg. Samples were collected and transported to the laboratory for processing following USDA-FSIS guidelines. Susceptibility and resistance profiles were also studied. Fifty-three percent (100/186) of samples were culture positive for Enterobacteriaceae, and 14% (14/100) were confirmed Salmonella positive by PCR with three of five farms having no PCR-positive samples. Salmonella serotype Braenderup was the most common serovar identified in environmental samples, while Salm. Infantis, Agona, and Montevideo were identified in fecal samples. Multidrug resistance patterns were only found in Farm 3, in fecal samples and in one floor sample. The observations reported in this study highlight areas of concern, such as locations prone to fecal contamination, to be considered when cleaning and sanitizing between groups of pigs to decrease presence of Salmonella spp. in farm environments.

RESUMO

Mapping the structural and functional connectivity of the central nervous system has become a key area within neuroimaging research. While detailed network structures across the entire brain have been probed using animal models, non-invasive neuroimaging in humans has thus far been dominated by cortical investigations. Beyond the cortex, subcortical nuclei have traditionally been less accessible due to their smaller size and greater distance from radio frequency coils. However, major neuroimaging developments now provide improved signal and the resolution required to study these structures. Here, we present an overview of the connectivity between the amygdala, brainstem, cerebellum, spinal cord and the rest of the brain. While limitations to their imaging and analyses remain, we also provide some recommendations and considerations for mapping brain connectivity beyond the cortex.

BACKGROUND: Current models of breathlessness often fail to explain disparities between patients' experiences of breathlessness and objective measures of lung function. While a mechanistic understanding of this discordance has thus far remained elusive, factors such as mood, attention and expectation have all been implicated as important modulators of breathlessness. Therefore, we have developed a model to better understand the relationships between these factors using unsupervised machine learning techniques. Subsequently we examined how expectation-related brain activity differed between these symptom-defined clusters of participants. METHODS: A cohort of 91 participants with mild-to-moderate chronic obstructive pulmonary disease (COPD) underwent functional brain imaging, self-report questionnaires and clinical measures of respiratory function. Unsupervised machine learning techniques of exploratory factor analysis and hierarchical cluster modelling were used to model brain-behaviour-breathlessness links. RESULTS: We successfully stratified participants across four key factors corresponding to mood, symptom burden and two capability measures. Two key groups resulted from this stratification, corresponding to high and low symptom burden. Compared with the high symptom burden group, the low symptom burden group demonstrated significantly greater brain activity within the anterior insula, a key region thought to be involved in monitoring internal bodily sensations (interoception). CONCLUSIONS: This is the largest functional neuroimaging study of COPD to date, and is the first to provide a clear model linking brain, behaviour and breathlessness expectation. Furthermore, it was possible to stratify participants into groups, which then revealed differences in brain activity patterns. Together, these findings highlight the value of multimodal models of breathlessness in identifying behavioural phenotypes and for advancing understanding of differences in breathlessness burden.

Introduction: Interoception, the perception of the internal state of the body, has been shown to be closely linked to emotions and mental health. Of particular interest are interoceptive learning processes that capture associations between environmental cues and body signals as a basis for making homeostatically relevant predictions about the future. One method of measuring respiratory interoceptive learning that has shown promising results is the Breathing Learning Task (BLT). While the original BLT required binary predictions regarding the presence or absence of an upcoming inspiratory resistance, here we extended this paradigm to capture continuous measures of prediction (un)certainty. Methods: Sixteen healthy participants completed the continuous version of the BLT, where they were asked to predict the likelihood of breathing resistances on a continuous scale from 0.0 to 10.0. In order to explain participants' responses, a Rescorla-Wagner model of associative learning was combined with suitable observation models for continuous or binary predictions, respectively. For validation, we compared both models against corresponding null models and examined the correlation between observed and modeled predictions. The model was additionally extended to test whether learning rates differed according to stimuli valence. Finally, summary measures of prediction certainty as well as model estimates for learning rates were considered against interoceptive and mental health questionnaire measures. Results: Our results demonstrated that the continuous model fits closely captured participant behavior using empirical data, and the binarised predictions showed excellent replicability compared to previously collected data. However, the model extension indicated that there were no significant differences between learning rates for negative (i.e. breathing resistance) and positive (i.e. no breathing resistance) stimuli. Finally, significant correlations were found between fatigue severity and both prediction certainty and learning rate, as well as between anxiety sensitivity and prediction certainty. Discussion: These results demonstrate the utility of gathering enriched continuous prediction data in interoceptive learning tasks, and suggest that the updated BLT is a promising paradigm for future investigations into interoceptive learning and potential links to mental health.

Introduction: Anxiety is one of the most prevalent mental health conditions worldwide, and psychotherapeutic techniques can be employed to help manage and mitigate symptoms. While the available therapies are numerous, key strategies often involve cognitive and/or embodiment techniques. Within body-centered methods, breathing-oriented approaches are particularly prevalent, using either attention towards or active control of breathing. As the perception of body states (i.e., interoception) is thought to be an integral component of emotion generation, these embodiment and breathing techniques may be key in addressing the miscommunication between the brain and body that is thought to exist with anxiety. Therefore, we conducted a systematic review and meta-analysis to assess the effects of acute administration of psychological interventions for state anxiety. Results: This systematic review was conducted in accordance with the PRISMA statement and registered prospectively in PROSPERO. A literature search for randomized controlled trials was conducted in PubMed, PsycINFO, and Scopus. We considered interventions that focused on cognitive, embodiment or breathing strategies, or a combination of these techniques. Twelve studies met our inclusion criteria, and study characteristics, quality and effect sizes were assessed. A single cognitive study was found to produce a moderate reduction in state anxiety, while moderate to large effects were found across studies assessing embodiment practices. In contrast, studies which utilized breathing-based interventions alone produced inconsistent results, with both attention towards and active control of breathing producing large to no effects depending on the technique employed. Finally, consistent moderate effects were found with combination techniques that involved passive attention (e.g., towards cognitions, body and/or breathing), with active combination techniques producing inconsistent results. Discussion: While study numbers are limited regarding brief interventions, cognitive and embodiment techniques are consistently helpful for reducing state anxiety, while breathing-based exercises need to consider the specific technique employed, and how successful this may be for each individual. Furthermore, combined practices such as mindfulness can also be successful, although care must be taken when introducing an active change to one or more elements. African swine fever virus (ASFV) is a highly infectious virus known to cause substantial mortality and morbidity in pigs. The transmissibility and severity of disease within pigs, as well as the potentially resultant catastrophic trade ramifications, warrant its status as a foreign animal disease of substantial concern to the United States. The ASFV virus can survive for extended periods of time outside its host, and its greatest concentration is often observed in blood and organs, products that are frequently used as raw materials to manufacture porcine-derived ingredients fed to animals in the United States. Unlike ruminant-based proteins that cannot be fed to ruminant animals, it is permissible to feed porcine-derived ingredients to pigs in the United States. However, the increased threat of ASFV entry into the United States and our evolving understanding of viral transmission by feedstuffs warrant further investigation into this practice. The objectives of this review are to describe the current knowledge of ASFV survival in raw materials used to produce porcine-based ingredients, identify priorities for future research, and summarize potential options for managing risk until additional knowledge can be gained. While limited data is available for ASFV-specific mitigation, the temperatures used in both spray-drying and rendering have proven to effectively reduce viral concentrations of multiple swine viruses below detectable limits. However, some of these procedures may not eliminate the risk of recontamination, which necessitates the need for additional prevention or mitigation measures. Most published research in this area relies on direct inoculation of raw ingredient, not the finished porcine-derived ingredient. Currently, three published studies report ASFV mitigation in either thermally processed conditions (>40 °C) or ingredient quarantine (<40 °C). Virus inactivation, or the reduction of viral concentrations below detectable levels, was observed in the thermally processed study and one of the two ingredient quarantine studies. In conclusion, there is little knowledge to eliminate the risk of recontamination in porcine-derived ingredients; therefore, future research should aim to support and validate the currently available literature for the continued and safe production of porcine-derived ingredients in the event of a foreign animal disease outbreak.

Formaldehyde has been found to decrease virus concentrations in feed and ingredient matrices. Continued research is needed to identify the appropriate inclusion levels and application time for different viruses in these matrices. The objective was to evaluate different inclusion levels of formaldehyde when applied either pre- or postinoculation of porcine epidemic diarrhea virus (PEDV), type 2 porcine reproductive and respiratory syndrome virus (PRRSV), and Seneca Valley virus 1 (SVV1) to complete feed or soybean meal. The experiment was designed in a 2 × 2 factorial with a formaldehyde-based product (Termin-8, Anitox Corporation, Lawrenceville, GA) applied either before virus inoculation (preinoculation) or after inoculation (postinoculation) at either a 2 or 3 kg/MT. On day 0, samples of the respective matrices were weighed in 50 g aliquots and added to 500 mL bottles. Formaldehyde was applied to the preinoculation samples at the respective inclusion levels and 50 µL of each virus were added to the postinoculation samples. All bottles were shaken and allowed to sit at room temperature for 24 h. On day 1, virus was added to the preinoculation samples and formaldehyde was added to the postinoculation bottles. Half of the samples were immediately processed (0 h) and the other half were incubated at room temperature for an additional 24 h. Samples were processed and aliquots were analyzed via triplex PCR. An application time × inclusion level interaction was observed for PEDV at 0 h and SVV1 and PEDV at 24 h in complete feed, where less viral RNA (P < 0.05) was detected in the postinoculation samples at either inclusion level as compared to the positive controls. In soybean meal, the same interaction was observed in PEDV and PRRSV at 0 h and SVV1 and PEDV at 24 h with less detectable RNA observed (P < 0.05) in the postinoculation samples regardless of inclusion level than the preinoculation counterparts and the controls. Overall, an application time effect was noticed in each matrix where less RNA was detected in the postinoculation samples at 0 h (P < 0.05) compared to the preinoculation samples and the control, and at 24 h, both the pre- and postinoculation samples had less detectable RNA (P < 0.05) than the control. Overall, formaldehyde can reduce detectable RNA immediately in contaminated complete feed and soybean meal, with greater decreases observed as mitigant contact time increases.

Acute exercise has been shown to induce a small reduction in state anxiety, yet the most beneficial exercise stimulus is not clear. This review provides an update on the papers published since the last comprehensive review in 2015, with specific emphasis on whether study quality has improved. Randomised control trials, conducted in samples of healthy adults with non-clinical anxiety, were sourced from PubMed, PsycInfo, and Scopus. Study characteristics and study quality were assessed in nine studies comprising thirteen exercise conditions. Acute exercise significantly reduced anxiety in 53% (N = 7/13) of the exercise conditions. In comparison to a control condition, four showed exercising to be more effective, and one was as effective as the control. Two of the effective studies did not contain a control group. Six conditions were ineffective in reducing anxiety. There was no clear pattern of what combination of exercise mode, duration, and intensity was most effective, suggesting a variety may be effective in reducing anxiety. Methodological limitations still exist within the research, e.g., participant recruitment not considering baseline anxiety; variations in the control condition content. Future research should include participant samples exhibiting moderate-to-high levels of anxiety and examine self-selected exercise intensities.

Genetic approaches that allow lineage tracing are essential to our future understanding of melanocytes and melanoma. To date, the approaches used to label melanocytes in mice have relied on random integration of transgenes driven by the promoters of the Tyrosinase and Dopachrome tautomerase genes, knock-in to the Dopachrome tautomerase locus or knock-in to the Mlana locus in a bacterial artificial chromosome. These strategies result in expression in other tissues such as telencephalon and other cell types such as nerves. Here we used homologous recombination in mouse embryonic stem cells to generate a targeted multicistronic allele of the Pmel locus that drives melanocyte-specific expression of CreERT2, nuclear localised H2B-Cerulean and membrane localised marcks-mKate2 allowing live imaging of melanocytes and activation of other conditional alleles. We combined this allele with R26R-EYFP mice allowing induction of EYFP expression on administration of tamoxifen or its metabolite 4-OHT. The fluorescent proteins H2B-Cerulean and marcks-mKate2 label the cell nucleus and plasma membrane respectively allowing live imaging and FACS isolation of melanoblasts and melanocytes as well as serving to provide an internal control allowing estimation of recombination efficiency after administration of tamoxifen. We demonstrate the utility of the transgene in embryonic and adult tissues.

Introduction: Impulsivity is a symptom of attention-deficit/hyperactivity disorder (ADHD) and variants in the Lphn3 (Adgrl3) gene (OMIM 616417) have been linked to ADHD. This project utilized a delay-discounting (DD) task to examine the impact of Lphn3 deletion in rats on impulsive choice. "Positive control" measures were also collected in spontaneously hypertensive rats (SHRs), another animal model of ADHD. Methods: For Experiment I, rats were given the option to press one lever for a delayed reward of 3 food pellets or the other lever for an immediate reward of 1 pellet. Impulsive choice was measured as the tendency to discount the larger, delayed reward. We hypothesized that impulsive choice would be greater in the SHR and Lphn3 knockout (KO) rats relative to their control strains - Wistar-Kyoto (WKY) and Lphn3 wildtype (WT) rats, respectively. Results: The results did not completely support the hypothesis, as only the SHRs (but not the Lphn3 KO rats) demonstrated a decrease in the percent choice for the larger reward. Because subsequent trials did not begin until the end of the delay period regardless of which lever was selected, rats were required to wait for the next trial to start even if they picked the immediate lever. Experiment II examined whether the rate of reinforcement influenced impulsive choice by using a DD task that incorporated a 1 s inter-trial interval (ITI) immediately after delivery of either the immediate (1 pellet) or delayed (3 pellet) reinforcer. The results of Experiment II found no difference in the percent choice for the larger reward between Lphn3 KO and WT rats, demonstrating reinforcement rate did not influence impulsive choice in Lphn3 KO rats. Discussion: Overall, there were impulsivity differences among the ADHD models, as SHRs exhibited deficits in impulsive choice, while the Lphn3 KO rats did not.

Research question: Pulmonary rehabilitation is the best treatment for chronic breathlessness in COPD but there remains an unmet need to improve efficacy. Pulmonary rehabilitation has strong parallels with exposure-based cognitive behavioural therapies (CBT), both clinically and in terms of brain activity patterns. The partial N-methyl-d-aspartate (NMDA)-receptor agonist d-cycloserine has shown promising results in enhancing efficacy of CBT, thus we hypothesised that it would similarly augment the effects of pulmonary rehabilitation in the brain. Positive findings would support further development in phase 3 clinical trials. Methods: 72 participants with mild-to-moderate COPD were recruited to a double-blind pre-registered (ClinicalTrials.gov identifier: NCT01985750) experimental medicine study running parallel to a pulmonary rehabilitation course. Participants were randomised to 250 mg d-cycloserine or placebo, administered immediately prior to the first four sessions of pulmonary rehabilitation. Primary outcome measures were differences between d-cycloserine and placebo in brain activity in the anterior insula, posterior insula, anterior cingulate cortices, amygdala and hippocampus following completion of pulmonary rehabilitation. Secondary outcomes included the same measures at an intermediate time point and voxel-wise difference across wider brain regions. An exploratory analysis determined the interaction with breathlessness anxiety. Results: No difference between d-cycloserine and placebo groups was observed across the primary or secondary outcome measures. d-cycloserine was shown instead to interact with changes in breathlessness anxiety to dampen reactivity to breathlessness cues. Questionnaire and measures of respiratory function showed no group difference. This is the first study testing brain-active drugs in pulmonary rehabilitation. Rigorous trial methodology and validated surrogate end-points maximised statistical power. Conclusion: Although increasing evidence supports therapeutic modulation of NMDA pathways to treat symptoms, we conclude that a phase 3 clinical trial of d-cycloserine would not be worthwhile.

A total of 350 weanling pigs (DNA 400 × 200; initially, 5.67 ± 0.06 kg BW) were used in a 42-day study with 5 pigs per pen and 14 replicate pens per treatment. At weaning, pigs were allotted to pens in a completely randomized design and pens of pigs were randomly assigned to one of five dietary treatments: 1) negative control (CON; standard nursery diet containing only 150 ppm Zn from trace mineral premix and no acidifier); 2) control diet with 3,000 ppm added zinc from ZnO included in phase 1 and 2,000 ppm added zinc from ZnO included in phase 2 (ZnO); 3) control diet with 0.70% formic acid (FA; Amasil NA; BASF, Florham, NJ); 4) control diet with 0.18% glycerol monolaurate (GML; Natural Biologics GML, Natural Biologics, Newfield, NY); and 5) control diet with a 1.0% blend of formic acid and glycerol monolaurate (FORMI; FORMI 3G, ADDCON GmbH, Bitterfeld-Wolfen, Germany). Pigs were fed treatment diets from d 0 to d 28 and were then fed a common diet from d 28 to d 42. From days 0 to 7, pigs fed ZnO or FORMI had increased (P = 0.03) ADG compared to pigs fed CON, with no difference in feed intake (P > 0.05). Overall, pigs fed GML had reduced (P < 0.0001) ADG compared with those fed the CON, ZnO, or FORMI diets. Fecal DM was evaluated from days 7 to 28 and there was a treatment × day interaction (P = 0.04). Pigs fed GML had a lower fecal DM % on day 7, but a higher fecal DM % on days 14 and 21; however, no differences in fecal DM were observed on day 28. Fresh fecal samples were collected from the same randomly selected pig on days 0 and 14 (70 pigs total;14 pigs per treatment) for analysis of fecal microbial populations using 16S rDNA sequencing. Dietary treatment did not significantly impact fecal microbiota at the phyla level, but pigs fed ZnO had an increased relative abundance (P < 0.01) of the family Clostridiaceae. A blood sample was also collected from one pig per pen on days 0 and 14 for analysis of serum IgA, IgG, and TNF-α. There was no evidence that dietary treatment effected IgA, IgG, or TNF-α concentrations. The effect of sampling day was significant (P < 0.05), where circulating IgA and TNF-α was increased and IgG was decreased from days 0 to 14. In summary, there is potential for a blend of formic acid and GML to improve growth performance immediately post-weaning without negatively impacting fecal consistency. Formic acid and GML alone or in combination did not impact fecal microbial populations or serum immune parameters.

Purpose: Hypercapnic chemosensitivity traditionally captures the ventilatory response to elevated pressures of carbon dioxide in the blood. However, hypercapnia also contributes to subjective breathing perceptions, and previously we demonstrated a closer matching of perception to changes in ventilation in athletes compared to controls. Here we investigated any potential underlying hypercapnic chemosensitivity differences between groups, and explored whether these measures relate to ventilatory and perceptual responses during exercise as well as trait levels of affect. Methods: A hypercapnic challenge, incremental maximal exercise test and affective questionnaires were completed by 20 endurance athletes and 20 age-/sex-matched sedentary controls. The hypercapnic challenge involved elevating end-tidal PCO2 by 0.8% (6.1 mmHg) and 1.5% (11.2 mmHg) for 3 min each (randomised), with constant end-tidal oxygen. Ventilatory and perceptual responses to hypercapnia were compared between groups, and within each group the relationships between hypercapnic chemosensitivity (slope analyses) and exercising ventilation and perceptions were calculated using Spearman's non-parametric correlations. Results: While absolute ventilation differences during hypercapnia and exercise were observed, no group differences were found across hypercapnic chemosensitivity (slope) measures. Correlation analyses revealed the anxiety hypercapnic response was related to maximal exercise anxiety, but only in sedentary individuals. Conclusion: Ventilatory and perceptual hypercapnic chemosensitivity do not differ between athletes and sedentary individuals. However, ventilatory and anxiety hypercapnic chemosensitivities were related to ventilatory and anxiety responses during exercise in untrained individuals only. Athletes may employ additional strategies during exercise to reduce the influence of chemosensitivity on ventilatory and perceptual responses.

ABSTRACT: Salmonella continues to be a significant cause of foodborne illnesses in human medicine. The Centers for Disease Control and Prevention reported Salmonella as the second leading cause of foodborne illness in the United States and the leading cause of both hospitalizations and deaths. Salmonella enterica 4,[5],12:i:- (STM) is a monophasic variant of Salmonella Typhimurium, and it is an emerging threat to both human and animal health. STM was first identified in the 1980s from poultry products and has become increasingly prevalent in meat products including pork. STM has also been identified in swine farms as well as in feed manufacturing environments and feed itself. Similar pulse-field gel electrophoresis profiles have been observed between human clinical cases and the STM samples originating from swine feed. These related profiles suggest a link between ingestion of contaminated feed by swine and the source of foodborne illness in human. The objective of this article was to better understand the history of STM and the possible pathway from swine feed to table. Continued research is necessary to better understand how STM can enter both the feed supply chain and the pork production chain to avoid contamination of pork products destined for human consumption.

The US Department of Agriculture (USDA) categorizes the risk of African swine fever virus (ASFV) entry into the United States through non-animal origin feed ingredients as 'negligible to moderate, with high uncertainty'. Both Canada and Australia have implemented policies that are suggested to reduce the risk of ASFV entry through feed ingredients, but the United States has not because of scientific limitations that have been addressed by recent publications. As regulators and industry consider a potential pathway forward, the objective of this manuscript is to describe a process to determine if a voluntary or regulatory import policy is warranted by the United States. Initially, the volume and types of non-animal origin feed ingredients imported from countries with ASFV were quantified and assigned a level of risk (high risk: unprocessed grains and oilseeds, moderate risk: soybean co-products (meals, oil, and oilcake), and low risk: amino acids, vitamins, and other synthetically produced products from countries that have ASFV). In 2020, moderate- and high-risk ingredients from ASFV-positive countries represented 3.1% of all ingredients imported into the United States. Policies from Canada and Australia were evaluated for practicality of implementation by US government officials. Industry representatives from both countries consistently stated their policies would not be feasible in the United States due to the differences in cost and complexity of the swine and feed industries. Overall, unprocessed, or high-risk, ingredients from ASFV-positive countries represent a low percentage of imported ingredients into the United States; however, cautionary procedures may still be warranted given industry demand.

The ability to sense, monitor, and control respiration - e.g., respiratory interoception (henceforth, respiroception) is a core homeostatic ability. Beyond the regulation of gas exchange, enhanced awareness of respiratory sensations is directly related to psychiatric symptoms such as panic and anxiety. Indeed, chronic breathlessness (dyspnea) is associated with a fourfold increase in the risk of developing depression and anxiety, and the regulation of the breath is a key aspect of many mindfulness-based approaches to the treatment of mental illness. Physiologically speaking, the ability to accurately monitor respiratory sensations is important for optimizing cardiorespiratory function during athletic exertion, and can be a key indicator of illness. Given the important role of respiroception in mental and physical health, it is unsurprising that there is increased interest in the quantification of respiratory psychophysiology across different perceptual and metacognitive levels of the psychological hierarchy. Compared to other more popular modalities of interoception, such as in the cardiac domain, there are relatively few methods available for measuring aspects of respiroception. Existing inspiratory loading tasks are difficult to administer and frequently require expensive medical equipment, or offer poor granularity in their quantification of respiratory-related perceptual ability. To facilitate the study of respiroception, we here present a new, fully automated and computer-controlled apparatus and psychophysiological method, which can flexibly and easily measure respiratory-related interoceptive sensitivity, bias and metacognition, in as little as 30 min of testing, using easy to make 3D printable parts.

ABSTRACT: Salmonella enterica serotype 4,[5],12:i:- (STM) has become an increasing problem for food safety and has been often detected in swine products. Weanling pigs were exposed to STM-contaminated feed, water, or air to determine possible STM transmission routes. A control group of pigs was included. STM was monitored daily in feces and rectal and nasal swabs. STM colonization was most prevalent in tissues from tonsil, lower intestine, and mesenteric lymph nodes. No differences in lesion severity were observed between inoculated and control pigs. Contaminated feed, water, and aerosolized particles caused infection in weaned pigs; however, no STM colonization was observed in skeletal muscle destined for human consumption. Based on the results from this study, STM contamination in pork products most likely results from cross-contamination of meat by digesta or lymph node tissue during processing.

Background: Interoception refers to an individual's ability to sense their internal bodily sensations. Acute mountain sickness (AMS) is a common feature of ascent to high altitude that is only partially explained by measures of peripheral physiology. We hypothesized that interoceptive ability may explain the disconnect between measures of physiology and symptom experience in AMS. Methods: Two groups of 18 participants were recruited to complete a respiratory interoceptive task three times at 2-week intervals. The control group remained in Birmingham (140 m altitude) for all three tests. The altitude group completed test 1 in Birmingham, test 2 the day after arrival at 2,624 m, and test 3 at 2,728 m after an 11-day trek at high altitude (up to 4,800 m). Results: By measuring changes to metacognitive performance, we showed that acute ascent to altitude neither presented an interoceptive challenge, nor acted as interoceptive training. However, AMS symptom burden throughout the trek was found to relate to sea level measures of anxiety, agoraphobia, and neuroticism. Conclusions: This suggests that the Lake Louise AMS score is not solely a reflection of physiological changes on ascent to high altitude, despite often being used as such by researchers and commercial trekking companies alike.

Maintaining biosecurity between swine barns is challenging, and boot baths are an easily implementable option some utilize to limit pathogen spread. However, there are concerns regarding their efficacy, especially when comparing wet or dry disinfectants. The objective of this study was to evaluate the efficacy of boot baths in reducing the quantity of detectable porcine epidemic diarrhea virus (PEDV) and porcine reproductive and respiratory syndrome virus (PRRSV) genetic material using wet or dry disinfectants. Treatments included 1) control, 2) dry chlorine powder (Traffic C.O.P., PSP, LLC, Rainsville, AL), and 3) wet quaternary ammonium/glutaraldehyde liquid (1:256 Synergize, Neogen, Lexington, KY). Prior to disinfection, rubber boots were inoculated with 1 mL of a co-inoculants of PRRSV (1 × 105 TCID50 per mL) and PEDV (1 × 105 TCID50 per mL) and dried for 15 min. After the drying period, a researcher placed the boot on the right foot and stepped directly on a stainless steel coupon (control). Alternatively, the researcher stepped first into a boot bath containing either the wet or dry sanitizer, stood for 3 s, and then stepped onto a steel coupon. After one minute, an environmental swab was then collected and processed from each boot and steel coupon. The procedure was replicated 12 times per disinfectant treatment. Samples were analyzed using a duplex qPCR at the Kansas State Veterinary Diagnostic Laboratory. Cycle threshold values were analyzed using SAS GLIMMIX v 9.4 (SAS, Inc., Cary, NC). There was no evidence of a disinfectant × surface × virus interaction (P > 0.10). An interaction between disinfectant × surface impacted (P < 0.05) the quantity of detectable viral RNA. As expected, the quantity of the viruses on the coupon was greatest in the control, indicating that a contaminated boot has the ability to transfer viruses from a contaminated surface to a clean surface. Comparatively, the dry disinfectant treatment resulted in no detectable viral RNA on either the boot or subsequent coupon. The wet disinfectant treatment had statistically similar (P > 0.05) viral contamination to the control on the boot, but less viral contamination compared to the control on the metal coupon. In this experiment, a boot bath with dry powder was the most efficacious in reducing the detectable viral RNA on both boots and subsequent surfaces.

It is poorly understood why asthma symptoms are often discordant with objective medical tests. Differences in interoception (perception of internal bodily processes) may help explain symptom discordance, which may be further influenced by mood and attention. We explored inter-relationships between interoception, mood and attention in 63 individuals with asthma and 30 controls. Questionnaires, a breathing-related interoception task, two attention tasks, and standard clinical assessments were performed. Questionnaires were analysed using exploratory factor analysis, and linear regression examined relationships between measures. K-means clustering also defined asthma subgroups. Two concordant asthma subgroups (symptoms related appropriately to pathophysiology, normal mood) and one discordant subgroup (moderate symptoms, minor pathophysiology, low mood) were found. In all participants, negative mood correlated with decreased interoceptive ability and faster reaction times in an attention task. Our findings suggest that interpreting bodily sensations relates to mood, and this effect may be heightened in subgroups of individuals with asthma.
